The trial of experimental vaccine consisting of protective protein antigens of P. aeruginosa cell wall was carried out on 114 volunteers. The vaccine proved to be faintly reactogenic and induced the formation of specific humoral immunity in 98% of the volunteers who retained a high level of anti-P. aeruginosa antibodies in their blood for up to 5 months (the term of observation after the course of immunization was over.

The preparations of the blood plasma of humans and the sera of animals immunized with enterobacterial vaccine proved to have elevated titers of antibodies to deep determinants of the core of the gram-negative bacterial endotoxin molecule (to glycolipid of chemotype Re) and protected animals from infection with live cultures or serogroup A, B, or C meningococcal endotoxins. Sera from nonimmunized animals and normal donor blood plasma showed no protective activity. In experiments carried out on different models immune plasma possessed both a pronounced protective activity and a curative effect. The efficacy of protection depended not on the serogroup of the infective agent, but on the virulence of the strain used in the experiment, being statistically significant in all cases.

Vaccines against Pseudomonas aeruginosa infection: 2. Clinico-immunological investigations.

A new multicomponent Pseudomonas vaccine 'pyoimmunogen' has been developed. It was tested for safety and immunogenicity by immunization of volunteer donors and burned patients. Immune plasma obtained from volunteers was used for treatment of severe forms of P. aeruginosa infections. Pyoimmunogen was shown to be of low toxicity for apparently healthy humans and to induce specific antibody formation. In 16 out of 20 burned patients it had a beneficial effect on the course of the disease and the period of hospitalization was shortened. As a result of complex treatment involving the use of anti-P. aeruginosa plasma 51 out of 60 patients recovered.

Vaccination of seronegative and seropositive donors with a concentrated purified vaccine (CV) against tick-borne encephalitis induced antibody synthesis in a concentration sufficient for preparation of donor immunoglobulin with antibody titres of 1:320 to 1:1280 by HI test. The resulting preparation showed a high antiviral activity both in vivo and in vitro against antigenically different strains of tick-borne encephalitis virus. The immune plasma derived from CV-immunized donors had a therapeutic effect in patients with tick-borne encephalitis. The optimal schedules for immunization of donors are discussed.

A rise in activity of natural cold isolymphocytotoxins after hemostimulation.

Natural cold isolymphocytotoxins (NCILCT) were studied in sera of patients with chronic renal insufficiency who had undergone 3-200 hemodialyses, in sera of healthy subjects before and after intramuscular injections of 5-9 ml of whole autologous blood, and in sera of hemophilia patients before and after a transfusion of 130-250 ml of freshly drawn donor plasma concentrate. It was noted NCILCT activity rose from 20-50% to 50-100% dead cells after injections of autologous blood and plasma transfusions in the majority of cases. In subjects who had undergone hemodialysis from 50 to 200 time NCILCT activity was considerably higher (50-80% dead cells) compared to patients with 3-10 hemodialyses (20-30% dead cells). It was suggested that the graft protection effect of blood transfusions and hemodialysis on kidney graft may be associated with an increase in NCILCT activity.

Activity of natural cold isolymphocytotoxins (NCILCT) was studied in the serum of 16 volunteers who received whole auto blood injections. Autohemostimulation was shown to result in the increased activity of NCILCT. The evidence obtained allowed the conclusion that autohemostimulation can be used for preparation of active NCILCT test sera.

[Results of a study of the safety, reactogenicity and immunologic effectiveness of pyoimmunogen in the immunization of volunteer donors]. / Rezul'taty izucheniia bezvrednosti, reaktogennosti i immunologicheskoi éffektivnosti pioimmunogena pri immunizatsii donorov-dobrovol'tsev.

The preliminary study of the safety, reactogenicity and immunological effectiveness of polyvalent vaccine prepared from Pseudomonas aeruginosa protective antigens was carried out. In this study the vaccine was used for the immunization of volunteer donors. No postvaccinal complications were revealed. After the immunization with pyoimmunogen the titer of antibodies to P. aeruginosa increased 2-fold in comparison with their initial level.

[Production of a hyperimmune antitoxic donor plasma against Pseudomonas aeruginosa]. / Poluchenie giperimmunnoi antitoksicheskoi donorskoi plazmy protiv Pseudomonas aeruginosa.

Research work has been carried out with the aim of obtaining hyperimmune antitoxic plasma against Pseudomonas aeruginosa. Hyperimmune plasma active against P. aeruginosa toxin has been obtained from donors immunized with P. aeruginosa toxoid. The preparation is highly specific and active, which is revealed by in vivo and in vitro tests. The clinical study of the preparation indicates its high efficacy in the treatment of diseases caused by different P. aeruginosa serotypes.
