RESUMO

Oral reference dose (RfD) is a key parameter for deriving the human health ambient water quality criteria (AWQC) for non-carcinogenic substances. In this study, a non-experimental approach was used to calculate the RfD values, which explore the potential correlation between toxicity and physicochemical characteristics and the chemical structure of pesticides. The molecular descriptors of contaminants were calculated using T.E.S.T software from EPA, and a prediction model was developed using a stepwise multiple linear regression (MLR) approaches. Approximately 95% and 85% of the data points differ by less than 10-fold and 5-fold between predicted values and true values, respectively, which improves the efficiency of RfD calculation. The model prediction values have certain reference values in the absence of experimental data, which is beneficial to the advancement of contaminant health risk assessment. In addition, using the prediction model constructed in this manuscript, the RfD values of two pesticide substances in the list of priority pollutants are calculated to derive human health water quality criteria. Furthermore, an initial assessment of the health risk was performed by the quotient value method based on the human health water quality criteria calculated by the prediction model.

BACKGROUND AND STUDY AIM: There is currently a lack of sensitive biomarkers for the diagnosis of hepatocellular carcinoma (HCC). Low expression of cylindromatosis (CYLD), a tumor suppressor gene that encodes a deubiquitinase, is associated with the development of HCC. The present study, therefore, aimed to determine the clinical utility of measuring CYLD expression in the early diagnosis of HCC. PATIENTS AND METHODS: The present study comprised 257 patients from the Affiliated Hospital of Qingdao University including 90 patients with HCC, 41 patients with liver cirrhosis (LC), 46 patients with hepatitis B (HB), and 80 healthy controls. qPCR was used to measure the amounts of CYLD mRNA in stored blood samples. The sensitivity and specificity of CYLD mRNA in diagnosing HCC was analyzed using receiver operator characteristic (ROC) curves. We also obtained HCC data from the Oncomine database to further verify our results. RESULTS: The relative levels of CYLD mRNA in peripheral blood from patients with HCC (median, 0.060; interquartile range [IQR], 0.019-0.260) was significantly lower than in blood from patients with LC (median, 3.732; IQR, 0.648-14.573), HB (median, 0.419; IQR, 0.255-1.809) and healthy controls (median, 1.262; IQR, 0.279-3.537; P < 0.05). CYLD mRNA levels in peripheral blood were significantly higher in patients with LC compared to healthy controls and patients with HB. Oncomine data demonstrated that CYLD mRNA expression levels in HCC tissues were significantly lower than in normal liver tissues. ROC analysis demonstrated that the combined use of peripheral blood levels of CYLD and AFP had the greatest diagnostic accuracy for HCC (area under the curve (AUC), 0.897; 95 % confidence interval [CI], 0.853-0.942). CYLD had utility as a supplementary marker to AFP for diagnosing HCC. CONCLUSION: Circulating levels of CYLD mRNA are significantly decreased in patients with HCC, indicating CYLD may have utility as a biomarker of HCC. Combined measurement of CYLD mRNA and AFP protein had the greatest diagnostic accuracy.

Pesticide pollution has been one serious ecological and environmental issue due to its wide application, high toxicity, and complex environmental behavior. The fugacity model has been widely used to quantify biogeochemical cycles of pesticides due to its clear compartments, simple structure, and easy-accessible data. However, the lack of detailed hydrological processes limits its application for large and heterogeneous watershed. In present study, a new framework was proposed through integration of hydrological processes of SWAT and pesticide fate of fugacity model, and was applied into a typical watershed in the Three Gorges Reservoir Area, China. The results showed that surface runoff, soil erosion, and percolation varied spatiotemporally, which highlighted the importance of considering regional and seasonal heterogeneity of pesticide transport variables in the fugacity model. The amount of dichlorvos (DDV) and chlorpyrifos (CHP) in air, water, soil, and sediment phase were estimated as 0.26 kg, 19.77 kg, 1.06 × 104 kg, and 0.55 kg, respectively. Spatiotemporally, pesticide concentrations in water phase peaked in summer, while the middle and southwest regions of the watershed were identified as the hotspots for pesticide pollution. Compared with the classical model, the new framework provided technical support for the pesticide assessment at watershed scale with heterogeneous hydrological conditions, which can be easily extended to other watersheds, and integrated with other models for comprehensive agricultural management.

