During chronic stress, there is an initial increase in glucocorticoid (GC) levels, but they then return to low, albeit not baseline, levels. Recent studies have renewed interest in cortisol in that it may also have important roles in the stress response. The purpose of our study was to test the hypothesis that chronic treatment with low levels of either corticosterone or cortisol would alter HLR and immune organ morphometrics. Further, we wanted to determine if chronic treatment with either GC would elicit an increase in cortisol levels in egg albumen. To test our hypotheses, we implanted silastic capsules that contained corticosterone, cortisol, or empty capsules as controls (N = 5/sex/treatment). Blood serum, smears, body weights, and egg quality data were collected. Ducks were then euthanized and body weight, weights of spleens, livers, and the number of active follicles were recorded. Albumen GC levels were assessed using mass spectrometry. Data were analyzed using a 2- or 3-way ANOVA as appropriate and post-hoc with Fishers PLSD. No treatment elicited differences in egg quality measures or body weight compared to controls. Corticosterone treatment did elicit an increase in serum corticosterone (p < 0.05), but not cortisol, levels compared to controls in both sexes. Both cortisol and corticosterone treatments increased (p < 0.05) serum levels of cortisol compared to controls. Relative spleen weights were higher (p < 0.05) in hens following corticosterone but not cortisol treatment. No other organs showed any differences among the treatment groups. Both GCs elicited an increase (p < 0.001) in HLR in hens at all time-points over the 2-week treatment period compared to controls. Cortisol, not corticosterone, only elicited an increase in HLR for drakes (p < 0.05) compared to controls but only at day 1 after implants. Chronic treatment with cortisol, but not corticosterone, elicited an increase (p < 0.01) in egg albumen cortisol levels compared to other groups. Corticosterone was not detected in any albumen samples. Our results suggest that glucocorticoids elicit differential effects and although corticosterone has been stated to be the predominant GC in avian species, cortisol may provide critical information to further understand bird welfare.

The environmental sampling of layer housing systems is essential to identifying potential pathogens that are of concern to human health. To identify the natural occurrence of pathogens (Listeria, Campylobacter, and Salmonella) at various locations in a cage-free aviary housing system, swabs were collected when hens were 22 to 39 wks of age. Duplicate environmental swabs were taken and inoculated with a low dose (101 cfu) Salmonella Enteritidis (SE) and examined for the recovery of SE from environmental samples. Detection of Listeria (P < 0.0001) and Campylobacter (P < 0.0001) varied between the environmental sample types taken: concrete dust, drag swabs, egg belt dust, manure belt scraper swabs, and wall dust. Detection of Listeria (P < 0.0001) was the highest (70.0%) at the beginning of the study (22 wk) and decreased over time. Detection of Campylobacter (P < 0.001) was also the highest at 22 wk, however the decrease over time was more gradual. Interestingly, detection of Campylobacter (P < 0.0001) was the greatest in concrete dust samples (96.25%), which can be attributed to the presence of rodent excreta in the samples. Drag swabs and manure belt scraper swabs were the best sampling types for high detection of Listeria and Campylobacter. It should be noted that Listeria recovered was not of human health concern. No naturally occurring Salmonella was identified in this study. The recovery of the SE inoculum increased over time, reaching the greatest recovery in drag (81.25%; P < 0.0001), egg belt dust (100.00%; P < 0.0001) and wall dust swabs (100.00%; P < 0.0001) by 39 wk. This high rate of SE recovery occurred just before US mandatory SE environmental monitoring at 40 to 45 wks of age. Based on this study, the use of drag and manure belt scraper swabs are effective in detecting Listeria and Campylobacter in cage-free aviary housing. Along with good pest management, the occurrence of pathogens could be monitored and reduced in laying hen flocks.

