RESUMO

Molding processes with molds containing topographical structures have been used for fabrication of hydrogel and cryogel particles. However, they can involve difficulties in separation of fabricated particles with complex shape from the molds or repeated fabrication of the particles although the overall processes do not require much skill and equipment. In this study, molds with etched superhydrophobic patterns have been developed by etching polytetrafluoroethylene (PTFE) blocks in user-defined designs with a femtosecond (FS) laser-based etching system. Lyophilized cryogel particles with various designs and sizes were fabricated by molding precursors with these PTFE molds. Additionally, the clean and easy separation of particles from the molds allowed repeated fabrication of the particles. For an application, relatively 'big' gelatin-norbornene (GelNB) cryogel particles prepared via molding with polydimethylsiloxane (PDMS) molds, swelling in phosphate buffered saline (PBS) and slicing height in half and 'small' GelNB cryogel particles fabricated with the PTFE molds were fabricated. Then, they were used to study scaffold size effect on calvarial bone regeneration. The molds generated with the FS laser-based etching system can be useful for various applications that require the mass production of cryogel particles in various geometries.

A diode-pumped Yb:Y2O3 ceramic thin-rod amplifier which operates in the femtosecond regime is studied here. In a single-stage and direct four-pass amplification scheme, the amplifier delivers maximum output power of 8.1 W at a center wavelength of 1030.5 nm and spectral bandwidth of 4.8 nm. Assume a sech2-shaped pulse, a pulse duration of 239 fs is measured, exhibiting a time-bandwidth product value of 0.324. To the best of our knowledge, our Yb:Y2O3 ceramic thin-rod femtosecond amplifier exhibits the shortest pulse duration with Watt-level output power among all Yb:Y2O3-based femtosecond amplifiers.

BACKGROUND: The aim of this study was to evaluate the feasibility of indocyanine green (ICG) fluorescent method for sentinel lymph node detection in early gastric cancer. METHODS: Between December 2012 and December 2014, 28 cases of pilot examination were performed at Seoul National University Bundang Hospital. Advanced version of multispectral fluorescence organoscope was used to identify sentinel node by quantitative estimation of ICG fluorescent signal intensity. Sensitivity, specificity, false positive value were analyzed and compared with dual tracer method. RESULTS: A total of 443 lymph nodes in 28 cases were examined and 184 sentinel nodes (41.5%) were identified by dual tracer method. The sensitivity using near-infrared ICG method was 98.9%. The specificity was 76.0% and false positive rate was 25.4% compared with dual tracer method. The adequate threshold for sentinel node detection was considered as 10% of maximum signal intensity. CONCLUSION: New near-infrared ICG fluorescent method could be a promising protocol for sentinel node navigation surgery in early gastric cancer.

Skin auto-fluorescence (SAF) has generated broad interest about the prospects for non-invasive advanced glycation end product assessment and its direct interplay with the development of microvascular complications, but clinical application of the existing SAF measuring of non-palmoplantar sites in non-Caucasian subjects with dark skin type is still controversial. Here, we tested the diabetic complication screening performance of a novel SAF measuring system in Asian type 2 diabetes mellitus (T2DM) subjects. A total of 166 Korean patients with T2DM were enrolled in this study and palmoplantar SAF was measured by a newly developed transmission-geometry noninvasive optical system. We found that transmitted SAF values of palmoplantar sites, 1st dorsal interossei muscles of the hand, in a complication group were significantly higher than in a non-complication group while no differences were observed between the two groups in reflected SAF of non-palmoplantar sites. The transmitted SAF values of palmoplantar sites were dramatically increased in subjects with multiple complications and were tightly correlated with the duration of microvascular complications. In conclusion, the SAF measurement in the palmoplantar sites with a non-invasive transmission-geometry optical system provided better microvascular complication screening performance compared to the SAF measurement of non-palmoplantar sites specifically in Asian T2DM subjects.

RESUMO

Optical measurement of skin auto-fluorescence (SAF), most likely emanating from accumulated advanced glycation end-products (AGEs), has been proposed for the noninvasive diagnosis of glucose intolerance in clinical settings. Here, we developed a novel imaging system with transmission geometry for SAF measurement and compared its diagnostic performance in a Korean population.

Light reflectance of blood is a complex phenomenon affected by hematocrit and red blood cell (RBC) aggregation (rouleaux formation). According to the hypothesis that RBC rouleaux are aligned with the direction of blood flow, the spatial alignment of RBC rouleaux, as well as their size and quantity in the blood, may also affect light reflectance. The present study aims to investigate the effect of the spatial alignment and distribution of RBC rouleaux on light reflection using optical coherence tomography (OCT). Blood flow velocity and reflectance profiles in a rat jugular-femoral bypass loop were simultaneously measured using a Doppler swept-source OCT system at various incident angles from -30 to +30 deg. The reflectance profiles of flowing blood show nonmonotonous decay with a local negative peak at the center of the tube. The profiles vary depending on the incident angle. This angular dependence is stronger at a higher angle of incidence. The anisotropic reflectance of flowing blood is consistent with the hypothesis on the spatial alignment of RBC rouleaux.

The combination of two-photon microscopy (TPM) and optical coherence tomography (OCT) is useful in conducting in-vivo tissue studies, because they provide complementary information regarding tissues. In the present study, we developed a new combined system using separate light sources and scanners for individually optimal imaging conditions. TPM used a Ti-Sapphire laser and provided molecular and cellular information in microscopic tissue regions. Meanwhile, OCT used a wavelength-swept source centered at 1300 nm and provided structural information in larger tissue regions than TPM. The system was designed to do simultaneous imaging by combining light from both sources. TPM and OCT had the field of view values of 300 µm and 800 µm on one side respectively with a 20x objective. TPM had resolutions of 0.47 µm and 2.5 µm in the lateral and axial directions respectively, and an imaging speed of 40 frames/s. OCT had resolutions of 5 µm and 8 µm in lateral and axial directions respectively, a sensitivity of 97dB, and an imaging speed of 0.8 volumes per second. This combined system was tested with simple microsphere specimens, and was then applied to image small intestine and ear tissues of mouse models ex-vivo. Molecular, cellular, and structural information of the tissues were visualized using the proposed combined system.
