RESUMO

BACKGROUND: 7α-Hydroxysteroid dehydrogenase (7α-HSDH) plays a pivotal role in vivo in the biotransformation of secondary bile acids and has great potential in industrial biosynthesis due to its broad substrate specificity. In this study, we expressed and characterized a novel thermostable 7α-HSDH (named Sa 7α-HSDH). METHODS: The DNA sequence was derived from the black bear gut microbiome metagenomic sequencing data, and the coding sequence of Sa 7α-HSDH was chemically synthesized. The heterologous expression of the enzyme was carried out using the pGEX-6p-1 vector. Subsequently, the activity of the purified enzyme was studied by measuring the absorbance change at 340 nm. Finally, the three-dimensional structure was predicted with AlphaFold2. RESULTS: Coenzyme screening results confirmed it to be NAD(H) dependent. Substrate specificity test revealed that Sa 7α-HSDH could catalyze taurochenodeoxycholic acid (TCDCA) with catalytic efficiency (kcat/Km) 3.81 S-1 mM-1. The optimum temperature of Sa 7α-HSDH was measured to be 75°C, confirming that it belongs to thermophilic enzymes. Additionally, its thermostability was assessed using an accelerated stability test over 32 hours. The catalytic activity of Sa 7α-HSDH remained largely unchanged for the first 24 hours and retained over 90% of its functionality after 32 hours at 50°C. Sa 7α-HSDH exhibited maximal activity at pH 10. The effect of metal ions-K+, Na+, Mg2+ and Cu2+-on the enzymatic activity of Sa 7α-HSDH was investigated. Only Mg2+ was observed to enhance the enzyme's activity by 27% at a concentration of 300 mM. Neither K+ nor Na+ had a significant influence on activity. Only Cu2+ was found to reduce enzyme activity. CONCLUSION: We characterized the thermostable 7α-HSDH, which provides a promising biocatalyst for bioconversion of steroids at high reaction temperatures.

The precise control of the spatiotemporal process in a roller kiln is crucial in the production of Ni-Co-Mn layered cathode material of lithium-ion batteries. Since the product is extremely sensitive to temperature distribution, temperature field control is of great significance. In this article, an event-triggered optimal control (ETOC) method with input constraints for the temperature field is proposed, which takes up an important position in reducing the communication and computation costs. A nonquadratic cost function is adopted to describe the system performance with input constraints. First, we present the problem description of the temperature field event-triggered control, where this field is described by a partial differential equation (PDE). Then, the event-triggered condition is designed according to the information of system states and control inputs. On this basis, a framework of the event-triggered adaptive dynamic programming (ETADP) method that is based on the model reduction technology is proposed for the PDE system. A critic network is used to approach the optimal performance index by a neural network (NN) together with that an actor network is used to optimize the control strategy. Furthermore, an upper bound of the performance index and a lower bound of interexecution times, as well as the stabilities of the impulsive dynamic system and the closed-loop PDE system, are also proved. Simulation verification demonstrates the effectiveness of the proposed method.

Temperature field control is crucial for the comprehensive performance of Ni-Co-Mn layered cathode material that is the most important part of lithium-ion batteries. Starting from the aspect of a class of distributed parameter systems described by highly dissipative partial differential equations (PDEs), an event-triggered optimal control (ETOC) method based on adaptive dynamic programming (ADP) for the roller kiln temperature field is proposed. First, we formulate the event-triggered control problem of the temperature field under the general framework of PDE systems. Then, an event-triggered condition is designed based on the stability of the closed-loop PDE system, which also guarantees the upper bound of the performance index. Subsequently, ADP technology is adopted to realize the ETOC, where the critic network is employed to approximate the optimal value function. Since the studied system can be regarded as an impulsive dynamic system with flow dynamics and jump dynamics simultaneously, the stability of the impulsive dynamic system combined with the ADP-based closed-loop PDE system is proved. Finally, simulation results on the temperature field verify the effectiveness of the proposed method.

We conducted the largest multiple-iteration retelling study to date (12,840 participants and 19,086 retellings) with two different studies that test how emotional appraisals are transmitted across retellings. We use a novel Bayesian model that tracks changes across retellings. Study 1 examines the preservation of appraisals of happy and sad stories and finds that retellings preserve the story's degree of happiness and sadness even when length shrinks and aspects of story coherence and rationalisation deteriorate. Study 2 compared the transmission of appraisals of happiness and sadness with embarrassment, disgust, and risk. Appraisals of happiness, sadness, and also embarrassment showed high appraisal preservation, while disgust and risk were not well preserved. We conclude that participants in our studies encoded happy and sad stories by encapsulating the events and details into an overall emotional appraisal of the story and that this processing strategy might also apply to stories involving other emotions like embarrassment. The emotional appraisal played a key role in retelling by helping to guide the selection, invention, and ordering of the story elements. Hence, we posit that emotion appraisals can operate as anchors for remembering and retelling stories, thus playing an important role in narrative communication.

We use an interactive story design in which participants read short stories and make two consecutive plot choices about whether protagonists commit low- or high-violence actions. Our study has four main findings. 1) People who choose high violence report greater satisfaction with the story, while those switching to or staying with no violence show lower satisfaction. 2) However, when participants encounter these stories without choices, they reliably rate higher-violence stories as less satisfying than lower-violence stories. 3) Regret seems to account for the low satisfaction of those who choose or switch to low violence. 4) There is a large segment of people (up to 66%) who can be persuaded by different story contexts (genre, perspective) to choose extreme violence in interactive fiction and as a consequence of their choice feel satisfaction. We hypothesize that people who opt for high violence enjoy the story as a result of their choice. Overall, we suggest that choosing violence serves as a gateway for enjoyment by creating an aesthetic zone of control detached from morality.

When people retell stories, what guides their retelling? Most previous research on story retelling and story comprehension has focused on information accuracy as the key measure of stability in transmission. This paper suggests that there is a second, affective, dimension that provides stability for retellings, namely the audience affect of surprise. In a large-sample study with multiple iterations of retellings, we found evidence that people are quite accurate in preserving all degrees of surprisingness in serial reproduction - even when the event that produced the surprisingness in the original story is dropped or changed. Thus, we propose that the preservation of affect is an implicit goal of retelling: merely do retellers not recall highly surprising events better, but rather they register all levels of surprisingness precisely and aim to surprise their implied audience to same degree. This study used 2,389 participants. Significance Statement: Story retelling is a process whereby cultural information is transmitted horizontally across social networks and vertically down generations. For the most part, retelling research has focused on the relevance and stability of factual information, "who did what, where, when, and why"; comparatively little is known about the transmission of affective information. We suggest that affect can serve as a second axis of stability for retelling, partially independent from factual information. In serial reproduction tasks modeled after the telephone game, we find that surprisingness of stories is well preserved across retellings - even when the facts and events of the story are not. The findings are significant for the communication of information, and thereby also the stability and transformation of culture in general.

The wavefront coding extension the depth of field (DOE) of an imaging system is by insert a phase plate into the pupil plane of a spatially incoherent imaging system, the encoded image can be digitally restored to produce a final image with improved depth dependent detail. To study the multicolor imaging performance of a logarithmic wavefront coding system, the phase and chromatic aberration variation with wavelength are analyzed, and characteristics such as shape, distribution and width of two wavelengths' PSF are compared. The research results show that if the chromatic aberration is excellently corrected then the technology can be used for multicolor even broadband spectrum DOE imaging system. Otherwise, the encode image can not be effectively decoded.