The fast growth rate of broiler chickens is a welfare concern that has increased consumer interest in chicken from slower growing (SG) broilers. Replacing conventional (CONV) broilers with SG broilers will reduce chicken supply and SG broilers require different management practices than CONV. This study evaluated the effects of 2 stocking densities on the carcass composition and meat quality of CONV broilers that reach market weight at 42 d and SG broilers that reach market weight at 63 d. Male broilers from each strain were exclusively stocked into 16 pens at a density of either 29 kg/m2 or 37 kg/m2. Live body, carcass without giblets (WOG), and part weights were recorded and used to calculate yield. Initial and 24-hour pH, color (L*, a*, and b*), cooking loss, and Warner Bratzler shear force of the breast and thigh muscles were measured. Birds from both strains reached similar live body and carcass WOG weights. CONV had 3.4%, 13.0%, and 2.8% greater (P ≤ 0.002), carcass WOG, breast, and tenderloin yields, while SG had 2.7%, 5.0%, 7.5%, and 1.2% greater (P < 0.0001) wings, leg quarters, frame, and skin yields, respectively. CONV breast 24-h pH and cooking loss were greater (P ≤ 0.04) than SG. While SG thigh shear force was greater than CONV (P = 0.008), breast shear force was the lowest for CONV stocked at 29 kg/m2 and the greatest for SG stocked at 37 kg/m2 (P = 0.04). SG had a paler breast than CONV, while CONV had a yellower breast than SG (P < 0.0001). While SG had a redder thigh than CONV (P = 0.002), SG stocked at 29 kg/m2 had a redder breast than SG stocked at 37 kg/m2, with both CONV groups intermediate (P = 0.04). These results indicate that differences in male broiler meat quality were affected more by strain than by stocking density. Compared with CONV, SG broiler meat quality was more affected by stocking density in this study.

Carotenoids, which are pigments known to have many health benefits, such as their antioxidant properties, are being researched for their potential as a feed additive for production animals. These pigments are found in varying quantities in different breeds of corn, and their impact on the chicken microbiome requires further investigation. This 35 d laying hen (Novagen White) feeding trial involved varying the levels and composition of carotenoids by changing the corn source: white (0.9 µg total carotinoids/g total diet), yellow (5.7 µg/g), and orange (24.9 µg/g). For each of the three corn diet treatments, 6 replicate cages were randomly assigned. The cecal microbial community composition of the hens was then studied by 16S rRNA gene amplicon sequencing. The composition of the cecal bacterial community, as determined by Bray-Curtis dissimilarity, was different (P < 0.05) in chickens fed the orange corn diet, compared to chickens on the white corn diet, but there was no statistical difference between animals fed yellow corn compared to the white or orange corn groups. There was no change in the alpha diversity between any of the groups. Within Lactobacillus, which is one of the most abundant genera, 2 amplicon sequence variants (ASVs) were decreased and one ASV was increased in the orange corn group compared to both the white and yellow corn groups. While previous studies showed that orange corn did not alter the community composition in broilers, it appears that orange corn based feed may alter the community composition of laying hens.

Footpad dermatitis (FPD), damage and inflammation of the plantar surface of the foot, is of concern for poultry because FPD affects the birds' welfare and production value. Footpad dermatitis is painful and causes costly chicken paw downgrades, carcass condemnations, and reduced live weights. However, a universal preventative has not been found. The hypothesis was that diets containing orange corn, when compared with diets containing yellow or white corn, would reduce the severity of footpad dermatitis in broiler chickens on wet litter. When compared with yellow and white corn, orange corn contains higher quantities of carotenoids, antioxidant pigments, believed to play a role in skin and feather health. This experiment was a randomized block, 3 × 2 factorial design: orange, yellow, and white corn diets with birds raised on wet or dry litter (control group). Female Ross 708 broilers (n = 960) were used to create 4 replicates of each diet x litter treatment combination. Footpads were scored at day 19, 27, 35, and 42, following the Global Animal Partnership standard's 0-2 scale of visual increasing severity: 0 indicates minimal damage and 1 and 2 indicate mild to severe lesions and ulceration, dark papillae, and/or bumble foot. At 42 d of age, birds on the wet litter had greater severity of FPD, scores 1 and 2, compared with the control group (88 vs. 13% respectively; P < 0.0001). At 42 d of age, prevalence of more severe footpad scores, 1 or 2, was lowest on the orange corn diet (33%), followed by white corn (56%) and yellow corn (63%). Birds fed the orange corn diet had higher BW throughout the study (P = 0.004) and had fat pads and livers with higher yellow pigment deposition (P < 0.005). Litter moisture content altered microbiome composition but corn type did not. In conclusion, the main determinant of FPD in this study was exposure to wet litter. When compared with yellow and white corn, orange corn was associated with improved bird growth and reduced severity of footpad dermatitis, especially at later time points.

