BACKGROUND: Olfaction plays an important role in host-seeking by parasitoids, as they can sense chemical signals using sensitive chemosensory systems. Psyttalia incisi (Silvestri) (Hymenoptera: Braconidae) is the dominant parasitoid of Bactrocera dorsalis (Hendel) in fruit-producing regions of southern China. The olfactory behavior of P. incisi has been extensively studied; however, the chemosensory mechanisms of this species are not fully understood. RESULTS: Bioinformatics analysis of 64,515 unigenes from the antennal transcriptome of both male and female adults P. incisi identified 87 candidate chemosensory genes. These included 13 odorant-binding proteins (OBPs), seven gustatory receptors (GRs), 55 odorant receptors (ORs), 10 ionotropic receptors (IRs), and two sensory neuron membrane proteins (SNMPs). Phylogenetic trees were constructed to predict evolutionary relationships between these chemosensory genes in hymenopterans. Moreover, the tissue expression profiles of 13 OBPs were analyzed by quantitative real-time PCR, revealing high expression of seven OBPs (1, 3, 6, 7, 8, 12, and 13) in the antennae. CONCLUSION: This study represents the first identification of chemosensory genes and the determination of their expression patterns in different tissues of P. incisi. These results contribute to a better understanding of the function of the chemosensory system of this parasitoid species.

Spinetoram wax-based bait station (SWBB) is a maintenance-free, long-lasting, and eco-friendly management measure for Bactrocera dorsalis. However, the impacts of low-concentration spinetoram on B. dorsalis have not yet been determined. Therefore, our study aimed to determine the impacts of low-concentration SWBBs on the biology, demographics, detoxifying enzymes, and gut microorganisms of B. dorsalis. Our results showed that low-concentration SWBBs posed dose-dependent effects on the lifespan and fecundity of B. dorsalis adults. Both the LC10 and LC30 treatments significantly reduced the fecundity, while only the latter led to significant deleterious effects on the longevity of adults. Transgenerational bioassays revealed that exposure to LC30 significantly affected the development period of larvae and pupae as well as the livability of pre-adult stage of the progeny. However, except for the ovipositional period, no significant effects on the biological traits of F1 adults were observed. In terms of the F1 demographic parameters, dose-dependent effects were observed. Moreover, both the LC10 and LC30 treatments significantly extended the mean generation time, while the latter remarkably decreased the finite and intrinsic rates. Additionally, the significant induction of CarE activity by the LC10 and LC30 treatment was maintained until 24 and 48 h respectively. The CYP450 O-deethylation activity in the LC30 treatment was significantly enhanced at 24 and 48 h intervals when compared to the control. Regarding the intestinal bacterial community, after B. dorsalis adults were exposed to low-concentration SWBBs, the relative abundances of Providencia and Vagococcus were significantly increased, whereas those of Lactococcus and Brachyspira experienced a significant decrease. The obtained results are expected to serve as a foundation for the application of spinetoram in "lure-and-kill" strategies against B. dorsalis.

As a vital pest control strategy, trapping plays an important role in the system of monitoring, catching and killing fruit flies. Cuelure (4-(4-acetoxyphenyl)-2-butanone, CL) is a male lure that attracts Zeugodacus tau and also stimulates feeding in this species. In this study, the attraction of Z. tau to CL and its subsequent feeding behavior were investigated. Under the significant influence of age and time of day, the attraction of CL to Z. tau was found to be optimal when flies were 14 days old, and the number of flies trapped increased with trapping duration. It was determined that consumption can improve the mating success and female adult fertility of Z. tau. After the observation period, the mating success rate of flies that ingested CL was significantly higher than that of the control group and was maintained at a higher level. It was found that parental consumption of CL could accelerate the development of eggs and larvae, resulting in increased pupation and emergence rates. The results of this study will further clarify the dynamic relationship between pest and lure, and provide a research basis for navigating the integrated management of Z. tau in the field.

