The ardA genes are present in a wide variety of conjugative plasmids and play an important role in overcoming the restriction barrier. To date, there is no information on the chromosomal ardA genes. It is still unclear whether they keep their antirestriction activity and why bacterial chromosomes contain these genes. In the present study, we confirmed the antirestriction function of the ardA gene from the Bifidobacterium bifidum chromosome. Transcriptome analysis in Escherichia coli showed that the range of regulated genes varies significantly for ardA from conjugative plasmid pKM101 and from the B. bifidum chromosome. Moreover, if the targets for both ardA genes match, they often show an opposite effect on regulated gene expression. The results obtained indicate two seemingly mutually exclusive conclusions. On the one hand, the pleiotropic effect of ardA genes was shown not only on restriction-modification system, but also on expression of a number of other genes. On the other hand, the range of affected genes varies significally for ardA genes from different sources, which indicates the specificity of ardA to inhibited targets. Author Summary. Conjugative plasmids, bacteriophages, as well as transposons, are capable to transfer various genes, including antibiotic resistance genes, among bacterial cells. However, many of those genes pose a threat to the bacterial cells, therefore bacterial cells have special restriction systems that limit such transfer. Antirestriction genes have previously been described as a part of conjugative plasmids, and bacteriophages and transposons. Those plasmids are able to overcome bacterial cell protection in the presence of antirestriction genes, which inhibit bacterial restriction systems. This work unveils the antirestriction mechanisms, which play an important role in the bacterial life cycle. Here, we clearly show that antirestriction genes, which are able to inhibit cell protection, exist not only in plasmids but also in the bacterial chromosomes themselves. Moreover, antirestrictases have not only an inhibitory function but also participate in the regulation of other bacterial genes. The regulatory function of plasmid antirestriction genes also helps them to overcome the bacterial cell protection against gene transfer, whereas the regulatory function of genomic antirestrictases has no such effect.

RESUMO

Probiotic supplements are currently widely used in cattle feeding practices. However, knowledge regarding the genomic landscape of cattle probiotic microorganisms is relatively scarce and is based on analogies with human probiotics. Here, we report on the draft genome sequences of two Enterococcus lactis strains, VKPM B-4989 and VKPM B-4992, which were isolated from the rumen of a healthy calf and utilized as a probiotic additive.

RESUMO

The draft genome sequence of Rhodococcus erythropolis VKPM Ac-1659, a putative oil-degrading strain, is reported. This genome sequence may provide better insights into the diversity and evolution of the genes responsible for hydrocarbon degradation in soil microorganisms.

The draft genome sequence of Caldanaerobacter sp. strain 1523vc, a thermophilic bacterium, isolated from a hot spring of Uzon Caldera, (Kamchatka, Russia) is presented. The complete genome assembly was of 2 713 207 bp with predicted completeness of 99.38%. Genome structural annotation revealed 2674 protein-coding genes, 127 pseudogenes and 77 RNA genes. Pangenome analysis of 7 currently available high quality Caldanaerobacter spp. genomes including 1523vc revealed 4673 gene clusters. Of them, 1130 clusters formed a core genome of genus Caldanaerobacter. Of the rest 3543 Caldanaerobacter pangenome genes, 385 were exclusively represented in 1523vc genome. 101 of 2801 Caldanaerobacter CDS were found to be encoding carbohydrate-active enzymes (CAZymes). The majority of CAZymes were predicted to be involved in degradation of beta-linked polysaccharides as chitin, cellulose and hemicelluloses, reflecting the metabolism of strain 1523vc, isolated on cellulose. 5 of 101 CAZyme genes were found to be unique for the strain 1523vc and belonged to GH23, GT56, GH15 and two CE9 family proteins. The draft genome of strain 1523vc was deposited at DBJ/EMBL/GenBank under the accessions JABEQB000000000, PRJNA629090 and SAMN14766777 for Genome, Bioproject and Biosample, respectively.

Data on the draft genome sequence of Bifidobacterium longumsubsp. longum strain Ac-1636 is presented in this report. This strain, isolated from the digestive tract of one-year old healthy infant, was deposited in the Russian National Collection of Industrial Microorganisms as a prospective candidate for development of probiotics and probiotic foods. The 2,321,741 bp draft genome consists of 73 scaffolds with N50 of 162,253 bp. Genome annotation revealed the presence of multiple determinants of probiotic properties of this strain. The draft genome sequence data of strainAc-1636 is available in DBJ/EMBL/GenBank under the accession nos. RZHL00000000, PRJNA511803 and SAMN10644101 for Genome, Bioproject and Biosample, respectively.

The draft genome sequence of Bacillus sp. VKPM B-3276, a mesophilic, gram-positive bacterium, isolated from dead Culex pipiens larvae is presented. This strain was deposited in the Russian National Collection of Industrial Microorganisms as a prospective candidate for development of new entomopathogenic agents. The genome of Bacillus sp. VKPM B-3276 was 6,126,346 bp in length with predicted completeness of 99.43%. Genome analysis identified 6518 protein-coding sequences and 111 RNAs genes. 13% (271) of the protein-coding genes were assigned to "Carbohydrates" subsystem category, according to RAST/SEED. Among them about 50 enzymes, responsible for chitin, peptidoglycan and related molecules decomposition, were found. The draft genome of strain VKPM B-3276 was deposited at DBJ/EMBL/GenBank under the accession nos. RZHM00000000, PRJNA511803 and SAMN10644103 for Genome, Bioproject and Biosample, respectively.

RESUMO

The introduction of acute leukemia chemotherapy programs in clinical practice improved effectiveness of treatment and patients' survival rate. However there is a high incidence of anticancer drugs toxicity leading to a decrease in therapeutic effect of treatment. Acute lymphoblastic leukemia (ALL) is a leader among oncohematological pathologies in childhood and adolescence. Its share is 20 % of all malignancies and up to 75% of all leukemias. Treatment for ALL in childhood is one of the most impressive achievements of medicine in recent years. Modern chemotherapy programs can achieve healing in 80 % of patients with ALL, and the researchers are facing new challenges to achieve a high safety profile of treatment. Chemotherapy toxicity frequency varies from 5% to 30 %. This review presents summarized literature analysis on the clinical symptoms of toxic reactions and the ways to prevent and correct developed adverse drug reactions.

RESUMO

Echocardiographic examination revealed in 184 (11%) of the 1680 persons an accessory chorda in the left cardiac ventricle (ACLV). It was found ACLV was often associated with paroxysms of cardiac of unclear etiology and ventricular fibrillation in myocardial infarction. The clinico-, phono-, electrocardiographic symptomatology and inheritability of ACLV are discussed. As defined by the authors the syndrome in ACLV includes: 1) cardialgia, 2) systolic murmur, 3) electrocardiographic syndrome of early ventricular repolarization, 4) paroxysms of cardiac fibrillation, 5) Predisposition to ventricular fibrillation in myocardial infarction.

Electrocardiographic symptoms were evaluated in 53 patients with an additional left-ventricular chorda (ALVC), detected by two-dimensional echocardiography. Signs of early ventricular repolarization (EVR) were identified in 45 (84.9%) patients. A transverse ALVC was more common in EVR-free patients (87.5%), as compared to patients with ALVC + EVR combinations (51.1%; p less than 0.05).