Consumer concern for broiler welfare has increased interest in chicken from slower growing (SG) broiler strains. Broilers from SG strains take longer to reach market weight, which may necessitate differences in management practices, such as stocking density. This study evaluated the effects of 2 stocking densities on production performance, body conformation, and welfare of broilers from 2 strains. Broilers from strains that reach market weight at age 42 D (CONV; N = 284) and at 63 D (SG; N = 284) were exclusively stocked into pens at a density of either 29 kg/m2 or 37 kg/m2. Birds were provided the same starter, grower, and finisher diets with diet phase changes occurring when SG bird body weight (BW) matched CONV. Live BW, body length, pelvic width, shank length, shank width, keel length, breast width, and breast depth were collected at 4 phases: Phase 1-chick placement, Phase 2-starter, Phase 3-grower, and Phase 4-finisher. At Phase 4, footpad dermatitis (FPD), hock burn (HB), and toe damage (TD) were scored. Feed conversion ratio (FCR) and mortality for each pen were recorded throughout the study. Final BW was similar (2.68 kg) for both strains and stocking densities of birds (P > 0.05). CONV bird FCR was 35% more efficient than SG (P < 0.0001). CONV birds had shorter bodies and shanks compared with SG birds at Phases 3 and 4 (P < 0.05). Slower growing birds stocked at 37 kg/m2 had the longest bodies and keel bones at Phase 4 (P < 0.01). Also at Phase 4, SG birds stocked at 29 kg/m2 had the lowest prevalence of HB (4%), yet the highest prevalence of TD (28%; P < 0.01). These results indicate differences in the effects of strain and stocking density on male broiler conformation, performance, and welfare and highlight the importance of tailoring management practices to the strain of broiler raised.

Laying hen skeletal health continues to be an industry priority. Bone ash and bone Ca quantification in laying hen long bones provides valuable information on skeletal health. Unfortunately, these measurements can only be accomplished by sacrificing hens, thus making longitudinal measurements on the same hen impossible. Quantitative computed tomography (QCT), used with a calcium hydroxyapatite phantom, has been used to determine bone density of wings and legs as well as live hens throughout the production cycle by scanning with a calcium hydroxyapatite phantom. QCT has also been used to scan live hens throughout the production cycle. The purpose of this study was to determine how QCT calculated bone mineral content (QCT BMC) corresponds to analytical bone Ca and bone ash. Wing and leg quarters from 72-wk-old W-36 hens were QCT scanned along with a QCT Phantom. After scanning, humeri, femurs, and tibias were cleaned, divided into epiphysis (E) and diaphysis (D), fat extracted, ashed, and digested under nitric acid, and Ca was determined by atomic absorption spectroscopy was used to determine E, D, and whole bone Ca. Four bones/type were used for E and D, while 6 bones/type were used for whole bone measurements. A second set of bones were prepared to determine correlation of BMC to bone ash. QCT scans were analyzed with Mimics software (Materialise NV, Leuven, Belgium) to calculate bone volume and density in Hounsfield units. Utilizing the QCT phantom and bone volume, BMC was calculated for E, D, and whole bone. Data were analyzed with regression analysis and Pearson correlation coefficients were determined. Analytical Ca was correlated to QCT BMC for E (R = 0.84, P < 0.01), D (R = 0.99, P < 0.01), and whole bone (R = 0.97, P < 0.01). Whole bone ash was highly correlated to QCT BMC for femur (N = 47, R = 0.97, P < 0.001), tibia (N = 50, R = 0.94, P < 0.001), and keel (N 50, R = 0.94, P < 0.001). Whole bone ash and QCT BMC values of femur and tibia were not different (P = 0.39 and 0.22 respectively). Based on this information, QCT could provide relative quantitative assessment of total bone mineral in live birds proving useful in long-term studies.

