A comparative study of the morpho-functional characteristics of the circulating neutrophils was carried out by the electron microscopy method on women with physiological pregnancy and pregnancy-induced hypertension or EPH gestosis. Some leukocyte reactions were observed, and their intensity depended on the clinical manifestation of the EPH gestosis. In neutrophils there were detected reformation in the nucleus in the form of redistribution of the ratio between heterochromatin and euchromatin, as well as changes in the ultrastructure of nucleolemma. Both an amount of glycogen and initial or azurophil granules in the cytoplasm decreased very much but solubilization of their enzymes in cytosol did not occur. Such degranulation was due to the features of exocytosis which determined an extraleukocytic release of the content from the primary granules. The data obtained evidence that neutrophils are functionally active in the vascular bed and are likely to take part in the humoral regulation of the whole organism.

Research of functional changes in the circulating thrombocytes at physiological pregnancy and EPH--gestosis in women was conducted with methods of an electron microscopy and microcoagulogy. Some peculiarities of the release reactions by thrombocytes which determine an extra thrombocyte release of the content from their cytoplasmatic granules have been determined.

Studied singularities of development platelet release reaction, as of functional activity platelets, at direct effect contents of cytoplasmic granular structures circulating neutrophile leucocytes (granulocytes), possessing a potential capability to selective exocytosis. The model researches were conducted in a system in vitro on isolated viable non active of platelets practically of healthy people. As possible activators the subpopulations them of cytoplasmic granules, distinguished on morphological and enzymetion to indications were applied, previously chosen by methods of an analytical centrifuging from donor neutrophile granulocytes. The development platelets of a response of a release was determined on dynamics quantitative reorganization in platelets an alpha-granules and change of activity in out of platelets to environment of the factor 4 platelets. The capability of a dominance lysosomal initiators neutrophile granulocytes in development of a response of a release of platelets was installed. The scheme of probable trigger participation circulating of neutrophile granulocytes in development syndrome disseminated intravascular coagulation.

We studied the activity of lysosomal enzymes (acid phosphatase and cathepsine D) and the status of lysosomal membranes of the liver, lung, heart and brain tissues of the male adult rats under acute hypoxia. We found that disturbances of lysosomal membranes permeability in these organs depend on the tissues hypoxia development, not yet on the genesis of hypoxic state. The lysosomal enzymes activation depends on the heaviness of the hypoxic influence.

An influence of animals immobilization with lack of insulin on the condition of systems depended on factor XII was investigated on the experiments under forty-five rabbits. It was determined that the immobilization causes deep violations in the "polysystem" of factor XII that was expressed in discordance of processes of coagulation, fibrinolysis, kininogenesis.

Development of experimental the syndrome of disseminated intravascular coagulation, DIC syndrome, in organism laboratory animals from affect preparation "EFA-2" by accompanied increase quantity of neutrophiles circulations, modifications of the lysosomal apparatus of neutrophiles the activity of the serum acid phosphatase increased, characteristic damage on hemostasis system and typical alteration in some organs. As a result limitation of number of neutrophiles was achieved of suppression, of granulocytopoiesis by means "Myelosan" pharmacy, did not increase activity in blood serum solution lysosomal enzyme of neutrophils and not development in organism DIC syndrome. Made conclusion, the neutrophils have influence of promotion generalization DIC syndrome.

By lysosomal ferments don't determine the can observer discordance of processes of coagulation, fibrinolysis, kininogenesis.

The experiments on 40 Wistar male rats with acute circulatory-hemic hypoxia induced by hemorrhage in amount of 15-20% of circulating blood volume were carried out to study the state of lysosomal apparatus of the liver and lung tissues as to changes in activity of acid phosphatase (AP), a marker enzyme of lysosomes. In the case of such a hemorrhage the common activity of AP decreased by 37% in the liver tissue and by 41% in the lung tissue, the free activity increased by 30% and 25%, and bound activity decreased by 84 and 70% in homogenates of the corresponding tissues. Under these conditions the activity of the above lysosomal enzyme in blood plasma became five times as higher as control. A conclusion is made concerning the circulatory-hemic hypoxia influence on the lysosomal membrane's structures of organ's tissues cells (as one can judge by the example of liver and lungs), which is observed in an increase of permeability of lysosome membrane and causes a sharp increase of AP enzymia in blood plasma.

Experiments on male rats have shown that the acid phosphatase activity increases in all the types of hypoxia (circulatory-hemic, hemic and hypoxic), in blood serum. An increase in the activity of this enzyme distinctly correlates with hypoxia gravity. A supposition is advanced that the blood enzyme level of lysosomal hydrolases can reflect the functional state of the lysosomal apparatus in cells of the organism.

An electron-microscopic examination of human leucocytes and thrombocytes under an effect of noninfectious stress factor (delivery) on organism shows that lysosomes of neutrophilic leucocytes move to plasmalemma, merge with it and secrete completely or partially their content to the blood plasma. Exocytosis of lysosomes results in a decrease of their amount in neutrophils. No changes in lysosomes of other leucocytes and thrombocytes are revealed. The fact that lysosomal neutrophilic enzymes enter the blood plasma is probably due to their participation in humoral regulation of organism functions.

Lysosomal enzymes of neutrophil leukocytes carried out both direct and indirect activation of prostaglandin synthesis via the kinin system, affecting the Hageman factor, which, except of kinins, activated blood coagulation, fibrinolysis and blood complement. Lysosomal enzymes participating in formation of biologically active substances may be among factors responsible for development of the absolute neutrophil leukocytosis in adaptation syndrome.
