RESUMO

In the tobacco industry, impurity detection is an important prerequisite for ensuring the quality of tobacco. However, in the actual production process, the complex background environment and the variability of impurity shapes can affect the accuracy of impurity detection by tobacco robots, which leads to a decrease in product quality and an increase in health risks. To address this problem, we propose a new online detection method of tobacco impurities for tobacco robot. Firstly, a BCFormer attention mechanism module is designed to effectively mitigate the interference of irrelevant information in the image and improve the network's ability to identify regions of interest. Secondly, a Dual Feature Aggregation (DFA) module is designed and added to Neck to improve the accuracy of tobacco impurities detection by augmenting the fused feature maps with deep semantic and surface location data. Finally, to address the problem that the traditional loss function cannot accurately reflect the distance between two bounding boxes, this paper proposes an optimized loss function to more accurately assess the quality of the bounding boxes. To evaluate the effectiveness of the algorithm, this paper creates a dataset specifically designed to detect tobacco impurities. Experimental results show that the algorithm performs well in identifying tobacco impurities. Our algorithm improved the mAP value by about 3.01% compared to the traditional YOLOX method. The real-time processing efficiency of the model is as high as 41 frames per second, which makes it ideal for automated inspection of tobacco production lines and effectively solves the problem of tobacco impurity detection.

In this work, the momentum mismatching based on which the acousto-optic (AO) transfer function and diffraction efficiency was acquired, was calculated considering the properties of AO crystals in AO interactions in acousto-optic tunable filter (AOTF). Transfer functions were obtained using a 4f optical system combined with AOTF and compared with theoretical calculations. It demonstrated the influence of acoustic energy shift on the AO interaction which should be considered in the design of AOTF.

Introduction: Global warming is caused by greenhouse gases (GHGs). It has been found that the release of methane (CH4) from Arctic permafrost, soil, ocean, and sediment is closely related to microbial composition and soil factors resulting from warming over several months or years. However, it is unclear for how long continuous warming due to global warming affects the microbial composition and GHG release from soils along Arctic glacial meltwater rivers. Methods: In this study, the soil upstream of the glacial meltwater river (GR) and the estuary (GR-0) in Svalbard, with strong soil heterogeneity, was subjected to short-term field incubation at 2°C (in situ temperature), 10°C, and 20°C. The incubation was carried out under anoxic conditions and lasted for few days. Bacterial composition and CH4 production potential were determined based on high-throughput sequencing and physiochemical property measurements. Results: Our results showed no significant differences in bacterial 16S rRNA gene copy number, bacterial composition, and methanogenic potential, as measured by mcrA gene copy number and CH4 concentration, during a 7- and 13-day warming field incubation with increasing temperatures, respectively. The CH4 concentration at the GR site was higher than that at the GR-0 site, while the mcrA gene was lower at the GR site than that at the GR-0 site. Discussion: Based on the warming field incubation, our results indicate that short-term warming, which is measured in days, affects soil microbial composition and CH4 concentration less than the spatial scale, highlighting the importance of warming time in influencing CH4 release from soil. In summary, our research implied that microbial composition and CH4 emissions in soil warming do not increase in the first several days, but site specificity is more important. However, emissions will gradually increase first and then decrease as warming time increases over the long term. These results are important for understanding and exploring the GHG emission fluxes of high-latitude ecosystems under global warming.

