BACKGROUND: The aim of this study was to determine the optimal vaccination strategies for the control of porcine respiratory disease complex (PRDC) caused by Mycoplasma hyopneumoniae, porcine reproductive and respiratory syndrome virus (PRRSV), and porcine circovirus type 2 (PCV2) in case of early mycoplasmal infection. METHODS: A total of 120 pigs were randomly divided into 6 groups (20 pigs per group). Four separate vaccine regimen groups were selected. Pigs from the four vaccinated groups were challenged with M. hyopneumoniae at 28 days old followed by a challenge of PRRSV or PCV2 at 49 days old. RESULTS: Regardless of PRRSV or PCV2 vaccination, pigs vaccinated with one of the M. hyopneumoniae vaccines at 7 days old had a significantly better growth performance over the whole length of the study compared to pigs vaccinated with a second M. hyopneumoniae vaccine at 21 days old. Vaccination of pigs with M. hyopneumoniae at 7 days and PRRSV at either 7, 14 or 21 days old resulted in significantly reduced PRRSV viremia and lung lesions compared to vaccination of pigs with M. hyopneumoniae and PRRSV at 21 days old. CONCLUSIONS: The efficacy of the PRRSV MLV vaccine is influenced by the different timing of M. hyopneumoniae vaccination whereas the efficacy of the PCV2 vaccine is not. This experiment study demonstrated that early vaccination with a M. hyopneumoniae vaccine should be the highest priority in order to control M. hyopneumoniae and PRRSV infection in cases of early M. hyopneumoniae infection.

Mycoplasma spp. are highly contagious pathogens and intramammary Mycoplasma infection is a serious issue for the dairy industry. As there is no effective vaccine for Mycoplasma infection, control depends on good husbandry and chemo-antibiotic therapy. In this study, antimicrobial susceptibility of Mycoplasma strains recently isolated from cases of bovine mastitis in Japan was evaluated by minimum inhibitory concentration (MIC). All Mycoplasma bovis strains were sensitive to pirlimycin, danofloxacin and enrofloxacin, but not kanamycin, oxytetracycline, tilmicosin or tylosin. M. californicum and M. bovigenitalium strains were sensitive to pirlimycin, danofloxacin, enrofloxacin, oxytetracycline, tilmicosin and tylosin, but not to kanamycin. This is the first report to describe the MIC of major antimicrobial agents for Mycoplasma species isolated from bovine mastitis in Japan.