Zeugodacus tau (Walker) (Diptera: Tephritidae) is a pest seriously harmful to Solanaceae crops and was found to oviposit on the pepino melon Solanum muricatum (Aiton). To date, the differences in the ability of the fruits of S. muricatum and other Solanaceae crops to attract gravid Z. tau females have seldom been reported. Oviposition and trapping bioassays were performed to clarify whether such differences existed. A combination of GC-MS and the Y-tube olfactometer system was used to identify and determine the compounds inducing behavioral responses in gravid Z. tau females to the volatile odors of S. muricatum. The results show that S. muricatum odors play a role in attracting gravid Z. tau females. The odors of Solanaceae crops influence their ability to attract these organisms. The nine compounds from the volatiles of S. muricatum induce tendency or repellency responses in gravid Z. tau females. Hexyl acetate, butyl acetate, amyl actate, and isoamyl acetate caused tendency behavior in gravid Z. tau females, while hexyl hexanoate, butyl isovalerate, butyl valerate, and isoamyl hexanoate caused repellency behavior. Heptyl acetate caused repellency behavior in gravid Z. tau females at higher concentrations (5 mg/mL) but caused tendency behavior at a low concentration (0.5 mg/mL). These results suggest that vigilance against the harm caused by Z. tau is required during the cultivation of S. muricatum. The nine compounds of the volatile odors of S. muricatum could help to develop attractants and repellents for gravid Z. tau females. These results are beneficial for preventing Z. tau females from harming S. muricatum, developing attractants and repellents for Z. tau females, and establishing a system of ecological control for Z. tau females.

The pupal parasitoid, Trichopria drosophilae Perkins (Hymenoptera: Diapriidae), is an ectoparasitoid of the genus Drosophila with great potential for application in biological control based on its excellent control efficiency for Drosophila suzukii Matsumura (Diptera: Drosophilidae), and it has has even been commercialized by biofactories. Due to its characteristics of short life cycle, large number of offspring, easy rearing, rapid reproduction, and low cost, Drosophila melanogaster (Diptera: Drosophilidae) is currently being utilized as a host to mass produce T. drosophilae. To simplify the mass rearing process and omit the separation of hosts and parasitoids, ultraviolet-B (UVB) was used as an irradiation source to irradiate D. melanogaster pupae, and the effects on T. drosophilae were studied. The results showed that UVB radiation significantly reduces host emergence and affects the duration of parasitoid development (female: F0 increased from 21.50 to 25.80, F1 from 23.10 to 26.10; male: F0 decreased from 17.00 to 14.10, F1 from 17.20 to 14.70), which has great significance for the separation of hosts and parasitoids as well as of females and males. Of the various studied conditions, UVB irradiation was ideal when the host was supplied with parasitoids for 6 h. The selection test results showed that the female-to-male ratio of emerging parasitoids in this treatment was highest at 3.47. The no-selection test resulted in the highest rates of parasitization and parasitoid emergence rate, maximized inhibition of host development, and allowed the omission of the separation step. Finally, the results of the semi-field test showed that the parasitoids bred in this treatment could search for their hosts normally and could therefore be directly applied in the biological control of Drosophila pests in the field.

Psyttalia incisi (Silvestri, 1916), an important solitary opiinae endoparasitoid, plays a crucial role in biological control programs against tephritid pests. In this study, the entire mitochondrial (mt) genome of P. incisi was sequenced and characterized. The whole mitogenome of P. incisi is 15,188 bp long with a G + C content of 14.80%, and encodes all 37 genes that are typically found in animal mt genomes, which contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and two ribosomal RNA genes (rRNAs). A maximum-likelihood (ML) tree demonstrates that P. incisi is closely related to Psyttalia lounsburyi, Psyttalia humilis, and Psyttalia concolor.

BACKGROUND: Bactrocera dorsalis is a notorious pest and spraying of insecticides has been the major approach to controlling its damage. However, insecticide abuse has led to negative impacts on public health and insecticide resistance; hence, sustainable strategies, such as bait stations, need to be developed and taken into account for B. dorsalis management. In this study, we integrated insecticide, sugar, olfactory and visual elements into a wax matrix to formulate a long-lasting bait station. We determined its efficacy against B. dorsalis under field conditions. RESULTS: The optimal bait station was a wax matrix incorporated with 1% spinetoram, 1% yellow pigment, and 3% ammonium acetate, and was spherical in shape with a 6 cm diameter. The longevity of this bait station was at least 10 weeks under field conditions. In a release and recapture experiment performed in Orchard 1, the bait stations were as effective as bait spray in controlling sterile fly populations, and more effective than the control. In a 2-year field trial conducted in Orchard 2, the number of female B. dorsalis and fruit infestation in the area where bait stations had been deployed was similar to the areas treated with cover spray. In Orchard 3, deployment of bait stations combined with chemical cover spray treatment decreased the population of female B. dorsalis and fruit damage more effectively than cover spray alone. CONCLUSIONS: Overall, wax-based bait stations can be regarded as a viable alternative to insecticides or a synergistic method for managing B. dorsalis. © 2022 Society of Chemical Industry.