AIM: To observe the role and mechanism of autophagy in retinal pigment epithelial cell (RPE) damaged by high glucose, so as to offer a new idea for the treatment of diabetic retinopathy (DR). METHODS: ARPE-19, a human RPE cell line cultured in vitro was divided into the normal control (NC), autophagy inhibitor 3-methyladenine (3-MA), high-glucose (HG), and HG+3-MA groups. Cell viability was detected by CCK-8 assay and the apoptosis rate was measured by flow cytometry. The protein expressions of apoptosis markers, including Bax, Bcl-2, and Caspase-3, as well as autophagy marker including microtubule-related protein 1 light chain 3 (LC3), p62, and mechanistic target of rapamycin (mTOR) were detected by Western blotting. Autophagic flux was detected by transfection with Ad-mCherry-GFP-LC3B. RESULTS: Under high glucose conditions, the viability of ARPE-19 was decreased, and the apoptosis rate increased, the protein expressions of Bax, Caspase-3, and LC3-II/LC3-I were all increased and the expressions of Bcl-2, p62 and p-mTOR decreased, and autophagic flux was increased compared with that of the controls. Treatment with 3-MA reversed all these changes caused by high glucose. CONCLUSION: The current study demonstrates the mechanisms of cell damage of ARPE-19 through high glucose/mTOR/autophagy/apoptosis pathway, and new strategies for DR may be developed based on autophagy regulation to manage cell death of RPE cells.

In recent years, excessive application and loss of pesticides have caused great risks to the aquatic systems, but the spatio-temporal variability in the ecological risk that agricultural pesticides pose to aquatic systems has not been explored at the national scale. In this study, an integrated assessment framework was proposed for the potential ecological risk of surface water caused by agricultural pesticide loss. The spatio-temporal variability in the potential ecological risk caused by agricultural pesticide runoff was evaluated. Based on the results, the total pesticide emissions increased from 165.47 tons in 2004 to 179.77 tons in 2017. Among the three pesticide types, insecticide had the largest application, but its runoff was estimated as the lowest. High-risk areas of insecticide runoff were concentrated in the east, south and central part of China, while the central region of China was identified as a hotspot due to the high and the ever-increasing ecological risk. This study provides an integrated method for potential ecological risk assessment of agricultural pesticide runoff to adjacent water bodies in large-scale regions and the results of the study have direct implications for environmental policies on pesticide management in China and around the world.

Pesticide loss during agricultural development has a serious effect on related water quality, and the critical concern is quantifying the potential exposure risks that pesticide loss pose to water quality at the national scale. In this study, an integrated assessment framework is proposed to scale emission factors from 232 monitoring plots to the national scale, while also considering the physicochemical properties of pesticides in dissolved or adsorbed forms. Based on the results of this study, the total pesticide emissions increased by 29.39% from 146.55 tons in 2004 to 189.62 tons in 2013 and the average loss intensities of insecticides, herbicides and fungicides were 35.25 g/km2, 44.24 g/km2 and 48.57 g/km2, respectively. Central and Southeastern China are identified as hotspots for pesticide loss, while the proportions of high or extremely high-risk areas mainly comprise >50% of farmland. In addition, single-field crops and single-crop rice are the major cropping patterns for pesticide loss in Northern and Southern China. Our results identify key areas for the management of pesticides at the national scale and have direct implications for environmental policies on reducing the potential exposure risk of agricultural pesticides to water quality.

Tungstate enrichment in aquatic systems may cause negative environmental and health effects. This study addresses tungstate removal from aqueous solution by nanocrystalline iowaite, an iron-bearing layered double hydroxide, which has not been used for treatment of tungstate-rich waters so far. Tungstate sorption experiments were conducted with various contact times, temperatures, initial tungstate concentrations (0.001-2 mM), and solution pH values (2-13), the results indicating that iowaite sorbed aqueous tungstate effectively and quickly, and the sorption maximum can be up to 71.9 mg/g. Moreover, the tungsten sorption capacity keeps nearly constant at a wide pH range from 3 to 11. Duo to its pH buffering effect, the alkaline conditions were generated by the addition of iowaite, which are favorable for the removal of aqueous tungstate because the polymerization of tungstate can be prohibited at alkaline pH values. Zeta potential, XRD and XPS analyses were employed to clarify the sorption mechanisms, and it was concluded that tungstate was sorbed via its exchange with the chloride originally intercalated into iowaite interlayers as well as its stronger inner-sphere complexation with the Fe atoms located in iowaite layers. Nanocrystalline iowaite is suitable for treating both tungstate-bearing natural waters with moderately high tungstate concentrations and industrial wastewaters extremely rich in tungstate.

RESUMO

With China's rapid economic growth, chemical-related environmental issues have become increasingly prominent, and the environmental management of chemicals has garnered increased attention from the government. This review focuses on the current situation and the application of risk assessment in China's environmental management of industrial chemicals. The related challenges and research needs of the country are also discussed. The Chinese government promulgated regulations for the import and export of toxic chemicals in 1994. Regulations for new chemical substances came into force in 2003, and were revised in 2010 based on the concept of risk management. In order to support the implementation of new regulations, Guidance for Risk Assessment of Chemicals is under development in an attempt to provide the concepts and techniques of risk assessment. With increasing concern and financial support from Chinese government, China is embarking on the fast track of research and development in environmental management of industrial chemicals.
