RESUMO

Malondialdehyde (MDA) has long been served as a crucial indicator for assessing cellular oxidative stress levels. In this study, we introduce a new approach to determine cellular MDA levels based on a methyl tert-butyl ether (MTBE) extraction, aimed at eliminating interferences from cellular components during thiobarbituric acid (TBA) derivatization of MDA. By leveraging the effective MTBE extraction, we identified that the determination of the MDA-TBA adduct formed from the MTBE extraction layer can effectively eliminate the interferences from cellular proteins and metabolites. This method demonstrated acceptable linearity and precision in cellular samples and showed significant differences in H2O2 treated cellular oxidative stress models. The MTBE extraction-based MDA-TBA approach provides a reliable, cost-effective, and feasible method to determine cellular MDA levels using batch microplate reader approach for the assessment of cellular oxidative stress.

The effects of blue-white, green-white, yellow-white, and red-white light combinations on the nutrient composition and antioxidant capacity of pseudo-stems and leaves of 'Yuanzang' green onion were investigated using light-emitting diodes (LEDs) with precise modulation of light quality, using white light as the control. The results showed that the leaf pigment, vitamin C, soluble sugar, organic acids, free amino acids, mineral elements, and antioxidant levels were significantly higher in green onion under blue-white combined light treatment, followed by white and red-white combined light, while green-white and yellow-white combined light significantly reduced fruit quality and antioxidant capacity. In conclusion, supplementation with blue LED light was the most effective light condition to improve palatability, nutritional value, and storage resistance of green onion by enhancing various nutrients in the plants, increasing antioxidant levels, and delaying plant aging.

Environmental health and food safety issues potentially caused by the dinitroaniline herbicide pendimethalin (PM) are a worldwide concern. The toxicity response of ginger and tissue accumulation effects of PM on ginger biomass were studied by utilizing PM (CK (clean water), PM1 (0.4%), PM2 (0.67%), PM3 (1.0%), and PM4 (1.67%)) in a dose-response study. It significantly reduced the biomass of ginger under PM4, which is attributed to root damage. The net photosynthetic rate of ginger under PM4 was 11.37% lower than that of CK, which is mainly caused by stomatal limitation. In addition, the ultrastructure of chloroplasts has changed. PM4 caused the accumulation of reactive oxygen species (ROS) in ginger. The activity of superoxide dismutase (SOD) and peroxidase (POD) increased accordingly, maintaining the dynamic balance of ROS content. PM had no significant effect on the expression of ginger α-tubulin genes. PM was significantly accumulated in ginger roots, but not rhizomes. Si increased the productivity of ginger under PM4, which is mainly related to the increase of root development (root application of silicon) and photosynthetic efficiency (foliar application of silicon). Si reduced the ROS content due to the increase in SOD, POD, and catalase (CAT) activity and photosynthetic efficiency.