Feline calicivirus (FCV) is one of the most important pathogens causing upper respiratory tract diseases in cats, posing a serious health threat to these animals. At present, FCV is mainly prevented through vaccination, but the protective efficacy of vaccines in China is limited. In this study, based on the differences in capsid proteins of isolates from different regions in China, as reported in our previous studies, seven representative FCV epidemic strains were selected and tested for their viral titers, virulence, immunogenicity, and extensive cross-protection. Subsequently, vaccine strains were selected to prepare inactivated vaccines. The whole-genome sequencing and analysis results showed that these seven representative FCV strains and 144 reference strains fell into five groups (A, B, C, D, and E). The strains isolated in China mainly fall into groups C and D, exhibiting regional characteristics. These Chinese isolates had a distant evolutionary relationship and low homology with the current FCV-255 vaccine strain. The screened FCV-HB7 and FCV-HB10 strains displayed desirable in vitro culture characteristics, with the highest virus proliferation titers (109.5 TCID50/mL) at 36 h post inoculation at a dose of 0.01 MOI. All five cats infected intranasally with FCV-HB7 or FCV-HB10 strains showed obvious clinical symptoms of FCV. The symptoms of cats infected with the FCV-HB7 strain were more severe than those infected with the FCV-HB10 strain. Both the single-strain inactivated immunization and combined bivalent inactivated vaccine immunization of FCV-HB7 and FCV-HB10 induced high neutralizing antibody titers in five cats immunized. Moreover, bivalent inactivated vaccine immunization protected cats from FCV-HB7 and FCV-HB10 strains. The cross-neutralizing antibody titer against seven representative FCV epidemic strains achieved by combined bivalent inactivated vaccine immunization was higher than that achieved by single-strain immunization, which was much higher than that achieved by commercial vaccine FCV-255 strain immunization. The above results suggest that the FCV-HB7 and FCV-HB10 strains screened in this study have great potential to become vaccine strains with broad-spectrum protective efficacy. However, their immune protective efficacy needs to be further verified by multiple methods before clinical application.

A multicarrier light source based on a recirculating frequency shift loop (RFSL) driven by a parity-time (PT)-symmetric optoelectronic oscillator (OEO) is proposed and experimentally demonstrated. The impact of the side-mode suppression ratio (SMSR) of the radio frequency (RF) signal on the multicarrier is studied for the first time, to our knowledge. The RFSL driven by PT-symmetric OEO significantly optimizes the phase noise and flatness of the multicarrier, facilitating the system miniaturization. In the experiment, a 10.019 GHz RF signal with a SMSR of 42 dB is generated with -98.63d B c/H z measured phase noise at 10 kHz offset frequency (actual phase noise should be lower than -122.87d B c/H z). Up to 120 subcarriers with 2.32 dB flatness are obtained successfully, covering the overall bandwidth of approximately 1.2 THz.

OBJECTIVES: Neoadjuvant immunotherapy can be used to treat early-stage non-small-cell lung cancer; however, their effects on pulmonary lymphoepithelioma-like carcinomas (LELC) remain unclear. MATERIALS AND METHODS: Thirty-nine patients with stages I-III LELC were treated with chemotherapy (Chemo) or neoadjuvant immune-checkpoint inhibitors (ICIs) with or without chemo (IO) before radical-intent surgery. Short-term outcomes included objective response rate (ORR), major pathologic response (MPR), pathologic complete response (PCR), and event-free survival. For comparison, we used IO to treat 63 patients with pulmonary squamous cell carcinomas (SQC) and 47 with adenocarcinomas (ADC). Propensity score matching was analyzed to minimize bias. RESULTS: ORRs of the LELC-IO and LELC-Chemo groups were 62.5% and 42.9%, respectively (odds ratio, 2.2, 95% confidence interval, 0.423-11.678, p = 0.346). Seven (21.9%) and zero patients in LELC-IO and LELC-Chemo groups, respectively, reached PCR. MPR was identified in five (15.6%) of the 32 patients with LELC-IO. The 1-year progression-free survival rates were 96.9% and 71.4% in IO and Chemo groups, respectively (p > 0.05). However, no difference was observed in ORR, PCR, and MPR between LELC and SQC groups (ORR, 63.2% vs. 68.4%, p > 0.05; PCR, 21.1% vs. 47.4, p > 0.05; MPR, 42.1% vs. 57.9%, p > 0.05) and LELC and ADC groups (ORR, 58.8% vs. 41.2%, p > 0.05; PCR, 17.6% vs. 23.5%, p = 0.672; MPR, 29.4% vs. 47.1%, p > 0.05). The plasma Epstein-Barr virus (EBV) DNA level in a patient was altered posttreatment. CONCLUSION: Patients with LELC could be benefit from neoadjuvant immunotherapy. Distinct histological subtypes demonstrated comparable efficacy with respect to neoadjuvant immunotherapy.

