BACKGROUND: Hepatitis C virus (HCV) infection screening and diagnosis are critical to control the hepatitis C epidemic. Testing for anti-HCV antibodies (Ab) in blood samples is the first step to screen people who have been infected with the virus. OBJECTIVES: To evaluate the performance of the MAGLUMI Anti-HCV (CLIA) Test for detection of HCV antibodies. STUDY DESIGN: To assess the diagnostic specificity, serum samples from 5053 unselected donors and 205 blood specimens from hospitalized patients were collected. To evaluate the diagnostic sensitivity, 400 positive HCV Ab samples were collected and 30 seroconversion panels were tested. All samples that met the test criteria were tested with the MAGLUMI Anti-HCV (CLIA) Test according to manufacturer's instruction. Results of the MAGLUMI Anti-HCV (CLIA) Test were compared with the Abbott ARCHITECT anti-HCV reference test. RESULTS: The specificity of the MAGLUMI Anti-HCV (CLIA) Test was 99.75% and 100.00% in blood donor and hospitalized patient samples, respectively. The sensitivity of the Test in HCV Ab positive samples was 100.00%. Seroconversion sensitivity was comparable between the MAGLUMI Anti-HCV (CLIA) Test and the reference assay. CONCLUSIONS: The performance of the MAGLUMI Anti-HCV (CLIA) Test makes it suited for HCV infection diagnosis.

Objective: This paper studies the mediating and interactive effects of social capital on psychological capital and the feeling of happiness from the impact of COVID-19. Since its emergence, the COVID-19 pandemic has taken a toll on people's mental health and affected their hopes for the future. Lifestyle and economic conditions have also been affected and have subsequently impacted people's sense of confidence in life. This could increase the likelihood of many people developing mental health issues, such as anxiety or depression. Therefore, it is vital to study the influence of psychological capital and social capital on people's subjective psychology and happiness experiences. Materials and Methods: Using an ordered probit model, this paper studied the independent influence and interaction between psychological capital and social capital on people's happiness. The ordered probit model was chosen because subjective well-being (SWB) is an ordered variable. We further used structural equation modeling (SEM) to study the mediating effects of social capital on psychological capital and happiness. Results: The regression results showed that both psychological capital and social capital were significantly positively correlated with happiness when controlling for other factors. In addition, psychological and social capital significantly interacted, in which the psychological capital promotes the effect of social capital on happiness. Moreover, the effect of psychological capital on happiness was greater than that of social capital, demonstrating that happiness is more greatly influenced by subjective psychological experience. The interaction coefficient of psychological and social capital was also significant, showing that the two have mutually reinforcing effects on happiness. Finally, health, income class, real estate, stranger trust, age, and urban household registration had significant positive effects on happiness, while the view of money, being female, education had a negative relationship with happiness. The SEM results showed that the mediating effect of psychological capital on happiness was partly transmitted through social capital: the total effect of psychological capital on happiness was highly significant (p < 0.0001), as was the total effect of social capital on happiness (p < 0.0001); however, the coefficient for psychological capital was greater than that for social capital. Through heterogeneity analysis, we found that the relationship between psychological capital, social capital, and happiness was significantly positive in each sub-sample group. There was also a significant interaction between psychological and social capital for men, women, urban and rural residents, and higher education background sample groups. However, the interaction was not significant in the sample group without higher education. In addition, the relationship between the happiness of rural residents and their educational background and gender was not significant. Conclusion: We found that psychological and social capital have significant positive relationships and effects on happiness. Psychological capital demonstrated both direct and indirect influences on happiness, and further strengthens the influence of social capital on happiness. These results support a scheme to emphasize psychological support during the COVID-19 pandemic period to enhance the mental health of citizens.