BACKGROUND: Non-fatal suicide attempt is the most important risk factor for later suicide. Emergency department visits for attempted suicide are increasingly recognised as opportunities for intervention. However, no strong evidence exists that any intervention is effective at preventing repeated suicide attempts. We aimed to investigate whether assertive case management can reduce repetition of suicide attempts in people with mental health problems who had attempted suicide and were admitted to emergency departments. METHODS: In this multicentre, randomised controlled trial in 17 hospital emergency departments in Japan, we randomly assigned people aged 20 years and older with mental health problems who had attempted suicide to receive either assertive case management (based on psychiatric diagnoses, social risks, and needs of the patients) or enhanced usual care (control), using an internet-based randomisation system. Interventions were provided until the end of the follow-up period (ie, at least 18 months and up to 5 years). Outcome assessors were masked to group allocation, but patients and case managers who provided the interventions were not. The primary outcome was the incidence of first recurrent suicidal behaviour (attempted suicide or completed suicide); secondary outcomes included completed suicide and all-cause mortality. This study is registered at ClinicalTrials.gov (NCT00736918) and UMIN-CTR (C000000444). FINDINGS: Between July 1, 2006, and Dec 31, 2009, 914 eligible participants were randomly assigned, 460 to the assertive case management group and 456 to the enhanced usual care group. We noted no significant difference in incidence of first recurrent suicidal behaviour between the assertive case management group and the enhanced usual care group over the full study period (log-rank p=0·258). Because the proportional hazards assumption did not hold, we did ad-hoc analyses for cumulative incidence of the primary outcome at months 1, 3, 6, 12, and 18 after randomisation, adjusting for multiplicity with the Bonferroni method. Assertive case management significantly reduced the incidence of first recurrent suicidal behaviour up to the 6-month timepoint (6-month risk ratio 0·50, 95% CI 0·32-0·80; p=0·003), but not at the later timepoints. Prespecified subgroup analyses showed that the intervention had a greater effect in women (up to 18 months), and in participants younger than 40 years and those with a history of previous suicide attempts (up to 6 months). We did not identify any differences between the intervention and control groups for completed suicide (27 [6%] of 460 vs 30 [7%] of 454, log-rank p=0·660) or all-cause mortality (46 [10%] of 460 vs 42 [9%] of 454, log-rank p=0·698). INTERPRETATION: Our results suggest that assertive case management is feasible in real-world clinical settings. Although it was not effective at reducing the incidence of repetition of suicide attempts in the long term, the results of our ad-hoc analyses suggested that it was effective for up to 6 months. The objective of this study was to investigate cystic ovarian disease (COD) in commercial Japanese Black cows and to evaluate the efficacy of 7-day insertion of an intravaginal progesterone insert (CIDR) combined with prostaglandin F(2alpha) (PGF(2alpha)) at CIDR removal. Experiment 1 was conducted to group cysts into 4 patterns based on alteration of plasma progesterone (P(4)) concentrations on d -7 and d 0 (=CIDR insertion) with 1.0 ng/ml as the cut-off level by ultrasonographic examination of 28 cows with COD that were >or=40 days postpartum and anoestrous after calving. In Experiment 2, a total of 55 cows under the same conditions as in Experiment 1 were utilized, and the same regimen as in Experiment 1 was performed without 7 days of pre-observation before treatment. As a result, 92.9% of CLs on d 21 were highly formed in Experiment 1 and 83.6% were highly formed in Experiment 2. The conception rates within 60 days after CIDR removal were also satisfactory high and were 71.4% and 54.5%, respectively. There were no differences in any overall reproductive parameters between Experiments 1 and 2 (P>0.05). The average days between CIDR removal and conception were 24.4 +/- 5.3 and 24.0 +/- 6.5 days, respectively (P>0.05); therefore, the conception dates of the cows in Experiment 2 were at least 7 days earlier compared with Experiment 1. In conclusion, treatment with a CIDR and PGF(2alpha) against COD could minimize the risk of incorrect treatment and provide sufficient reproductive performance in Japanese Black cows.

Reproductive performance of two types of timed artificial insemination (TAI) protocols with or without intravaginal progesterone insert (CIDR) was investigated in a commercial herd of Holstein heifers. A total of seventy-four heifers with 14.4 months of age were allocated to two groups; Ovsynch (n=44) and estradiol benzoate (EB) used Heatsynch (EB-Heatsynch, n=30), and each group was additionally divided into two subgroups with CIDR insertion from day 0 to 7 (n=36) and without CIDR group (n=38). Blood was collected for progesterone (P4) analysis and ovarian finding was monitored with ultrasonography. Heifers in CIDR-treated group resulted in higher pregnancy rate as compared to No-CIDR-treated group (63.9% vs 21.1%, P<0.01). Heifers with functional corpus luteum (CL) on day 0 resulted in significantly higher pregnancy rate in CIDR-treated group than No-CIDR-treated group (day 0: 67.9% vs 13.0%, P<0.01). CIDR insertion suppressed the intermediate ovulation during the first 7 days and the period from the second GnRH or EB administration to TAI as compared to No-CIDR-treated group (first 7 days: 33.3% vs. 52.6%; P<0.05, before TAI: 11.1% vs. 37.0%; P<0.05). In conclusion, the selected TAI protocols with CIDR provided acceptable pregnancy rate and contributed to the economical improvement by shortening the average age of first calving approximately for 2.5 months as compared to the previous management without TAI protocols.