In this paper, we propose a novel, to the best of our knowledge, method to our knowledge for generating and accurately measuring Nyquist pulse sequences with an ultra-low duty cycle of only 0.037, which breaks the limitations caused by the noise and bandwidth of the optical sampling oscilloscope (OSO) by using a narrow-bandwidth real-time oscilloscope (OSC) and an electrical spectrum analyzer (ESA). By this method, it is found that the bias point drift of the dual parallel Mach-Zehnder modulator (DPMZM) is the main cause of the distortion of the waveform. In addition, we increase the repetition rate of Nyquist pulse sequences by a factor of 16 by multiplexing the unmodulated Nyquist pulse sequences.

Rodents have been extensively used as animal models in microbiome studies. However, all rodents have a habitual nature called coprophagy, a phenomenon that they self-reinoculate feces into their gastrointestinal tract. Recent studies have shown that blocking coprophagy can alter rodents' diversity of gut microbiota, metabolism, neurochemistry, and cognitive behavior. However, whether rodents' coprophagy behavior affects the levels of inflammation and depression is unclear. In order to address this problem, we first blocked coprophagy in healthy mice. It displayed an increase in the levels of depression, verified by depressive-like behaviors and mood-related indicators, and inflammation, verified by the increased levels of the pro-inflammatory cytokine, in coprophagy-blocked mice. Furthermore, we transplanted fecal microbiota from chronic restraint stress (CRS) depression model mice and lipopolysaccharide (LPS) inflammation model mice to healthy recipient mice, respectively. It showed that the disease-like phenotypes in the coprophagy-blocked group were worse than those in the coprophagy-unblocked group, including severer depressive symptoms and higher levels of pro-inflammatory cytokines (IL-1ß, IL-6, TNF-α and IFN-γ) in serum, prefrontal cortex (PFC), and hippocampus (HIP). These findings showed that blocking coprophagy in mice not only increased the levels of inflammation and depression in healthy mice but also aggravated inflammation and depression induced by fecal microbiota from disease donors. The discovery may provide a vital reference for future research involving FMT in rodents.

The design of this study was to investigate the solid-aqueous migration and chemical speciation transformation of heavy metals (HMs) in the sewage sludge during the combined process of thermal hydrolysis, anaerobic digestion and heat-drying. The results showed that most of the HMs were still accumulated in the solid phase of various sludge samples after treatment. After thermal hydrolysis, the concentrations of Cr, Cu and Cd increased slightly. All the HMs measured after anaerobic digestion were concentrated obviously. While the concentrations of all HMs decreased slightly after heat-drying. The stability of HMs in the sludge samples was enhanced after treatment. The environmental risks of various HMs were also relieved in the final dried sludge samples.

Microorganisms have successfully predominated deep-sea ecosystems, while we know little about their adaptation strategy to multiple environmental stresses therein, including high hydrostatic pressure (HHP). Here, we focused on the genus Halomonas, one of the most widely distributed halophilic bacterial genera in marine ecosystems and isolated a piezophilic strain Halomonas titanicae ANRCS81 from Antarctic deep-sea sediment. The strain grew under a broad range of temperatures (2 to 45°C), pressures (0.1 to 55 MPa), salinities (NaCl, 0.5 to 17.5%, wt/vol), and chaotropic agent (Mg2+, 0 to 0.9 M) with either oxygen or nitrate as an electron acceptor. Genome annotation revealed that strain ANRCS81 expressed potential antioxidant genes/proteins and possessed versatile energy generation pathways. Based on the transcriptomic analysis, when the strain was incubated at 40 MPa, genes related to antioxidant defenses, anaerobic respiration, and fermentation were upregulated, indicating that HHP induced intracellular oxidative stress. Under HHP, superoxide dismutase (SOD) activity increased, glucose consumption increased with less CO2 generation, and nitrate/nitrite consumption increased with more ammonium generation. The cellular response to HHP represents the common adaptation developed by Halomonas to inhabit and drive geochemical cycling in deep-sea environments. IMPORTANCE Microbial growth and metabolic responses to environmental changes are core aspects of adaptation strategies developed during evolution. In particular, high hydrostatic pressure (HHP) is the most common but least examined environmental factor driving microbial adaptation in the deep sea. According to recent studies, microorganisms developed a common adaptation strategy to multiple stresses, including HHP, with antioxidant defenses and energy regulation as key components, but experimental data are lacking. Meanwhile, cellular SOD activity is elevated under HHP. The significance of this research lies in identifying the HHP adaptation strategy of a Halomonas strain at the genomic, transcriptomic, and metabolic activity levels, which will allow researchers to bridge environmental factors with the ecological function of marine microorganisms.