Photoreduction of CO2 into CO, CH4 or hydrocarbons is attractive, due to environmental compatibility and economic feasibility. Optimizing the reaction engineering of CO2 reduction is an effective and general strategy that should be given special consideration. In this article, the photocatalytic CO2 reduction performances are originally investigated in a low vacuum in both dilute (10%) and pure CO2. We discover that the CH4 yield increased above one hundred times as the vacuum degree increased from barometric pressure to -80 kPa in dilute CO2. It also reveals long-term stability and good cycling performance in a low vacuum. The enhanced CO2 photoreduction performance in a low vacuum comes from better accumulation of photogenerated electrons, less intense Brownian movement of gas molecules in the environment and hindrance of the active site-blocking of gas molecules in the environment. Improved photocatalytic CO2 reduction in a low vacuum is further verified by Pt-TiO2 catalysts. This research presents a general route for producing clean fuels by photocatalytic CO2 reduction in a more effective way.

Objective: This study aimed to investigate the feasibility of using indocyanine green (ICG) near-infrared (NIR) imaging during lymphadenectomy for oesophageal cancer. Methods: Eighty-seven patients with primary oesophageal cancer were enrolled in this study. All the enrolled patients received an endoscopic injection of ICG between 40 min and 23 h before surgery. Nodal dissection during surgery was performed under fluorescence imaging visualisation, with the NIR signal shown in purple. ICG+ or ICG- nodes were recorded station by station and were microscopically evaluated. Results: Endoscopic peritumoral ICG injection was successfully performed in all patients. Major post-surgery complications included wound infection, pleural effusion, dysphonia, pneumonia and anastomotic fistula. No patients experienced ICG-related adverse events. A total of 2,584 lymph nodes were removed, and the mean number of lymph nodes for each patient was 29.70 ± 9.24. Most of the removed nodes (97.83%) were ICG+, and 3.32% of the ICG+ nodes were metastatic. No metastatic nodes were ICG- or belonged to an ICG- lymph node station. The time from ICG injection to surgery did not affect the number of harvested lymph nodes. Conclusions: The use of ICG-NIR imaging during oesophageal cancer surgery can enhance the visualisation of lymph nodes during surgery. It is a feasible, safe and helpful technique for lymphadenectomy.

Surface enhanced Raman spectroscopy (SERS) is a highly sensitive analytical detection technique that provides unique chemical and structural information on target molecules. Here, simultaneous extraction and SERS detection of nicotine for the rapid and reliable identification of nicotine released from snus products were performed based on a nano-Au assembly hierarchy structure in the capillary. Based on this strategy, the time evolution of the concentrations of nicotine released from the snus products was measured. Through comparison of the intensities of the spectral peaks of the symmetrical breathing of the pyridine moiety of nicotine molecules, with the prolongation of time, the concentration of nicotine released decreased significantly, which is helpful for establishing a method for the rapid evaluation of the processing and selection of excipients of snus products, and provides a new idea for further study of the production of snus pouches and related tobacco products. Moreover, based on data fitting, it can be calculated that the concentration of nicotine in the extraction presented an obvious quadratic relationship with time, and the release of most of the nicotine in the snus pouch, which is held through the gums and palate, was basically completed after ∼15 min. Such destruction-free simultaneous measurements of snus products are opening up new perspectives for further research about the impact of nicotinoids on smokers' health and cessation programs.

Colitis induced by C. difficile is one of the most common and costly healthcare-related infections for humans. Probiotics are one of the most promising approaches for controlling CDI. Here, we presented the isolation, safety, and probiotic property evaluation of a novel E. thailandicus strain, d5B, with effective antimicrobial activity against C. difficile. Strain d5B showed strong bactericidal effects on at least 54C. difficile strains. Safety tests showed that strain d5B was sensitive to clinically important antibiotics, and had no haemolytic and cytotoxic activities. Whole genomic analysis showed strain d5B only contained one aminoglycoside resistance gene located in the chromosome. Moreover, d5B was devoid of functional virulence genes. Finally, strain d5B exhibited probiotic properties, such as tolerance to the gastrointestinal tract, and adhered well to HT-29 cells. In conclusion, the E. thailandicus strain d5B should be investigated further for useful properties as a novel candidate probiotic for controlling CDI.

