RESUMO

With the increasing environmental and ecological problems caused by petroleum-based packaging materials, the focus has gradually shifted to natural resources for the preparation of functional food packaging materials. In addition to biodegradable properties, nanocellulose (NC) mechanical properties, and rich surface chemistry are also fascinating and desired to be one of the most probable green packaging materials. In this review, we firstly introduce the recent progress of novel applications of NC in food packaging, including intelligent packaging, nano(bio)sensors, and nano-paper; secondly, we focus on the modification techniques of NC to summarize the properties (antimicrobial, mechanical, hydrophobic, antioxidant, and so on) that are required for food packaging, to expand the new synthetic methods and application areas. After presenting all the latest advances related to material design and sustainable applications, an overview summarizing the safety of NC is presented to promote a continuous and healthy movement of NC toward the field of truly sustainable packaging.

RESUMO

Flash Joule heating (FJH) is an emerging and profitable technology for converting inexhaustible biomass into flash graphene (FG). However, it is challenging to produce biomass FG continuously due to the lack of an integrated device. Furthermore, the high-carbon footprint induced by both excessive energy allocation for massive pyrolytic volatiles release and carbon black utilization in alternating current-FJH (AC-FJH) reaction exacerbates this challenge. Here, we create an integrated automatic system with energy requirement-oriented allocation to achieve continuous biomass FG production with a much lower carbon footprint. The programmable logic controller flexibly coordinated the FJH modular components to realize the turnover of biomass FG production. Furthermore, we propose pyrolysis-FJH nexus to achieve biomass FG production. Initially, we utilize pyrolysis to release biomass pyrolytic volatiles, and subsequently carry out the FJH reaction to focus on optimizing the FG structure. Importantly, biochar with appropriate resistance is self-sufficient to initiate the FJH reaction. Accordingly, the medium-temperature biochar-based FG production without carbon black utilization exhibited low carbon emission (1.9 g CO2-eq g-1 graphene), equivalent to a reduction of up to ~86.1% compared to biomass-based FG production. Undoubtedly, this integrated automatic system assisted by pyrolysis-FJH nexus can facilitate biomass FG into a broad spectrum of applications.

RESUMO

Two-dimensional (2D) ferroelectric metals (FEMs) possess intriguing characteristics, such as unconventional superconductivity and the nonlinear anomalous Hall effect. However, their occurrence is exceedingly rare due to mutual repulsion between ferroelectricity and metallicity. In addition, further incorporating other features like ferromagnetism into FEMs to enhance their functionalities poses a significantly greater challenge. Here, via first-principles calculations, we demonstrate a case of an FEM that features a coexistence of room-temperature ferromagnetism, ferroelectricity, and metallicity in a thermodynamically stable 2D Os2Se3. It presents a vertical electric polarization of 3.00 pC/m that exceeds those of most FEMs and a moderate polarization switching barrier of 0.22 eV per formula unit. Moreover, 2D Os2Se3 exhibits robust ferromagnetism (Curie temperature TC ≈ 527 K) and a sizable magnetic anisotropy energy (-30.87 meV per formula unit). Furthermore, highly magnetization-dependent electrical conductivity is revealed, indicative of strong magnetoelectric coupling. Berry curvature calculation suggests that the FEM might exhibit nontrivial band topology.

RESUMO

Churches in China are material witnesses of cultural dissemination, and their architectural forms are in the process of localization. In order to determine the optimal degree of localization of church facades as well as to study the correlation between visual behavior and subjective cognition, five church facades with different degrees of localization were selected in present study, and the questionnaire survey as well as eye-tracking technology were used to collect data from two aspects: subjective cognition (the impression and acceptance levels) and objective eye movement (the first fixation duration, total fixation duration, fixation count, and visit count). The results showed the differences in public perceptions of church facades, and the impression of participants was continuously enhanced with the increasing of localization degree of church facade, while the acceptance level showed a U-shaped change. What's more, the correlation between the impression level and the first fixation duration was found to be 0.910, the Pearson coefficient between the acceptance level and the total fixation duration was found to be 0.928, indicating that eye-tracking indicators could accurately reflect the subjective cognition of the public. Performed analyses demonstrated that eye-tracking technology would provides an important technical mean for the design, conservation, and renewal of building facades.