In this study, rice straw, soybean straw, wheat straw, and corn straw were chosen as raw materials, and biochars were prepared through the pyrolysis method at 550℃ under oxygen-limited conditions to investigate the physicochemical properties of biochars derived from the straws, the migration and transformation characteristics of heavy metals (HMs) (Cr, Ni, Cu, As, Cd, and Pb) after pyrolysis, and their leaching behaviors in different leaching solutions. The results showed that the physicochemical properties and elemental composition of the biochars were basically consistent. However, compared with that of biochars derived from other straws, biochar derived from wheat straw had a higher ash content (22.48%) and H/C radio (0.06). Meanwhile, biochar derived from corn straw had a smaller micropore volume (0.006 cm3·g-1) and a correspondingly smaller specific surface area (110.120 m2·g-1), which was consistent with the SEM image. After pyrolysis, the content of HMs (except Cd) increased by 14.04% to 410.81%, especially that of Cu and As. However, the content of Cd in soybean straw and corn straw decreased by 20.49% and 8.20% after pyrolysis, respectively, due to the low boiling point of Cd. Furthermore, most of the HMs (except Cd and Pb) tended to transform from unstable (acid-soluble/exchangeable and reducible forms) to stable forms (oxidizable and residual forms), implying that pyrolysis facilitated the stabilization of the HMs. The HMs in biochar were not leached or were leached in small amounts in ultra-pure water and buffered salt solutions, as opposed to leaching in relatively larger amounts in acetic acid solution and humic acid solution. Cr and Ni showed low leaching capacity in all leaching solutions. Cu showed relatively high leaching capacity in acetic acid solution, with the leaching amount ranging from 2.601 mg·kg-1 to 4.224 mg·kg-1, and As showed a relatively high leaching capacity in humic acid solution, with the leaching amount ranging from 0.074 mg·kg-1to 0.166 mg·kg-1. After pyrolysis, the environmental quality index (PIi) and the Nemerow pollution index (NPI) values of various HMs increased by different degrees. However, the pollution of single HMs remained at a safe level, and the integrated pollution of biochars was at the level of "clean". Due to the significant increase in potential ecological risk factors (Er) of Ni, Cd, and Pb after pyrolysis, the potential ecological risk index (RI) of biochar derived from the rice straw increased slightly. However, the potential ecological risk indexes of biochars derived from other straws significantly decreased after pyrolysis, owing to the stabilization of HMs.

Algae-derived marine oligosaccharides have been reported to be promising bioactive compounds because of their various properties with health benefits and potential significance in numerous applications in industrial biotechnology. In this study, laminaran oligosaccharides (LOs) with varying degrees of polymerization were obtained through partial acid hydrolysis of laminaran derived from Laminaria digitata. Based on response surface methodology, the optimum LOs yield was obtained for acid hydrolysis laminaran at a hydrolysis time of 55 min, temperature of 71°C, and acid concentration of 1.00 mol/L. The size-exclusion resin Bio-Gel P-2 was considered to be a better option for LOs purification. The structure of the purified oligosaccharides was analyzed through mass spectrometry and nuclear magnetic resonance. They demonstrated the main oligosaccharide structure corresponding to the connection of glucose with ß-D-Glcp-(1→3)-ß-D-Glcp, which was identified as laminaribiose (DP2), laminaritriose (DP3), laminaritetrose (DP4), and laminaripentaose (DP5). LOs demonstrate excellent antioxidant activities, as evidenced from their reactions with oxidizing free radicals, 1, 1-diphenyl-2-picryl-hydrazyl, and 2, 2'-azino-bis (3-etilbenzotiazoline-6-sulfonic acid) radicals. LOs exhibited a prebiotic effect on the growth of Bifidobacterium adolescentis and Lactobacillus plantarum. Therefore, we propose the development of LOs as natural antioxidants and prebiotics in the functional food and pharmaceutical industries.