Many laying hen companies in the United States are pledging to move away from intensive conventional cages to extensive housing systems. Enriched colony cages (ECC) are a practical alternative to conventional cage systems. Scientific research is limited on the effects of ECC on hen production and welfare. Therefore, the objective of this study was to evaluate the effects of stocking density on welfare and performance with the overall outcome to provide guidance on stocking density for ECC. At 16 wk, W-36 pullets were placed into 2 commercial ECC housing systems. Within each ECC enclosure, hens were allocated into 1 of 6 stocking densities: A) 465 to 484 cm2/bird, B) 581 to 606 cm2/bird, C) 652 to 677 cm2/bird, D) 754 to 780 cm2/bird, E) 799 to 832 cm2/bird, and F) 923 to 955 cm2/bird. Body weight, egg production, mortality, and Welfare Quality data were collected each 28 d period from 17 to 68 wk. The 6 ECC stocking densities had several transient effects on production measures within age periods with no difference in hen-day production (P > 0.05). Body weight was affected by stocking density (P < 0.05) where hens raised at stocking density A (465 to 484 cm2/bird) weighed at least 25 g less than hens from other stocking densities. Stocking density differences for Welfare Quality assessments were only apparent for feather coverage. Hens raised at stocking density A (465 to 484 cm2) consistently had the poorest (P < 0.05) crop, keel, belly, back, and rump feather coverage. The keel, neck, and back body regions had poorer feather coverage when hens were raised at stocking densities B (581 to 606 cm2) and C (652 to 677 cm2) compared to hens from lower stocking densities (P < 0.05). Therefore, the minimum area per hen housed in commercial ECC systems should be 754 cm2 per bird for greater feather coverage.

Multi-tiered aviaries for laying hens are designed to provide resources, such as perches, that allow birds to perform natural behaviors, thus improving their welfare. This research examined nighttime roosting heights and substrates used by laying hens of 4 genetic strains (Dekalb White: W1, Hy-Line W36: W2, Hy-Line Brown: B1, Bovans Brown: B2), in multitier aviaries (144 hens/unit, 4 units/strain) at 25 to 28 wk of age (peak lay). Influence of litter provision on roosting patterns of the strains was also tested. Direct observations of hens' nighttime roosting patterns on wire floors, ledges and perches across tiers were conducted before (PRE), immediately after (IMM), and 3 wk after (ACC) hens gained access to litter. During all periods, more W1 and W2 hens roosted on middle and upper ledges than B1 and B2 hens (all P≤0.05), while more B1 and B2 hens used perches throughout the aviary than W1 and W2 hens (all P≤0.05). W1 (15±1.9, 14±3.36) and W2 (19±2.1, 18±2.6) hens occupied perches in the upper tier in greater numbers than B1 (7±3.2, 3±4.6) and B2 (11±2.1, 5±3.36) hens during PRE (P = 0.01) and ACC (P = 0.02) periods, respectively. B1 and B2 hens occupied wire floors in larger numbers than W1 and W2 hens during PRE (P = 0.02) and IMM (P = 0.03) periods, though this difference disappeared in the ACC period. During the IMM period, more W1 and W2 roosted in the lower tier, while more B1 and B2 hens were observed in the middle and upper tiers (all P ≤ 0.05). These findings demonstrate the importance of perches for B1 and B2 hens and space to roost higher in aviary units for W1 and W2 hens during the night, and underscore the need to consider aviary design, management practices, and preferences of different hen strains to ensure good hen welfare in aviaries.

More than 90% of the commercial egg production in the United States is pledged to be in cage-free systems by 2025. Management practices like induced molting and litter area management have come under scrutiny because of the housing system change. The aim of this study was to determine the welfare and production implications of different litter substrates and also evaluate induced molting of hens in a cage-free system. Bovan White hens were housed in a multi-tier aviary system with daily access to open litter area of either Astroturf (AT), wood shavings (SH), or straw (ST) and bare concrete floor (CO) serving as control. At 68 wk of age, molt was induced in half of the hens whereas the other half continued without molting to 116 wk. Production and welfare parameters were measured periodically throughout first and second cycles. Litter substrate did not influence hen-day production and case-weight measurements. However, CO had the lowest total number of eggs produced during the first cycle (P < 0.05). Hen-day percentage was approximately 14% greater in molted hens during the second cycle with egg case weight being heavier in non-molt hens toward the end of second cycle (P < 0.05). The only welfare parameter influenced by litter substrate during the first cycle was a greater crop feather loss in AT than ST at mid-lay (P < 0.05). Keel deformations increased with age irrespective of the litter substrate with 91.5% of palpated hens having keel deformations at the end of first cycle (P < 0.05). Molting did not influence the keel palpation and footpad scores whereas frequency of moderate comb wound was greater in molt hens during molt (P < 0.05). Severe feather loss was seen in non-molt hens during the second cycle (P < 0.05). Litter substrate does not affect production and physical parameters of welfare of hens in a multi-tier aviary system. Additionally, induced molting can be successfully carried out in the multi-tier cage-free system.