RESUMO

The emergence and rapid spread of Mpox (formerly monkeypox) have caused significant societal challenges. Adequate and appropriate diagnostics procedures are an urgent necessity. Herein, we discover a pair of aptamers through the systematic evolution of ligands by exponential enrichment (SELEX) that exhibit high affinity and bind to different sites towards the A29 protein of the Mpox virus. Subsequently, we propose a facile, sensitive, convenient CRISPR/Cas12a-mediated aptasensor for detecting the A29 antigen. The procedure employs the bivalent aptamers recognition, which induces the formation of a proximity switch probe and initiates subsequent cascade strand displacement reactions, then triggers CRISPR/Cas12a DNA trans-cleavage to achieve the sensitive detection of Mpox. Our method enables selective and ultrasensitive evaluation of the A29 protein within the range of 1 ng mL-1 to 1 µg mL-1, with a limit of detection (LOD) at 0.28 ng mL-1. Moreover, spiked A29 protein recovery exceeds 96.9%, while the detection activity remains above 91.9% after six months of storage at 4 °C. This aptasensor provides a novel avenue for exploring clinical diagnosis in cases involving Mpox as facilitating development in various analyte sensors.

RESUMO

The trophoblast epithelial-to-mesenchymal transition (EMT) is a procedure related to embryo implantation, spiral artery establishment and fetal-maternal communication, which is a key event for successful pregnancy. Inadequate EMT is one of the pathological mechanisms of recurrent miscarriage (RM). Whole-exome sequencing revealed that the mutation of bromodomain PHD-finger transcription factor (BPTF) was strongly associated with RM. In the present study, the effects of BPTF on EMT and the underlying mechanism were investigated. We found that the expression of BPTF in the villi of RM patients was significantly downregulated. Gene Ontology (GO) analysis revealed that BPTF participated in cell adhesion. The knockdown of BPTF prevented EMT and attenuated trophoblast invasion in vitro. BPTF activated Slug transcription by binding directly to the promoter region of the Slug gene. Interestingly, the protein levels of both Slug and BPTF were decreased in the villous cytotrophoblasts (VCTs) of RM villi. In conclusion, BPTF participates in the regulation of trophoblast EMT by activating Slug expression, suggesting that BPTF defects are an important factor in RM pathogenesis.

