RESUMO

Korea is currently developing country-specific emission factors to support the 2050 zero-carbon campaign. Dairy cattle represent one of the largest livestock industries in Korea, and the industry is estimated to continue increasing because of an increase in milk demand. However, country-specific emission factors for dairy cattle are currently only available for calculating methane (CH4) emissions from enteric fermentation. Two experiments were conducted to evaluate CH4 and nitrous oxide (N2O) fluxes from sawdust-bedded barn in dairy cow and steer, as well as dairy cattle manure composting lots. The greenhouse gas (GHG) fluxes were quantified using the open-chamber method and gas chromatography. CH4 fluxes from steer, dairy cow, and manure compost were 27.88 ± 5.84, 36.12 ± 10.85, and 259.44 ± 61.78 µg/head/s, respectively. N2O fluxes from steer, dairy cow, and manure compost were 14.04 ± 1.27, 4.11 ± 1.57, and 3.97 ± 1.08 µg/head/s, respectively. The result of this study can be used to construct country-specific data for GHG emissions from manure management. Thus, the application of mitigation strategies can be prioritized based on the GHG profile and targeted source.

In this study, we examined the effect of diets supplemented with organic and inorganic mineral premixes on the laying performance and eggshell quality of aged laying hens. A total of 600 68-week-old Hy-Line Brown laying hens were randomly assigned to 1 of 3 dietary treatments, repeated 5 times: Mash type basal diet, basal diet supplemented with an inorganic mineral premix (1.0 g/kg), and basal diet supplemented with an organic mineral premix (1.8 g/kg). The results showed that eggshell strength was higher (p < 0.01) in the inorganic mineral diet group than in the organic mineral and basal diet groups. Further, the levels of Fe and Mn in the liver were higher (p < 0.05) in the inorganic and organic mineral diet groups than in the basal diet group. The concentrations of Fe and Mg in the spleen were different (p < 0.05) among the treatment groups, with the highest levels reported in the organic mineral premix group. The concentrations of Cu, Zn, and Mn in the eggshell were different (p < 0.05) among the groups, with the highest levels reported in the inorganic and organic mineral premix diet groups. In conclusion, a diet containing organic mineral premix improved eggshell strength and had no detrimental effect on the laying performance of aged laying hens.

This study was performed to development the alternative farrowing pen (AFP) and to investigate performance and behavior of lactating sows and their litter. A total of 64 multiparous sows were randomly divided into two groups and were allocated to farrowing crates (FCs) and AFPs. The AFPs contained a crate and support bars that could be folded to provide the sows with extra space on day 5 postpartum. Behavior was recorded by charge-coupled device cameras and digital video recorders, and the data were scanned every 2 min to obtain an instantaneous behavioral sample. Farrowing systems did not affect feed intake, back-fat thickness, litter size and piglet weight at birth and weaning (p > 0.05). In addition, there were no differences in the number of crushed piglets between the two farrowing systems (p > 0.05). However, the weaning-to-estrus interval was shorter in the sows of the AFPs than in thous of the FCs (p < 0.05). The sows spent most of their time lying down during the lactating period, at about 80% lateral recumbency and 10%-15% ventral recumbency. The only significant differences were in the feeding and drinking behavior between sows in the two farrowing systems (p < 0.05). The FC sows displayed more feeding and drinking behavior than the AFP sows, especially in the late lactating period (p < 0.05). Piglets in the FCs tended to spend more time walking than piglets in the AFPs (p < 0.05), whereas there were no differences in suckling and lying behavior between piglets in the two farrowing systems (p > 0.05). It is concluded that the AFPs with temporary crating until day 4 postpartum did not negatively affect performance and crushed piglet compared with the FCs. It also may improve animal welfare by allowing sows to move and turn around during the lactating period. Further research is needed to find suitable housing designs to enhance productivity and animal welfare.

BACKGROUND: Non-adherence and comorbidities are prevalent among hemodialysis patients and are associated with increased mortality and financial burden. We aimed to investigate the influence of major coping strategies (CSs) on non-adherence and comorbidities in hemodialysis patients. METHODS: A total of 49 patients were enrolled. We collected participant data including CS measured by a Korean version of the ways of coping questionnaire (K-WCQ), comorbidities measured by age-adjusted Charlson comorbidity index (CCI), and adherence measured by the 8-item Morisky medication adherence scale (MMAS-8). RESULTS: Regarding major CS, 61.2% of participants reported use of support-seeking CS (SUP group), 14.3% reported use of problem-focused CS (PRO group), and 24.5% reported use of hopeful-thinking CS (HOP group). The mean MMAS-8 score was higher in the PRO group than in the HOP group (P = 0.024). The mean CCI score was lower in the PRO group than in the HOP group (P = 0.017). In the HOP group, the severity of somatic symptoms was positively correlated with the scores for the emotion-focused CS subscale (r = 0.39, P = 0.029) and the hopeful-thinking CS subscale (r = 0.38, P = 0.036) of the K-WCQ. The level of life satisfaction positively correlated with the score for the problem-focused CS subscale in the HOP group (r = 0.40, P = 0.027). CONCLUSION: We should pay more attention to the CSs of hemodialysis patients and provide interventions that promote problem-focused CSs, especially for nonadherent patients with high comorbidity rates who mainly use a hopeful-thinking CS.