Sexual dimorphism occurs widely throughout insects and has profound influences on evolutionary path. Sex-biased genes are considered to account for most of phenotypic differences between sexes. In order to explore the sex-biased genes potentially associated with sexual dimorphism and sexual development in Drosophila suzukii, a major devastating and invasive crop pest, we conducted whole-organism transcriptome profiling and sex-biased gene expression analysis on adults of both sexes. We identified transcripts of genes involved in several sex-specific physiological and functional processes, including transcripts involved in sex determination, reproduction, olfaction, and innate immune signals. A total of 11,360 differentially expressed genes were identified in the comparison, and 1,957 differentially expressed genes were female-biased and 4,231 differentially expressed genes were male-biased. The pathway predominantly enriched for differentially expressed genes was related to spliceosome, which might reflect the differences in the alternative splicing mechanism between males and females. Twenty-two sex determination and 16 sex-related reproduction genes were identified, and expression pattern analysis revealed that the majority of genes were differentially expressed between sexes. Additionally, the differences in sex-specific olfactory and immune processes were analyzed and the sex-biased expression of these genes may play important roles in pheromone and odor detection, and immune response. As a valuable dataset, our sex-specific transcriptomic data can significantly contribute to the fundamental elucidation of the molecular mechanisms of sexual dimorphism in fruit flies, and may provide candidate genes potentially useful for the development of genetic sexing strains, an important tool for sterile insect technique applications against this economically important species.

Diachasmimorpha anshunensis sp. nov., a koinobiont endoparasitoid of larvae of Zeugodacus tau (Walker) (Tephritidae: Diptera), is discovered from Guizhou (Southwest China) and is described by multiple forms of evidence. Morphological characteristics, photographs, and molecular data differentiating it from similar species are provided. Several biological characteristics of this new parasitoid, observed in a laboratory setting, are also provided as evidence to separate it from the most similar species in appearance.

Fopius arisanus (Sonan, 1932), an important egg parasitoid of several notorious tephritid pests, plays a key role in biological control programs. In the present study, the whole mitochondrial genome of F. arisanus was sequenced and characterized. The mitogenome of F. arisanus is 16,425 bp in length with 14.94% GC content, and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and two ribosomal RNA genes (rRNAs). The phylogenetic trees demonstrated that F. arisanus is sister group to Psyttalia concolor, P. humilis, P. lounsburyi and Diachasmimorpha longicaudata.

Psyttalia incisi (Silvestri) is the dominant parasitoid against Bactrocera dorsalis (Hendel) in fruit-producing regions of southern China. Prior to a large-scale release, it is important to generate a sufficient stockpile of P. incisi whilst considering how best to maintain their quality and performance; cold storage is an ideal method to achieve these aims. In this study, the impacts of temperature and storage duration on the developmental parameters of P. incisi pupae at different age intervals were assessed. Then, four of the cold storage protocols were chosen for further evaluating their impacts on the quality parameters of post-storage adults. Results showed that the emergence rate of P. incisi was significantly affected by storage temperature, storage duration, and pupal age interval and their interactions. However, when late-age P. incisi pupae developed at a temperature of 13 °C for 10 or 15 d, no undesirable impacts on dry weight, flight ability, longevity, reproduction parameters of post-storage adults, emergence rate, or the female proportion of progeny were recorded. Our findings demonstrate that cold storage has the potential for enhancing the flexibility and effectiveness of the large-scale production and application of P. incisi.

One environmentally friendly method used to manage Bactrocera dorsalis (Hendel), a key agricultural pest of substantial economic importance, is the sterile insect technique (SIT). Nevertheless, several deficiencies related to this strategy impair the success of the SIT, including the inferior performance of released sterile males compared with wild males, which could be partly solved by the utilization of gut symbionts as probiotic dietary components. In this study, a culture-dependent method was used to isolate and characterize gut-associated bacterial species in adult B. dorsalis genetic sexing strain (GSS) males. In addition, three bacterial isolates from the Enterobacteriaceae family, namely, Enterobacter sp., Morganella morganii, and Moellerella wisconsensis, were used as supplements in larval and adult diets to assess their effects on the life-history traits of irradiated males. Consistent with many previous studies, Enterobacter spp. was shown to be beneficial, with some quality control indices, such as adult size, pupal weight, survival rate under stress and nutritionally rich conditions, and mating competitiveness, being significantly increased, while slight nonsignificant increases in emergence rate and flight ability were observed. Conversely, the M. morganii and M. wisconsensis strains both had negative effects on irradiated male fitness and mating competitiveness. Our results, in combination with those of earlier studies, can contribute to improving the effectiveness of SIT application by enhancing the different aspects of augmentative rearing and biological traits of pests under laboratory rearing conditions.