RESUMO

OBJECTIVE: To investigate the independent risk variables associated with the potential invasiveness of ductal carcinoma in situ (DCIS) on multi-parametric ultrasonography, and further construct a nomogram for risk assessment. METHODS: Consecutive patients from January 2017 to December 2022 who were suspected of having ductal carcinoma in situ (DCIS) based on magnetic resonance imaging or mammography were prospectively enrolled. Histopathological findings after surgical resection served as the gold standard. Grayscale ultrasound, Doppler ultrasound, shear wave elastography (SWE), and contrast-enhanced ultrasound (CEUS) examinations were preoperative performed. Binary logistic regression was used for multifactorial analysis to identify independent risk factors from multi-parametric ultrasonography. The correlation between independent risk factors and pathological prognostic markers was analyzed. The predictive efficacy of DCIS associated with invasiveness was assessed by logistic analysis, and a nomogram was established. RESULTS: A total of 250 DCIS lesions were enrolled from 249 patients, comprising 85 pure DCIS and 165 DCIS with invasion (DCIS-IDC), of which 41 exhibited micro-invasion. The multivariate analysis identified independent risk factors for DCIS with invasion on multi-parametric ultrasonography, including image size (>2cm), Doppler ultrasound RI (≥0.72), SWE's Emax (≥66.4 kPa), hyper-enhancement, centripetal enhancement, increased surrounding vessel, and no contrast agent retention on CEUS. These factors correlated with histological grade, Ki-67, and human epidermal growth factor receptor 2 (HER2) (P < 0.1). The multi-parametric ultrasound approach demonstrated good predictive performance (sensitivity 89.7 %, specificity 73.8 %, AUC 0.903), surpassing single US modality or combinations with SWE or CEUS modalities. Utilizing these factors, a predictive nomogram achieved a respectable performance (AUC of 0.889) for predicting DCIS with invasion. Additionally, a separate nomogram for predicting DCIS with micro-invasion, incorporating independent risk factors such as RI (≥0.72), SWE's Emax (≥65.2 kPa), and centripetal enhancement, demonstrated an AUC of 0.867. CONCLUSION: Multi-parametric ultrasonography demonstrates good discriminatory ability in predicting both DCIS with invasion and micro-invasion through the analysis of lesion morphology, stiffness, neovascular architecture, and perfusion. The use of a nomogram based on ultrasonographic images offers an intuitive and effective method for assessing the risk of invasion in DCIS. Although the nomogram is not currently considered a clinically applicable diagnostic tool due to its AUC being below the threshold of 0.9, further research and development are anticipated to yield positive outcomes and enhance its viability for clinical utilization.

RESUMO

Key Clinical Message: Sexual dysfunction induced by hyperprolactinemia accompanied by reduced luteinizing hormone (LH) is common in anrology clinics. A low dose of bromocriptine is helpful for restoring penile erectile function and libido in patients. Abstract: Sexual dysfunction is closely related to hormonal disorders, of which prolactin (PRL) and luteinizing hormone (LH) disorders are common. How to treat sexual dysfunction induced by hyperprolactinemia accompanied by reduced LH levels is worth discussing. In this study, we aimed to present the case of a 35-year-old male patient with sexual dysfunction. The treatment process and physical and laboratory examination results were recorded. Before treatment, the PRL and LH levels in this patient were 31.27 ng/mL and 1.62 mIU/mL, respectively. The International Index of Erectile Function-5 (IIEF-5) score was initially 14 points. After regular treatment with low doses of bromocriptine and tadalafil, the hormonal disorder was corrected (PRL: 11.16 ng/mL and LH: 2.28 mIU/mL) and sexual function was recovered (IIEF-5: 23 points). This case report suggested a sufficient exposure to low-dose bromocriptine for such patients. Conversely, the exogenous supplementation of human chorionic gonadotropin may not be appropriate.

RESUMO

Advanced portable healthcare devices with high efficiencies, small pressure drops, and high-temperature resistance are urgently desired in harsh environments with high temperatures, high humidities, and high levels of atmospheric pollution. Triboelectric nanogenerators (TENGs), which serve as energy converters in a revolutionary self-powered sensor device, present a sustainable solution for meeting these requirements. In this work, we developed a porous negative triboelectric material by synthesizing ZIF-8 on the surface of a cellulose/graphene oxide aerogel, grafting it with trimethoxy(1H,1H,2H,2H-heptadecafluorodecyl)silane, and adding a negative corona treatment, and it was combined with a positive triboelectric material to create a cellulose nanofiber-based TENG self-powered filter. The devices achieved a balance between a small pressure drop (53 Pa) and high filtration efficiency (98.97%, 99.65%, and 99.93% for PM0.3, PM0.5, and PM1, respectively), demonstrating robust filtration properties at high temperatures and high humidities. Our work provides a new approach for developing self-powered wearable healthcare devices with excellent air filtration properties.