Standard procedure for most conventional bone assays rely on bones being free of attached muscle or integumentary tissue. Use of an autoclave for bone cleaning is advantageous, as parts may be cleaned afterward by peeling the muscle away as opposed to tediously scrapping muscle tissue from the bone by hand. However, autoclave use for tissue removal has not been validated and published studies typically do not specify the cleaning method. One concern is that autoclave usage could cause mineral leaching out of the bone. The objective was to determine any change in bone mineral content as a result of autoclaving bone samples to remove muscle tissue. Ten pairs of frozen chicken legs were randomly selected and thawed from 72-wk-old W36 hens. Right legs were autoclaved at 121°C for 25 min in individual trays. Left legs were thawed and cleaned by hand. The tibia, meat, and exudate were collected from each leg. Cleaned bones were placed in a soxhlet to extract the fat for 30 h and ashed at 600°C for 8 h. Bone and muscle samples underwent microwave digestion in 10 mL of 70% nitric acid. Digested samples were analyzed for calcium using a flame atomic absorption spectrophotometer. Phosphorus was determined by a colorimetric assay measuring phosphate ion complexes. Statistical analysis was completed by paired t-tests. We found no significant calcium (P = 0.6319) or phosphorus (P = 0.1698) loss from bones autoclaved as compared with bones that were hand cleaned. This study provides evidence that affirms that the use of the autoclave on bones is a suitable method for tissue removal from the leg bones of adult laying hens.

In the United States, there is an increase in need for cage-free eggs in retail and food manufacturing sectors. Understanding the impact of cage-free systems and the corresponding management on egg quality is pertinent as the U.S. industry adapts existing housing and builds new cage-free housing structures. A study was conducted comparing 2 brown shell and 2 white shell hen strains housed in a cage-free aviary system. Each set of eggs were placed in cold storage and assessed at 0, 2, 4, 8, and 12 wk. Eggs were collected at 21, 31, 42, and 60 wk of hen age. A full profile of physical quality measurements was conducted on up to 18 intact eggs for each hen strain/egg storage/hen age combination. Egg weight increased approximately 10 g for brown shell and 14 g for white shell eggs as hens aged. Many of the properties monitored were significantly impacted by all 3 main effects (hen strain, egg storage, and hen age) resulting in 3-way interactions. A brown and a white shell strain had stronger shells (44 N; P < 0.0001) than the remaining brown and white shell strains (42 N and 39 N, respectively). The current study also determined volume of shell, total length, maximum width, and percent length at maximum width to more accurately indicate egg shape than shape index. One brown shell strain produced eggs with the most consistent shape characteristics over the hen ages monitored. White shell eggs from the cage-free aviary housing produced the highest whole-egg total solids between 31 to 60 wk of hen age, whereas brown shell eggs resulted in the most consistent level of whole-egg total solids (22-23.5%). The brown and white shell strains in the current study produce cage-free aviary eggs with distinctive physical quality attributes. The outcomes from this study can be utilized by the U.S. egg industry in planning management strategies and market placement of cage-free eggs.

