RESUMO

The numerical density of secretory granules dense-core vesicles (DCV) in the cytoplasm of pinealocytes of the pineal gland was estimated by transmission electron microscopy in male white mice and Wistar rats. The 3-day biorhythm and lunaphase changes in the DCV content in the perikaryon and the processes of pinealocytes, which are manifested significantly in different seasons of the year, are established. The three-day biorhythm in adult male mice in comparison with younger male rats is not expressed uniformly in different phases of the moon. The in-phase manifestation of infradian biorhythms in different species of animals during the year with an unchanged daily photophase indicates the existence of common external synchronizers for mammals of these biorhythms that are not associated with the light/dark cycle.

A preclinical study of the immunocorrection action of the sum of active substances isolated from ethereal-oil plants Coluria geoides (Pall.) Ledeb. (Rosaceae family) with respect to experimental immunodeficiency showed that preparations relieve symptoms of immunodeficiency caused by the administration of cyclophosphan: suppressed synthesis of anti-erythrocyte antibodies (agglutinine) and proliferative processes in the spleen. Under the influence of C. geoides preparations, the absolute numbers of cariocytes and antibody forming cells in spleen significantly increased (compared to the group of animals with experimental immunodeficiency) and in some cases reached the background level. The drugs studied produced a more pronounced stimulating effect on the synthesis of specific immunoglobulins and proliferation of antibody forming cells of spleen as compared to the effect of Echinacea tincture. Preparation C-2 (extract from underground organs and grass of C. geoides obtained by percolation method with 70% ethanol) is most promising for in-depth research and the development of new effective drugs with immunocorrecting properties.

Mechanisms of interaction between ciprofloxacin-based radiopharmaceutical labeled with technetium-99m and cultured Staphylococcus aureus were studied. The substance, reagent for radiopharmaceutical, and the radiopharmaceutical demonstrated similar bactericidal activity against the test bacterial cultures, which confirmed specific antibacterial activity of technetium-99m-labeled ciprofloxacin. The technetium-99m-labeled antibiotic reversibly interacts with inactivated Staphylococcus aureus culture, which indicates the existence of an additional non-specific pathway for radiopharmaceutical accumulation in the bacterial inflammation focus. Our experiment ruled out the possibility of binding free technetium-99m at concentrations <5% (its concentration in the radiopharmaceutical) with Staphylococcus aureus.

RESUMO

The characteristics of the functional status of peripheral blood monocytes/macrophages in patients with Ixodes tick-borne acute borreliosis accompanied by opisthorchiasis invasion were studied. The study revealed a decrease in the phagocytic activity of monocytes and in the level of expression of cell receptors Fcgamma with the expression of cell receptors C3beta being normal.

An appreciable increase in the number of aberrant lymphocytes was detected in the peripheral blood of patients with febrile form of tick-borne encephalitis (TBE). This increase peaked during week 2 of the infectious process and was paralleled by a decrease in the count of natural killer cells. By the end of the acute period of neuroinfection the number of cells with structural chromosome aberrations decreased, but still surpassed the control (donor blood). Genetically unstable cells were eliminated predominantly at the expense of lymphocytes with chromatid disorders, while the count of CD16-expressing lymphocytes increased. The intensity of reparative DNA synthesis was suppressed during the entire acute period of tick-borne encephalitis.

RESUMO

We studied metabolic and functional activity of peripheral blood phagocytes from patients with febrile tick-borne encephalitis. Phagocytic activity of neutrophils determining nonspecific resistance of the organism progressively increased, while metabolic activity decreased. Secretion of nitric oxide by peripheral blood mononuclear phagocytes was suppressed, while phagocytic functions of these cells remained unchanged.

RESUMO

Mast cells are involved in the reaction of the organism to herpesvirus infection, which manifests by changes in their count, morphology, and function. The onset of infection is characterized by primary activation of mast cells, the peak of disease by suppressed function, and convalescence by secondary activation and stabilization of cell morphology and function. Viral infection running in the presence of immunosuppression caused by cyclophosphamide is characterized by deep persistent suppression of the morphology and functions of mast cells at the peak of disease.

RESUMO

A method for morphological estimation of the thymus cell loss during apoptosis proposed, which can be used as a screening test in the stage of preclinical characterization of new biologically active compounds. Efficacy of the proposed method is demonstrated by evaluating the degree of apoptosis-inducing effect of some antitumor preparations.

RESUMO

The action of Staphylococcus aureus, Escherichia coli and Herpes simplex virus on the hemopoiesis of mice with cytostatic myelosuppression was studied. The study revealed that the infection of the animals simultaneously with the action of cyclophosphamide considerably activated the processes leading to the restoration of hemopoiesis due to an increase in the mitotic activity of hemopoietic cells, the accelerated differentiation of hemopoietic precursor cells which could survive the cytostatic action and an increase in the functional activity of the hemopoiesis-producing microenvironment.

RESUMO

The influence of L. monocytogenes on the hemopoietic system was studied in mouse experiments. All elements of hemopoiesis were damaged. The most pronounced changes developed in erythropoiesis. This was testified by a decrease in erythrocyte count and hemoglobin content in peripheral blood, by the hypoplasia of the hemopoietic erythroid germ in the marrow, as well as by the decrease of yield in culture CFU-E. These changes in the hemopoietic tissue supposedly caused by the direct action of L. monocytogenes and its toxins.

RESUMO

The stability of the antigenic structure of tick-borne encephalitis (TBE) virus strain 205 in the process of passage through the brain of BALB/c mice has been studied. Its relationships with other viruses of the TBE complex have been analyzed with the use of monoclonal antibodies to virus proteins E and NS3. The stability of the protein structure of the virus has been determined by the immunofluorescence test and the enzyme immunoassay.

Comparative analysis of the characteristics of tick-borne encephalitis (TBE) strain 205 used for the production of vaccine against TBE and of its variants obtained by passages in mouse brain showed the stability of such properties as infective activity, neurovirulence, sensitivity to physical (heating to 50 C) and chemical (sodium deoxycholate treatment) factors. At the same time increased neurovirulence of variants 205/M10 and 205/M20, which undergone through 10 and 20 passages in white mouse brain, for low-sensitive Syrian hamsters was revealed. Use of a panel of 5 monoclonal antibodies to protein E and of 4 monoclonal antibodies to protein E and of 4 monoclonal antibodies to protein NS3 helped differentiate between not only strains 205 and Sofyin, but between variants of strain 205 as well.