RESUMO

Background: Systemic lupus erythematosus (SLE)-associated hepatitis ("lupus hepatitis") was one of the most frequent causes of liver function abnormalities in patients with SLE. Lupus hepatitis (LH) is commonly treated with conventional treatment, including non-steroidal anti-inflammatory drugs, corticosteroids, and immunomodulators. However, in refractory cases, other treatment options may be required.Methodology: We report the case of a patient with lupus hepatitis refractory to both conventional therapy and belimumab who was successfully treated with telitacicept, a new dual B lymphocyte stimulator (BLyS)/APRIL (a proliferation-inducing ligand) inhibitor.Literature review was performed on PubMed search forum.Result: The specific search term was "telitacicept", 23 papers were searched, among them 10 case reports/series articles reporting telitacicept treatment were elected.Apart from our literature reporting the effectiveness of telitacicept in treating LH, there is no report on it in treating LH.Conclusion: This case suggests that telitacicept should be an effective and safe treatment for LH refractory, even to those who failed to belimumab based on the standard treatment, and can reduce the dosage of glucocorticoids. However, further investigations, particularly prospective randomized controlled trials, are warranted to verify our findings and ensure patient safety.

RESUMO

PURPOSE: The objective of this study was to investigate the independent risk factors and associated predictive values of contrast-enhanced ultrasound (CEUS), shear wave elastography (SWE), and strain elastography (SE) for high-risk lesions (HRL) and malignant tumors (MT) among nonpalpable breast masses classified as BI-RADS category 4 on conventional ultrasound. METHODS: This prospective study involved consecutively admitted patients with breast tumors from January 2018, aiming to explore the management of BI-RADS category 4 breast tumors using CEUS and elastography. We conducted a retrospective review of patient data, focusing on those with a history of a nonpalpable mass as the primary complaint. Pathologic findings after surgical resection served as the gold standard. The CEUS arterial-phase indices were analyzed using contrast agent arrival-time parametric imaging processing mode, while quantitative and qualitative indices were examined on ES images. Independent risk factors were identified through binary logistic regression multifactorial analysis. The predictive efficacy of different modalities was compared using a receiver operating characteristics curve. Subsequently, a nomogram for predicting the risk of HRL/MT was established based on a multifactorial logistic regression model. RESULTS: A total of 146 breast masses from 146 patients were included, comprising 80 benign tumors, 12 HRLs, and 54 MTs based on the final pathology. There was no significant difference in pathologic size between the benign and HRL/MT groups [8.00(6.25,10.00) vs. 9.00(6.00,10.00), P = 0.506]. The diagnostic efficacy of US plus CEUS exceeded that of US plus SWE/SE for BI-RADS 4 nonpalpable masses, with an AUC of 0.954 compared to 0.798/0.741 (P ï¼œ 0.001). Further stratified analysis revealed a more pronounced improvement for reclassification of BI-RADS 4a masses (AUC: 0.943 vs. 0.762/0.675, P ï¼œ 0.001) than BI-RADS 4b (AUC:0.950 vs. 0.885/0.796, P>0.05) with the assistance of CEUS than SWE/SE. Employing downgrade CEUS strategies resulted in negative predictive values ranging from 95.2 % to 100.0 % for BI-RADS 4a and 4b masses. Conversely, using upgrade nomogram strategies, which included the independent predictive risk factors of irregular enhanced shape, poor defined enhanced margin, earlier enhanced time, increased surrounding vessels, and presence of contrast agent retention, the diagnostic performance achieved an AUC of 0.947 with good calibration. CONCLUSION: After investigating the potential of CEUS and ES in improving risk assessment and diagnostic accuracy for nonpalpable BI-RADS category 4 breast masses, it is evident that CEUS has a more significant impact on enhancing classification compared to ES, particularly for BI-RADS 4a subgroup masses. This finding suggests that CEUS may offer greater benefits in improving risk assessment and diagnostic accuracy for this specific subgroup of breast masses.