The international trade of shell eggs has become more important in recent years in order to feed a growing worldwide population, meet food manufacturing demands, and address supply issues during disease outbreaks or product recalls. The primary barriers for the export and import of shell eggs are: whether to wash eggs and egg storage temperature. The current study was undertaken to compare egg quality factors as influenced by egg washing and storage temperature. Three lots of nest run white shell eggs were collected on consecutive d from a commercial in-line egg production facility. The treatment and storage conditions were selected to encompass the primary egg handling and storage conditions utilized throughout the world: washed; washed, oiled; and unwashed stored at 4°C; and unwashed stored at 22°C. Eggs were assessed weekly from 0 to 15 wk. Percent egg weight loss was greatest for the unwashed 22°C eggs (15.72%) and least for washed, oiled 4°C (0.33%, P < 0.0001). Less than 24 h at 22°C had a greater impact on yolk shape measurements decline than 15 wk at 4°C (P < 0.05). After 15 wk, average Haugh unit scores for all refrigerated treatments were still Grade A, and unwashed 22°C dropped from Grade AA to almost Grade B in one week. Room temperature storage of eggs rapidly declines egg quality. Egg treatment did not impact egg quality factors when stored at 4°C. Washing and oiling eggs before refrigerated storage did suppress the rate of egg weight loss.

The tiered aviary for laying hens includes a floor litter area to promote foraging and dust bathing. Data are needed on hens' use of different litter substrates and effectiveness of substrates in removing excess feather lipids to ensure a suitable litter area. Bovans White hens were housed in commercial-style aviaries with access to one of 3 litter substrates (wood shavings, straw, or plastic turf mats-AstroTurf®, n = 4 aviary pens per substrate, 144 cage-reared hens populated per pen). Litter areas were videoed across 2 d each at 4 ages: immediately following first aviary opening (25 wk), then at 28, 50, and 68 weeks. Observations of hens throughout the d included percentages of all hens in each pen on the litter area, foraging and transitioning between the tiered enclosure and litter area. Percentages of hens dust bathing were observed from 11:00 to 15:00. Breast and back feather samples from 7 birds per pen at 28, 50, and 68 wk were analyzed for lipid content. Overall, fewer hens simultaneously accessed the AstroTurf® (P < 0.0001), but flocks showed relatively balanced transitions between the tiered enclosure and the litter area throughout the d, regardless of substrate. On average, less than 5% of all hens were observed dust bathing (peaks up to 15% of hens) with no differences among litter substrates or ages (P ≥ 0.18). On average, less than 2% of hens were observed foraging (peaks up to 4% of hens) with fewer hens foraging on AstroTurf® (P < 0.0001). Feather lipid differences among litter substrates (P < 0.0001) were inconsistent across sampling periods, possibly due to different birds sampled across time. At all ages, lipid levels were higher on the back over breast feathers (P < 0.0001) for hens housed with AstroTurf®. AstroTurf® may be suitable for nest boxes, but straw and shavings are more ideal litter substrates. Further study should investigate alternative substrates or regular substrate addition to encourage more foraging and dust bathing.

This study was aimed at analyzing bone properties of Lohmann White hens in different commercial housing systems at various points throughout production. Pullets reared in conventional cages (CC) were either continued in CC or moved to enriched colony cages (EN) at 19 weeks. Pullets reared in cage-free aviaries (AV) were moved to AV hen houses. Bone samples were collected from 60 hens at each of 18 and 72 wk and 30 hens at 26 and 56 wk from each housing system. Left tibiae and humeri were broken under uniform bending to analyze mechanical properties. Cortical geometry was analyzed using digital calipers at the fracture site. Contralateral tibiae and humeri were used for measurement of ash percentage. AV pullets' humeri had 41% greater cortical areas, and tibiae had 19% greater cortical areas than the CC pullets (P < 0.05). Average humeri diameter was greater in AV pullets than in CC pullets (P < 0.05), whereas the tibiae outer dimensions were similar. Aviary pullet bones had greater stiffness (31 and 7% greater for tibiae and humeri, respectively) and second moment of inertia (43 and 13% greater for tibiae and humeri, respectively) than CC pullets (P < 0.05). The differences between bones of AV and CC hens persisted throughout the laying cycle. Moving CC pullets to EN resulted in decreased endosteal resorption in humeri, evident by a 7.5% greater cortical area in the EN hens (P < 0.05). Whole-bone breaking strength did not change with age. Stiffness increased with age, while energy to failure decreased in both the tibiae and humeri. These results indicated that tibiae and humeri of laying hens become stiffer but lose toughness and become brittle with age. Furthermore, AV and EN systems can bring positive changes in mechanical and structural properties that are more pronounced in the humerus than the tibia.