PURPOSE. To solve the shortage of donor corneas, a decellularizing method based on hypertonic saline treatment was introduced, and a favorable outcome was observed in pig-to-rabbit lamellar corneal transplantation. This study was an investigation of the efficacy of pig-to-nonhuman primate lamellar corneal transplantation, using both decellularized and fresh porcine corneas to assess feasibility as a substitute for human corneas. METHODS. Nine Chinese rhesus macaques underwent lamellar corneal transplantation using both decellularized (n = 5) and fresh (n = 4) porcine corneas. Clinically acceptable graft size (7.5 mm in diameter) and minimal immunosuppression based on topical and systemic corticosteroids were applied. Rejection signs, histology of porcine grafts, and serial changes in recipients' blood profile, including memory T-cell subset, anti-α-Gal and donor pig-specific antibodies, and complement were evaluated. Changes in aqueous complement concentration were also assessed at 4 weeks after transplantation. RESULTS. Of the decellularized porcine lamellar grafts, 80% remained transparent for more than 6 months, whereas half of the fresh porcine lamellar grafts developed chronic rejection. Rejected grafts showed extensive cellular infiltration, predominantly CD8(+) T lymphocytes and macrophages. Immunologic profiles of the recipients with rejected grafts showed a significant increase in the concentration of aqueous complement, an enhancement of memory T cells, and an abrupt increase in donor pig-specific antibodies. CONCLUSIONS. The findings suggested that decellularized porcine cornea could be a promising substitute for human corneal allograft. Fresh porcine cornea may be a feasible option for a substitute if combined with more potent immunosuppression or if obtained from transgenic pigs with complement-regulatory proteins.

BACKGROUND: It has been reported that hyperacute rejection (HAR) does not occur after pig-to-nonhuman corneal xenotransplantation. However, considering that immune privilege is already disrupted in most human corneal recipients, and the expression of αGal can be gradually reduced after pig-to-rat corneal transplantation, the long-term survival of corneal grafts from wild-type pigs cannot be guaranteed. Accordingly, we aimed to investigate the effect of αGal on the change in anti-Gal antibodies, using sensitized α1,3-galactosyltransferase gene-knockout (GTKO) mice recipients. METHODS: C57BL/6 (B6) and GTKO mice were divided into 5 groups and underwent orthotopically full thickness cormeal transplantation as follows (n=4 for each group): (1) group 1: B6 to B6; (2) group 2: fresh porcine posterior corneal lamella to B6; (3) fresh porcine posterior corneal lamella to GTKO; (4) group 4: decellularized porcine posterior corneal lamella to GTKO, and (5) group 5: B6 to GTKO. Before transplantation, all GTKO recipients were sensitized using intraperitoneal injections of rabbit blood cells. Median survival times (MST) for the corneal grafts of the different groups were compared and plasma concentrations of IgG/IgM anti-Gal antibodies were evaluated at 1 week, 2 weeks and 3 weeks post-transplantation. RESULTS: There were no differences in MSTs between groups. Although there was no HAR of fresh porcine posterior corneal grafts even in sensitized GTKO recipients, αGal expression was induced in the transplanted fresh porcine corneal grafts and plasma concentration of IgG anti-Gal antibody was gradually increased in fresh porcine cornea-grafted GTKO recipients. On the contrary, αGal expression did not increase in the grafts and plasma concentration of anti-Gal antibodies did not change after transplantation using decellularized porcine corneas. CONCLUSIONS: Our findings suggest that αGal may affect the long-term survival of porcine corneal xenografts via antibody-mediated rejection, although αGal does not have an effect on acute rejection and decellularized porcine corneas may enable the long-term survival of porcine corneal xenografts.

Anti cancer agent 5-FU (Fluoro Uracil) is a prodrug that can be metabolized and then activated to interfere with RNA and DNA homeostasis. However, the majority of administered 5-FU is known to be catabolized in vivo in the liver where Dihydropyrimidine dehydrogenase (DPD) is abundantly expressed to degrade 5-FU. The biological factors that correlate with the response to 5-FU-based chemotherapy have been proposed to include uridine phosphorylase (UPP), thymidine phosphorylase (TPP), p53 and microsatellite instability. Among these, the expression of UPP is known to be controlled by cytokines such as TNF-alpha, IL1 and IFN-gamma. Our preliminary study using a DNA microarray technique showed that basic fibroblast growth factor (bFGF) markedly induced the expression of UPP1 at the transcription level. In the present study, we investigated whether bFGF could modulate the expression of UPP1 in osteo-lineage cells and examined the sensitivity of these cells to 5-FU mediated apoptosis.