The abnormal level of Cu2+ or GSH can cause variety of neurodegenerative diseases in humans. Thus, the selective and sensitive detection of Cu2+ and GSH has inspired intensive research efforts in biological sample analysis fields. Herein, an "on-off-on" fluorescent probe based on nitrogen and sulfur co-doped carbon dots (N,S-CDs) has been successfully prepared for the detection of Cu2+ and GSH. The "turn-off" process of fluorescence in the presence of Cu2+ ions was induced by forming a non-luminescent ground state complex due to the interaction between surface groups of the probe and Cu2+ ions. Moreover, the strong coordination between GSH and Cu2+ could destroy the structure of the complex and restore the fluorescence to "turn-on". This fluorescent probe had excellent selectivity and high sensitivity toward Cu2+ and GSH with the limits of detection (LODs) of 38 nM and 41 nM. More importantly, the as-prepared N,S-CDs served as an efficient fluorescent probe for not only detecting Cu2+ ions in lake water and tap water, and GSH in BSA solution, but also sensing Cu2+ and GSH in living cells. Therefore, these N,S-CDs could be considered as a promising fluorescence probe candidate for environmental monitoring and biological imaging.

Insights into the interaction between phages and their bacterial hosts are crucial for the development of phage therapy. However, only one study has investigated global gene expression of Clostridioides (formerly Clostridium) difficile carrying prophage, and transcriptional reprogramming during lytic infection has not been studied. Here, we presented the isolation, propagation, and characterization of a newly discovered 35,109-bp phage, JD032, and investigated the global transcriptomes of both JD032 and C. difficile ribotype 078 (RT078) strain TW11 during JD032 infection. Transcriptome sequencing (RNA-seq) revealed the progressive replacement of bacterial host mRNA with phage transcripts. The expressed genes of JD032 were clustered into early, middle, and late temporal categories that were functionally similar. Specifically, a gene (JD032_orf016) involved in the lysis-lysogeny decision was identified as an early expression gene. Only 17.7% (668/3,781) of the host genes were differentially expressed, and more genes were downregulated than upregulated. The expression of genes involved in host macromolecular synthesis (DNA/RNA/proteins) was altered by JD032 at the level of transcription. In particular, the expression of the ropA operon was downregulated. Most noteworthy is that the gene expression of some antiphage systems, including CRISPR-Cas, restriction-modification, and toxin-antitoxin systems, was suppressed by JD032 during infection. In addition, bacterial sporulation, adhesion, and virulence factor genes were significantly downregulated. This study provides the first description of the interaction between anaerobic spore-forming bacteria and phages during lytic infection and highlights new aspects of C. difficile phage-host interactions.IMPORTANCE C. difficile is one of the most clinically significant intestinal pathogens. Although phages have been shown to effectively control C. difficile infection, the host responses to phage predation have not been fully studied. In this study, we reported the isolation and characterization of a new phage, JD032, and analyzed the global transcriptomic changes in the hypervirulent RT078 C. difficile strain, TW11, during phage JD032 infection. We found that bacterial host mRNA was progressively replaced with phage transcripts, three temporal categories of JD032 gene expression, the extensive interplay between phage-bacterium, antiphage-like responses of the host and phage evasion, and decreased expression of sporulation- and virulence-related genes of the host after phage infection. These findings confirmed the complexity of interactions between C. difficile and phages and suggest that phages undergoing a lytic cycle may also cause different phenotypes in hosts, similar to prophages, which may inspire phage therapy for the control of C. difficile.