RESUMO

As important chemical raw materials and organic solvents, halogenated hydrocarbons not only play an important role in economic development, but are also the main source of environmental pollution. This study proposed an improved groundwater risk assessment model system, aimed at identifying and treating contaminants at leak sites. Groundwater ubiquity score (GUS) was used to evaluate the leachability of organic pollutants. The entropy-weighted water quality index (EWQI) method was used to assess the comprehensive quality of groundwater at the site. An improved groundwater health risk assessment model was constructed to analyze the health risks of groundwater. The sources of organic pollutants were identified based on the positive matrix factorization (PMF) model. Self-organizing mapping (SOM) and the K-means algorithm were integrated to classify and manage pollution source areas. The results showed that groundwater in the study area was strongly affected by human activities. The pollution source was located in a factory near S05. Different organic pollutants were highly leachable and had high potential to contaminate surrounding groundwater. 1,2-dichloropropane and 1,2,3-trichloropropane caused the largest range of contamination. The groundwater pollution index in the study area was high, and 72% of the monitoring points were non-drinkable. Both the carcinogenic and non-carcinogenic indexes of groundwater far exceeded the international standard limits and had a great impact on human health. 1,2,3-trichloropropane and 1,2-dichloropropane were major non-carcinogenic risk factors. The leakage of pollutants and pesticide solvents were the main causes of groundwater pollution. Cluster areas III and II were areas with significant pollution impacts and needed to be monitored intensively. Most areas were cluster I, with relatively low risk. This study can provide technical support for groundwater pollution risk assessment and management in similar industrial parks.

RESUMO

BACKGROUND: The consolidation tumor ratio (CTR) is a predictor of invasiveness in peripheral T1N0M0 lung adenocarcinoma. However, its association with spread through air spaces (STAS) remains largely unexplored. We aimed to explore the correlation between the CTR of primary tumors and STAS in peripheral T1N0M0 lung adenocarcinoma. METHODS: We collected data from patients who underwent surgery for malignant lung neoplasms between January and November 2022. Univariate and multivariate analyses following propensity-score matching with sex, age, BMI, were performed to identify the independent risk factors for STAS. The incidence of STAS was compared based on pulmonary nodule type. A smooth fitting curve between CTR and STAS was produced by the generalized additive model (GAM) and a multiple regression model was established using CTR and STAS to determine the dose-response relationship and calculate the odds ratio (OR) and 95% confidence interval (CI). RESULTS: 17 (14.5%) were diagnosed with STAS. The univariate analysis demonstrated that the history of the diabetes, size of solid components, spiculation, pleural indentation, pulmonary nodule type, consolidation/tumor ratio of the primary tumor were statistically significant between the STAS-positive and STAS-negative groups following propensity-score matching(p = 0.047, 0.049, 0.030, 0.006, 0.026, and < 0.001, respectively), and multivariate analysis showed that the pleural indentation was independent risk factors for STAS (with p-value and 95% CI of 0.043, (8.543-68.222)). Moreover, the incidence of STAS in the partially solid nodule was significantly different from that in the solid nodule and ground-glass nodule (Pearson Chi-Square = 7.49, p = 0.024). Finally, the smooth fitting curve showed that CTR tended to be linearly associated with STAS by GAM, and the multivariate regression model based on CTR showed an OR value of 1.24 and a p-value of 0.015. CONCLUSIONS: In peripheral stage IA lung adenocarcinoma, the risk of STAS was increased with the solid component of the primary tumor. The pleural indentation of the primary tumor could be used as a predictor in evaluating the risk of the STAS.

