RESUMO

Anti-EGFR antibodies combined with chemotherapy doublets are a cornerstone of the upfront treatment of colorectal cancer. RAS and BRAF mutations are established negative predictive factors for such therapy. The primary tumour located in the proximal colon has recently emerged as another negative predictive factor. We have conducted a retrospective multicentre study to collect data on real-world population characteristics, practice patterns, and outcomes in patients with metastatic colorectal cancer treated in a first-line setting with either cetuximab or panitumumab in combination with either FOLFOX or FOLFIRI chemotherapy. The presented analysis focuses on the impact of the primary tumour location. 126 of 842 patients analysed (15.0%) had proximal primary. It was associated with a lower BMI at diagnosis, mucinous histology, and peritoneal metastases. It was also associated with inferior treatment outcomes in terms of response ratio: 59.4% vs. 74.22% (odds ratio [OR] 0.51, 95% CI 0.33-0.78, p = 0.010), and median depth of response: -36.7% vs. -50.0% (p = 0.038). There was only a borderline non-significant trend for inferior PFS in patients with proximal tumours. OS data was incomplete. The presented analysis confirms the negative impact of tumour sidedness on the efficacy of an upfront anti-EGFR-chemotherapy combination and provides valuable data on real-world population characteristics.

Physical activity (PA) is defined as any bodily movement produced by skeletal muscles that requires energy expenditure. Due to civilization's development, we can observe a global decline in physical activity which negatively affects the state of physical and mental health. The physical activity of children and adolescents is a counterpart to their physical fitness. There is also more frequent spending of free time in a passive way rather than actively. The aim of the study was to determine whether there are differences in the physical fitness of young people who rest passively in relation to those who rest actively. In addition, it was checked whether factors, such as age, weight, body height and BMI differentiate the level of fitness in adolescents. Study group: 25 boys and 25 girls declaring active leisure activities. Control group: 25 boys and 25 girls declaring passive leisure activities. Age of the respondents ranged from 11 to 15 years (Me = 13; SD = 1.23). The research used: the author's questionnaire and the Index of Physical Fitness of K. Zuchora. The results were statistically developed. The youth who spend their free time actively were characterised by a higher level of physical fitness than their peers who choose passive recreation. The students with a higher BMI obtained worse results than the children with a lower body mass index. In both groups, slightly better results were obtained by girls. A significant relationship between age and results has been observed in the control group-the results increased with increasing age. The level of physical fitness is higher in active forms of recreation than in passive rest. Physical fitness tends to increase with age but decreases with increasing BMI. Girls are characterised by a higher level of physical fitness than boys.

RESUMO

Gastric cancer is one of the most frequent neoplasms. Although the incidence of gastric cancer worldwide has declined, there is still high mortality. Treatment of inoperable disease is under evaluation in clinical trials. In palliative treatment chemotherapy containing cisplatin and 5-fluorouracil is the most widely used. In the past years progress in tumour biology has advanced greatly and has led to development of new molecules aimed at targets important for cancer expansion. There are several randomized trials under targeted therapies for gastric cancer patients. One of them led to approval of trastuzumab. In the current paper the authors illustrate new possibilities in systemic treatment with particular attention to targeted therapy and personalization in medicine.
