Identifying and analysing distinct blood cells is crucial for the diagnosis and treatment of diseases in the field of biomedicine. The present study was undertaken to study the cytomorphological and cytochemical characteristics of the blood cells of Zoar, a non-descript indigenous breed of chicken extensively reared under backyard poultry farming in Mizoram, India. For this study, 2 mL of blood samples were aseptically collected from the wings veins of 12 chickens and were processed for light microscopic study under standard protocols. The matured erythrocytes were elliptical, while the immature erythrocytes appeared oval. The heterophils were positive for SBB (SBB), Periodic Acid Schiff (PAS), acid phosphatase, alkaline phosphatase and Arylsulphatase while the eosinophils were positive for SBB, PAS, alkaline phosphatase, cytochrome oxidase and peroxidase. The basophils of were positive for toluidine blue while the thrombocytes were positive for PAS. These cytochemical and cytoenzymatic staining properties plays a very important role in diagnosis, differentiation, and classification of leukaemias.

BACKGROUND: Ultrastructural information regarding the peripheral blood cells of local (Zovawk) pigs from Mizoram, India, is not available in the scientific literature. OBJECTIVES: The present study was designed to reveal the fine structural details of the blood cells from these local pigs using a transmission electron microscope (TEM). METHODS: Blood samples were collected from 12 healthy Zovawk pigs of either sex and processed according to a standard protocol. Processed blood samples were then sent to the All India Institute of Medical Sciences, New Delhi, for further processing and imaging under TEM. Different types of blood cells were viewed under TEM, and different characteristics of these cells were assessed. RESULTS: In the present study, erythrocytes are elongated, biconcave, and nucleated without cytoplasmic organelles. Neutrophils are round with 2-5 lobed nuclei surrounded by cytoplasm with an indistinct bilayered nuclear membrane. The cytoplasm is packed with membrane bound round, oval, and elongated cytoplasmic granules. Eosinophils are round to oval with 2-3 lobed nuclei with distinct nuclear membranes. Basophils are spherical and contained small, medium, and large electron-dense granules. Lymphocytes are small, medium, and large and contained all cellular components. Monocytes are irregularly spherical with slight nuclear indentations. The platelets are elongated, oval, or rounded, with a few pseudopods at the cell surface. CONCLUSIONS: From the present study, we can conclude that the ultrastructural morphology of blood cells from Zovawk pigs resembles those of other domestic animals. However, a few differences have been observed.

The literature on the blood cells of Zovawk pigs is scanty; thus, this study was designed to elaborate the light microscopy (LM) and scanning electron microscopy (SEM) characterization of blood cells of Zovawk pigs by LM and SEM. Twelve different blood samples were aseptically obtained from adult healthy Zovawk pigs, irrespective of sex. To determine the cytomorphological and cytochemical structures of the many produced constituents of blood, blood smears were stained with various stains. The blood samples were treated with various substrates for cytoenzymatic research, and the alterations were noted. A 1000× magnification Olympus Trinocular Research microscope was used to examine the smears. The blood samples were prepared for electron microscopy according to the standard procedure. The prepared samples were delivered to the Sophisticated Analytical Instrumentation Facility (SAIF), All India Institute of Medical Sciences (AIIMS), New Delhi, for SEM imaging. On LM, pig erythrocytes were spherical and nonnucleated. The cytoplasm of the neutrophils was spherical and included cytoplasmic granules. The eosinophils had prominent cytoplasmic granules and were round. Basophils were infrequently present and had cytoplasmic granules that were clear blue. The sizes of small, medium and large lymphocytes were noted. The monocytes were oval or circular. The platelets ranged in form from asymmetric to round. The blood samples were stained for cytochemical analyses using acid ferrocyanide stain for iron, Sudan black blue stain for lipids, toluidine blue stain for mucopolysaccharides, and periodic acid Schiff's stain for glycogen. The cytoenzymatic characteristics were evaluated and compared with substrates treated with acid phosphatase, alkaline phosphatase, peroxidase, arylsulfatase, cytochrome oxidase, beta-glucuronidase, and succinate dehydrogenase. Erythrocytes appeared as biconcave disks under SEM. Two forms of leukocytes were observed, having a rough and pointed cell surface like a flower. It can be concluded that the LM and SEM morphology of blood cells of Zovawk pigs resembled other domestic animals, however, few differences were observed among the discussed animals.