RESUMO

BACKGROUND: A compact PET/SPECT/CT system Inliview-3000B has been developed to provide multi-modality information on small animals for biomedical research. Its PET subsystem employed a dual-layer-offset detector design for depth-of-interaction capability and higher detection efficiency, but the irregular design caused some difficulties in calculating the normalization factors and the sensitivity map. Besides, the relatively larger (2 mm) crystal cross-section size also posed a challenge to high-resolution image reconstruction. PURPOSE: We present an efficient image reconstruction method to achieve high imaging performance for the PET subsystem of Inliview-3000B. METHODS: List mode reconstruction with efficient system modeling was used for the PET imaging. We adopt an on-the-fly multi-ray tracing method with random crystal sampling to model the solid angle, crystal penetration and object attenuation effect, and modify the system response model during each iteration to improve the reconstruction performance and computational efficiency. We estimate crystal efficiency with a novel iterative approach that combines measured cylinder phantom data with simulated line-of-response (LOR)-based factors for normalization correction before reconstruction. Since it is necessary to calculate normalization factors and the sensitivity map, we stack the two crystal layers together and extend the conventional data organization method here to index all useful LORs. Simulations and experiments were performed to demonstrate the feasibility and advantage of the proposed method. RESULTS: Simulation results showed that the iterative algorithm for crystal efficiency estimation could achieve good accuracy. NEMA image quality phantom studies have demonstrated the superiority of random sampling, which is able to achieve good imaging performance with much less computation than traditional uniform sampling. In the spatial resolution evaluation based on the mini-Derenzo phantom, 1.1 mm hot rods could be identified with the proposed reconstruction method. Reconstruction of double mice and a rat showed good spatial resolution and a high signal-to-noise ratio, and organs with higher uptake could be recognized well. CONCLUSION: The results validated the superiority of introducing randomness into reconstruction, and demonstrated its reliability for high-performance imaging. The Inliview-3000B PET subsystem with the proposed image reconstruction can provide rich and detailed information on small animals for preclinical research.

RESUMO

The placenta plays a pivotal role in the maintenance of normal pregnancy, but how it forms, matures, and performs its function remains poorly understood. Here, we describe a novel mouse line (Prl3d1-iCre) that expresses iCre recombinase under the control of the endogenous prl3d1 promoter. Prl3d1 has been proposed as a marker for distinguishing trophoblast giant cells (TGCs) from other trophoblast cells in the placenta. The in vivo efficiency and specificity of the Cre line were analyzed by interbreeding Prl3d1-iCre mice with B6-G/R reporter mice. Through anatomical studies of the placenta and other tissues of Prl3d1-iCre/+; B6-G/R mouse mice, we found that the tdTomato signal was expressed in parietal trophoblast giant cells (P-TGCs). Thus, we report a mouse line with ectopic Cre expression in P-TGCs, which provides a valuable tool for studying human pathological pregnancies caused by implantation failure or abnormal trophoblast secretion due to aberrant gene regulation.

RESUMO

The realization of the electrical control of spin is highly desirable. One promising approach is by regulating the Rashba spin-orbit coupling effect of materials through external electric fields. However, this method requires materials to possess either a high electric field response and a large Rashba constant or the simultaneous presence of Rashba splitting and ferroelectric polarization. These stringent requirements result in a scarcity of suitable materials. In order to surpass these limitations and exploit a new prospect for spin manipulation via the Rashba effect, a conceptual class of materials named bipolar Rashba semiconductors (BRS) is proposed, whose valence band and conduction band possess opposite spin texture directions when approaching the Fermi level. The unique electronic structure of BRS makes it feasible to reverse the spin precession by simply applying a gate voltage. The existence of BRS is confirmed through first-principles calculations on the two-dimensional (2D) material AlBiS3.

RESUMO

Cellulose-based triboelectric nanogenerators (TENGs) have attracted widespread attention due to the low cost and environmentally friendly characteristics of cellulose. However, achieving high electrical energy output from these generators still presents significant challenges. Here, cellulose is dissolved-regenerated to form a composite aerogel with high specific surface area, in which cellulose-based composites with excellent negative triboelectric properties are developed by coupling the rich 3D network structure of the regenerated cellulose aerogel, modified barium titanate, and poly(vinylidene fluoride). The TENGs assembled from the composite materials exhibit an output voltage of 1040 V and a current of 1.165 mA at an external force of 8 N and a frequency of 4 Hz, outperforming all cellulose-based negative triboelectric materials. In addition, the nanogenerators have a stable electrical energy output capacity, with no significant property degradation in 100 000 contact-separation tests. The excellent electrical output property of the composite materials enables them to harvest energy from human movement and waterdrops, demonstrating their great application prospects in wearable devices, energy harvesting devices, self-powered sensors, and other fields.