Peripheral quantitative computed tomography (QCT) has been used in poultry bone research in recent years to analyze cortical and cross-sectional geometry. For QCT to be used as a standard research tool for analysis of bones of laying hens (cortical thickness <2 mm), the accuracy of the scans must be assessed. The primary difference between the QCT and micro-computed tomography (micro-CT) is image resolution. Image resolution is inversely related to the pixel size. The aim of the current study was to correlate the cortical parameters measured using clinical CT scans with the measurements from micro-CT, the current gold standard. A total of 15 tibiae and 14 humeri of Lohmann White hens was scanned using clinical CT and micro-CT. Reconstruction of the scans generated images with final voxel resolution of 195 µm for clinical CT scans and 46 µm for micro-CT scans. Cortical and total area were measured using MIMICS® software at proximal, middle, and distal locations of 20 mm sections of humerus diaphysis and 30 mm sections of tibia diaphysis. The total area for proximal and middle locations as well as proximal cortical area measurements for humeri produced strong correlation coefficients (R ≥ 0.70). Moderate strength correlation coefficients (R = 0.40 to 0.60) in humeri were seen in middle and distal cortical areas. Distal total area in humeri displayed a weak correlation coefficient (R ≤ 0.3; P = 0.25). Overall, tibiae demonstrated a weaker correlation. Proximal and middle cortical areas indicated moderate correlation coefficients (R = 0.40 to 0.60), while proximal and middle total areas accompanied by distal cortical and total area displayed weak correlation coefficients (R ≤ 0.3). Only the middle cortical area measurement for tibiae was significant (P = 0.03). These results indicate stronger correlation for humeri measurements among the scans than tibia. Overall, cross-sectional area measurements were only low to moderately correlated between clinical and micro-CT scans.

Many laying hen producers are transitioning from conventional cages to new housing systems including multi-tier aviaries. Aviary resources, such as litter areas, are intended to encourage hens' expression of natural behaviors to improve their welfare. Little research has examined the influence of laying hen strain on distribution and behavior inside aviaries, yet differences could influence a strain's suitability for an aviary design. This research examined how laying hens of 4 strains (Hy-Line Brown [HB], Bovans Brown [BB], DeKalb White [DW], and Hy-Line W36) distributed themselves among 3 enclosed aviary tiers and 2 litter areas at peak lay (25 to 28 wk of age) and after gaining access to litter on the floor (26 wk). Observations of hens' spatial distribution were conducted immediately before and after, and 3 wk after hens gained access to litter. More HB and BB hens were in upper tiers in morning compared to DW and W36 (all P ≤ 0.05). However, DW and W36 hens roosted in upper tiers in larger numbers than HB and BB during evening (all P ≤ 0.05). More DW and W36 hens were on litter compared to BB and HB, particularly when litter was first accessible (all P ≤ 0.05). The number of hens on litter increased over time for all strains (P ≤ 0.06). White hens on litter occupied open areas in higher numbers (P ≤ 0.05), while more brown hens occupied litter under the aviary after acclimation (P ≤ 0.05). In the dark period, W36 and DW hens were present in higher numbers in upper tiers than HB and BB, while HB and BB showed higher tier-to-tier movement than DW and W36 (P ≤ 0.05). In general, more white hens roosted higher at night and explored litter sooner, while more brown hens were near or in nests in the morning and moved at night. Distinct strain differences indicate that attention should be paid to the match between configuration of the aviary design and strain of laying hen.