The present study was undertaken to characterize the ultrastructural morphology of the blood cells of commonly reared chickens in the state of Mizoram, India under backyard poultry farming. For this study, 2 ml of whole blood was aseptically collected from the wings veins of 12 chickens of three different breeds namely the Zoar, Aseel and Rhode Island Red and processed for ultrastructural imaging under standard protocols. Under scanning electron microscopy (SEM) the matured erythrocytes of Zoar, Aseel and Rhode Island Red appeared elliptical in shape while the leukocytes and thrombocytes appeared round in shape with variable surface modifications. Under transmission electron microscopy (TEM) the granules of the heterophils of Zoar, Aseel and Rhode Island Red appeared predominantly fusiform in shape, the granules of the eosinophils appeared round in shape and that of the basophils appeared pleomorphic in shape. The cytoplasm of the monocytes, medium-to-large lymphocytes and thrombocytes of Zoar, Aseel and Rhode Island Red under TEM appeared to be vacuolated and granular while that of the small lymphocytes appeared to be granular but non-vacuolated. The study concluded that the ultrastructural characteristics of the blood cells of the three breeds of chicken studied were almost similar to the blood cells of other birds reported earlier.

This study aims to characterize the cytomorphology and ultrastructure of blood cells of native cattle of Mizoram. Twelve numbers of blood samples (10 ml) were collected from the Zobawng cattle, irrespective of sex. Blood smears were prepared and stained with different stains for cytomorphological study. The standard protocol has been followed for preparing blood samples for electron microscopy. Under a light microscope, erythrocytes of cattle were non-nucleated and round. The neutrophils were round, and the cytoplasm contained cytoplasmic granules. The eosinophils were rounded in outline with distinct cytoplasmic granules. The presence of basophils was infrequent with distinct blue color cytoplasmic granules. Small, medium, and large types of lymphocytes were recorded. The monocytes were round to oval in outline. Platelets were irregular to round. The reticulocytes were recorded occasionally, like small blue thin rods or granules. The cytoplasm and nucleus of granulocytes fluoresced greenish-yellow and red, respectively, with supravital stain. Under scanning electron microscopy (SEM), the erythrocytes appeared as biconcave discs. Different leukocytes were observed with their finger-like, plate-like, and narrow cell processes on their surface. Platelets were irregular structures. In transmission electron microscopy (TEM), the erythrocytes appeared anucleated biconcave elongated, neutrophils were roughly rounded with small cytoplasmic processes, and eosinophils were roughly circular with small cytoplasmic processes, the basophils were roughly circular with oval to elongated cytoplasmic granules, lymphocytes were roughly circular with centrally placed well-marked oval indented nucleus and some cytoplasmic processes, monocytes appeared spherical with long thick cytoplasmic processes and the non-nucleated platelets appeared roughly round to elongated. RESEARCH HIGHLIGHTS: Blood tests are a very good tool for determining animal health and diagnosing hematological illnesses. This study showed the cytomorphological and ultrastructural features of the blood cells of native cattle (Zobawng) of Mizoram state of India. The cytomorphological studies revealed that the morphological features of the blood cells of Zobawng cattle resembled to other domestic and wild animals, however, there were some differences in the cellular components. Nuclear DNA appeared orthochromatically greenish-yellow, but cytoplasmic RNA was observed to be metachromatically red under the fluorescence microscope. scanning electron microscopic (SEM) and transmission electron microscopic (TEM) results revealed that the ultrastructural features of the blood cells of Zobawng cattle contain certain cellular morphological differences as compared to the other domestic and wild animals.

The present study reports data on the skull bone morphometry of barking and sambar deer. The skulls of adult barking deer (n = 6) and sambar deer (n = 6) of either sex (n = 3 males and n = 3 females) were collected from the Aizawl Zoological Park, Aizawl, Mizoram, India, with official permission from the Government of Mizoram. Anatomically, barking and sambar deer's skulls were elongated, pyramid-like, dolichocephalic and consisted of thirty-two cranial and facial bones. The cranial bones were eleven (three single and four paired), comprising of occipital, sphenoid, ethmoid, frontal, interparietal, parietal and temporal. The facial bones were twenty-one (one single and ten were paired), consisting of the maxilla, premaxilla (incisive), palatine, pterygoid, nasal, lacrimal, zygomatic (malar), vomer, turbinates, mandible and hyoid. In the present study, altogether 41 different measurements were taken morphologically and 6 different indices were applied. The obtained morphometrical parameters were significantly (p < .01, p < .05) higher in males than females of both species. Species wise, all obtained parameters were higher in sambar deer than barking deer. The obtained 41 different skull parameters and 6 indices showed statistically significant differences (p < .01 and p < .05) between both sexes of barking and sambar deer; however, practically these differences were meagre. The present morphometrical study on the skull of both species can help the wildlife professionals and zoo veterinarians determine the sex of these animals and differentiate it from other domestic and wild small ruminants for solving veterolegal cases. This study's findings will also motivate and assist other comparative studies with various domestic and wild small ruminants.