Cold storage and heat exposure are crucial components of tephritid fruit fly mass-rearing programs, as they influence the development and fitness traits of produced flies. This work investigated the effects of cold storage on the pupal developmental parameters and quality of Bactrocera dorsalis (Hendel) genetic sexing strain (GSS) adults. Furthermore, the impact of short-term thermal exposure on the fecundity of B. dorsalis (GSS) that also underwent pupal cold storage was examined. Our results show that pupal development time, emergence rate, partial emergence rate, flight ability and fecundity were significantly affected by low temperature and pupal age and their interaction. Pupal cold storage did not pose negative impacts on the mating competition and response to methyl eugenol (ME) in the males. In addition, compared with the adults that were subjected to the same pupal storage protocol (five-day-old pupae stored at 13 °C), adult exposure to 41 °C for 1 h showed significant reparative effects on fecundity. In summary, the cold storage procedure of B. dorsalis (GSS) pupae has the potential to improve the flexibility and efficiency of mass-rearing schedules. Furthermore, short-term thermal exposure showed reparative effects on the fecundity costs induced by pupal cold storage in B. dorsalis (GSS).

Fopius arisanus is a solitary endoparasitoid that parasitizes a variety of tephritid species. Native to the Indo-Australian region, it is currently exploited worldwide as a biological control agent due to its exceptional efficiency in reducing pest populations. The efficiency of any biological control program is affected by the host location ability of the parasitoids. The present study used a Y-tube olfactometer to test the behavioural responses of female F. arisanus to four fruit species which had undergone different types of damages: undamaged, damaged through Bactrocera dorsalis ovipositioning (i.e., infested), or different levels of mechanical damage. Our results suggest that F. arisanus females were significantly attracted to mangoes and pears (vs. purified air), regardless of their condition; however, whilst infested mangoes did not attract more female parasitoids compared to healthy or mechanically damaged fruits, infested pears attracted significantly more. For citrus fruits and peaches, oviposition damage caused them to be more attractive to parasitoid females. In terms of the longevity of the effects, infested mango fruits remained attractive for up to 5 days after infestation, whereas for infested peaches, pears, and citrus fruits, the attractiveness tended to decrease as time passed. Regarding mechanical damage, mango fruits that had undergone any intensity of damage were equally attractive to parasitoid females; however, peach and citrus fruits with high levels of mechanical damage were more attractive, and pears were found to be most attractive with slight mechanical damage. Additional to the above, we also tested the effect of insecticides on behavioural responses using mangoes. We found that the treatment of infested fruits with lambda-cyhalothrin and cypermethrin remained attractive to F. arisanus females, albeit to different extents, which is in contrast to spinosad, cyantraniliprole, and acetamiprid. Finally, we suggest that the host-searching behaviour of F. arisanus females is mainly mediated by oviposition-induced volatiles, either emitted from the fruit or left by the fruit fly.

This study aims to evaluate several life-history traits of a T. drosophilae population from southern China and its parasitic preference of three Drosophila species. For mated T. drosophilae females, the mean oviposition and parasitization period were 27.20 and 37.80 d, respectively. The daily mean parasitization rate was 59.24% per female and the lifetime number of emerged progeny was 134.30 per female. Trichopria drosophilae females survived 37.90 and 71.61 d under host-provided and host-deprived conditions, respectively. To assess the potential for unmated reproduction in T. drosophilae, the mean oviposition and parasitization period of unmated females was 22.90 and 47.70 d, respectively. They had a daily mean parasitization rate of 64.68%, produced a total of 114.80 offspring over their lifetime, and survived 52 d. Moreover, T. drosophilae showed a preference towards D. suzukii based on the total number of emerged offspring under a choice test. Our findings indicate that T. drosophilae from southern China appears to be suitable for the control of D. suzukii in invaded areas, due to its reproductive potential.

