RESUMO

Environmental Sensitivity, which explains individual differences in the tendency to respond more to both positive and negative environmental influences, can be measured by the self-reported Highly Sensitive Person scale. This paper introduced psychometric properties of a brief Japanese version of a 10-item measure of sensitivity (HSP-J10) developed by four studies involving 2,388 adults. The results showed that (1) the newly created HSP-J10 supported the bifactor structure (i.e., Ease of Excitation, Low Sensory Threshold, Esthetic Sensitivity, plus General Sensitivity factor), (2) the HSP-J10 correlated with but discriminated against other personality traits and affects, (3) it had high temporal stability, and (4) participants who scored higher on the HSP-J10 showed significant increases in positive emotion from before watching a video with positive content to after, while those who scored low showed no significant change in positive emotion. It demonstrated the new scale's good psychometric properties in that it moderated outcomes as theoretically expected when the environment was experimentally manipulated. The four studies suggested that the newly created HSP-J10 might adequately measure individual differences in adults' Environmental Sensitivity.

Luteolin, an abundant flavonoid in the leaves of Capsicum annuum, has antioxidant activity and is, thus, a key chemical for promoting plant residue utilization, especially for the development of healthcare products. We assessed the inhibitory effect of luteolin and its glycosides on osteoclastic differentiation in human cells and found that the differentiation was effectively inhibited at noncytotoxic concentrations. We also screened 47 varieties of C. annuum for the accumulation of luteolin and apigenin to determine the prevalence of luteolin in diverse cultivars and identify varieties with high and/or selective luteolin production. The glycosides of luteolin and apigenin were found in all the tested varieties, with luteolin predominant over apigenin in most varieties. The identification and characterization of highly productive varieties of C. annuum is expected to be beneficial for the effective development of useful luteolin-based products from plant residues.

BACKGROUND: Glioblastoma-initiating cells (GICs) comprise a tumorigenic subpopulation of cells that are resistant to radio- and chemotherapies and are responsible for cancer recurrence. The aim of this study was to identify novel compounds that specifically eradicate GICs using a high throughput drug screening approach. METHODS: We performed a cell proliferation/death-based drug screening using 10 560 independent compounds. We identified dihydroorotate dehydrogenase (DHODH) as a target protein of hit compound 10580 using ligand-fishing and mass spectrometry analysis. The medical efficacy of 10580 was investigated by in vitro cell proliferation/death and differentiation and in vivo tumorigenic assays. RESULTS: Among the effective compounds, we identified 10580, which induced cell cycle arrest, decreased the expression of stem cell factors in GICs, and prevented tumorigenesis upon oral administration without any visible side effects. Mechanistic studies revealed that 10580 decreased pyrimidine nucleotide levels and enhanced sex determining region Y-box 2 nuclear export by antagonizing the enzyme activity of DHODH, an essential enzyme for the de novo pyrimidine synthesis. CONCLUSION: In this study, we identified 10580 as a promising new drug against GICs. Given that normal tissue cells, in particular brain cells, tend to use the alternative salvage pathway for pyrimidine synthesis, our findings suggest that 10580 can be used for glioblastoma therapy without side effects.Key Points1. We have developed a novel method, antagonistic template-based biopanning, for screening peptide ligands specifically recognizing local tertiary protein structures. We chose water-soluble pyrroloquinoline quinone (PQQ) glucose dehydrogenase (GDH-B) as a model enzyme for this screening. Two GDH-B mutants were constructed as antagonistic templates; these have some point mutations to induce disruption of local tertiary structures within the loop regions that are located at near glucose-binding pocket. Using phage display, we selected 12-mer peptides that specifically bound to wild-type GDH-B but not to the antagonistic templates. Consequently, a peptide ligand showing inhibitory activity against GDH-B was obtained. These results demonstrate that the antagonistic template-based biopanning is useful for screening peptide ligands recognizing the specific local tertiary structure of proteins.