SUMMARY: Zovawk is newly identified breed of pig of Mizoram approved by breed registration committee of Indian Council of Agricultural Research (ICAR), New Delhi. This study was designed to provide maximum number of morphometrical parameters of skull and some valuable information on clinically important parameters of Zovawk. The skull of the Zovawk was dolichocephalic according to the cephalic index (53.56±0.11). The supraorbital foramina distance, infraorbital foramina distance, skull length, skull width, cranial length and nasal length of the Zovawk were 3.49±0.01 cm, 6.55±0.01 cm, 28.26±0.03 cm, 15.11±0.26 cm, 13.17±0.04 cm and 13.79±0.02 cm, respectively. The orbital margins were found to be incomplete with bilateral variation among the orbits of both the sides. The distance from the process of alveolar socket of canine tooth to the infraorbital canal and from the latter to the root of the fourth upper premolar alveolar tooth directly ventral to it was 4.77±0.04 cm and 1.20±0.01 cm, respectively in Zovawk. The data are of clinical importance as a guide for tracking the infra-orbital nerve, and necessary for its desensitization during the manipulations in the skin of the upper lip, nostril and face at the level of the foramen. The distance between the lateral end of the alveolus of the third incisor tooth to the mental foramen was 3.57±0.04 cm in Zovawk, which is an important landmark for achieving the location of the mental nerve for the regional nerve block Zovawk. The length and height of the mandible were 25.02±0.09 cm and 10.54±0.07 cm, respectively in Zovawk. The morphometric measurements of the skull and applied anatomy of the head region of the Zovawk provide an important baseline data for further research in the field of applied anatomy.


RESUMEN: Zovawk es una raza de cerdo de Mizoram recientemente identificada, aprobada por el comité de registro de razas del Consejo Indio de Investigación Agrícola (ICAR), Nueva Delhi. Este estudio fue diseñado para proporcionar el número máximo de parámetros morfométricos del cráneo y cierta información valiosa sobre parámetros clínicamente importantes de Zovawk. El cráneo de Zovawk es dolicocefálico según el índice cefálico (53,56 ± 0,11). La distancia de foramina supraorbital, la distancia de foramina infraorbital, la longitud del cráneo, el ancho del cráneo, la longitud craneal y la longitud nasal del Zovawk fueron 3.49 ± 0.01 cm, 6.55 ± 0.01 cm, 28.26 ± 0.03 cm, 15.11 ± 0.26 cm, 13.17 ± 0.04 cm y 13.79 ± 0,02 cm, respectivamente. Se encontró que los márgenes orbitales estaban incompletos con una variación bilateral entre las órbitas de ambos lados. La distancia desde el proceso de la cavidad alveolar del diente canino al canal infraorbitario y desde este último a la raíz del cuarto diente alveolar premolar superior directamente ventral fue de 4.77 ± 0.04 cm y 1.20 ± 0.01 cm, respectivamente, en Zovawk. Los datos son de importancia clínica como guía para el seguimiento del nervio infra-orbital y necesarios para su desensibilización durante las manipulaciones en la piel del labio superior, fosa nasal y cara a nivel del foramen. La distancia entre el extremo lateral del alvéolo del tercer diente incisivo y el foramen mental fue de 3.57 ± 0.04 cm en Zovawk, que es un hito importante para lograr la ubicación del nervio mental para el bloqueo nervioso regional Zovawk. La longitud y la altura de la mandíbula fueron 25.02 ± 0.09 cm y 10.54 ± 0.07 cm, respectivamente, en Zovawk. Las mediciones morfométricas del cráneo y la anatomía aplicada de la región de la cabeza de Zovawk proporcionan datos de referencia importantes para futuras investigaciones en el campo de la anatomía aplicada.