Theranostic agents that integrate diagnostic and therapeutic modalities have drawn extensive attention due to their ability to deliver real-time imaging-guided tumor treatment. Herein, a novel core-shell polydopamine (PDA)-based theranostic agent (PDA@TA-Fe) was fabricated via a two-step strategy. Upon 808 nm and 1064 nm laser irradiation, this agent exhibited high photothermal conversion efficiencies of 29% and 41%, respectively. After endocytosis into tumor cells, the TA-Fe shell of PDA@TA-Fe gradually disintegrated in the weakly acidic tumor microenvironment (TME), and released the TA as an acidity-activated reductant that could reduce Fe3+ to Fe2+. Subsequently, the generated Fe2+ reacted with H2O2 to generate toxic hydroxyl radicals (˙OH) via the Fenton reaction, which induced the apoptosis of tumor cells and achieved the chemodynamic therapy (CDT). The heat produced by photothermal therapy (PTT) accelerated the ˙OH generation to achieve a synergetic effect of CDT/PTT. In vivo tumor-xenograft imaging and therapeutic assays demonstrated obvious contrast enhancement at the tumor site in the T1/T2-weighted MR imaging and efficient tumor suppression achieved after the intravenous injection of this agent because of the enhanced permeation and retention (EPR) effect. This study offered a new strategy to design an "all-in-one" nanoplatform for T1/T2 MR imaging-guided synergistic cancer treatment of CDT/PTT.

Fe2+ is a trace metal ion required by the human body, and its abnormal metabolism can cause serious diseases. Herein, we report the development of a highly efficient "ON-OFF" fluorescent probe based on carbon dots (CDs), prepared by a simple one-step hydrothermal method. The CDs exhibited exceptional water dispersibility and stability, superior luminescence performance and low cytotoxicity. The fluorescence could be efficiently quenched by Fe2+ through an electronic transfer process. And under the optimized experimental conditions, this probe shows excellent selectivity and high sensitivity towards Fe2+ with a detection limit of 51 nmol. More interestingly, this probe could realize the visual detection of Fe2+ when Fe3+ and Cu2+ ions were efficiently shielded by tartaric acid in the presence of ascorbic acid. Furthermore, the developed fluorescent probe has been successfully applied for the detection of Fe2+ in tap water and BSA solution as well as for the biosensing of Fe2+ in living cells.

Nanotheranostic agents that can simultaneously provide real-time tracking and accurate treatment at tumor sites are playing an increasingly important role in medicine. Herein, a novel polypyrrole (PPy)-based theranostic agent containing double rare-earth elements (PPy@BSA-Gd/Dy NPs) was successfully synthesized via an integrated strategy combining biomineralization and oxidation polymerization. The obtained PPy@BSA-Gd/Dy NPs with a diameter of approximately 59.48 ± 6.12 nm exhibited excellent solubility, long-term stability, superior biocompatibility, and negligible toxicity. Importantly, due to its intrinsic paramagnetic and strong X-ray attenuation ability, this agent demonstrated brilliant imaging performance in both T1/T2-weighted magnetic resonance imaging (MRI) and X-ray computed tomography (CT) imaging in vitro and vivo. Additionally, with an excellent photothermal conversion efficiency (26.61%) upon irradiation by an 808 nm laser, this theranostic agent showed significant photothermal cytotoxicity against HeLa cells and 4T1 cells in vitro and antitumor efficacy through intravenous injection in vivo. Meanwhile, biodistribution and blood circulation were also used to explore its fate in vivo. In summary, this study highlighted the versatility and practicability of PPy@BSA-Gd/Dy NPs and also suggested that the agent may be a promising candidate for T1/T2-weighted MRI/CT tri-modal imaging guided photothermal cancer therapy.