PURPOSE: To investigate the potential use of Prussian blue-coated magnetic nanoparticles, termed "Prussian blueberry", to bring about the magnetic elimination of cesium. METHODS: Prussian blueberry were prepared by a layer-by-layer assembly method. The morphology, structure and physical properties of the Prussian blueberry were investigated as was their ability to magnetically eliminate cesium. RESULTS: We confirmed that Prussian blueberry were composed of a magnetite nanoparticle-core and a Prussian blue-shell. Under a magnetic field, Prussian blueberry (5 mg) reduced the cesium concentration of seawater (3 ml) from 150 ppm to about 50 ppm; but regular Prussian blue could not magnetically eliminate cesium. Moreover, Prussian blueberry removed a similar proportion of cesium from a larger volume of seawater, and from fetal bovine serum and cow's milk. CONCLUSIONS: Under a magnetic field, Prussian blueberry was able to rapidly eliminate cesium from seawater and from biological matrices such as serum and milk.

Perceptual asymmetries for tasks involving aesthetic preference or line bisection can be affected by asymmetrical neurological mechanisms or left/right reading habits. This study investigated the relative contribution of these mechanisms in 100 readers of Japanese and English. Participants made aesthetic judgments between pairs of mirror-reversed pictures showing: (a) static objects, (b) moving objects and (c) landscapes. A line bisection task was also administered. There was a strong effect of reading direction for static and mobile objects whereby Japanese readers preferred objects with a right-to-left directionality (and vice versa for English readers). In contrast, similar patterns were observed for the Japanese and English readers for the landscape and line bisection tasks. The results show that reading habits affect aesthetic judgments for static and moving object tasks, but not the landscape and line bisection tasks. The difference between the tasks may be related to the horizontal/vertical geometry of the stimuli, which makes the landscape and line bisection tasks more prone to universal effects related to cerebral dominance.

Cancer gene therapy requires a safe and effective gene delivery system. Polymer- and lipid-coated magnetic nanocrystals have been used to deliver silencing RNA, but synthesizing these magnetic vectors is difficult. Here, we show that a new nanoparticle formulation can be magnetically guided to deliver and silence genes in cells and tumours in mice. This formulation, termed LipoMag, consists of an oleic acid-coated magnetic nanocrystal core and a cationic lipid shell. When compared with the commercially available PolyMag formulation, LipoMag displayed more efficient gene silencing in 9 of 13 cell lines, and better anti-tumour effects when systemically administered to mice bearing gastric tumours. By delivering an optimized sequence of a silencing RNA that targets the epidermal growth factor receptor of tumour vessels, the intended therapeutic benefit was achieved with no evident adverse immune reaction or untoward side effects.

The heart of the ostracod crustacean Vargula hilgendorfii has a single intrinsic neuron that morphologically appears to innervate the myocardium. We, therefore, examined the heart activity electrophysiologically to determine whether the heartbeat is neurogenic. Each heartbeat is associated with a myocardial action potential composed of a spike potential followed by a plateau potential. The frequency of the action potential is not stable but changes successively over a wide range. The action potential is not preceded by a pacemaker potential and has an inflection in its rising phase. The myocardial cells couple electrically and fire almost simultaneously. The frequency of the action potential was unchanged by injection of depolarizing or hyperpolarizing current into the myocardium. However, slow oscillatory potentials appeared during the depolarization and its frequency was higher with increasing current intensity. Application of 1-microM tetrodotoxin (TTX) depolarized the myocardial membrane and completely prevented the action potential. During this depolarization, slow oscillatory potentials often appeared spontaneously. These results suggest that, although the myocardium has a property of conditional oscillator, the heartbeat is driven by the single cell cardiac ganglion that has both pacemaker and motor functions.

RESUMO

Double-chambered right ventricle is a congential malformation caused by an anomalous muscle band obstructing the right ventricular outflow tract. Most reported cases of this condition have been diagnosed in infants, or adolescents. We encountered a 61-year-old woman with a double-chambered right ventricle, associated with a large interventricular septal aneurysm, which is a rare complication. The right ventricular obstruction was corrected with surgery.