Tyrosine hydroxylase (TH) is the initial enzyme responsible for cuticle sclerotization and pigmentation in many insect species, but to date, no direct functional studies have focused on TH in Zeugodacus tau. Here, the 3336-bp full-length cDNA of TH was isolated from Z. tau, a notorious horticultural pest infesting fruits and vegetables. qRT-polymerase chain reaction revealed that ZtTH transcripts were highly abundant at the time of pupal tanning and during adult emergence and were expressed in the midgut, integument and head of molting larvae. The pupation and eclosion rates gradually decreased when the 1st-instar larvae were fed diets containing higher concentrations of the TH inhibitor 3-iodo-tyrosine (3-IT). Moreover, pupal weights were significantly decreased, and abnormal uncolored phenotypes were observed after 20 mg/g 3-IT was incorporated into the diet. In addition, the suppression of TH function (mediated by RNA interference) led to a decrease in TH mRNAs and eclosion rates, accompanied by less-pigmented phenotypes. There was a severe impairment of larval-pupal cuticle tanning, leading to pupae with less yellowish pigment or that were completely white and transparent, when we injected 2 µL of 24.4 mM or 73.27 mM 3-IT into 3rd-instar larvae or prepupae. These results suggest that TH is an important enzyme for the normal growth and pupal pigmentation of Z. tau and that TH is a potential gene target for use in the control of Z. tau.

We evaluated a protein bait based on an enzymatically hydrolyzed beer yeast and two widely used baits including a sugar + vinegar + wine mixture and apple cider vinegar for their ability to trap Drosophila suzukii (Matsumura) in the laboratory and outdoors. The protein bait was a more attractive lure than the other tested baits, with the protein bait capturing significantly more female and male adults at different developmental stages than the other baits. Furthermore, the protein bait with 20% vinegar attracted significantly more adult flies than the other baits, and the protein bait without dilution attracted the most adults. Except for the addition of 0.05% spinosad, increased insecticide content in protein bait reduced its attractiveness to adult flies. Moreover, we found that D. suzukii has a bimodal activity pattern in visiting protein bait, where maximum activity occurs during 8:00-10:00 a.m. and 14:00-16:00 p.m. The antennae and maxillary palpi played an important role in detecting the chemicals emitted from the protein bait, as the number of intact flies that responded to new protein bait was significantly higher than the number of flies without antennae that responded, but almost 15% of defective flies still responded to the protein bait. Our results suggested that the protein bait based on spent brewery yeast could be a promising alternative for D. suzukii population monitoring and for managing this pest when combined with bioinsecticides, providing guidance for using this protein bait as a component of integrated pest management.

This study explored the potential for Bactrocera dorsalis (Hendel) (Diptera: Tephritidae) larvae hatched from irradiated eggs as hosts for Diachasmimorpha longicaudata (Ashmead) (Hymenoptera: Braconidae). B. dorsalis eggs of three different ages (12-, 24-, and 36-h old) were analyzed for hatchability, pupation rate, pupal weight, emergence rate, and sex ratio after exposure to different doses of radiation (5 and 10 Gy) at different dose rates (1 and 6 Gy/min). For the eggs of different ages exposed to radiation, only the hatchability and pupal weight of 36-h-old eggs exposed to the dose rate of 1 Gy/min were not affected; therefore, 6 Gy/min was not suitable for irradiating eggs. The viability of the parents and progenies of D. longicaudata when the parents were reared from 36-h-old eggs irradiated at nine different doses (0, 5, 10, 15, 20, 25, 30, 35, and 40 Gy) under laboratory conditions were investigated. The emergence percentage, sex ratio, and longevity of parasitoids developed from irradiated eggs were similar to those reared from nonirradiated hosts. A significant increase in larva mortality was observed for the eggs irradiated at doses above 25 Gy, and no redundant adult flies emerged at doses above 15 Gy. Hence, for B. dorsalis eggs to be applied in the mass rearing of D. longicaudata, the age of 36 h and a dose of 20-25 Gy are the optimal parameters. The results reveal that hosts and parasitoids need not be separated, enabling a reduction in cost, labor, and time and resulting in an improved mass rearing procedure for D. longicaudata.

RESUMO

OBJECTIVE: Evaluating the accuracy and safety as well as the equivalence compared with the control kit of RIDA QUICK Norovirus detection kit(R-Biopharm, Germany). METHODS: Based on the results of commercially available IDEA Norovirus detection kit (ELISA), the sensitivity and specificity and accuracy of RIDA QUICK Norovirus detection kit (immunochromatographic assay) were evaluated. RESULTS: The sensitivity and specificity of RIDA QUICK Norovirus detection kit were 98.4% and 92.4%, and the accuracy was 97.6% compared with the control kit. CONCLUSION: RIDA QUICK Norovirus detection kit has good sensitivity and specificity for the detection of norovirus antigens.