Susceptibility of caged layers to osteoporosis and cage layer fatigue has generated interest in newer housing systems that favor increased load-bearing activities. However, high incidences of fractures incurred during lay period have been reported in these newer systems. This study is aimed at determining the housing and strain effects on bone properties: dry weight, percentage ash content, cortical density (CBD), cortical thickness (CBT), and keel bone deformities. Tibia, femur, and keel from Hy-Line Brown (HB), Hy-Line Silver Brown (SB), and Barred Plymouth Rock (BR) hens housed in conventional cages (CC), cage-free (CF), and cage-free with range (outdoor access; R) were studied. At 78 wk, 60 hens from each strain and housing system combination were euthanized and bones were excised for analysis. Quantitative computed tomography (QCT) was used to measure CBD and CBT in each bone. Three-dimensional images of keels were generated from software using QCT scans to analyze the deformities. Tibiae CBT was greater (P < 0.01) in BR compared to other two strains. Between housing systems, CBT was greater (P < 0.05) for mid and distal tibia of R and CF compared to CC. Tibiae and femoral cortex were denser (P < 0.05) in BR compared to HB and SB. There was no effect of housing system for femur CBD, but CBD was greater (P < 0.05) for middle and distal tibia of birds housed in R compared to CC. CBD for keel bone was greater (P < 0.05) in CF and R birds compared to CC birds. The housing system did not influence the dry bone weight and ash percentage of tibiae and femur. Each housing system was associated with high prevalence (>90%) of keel deformities and the housing and genotype influenced the type of deformity. These findings indicate that range and cage-free housing may have beneficial impact on tibia and keel bone integrity compared to conventional cages but the improvement may not be sufficient to prevent fractures or deformities of keel.

The housing of laying hens is important for social, industrial, and regulatory aspects. Many studies have compared hen housing systems on the research farm, but few have fully examined commercial housing systems and management strategies. The current study compared hens housed in commercial cage-free aviary, conventional cage, and enriched colony cage systems. Environmental and eggshell pool samples were collected from selected cages/segments of the housing systems throughout the production cycle and monitored for Salmonella and Campylobacter prevalence. At 77 wk of age, 120 hens per housing system were examined for Salmonella and Campylobacter colonization in the: adrenal glands, spleen, ceca, follicles, and upper reproductive tract. All isolates detected from environmental swabs, eggshell pools, and tissues were identified for serotype. Two predominant Salmonella were detected in all samples:S.Braenderup andS.Kentucky.Campylobacter coli and C. jejuni were the only Campylobacter detected in the flocks. Across all housing systems, approximately 7% of hens were colonized with Salmonella, whereas >90% were colonized with Campylobacter Salmonella Braenderup was the isolate most frequently detected in environmental swabs (P<0.0001) and housing system impacted Salmonella spp. shedding (P<0.0001).Campylobacter jejuni was the isolate most frequently found in environmental swabs (P<0.01), while housing system impacted the prevalence of C. coli and jejuniin ceca (P<0.0001). The results of this study provide a greater understanding of the impact of hen housing systems on hen health and product safety. Additionally, producers and academia can utilize the findings to make informed decisions on hen housing and management strategies to enhance hen health and food safety.

Osteoporosis in caged hens is one driving factor for the United States egg industry to explore options regarding alternative housing systems for laying hens. The aim of our research was to study the influence of housing systems on tibiae and humeri of 77-week-old Lohmann White hens. Pullets raised in an aviary system were either continued in aviary hen systems (AV) or conventional cages (AC) whereas pullets reared in conventional cages continued in conventional hen cages (CC) or enriched colony cages (EN) at 19 weeks. From each group, 120 hens were randomly euthanized and right and left tibae and humeri were excised for structural and mechanical analysis. Volumetric density of the cortical bone was measured using quantitative computed tomography (QCT). Aviary (AV) hens had greater cortical thickness and density but similar outer dimensions to AC hens (P < 0.05). Hens in EN system had humeri with similar cortical thickness and density but wider outer dimensions than the humeri of CC hens (P < 0.05). Cortical geometry of the tibiae was the same for the EN and CC hens, whereas EN hens had denser tibial cortex than CC hens (P < 0.05). Geometrical changes in the humeri suggest that hens in the AV system were better able to protect their structure from endosteal resorption during the laying phase. Humeri of AV and EN hens had increased second moment of area compared to the AC and CC hens; however, the changes were not observed in tibiae. Mechanical property differences were observed, with bones of AV hens having greater failure moment and stiffness than AC hens and the same difference was observed between the EN and CC hens, (P < 0.05). These findings indicate that movement limitation causes loss of bone mass and density whereas provision of moderate movement increases certain bone quality parameters during adulthood in laying hens.