The Gram-negative rod-shaped bacterium Serratia marcescens is an opportunistic pathogen of many organisms, including insects. We report the identification and optimal in vitro chitinase production conditions of a novel chitinolytic S. marcescens strain TC-1 isolated from a naturally infected white grub (Anomala corpulenta) collected from a peanut field at Nanyang city, Henan province, China. Strain identification was conducted by morphological, physiological, biochemical and molecular analyses. The amplified 16S rRNA gene of TC-1 showed a similarity greater than 99% with multiple strains of S. marcescens. Based on Neighbor-joining phylogenetic tree analysis of bacterial 16S rRNA gene sequences, TC-1 formed a clade with S. marcescens, clearly separated from other Serratia spp. The strain TC-1 showed larvicidal activities against five insect species (A. corpulenta, Plutella xylostella, Spodoptera exigua, Helicoverpa armigera, Bombyx mori) and the nematode Caenorhabditis elegans, but not against S. litura. The operating parameters of chitinase production by TC-1 were optimized by response surface methodology using a three-factor, three-level Box-Behnken experimental design. The effects of three independent variables i.e. colloidal chitin concentration (7-13 g l-1), incubation time (24-72 h) and incubation temperature (24-32 °C) on chitinase production by TC-1 were investigated. A regression model was proposed to correlate the independent variables for an optimal chitinase activity predicted as 20.946 U ml-1, using a combination of colloidal chitin concentration, incubation time and incubation temperature of 9.06 g l-1, 63.83 h and 28.12 °C, respectively. The latter agreed well with a mean chitinase activity of 20.761 ± 0.102 U ml-1 measured in the culture supernatants of TC-1 grown under similar conditions with a colloidal chitin concentration, incubation time and incubation temperature of 9 g l-1, 64 h and 28 °C, respectively. Our study revealed the S. marcescens strain TC-1 with potential as a biocontrol agent of insect pests and nematodes and demonstrated the proposed regression model's potential to guide chitinase production by this strain.

BACKGROUND: Response to immune checkpoint inhibitors (ICIs) is affected by multiple factors. This study aimed to explore whether sites of metastasis are associated with clinical outcomes of ICIs in advanced non-small-cell lung cancer (NSCLC) patients. METHODS: The data of NSCLC patients with high programmed death-ligand 1 expression and good performance status receiving first-line ICIs monotherapy from Guangdong Provincial People's Hospital between May 2019 and July 2020 were retrospectively analyzed. Metastatic sites included liver, bone, brain, adrenal gland, pleura, and contralateral lung. Progression-free survival (PFS) and overall survival (OS) were compared between different metastatic sites and metastatic burden by the Kaplan-Meier method. Organ-specific disease control rate (OSDCR) of different individual metastatic sites was evaluated. RESULTS: Forty NSCLC patients meeting the criteria were identified. The presence of liver metastasis was significantly associated with shorter PFS (3.1 vs . 15.5 months, P  = 0.0005) and OS (11.1 months vs . not reached, P  = 0.0016). Besides, patients with bone metastasis tend to get shorter PFS (4.2 vs . 15.5 months, P  = 0.0532) rather than OS ( P  = 0.6086). Moreover, the application of local treatment could numerically prolong PFS in patients with brain metastasis (15.5 vs . 4.3 months, P  = 0.1894). More metastatic organs involved were associated with inferior PFS ( P  = 0.0052) but not OS ( P  = 0.0791). The presence of liver metastasis or bone metastasis was associated with more metastatic organs (Phi[ϕ]: 0.516, P  = 0.001). The highest OSDCR was observed in lung (15/17), and the lowest in the liver (1/4). CONCLUSIONS: Metastases in different anatomical locations may be associated with different clinical outcomes and local tumor response to ICIs in NSCLC. ICIs monotherapy shows limited efficacy in patients with liver and bone metastasis, thus patients with this type of metastasis might require more aggressive combination strategies.