RESUMO

The purpose of this study was to determine which shrub species will enhance soil anti-scouring resistance on an ecological slope. Root traits and soil anti-scouring resistance of three shrubs (Amorpha fruticosa Linn (AFL), Swida alba Opiz (SAO) and Lespedeza bicolor Turcz (LBT)) were measured. Results showed that root biomass and root morphological traits of three shrubs were significantly correlated with the soil anti-scouring resistance index. According to the composition characteristic values, root morphological traits among the three shrubs had a high contribution rate. Under two slopes and two rainfall conditions, when root biomass and root morphological traits (e.g., root length, root volume and root surface area) were identical, AFL had the highest soil anti-scouring resistance index. These results suggested that root biomass and morphological traits of AFL had more significant effects on soil anti-scouring resistance comparing with SAO and LBT. Therefore, in engineering practice, AFL with stronger soil anti-scouring resistance can be selected as slope plants.

RESUMO

Bio-based carbon fibers derived from lignin have gained significant attention due to their diverse and renewable sources, ease of extraction, and low cost. However, the current limitations of low specific surface area and insufficient electrical conductivity hinder the widespread application of lignin-derived carbon fibers (LCFs). In this work, highly conductive and porous LCFs are developed through melt-blowing, pretreatment, and carbonization processes. The effects of the carbonization temperature and heating rate on the structures and properties of the LCFs are systematically investigated. The resultant LCFs exhibit high electrical conductivity (71 400 S m-1) and a large specific surface area (923 m2 g-1). The assembled lithium-ion battery based on the LCF anodes demonstrates a long cycle life of >800 cycles and a high specific capacity of 466 mA h g-1. The findings of this study hold practical significance for promoting the utilization of lignin in the fields of energy storage, adsorption, and beyond.

RESUMO

OBJECTIVE: To investigate the effect of phorbol-12-myristate-13-ace-tate (TPA) on the proliferation and apoptosis of acute promyelocytic leukemia cell line NB4 and its molecular mechanism. METHODS: The effect of different concentrations of TPA on the proliferation of NB4 cells at different time points was detected by CCK-8 assay. The morphological changes of NB4 cells were observed by Wright-Giemsa staining. The cell cycle and apoptosis of NB4 cells after TPA treatment were detected by flow cytometry. The mRNA expressions of NB4 cells after TPA treatment were analyzed by high-throughput microarray analysis and real-time quantitative PCR. Western blot was used to detect the protein expression of CDKN1A, CDKN1B, CCND1, MYC, Bax, Bcl-2, c-Caspase 3, c-Caspase 9, PIK3R6, AKT and p-AKT. RESULTS: Compared with the control group, TPA could inhibit the proliferation of NB4 cells, induce the cells to become mature granulocyte-monocyte differentiation, and also induce cell G1 phase arrest and apoptosis. Differentially expressed mRNAs were significantly enriched in PI3K/AKT pathway. TPA treatment could increase the mRNA levels of CCND1, CCNA1, and CDKN1A, while decrease the mRNA level of MYC. It could also up-regulate the protein levels of CDKN1A, CDKN1B, CCND1, Bax, c-Caspase 3, c-Caspase 9, and PIK3R6, while down-regulate MYC, Bcl-2, and p-AKT in NB4 cells. CONCLUSION: TPA induces NB4 cell cycle arrest in G1 phase and promotes its apoptosis by regulating PIK3/AKT signaling pathway.