This study investigated the prevalence of antimicrobial resistant Enterococcus faecalis (E. faecalis) from ducks at slaughterhouses, analyzed antimicrobial resistance genes and virulence-associated genes of the isolates. Multilocus sequence typing (MLST) was performed to characterize their molecular characteristics. A total of 227 E. faecalis isolates (67.8%) were obtained from cecum (n = 114), cloaca (n = 50), skin (n = 59), and rinsed water (n = 4). These E. faecalis exhibited high level of resistance against tetracycline (95.6%), doxycycline (94.3%), linezolid (75.8%), erythromycin (72.2%), followed by norfloxacin (56.8%), vancomycin (38.3%), penicillin (36.1%), teicoplanin (30.8%). Lower level of resistance was found to high-level streptomycin (19.8%), imipenem (15.9%) and high-level gentamicin (5.7%). The vast majority of isolates (90.3%) were multidrug resistant (MDR). Moreover, the commonly observed resistance genes were optrA (90.7%) and ermB (90.3%), followed by aph(3')-Ⅲ (86.8%), tetM (84.6%), acc(6')-aph(2) (77.5%), blaZ (76.7%) and aac(6')-Ie-aph(2")-Ia (75.8%). The less frequently observed genes were vanC (19.8%), blaTEM (4.8%), vanM (2.6%), and vanA (0.4%). None of the strains carried aph(2")-Ic and vanB genes. Furthermore, a high prevalence of ten virulence determinants was identified, and efaA (99.1%) was predominant, followed by eep (97.4%), srtA (96.9%), asa1 (95.6%), fsrB (92.1%), sprE (89.9%), aggA (63.9%), gelE (56.4%), esp (33.9%), and cylL (15.4%). Eleven isolates (4.9%) co-carried all of the tested virulence-associated genes. MLST analysis demonstrated that, E. faecalis isolates consisted of 12 known STs and 5 new STs, among which 6 of the identified STs were associated with nosocomial infection. Our data indicated that retail ducks serve as an important source of MDR E. faecalis with high pathogenicity potential, and suggested that transmission to humans could not be excluded.

Topsoil (0-20 cm) samples (n=101) in 5 different land use types in Shenyang North New Area (SNNA), Shenyang, China were collected using the uniform grid layout method to investigate the spatial distribution characteristics, composition spectrum, and source analysis of 16 polycyclic aromatic hydrocarbons (PAHs) listed as priority pollutants by the Environmental Protection Agency of the United States. Results showed that the total concentration of the 16 PAHs (ΣPAHs) in soils of SNNA ranged from 123.7 µg·kg-1 to 932.5 µg·kg-1. The PAH components were mainly dominated by 3-ring and 4-ring PAHs, of which the proportion of 3-ring PAHs was the highest. The spatial distribution of the ΣPAHs concentration was obvious, showing a decreasing tendency from south to north and from east to west. In the five soil types, the average concentrations of the ΣPAHs were relatively higher in the urban green space and the artificial forest, followed by the vegetable land, while the total PAH concentrations in paddy fields and corn fields were relatively lower and had no obvious spatial distribution differences. Source apportionment results studied using characteristic ratio analysis and factor analysis/multivariate linear regression showed that the main sources of PAHs in the topsoil of SNNA were mixed sources. Industrial coal combustion and motor vehicle exhaust were the main PAH contributors, with a combined contribution rate of 79.6%. The oil spill and coke oven contribution rate was about 16.2%, and the biomass fuel combustion was about 4.2%.

OBJECTIVE: To evaluate the clinical efficacy and security of super crush-run tong xinluo capsule (SCTXLC) for apoplexy due to energy-deficiency and blood-stasis. METHOD: The randomised controlled double blind non-inferiority trial versus paroxetine, parallel contrast, different Kinds of Techniques and dosage, the clinical trial design was adopted, 144 patients with stroke of convalescent stage were selected by 2 group, which course of diseases was in 2 weekens to 3 months, neurological deficit scores was 8 to 30, grade of acaties of daily living scores was 2 to 5. the treatment group (n = 72) received SCTXLC 0.26 g (a capsule), 4 capsules at a time, three times a day, while that of the control group (n = 72) received common crush-run tong xinluo capsule (CCTXLC) 0.38 g (a capsule), 4 capsules at a time, three times a day, the therapeutic course for both groups was 28 d. RESULT: The synthesis total effective rates of the stroke in treatment group and control group were 91.3% and 87.3% respectively, showing no significant difference. The Lower Bound Upper Bound of Asymptotic 95% Confidence Interval of the total effective rates difference is -4.57%, over the beforehand Lower Bound of 15%, non-inferiority trial versus paroxetine was eligible. The adverse reactions occurred was 1 patient in the treatment group and 2 patients in control group in clinical trial. CONCLUSION: SCTXLC has definite effect for apoplexy due to energy-deficiency and blood-stasis, the efficacy in the treated group was equal to that in the control group, and favourable satety for usage.