An anticorrosive composite coating with enhanced corrosion resistance in acid, alkaline and salt solutions was fabricated by compounding micro- and nanoporous inorganic structure and organic corrosion inhibitor, which was used to improve the corrosion resistance of amphoteric metal and its oxides in various corrosive medium. Micro- and nanoporous structure was prepared by microarc oxidation (MAO) coatings on 2024 aluminium alloy, which was used both as the inorganic anticorrosion coating and the container for organic corrosion inhibitor (M16). Electrochemical impedance spectroscopy, Tafel plots and salt spray resistance were measured to research the anticorrosion performance of the MAO/M16 composite coating. Enhanced corrosion resistance was observed for the MAO/M16 coating compared to the MAO by itself. When the concentration of corrosion inhibitor M16 is at 2 w%, the best anticorrosive properties of the composite coating were obtained. Moreover, the MAO/M16 composite coating showed better corrosion-resistant performance than pure MAO coating and Al alloy substrate in the corrosion environment of 1 M HCl, 0.1 M NaOH and 3.5 w% NaCl solutions, respectively. The enhancement of corrosion resistance for MAO/M16 composite coating was achieved by a unique synergy between the microarc oxidation layer and the corrosion inhibitor. The composite coating indicates its promising applications in acid, alkaline and salt solutions environments and other harsh environments.

An effective acupuncture treatment must comprehend the influence of various factors, but studies in this aspect remain limited. This study aimed to identify relevant factors and search for the best practical method of acupuncture for patients with tinnitus. The study was a retrospective review of patients' data with a prospective design who had subjective idiopathic tinnitus and received acupuncture between May 2012 and August 2017. Patients' demographics, tinnitus characteristics, previous diseases, underlying diseases, oral habits, audiograms, acupuncture sessions, and acupoints were recorded and analyzed. A visual analog scale (VASloudness) was used for measuring the loudness of tinnitus, and the Clinical Global Impression-Improvement scale (CGI-I) was used for assessing the suffering of patients. Good treatment responses in patients were defined as the magnitude of change from the baseline VASloudness for ≥ 30% plus CGI-I ≤ 3 points. In total, 107 patients were enrolled. Most factors were not significantly associated with the treatment effectiveness of acupuncture in tinnitus patients. Only the combination of acupoints and the number of acupuncture sessions reached statistically significant differences. Further analyzing these two factors, we confirmed that the combination of periauricular and distal acupuncture and 17 to 24 acupuncture sessions contributed to a considerably better outcome. This result would serve as a reference for clinical acupuncturists to select an appropriate acupuncture strategy in the treatment of tinnitus.

A novel homogeneous illusion device with arbitrary polygonal cross section that acts as either invisible cloak or shifting medium has been proposed and designed based on coordinate transformation method. The material parameters of the device are derived and the effectiveness is verified by full-wave simulation. Results show that whether the illusion device acts as invisible cloak or shifting medium depends on a value of shifting distance which is about 2a (ais circum-radius of the outer polygon).When the shifting distance is larger than 2a, the illusion device acts as an invisible cloak, and otherwise it acts as shifting medium. The stealth effect of all kinds of illusion devices are investigated, including identical-size mapped polygonal devices or non-identical size mapped polygonal devices. The results show that the device is a novel interactive cloak and is different from pre-proposed cloaks. The shifting properties of the device are validated by two examples, including moving the target object virtually and generating illusionary image of a line source. The material parameters of the device are homogeneous, which makes it more practicable in reality. It is hoped that our works may open an avenue for designing novel invisible cloaks, and are helpful for speeding up the potential applications of the illusion devices, such as aircraft or military equipment stealth, target objects camouflage or protection.

Metamaterials are artificial multifunctional materials that acquire their material properties from their structure, rather than inheriting them directly from the materials they are composed of, and they may provide novel tools to significantly enhance the sensitivity and resolution of sensors. In this paper, we derive the dispersion relation of a cylindrical dielectric waveguide loaded on a negative permeability metamaterial (NPM) layer, and compute the resonant frequencies and electric field distribution of the corresponding Whispering-Gallery-Modes (WGMs). The theoretical resonant frequency and electric field distribution results are in good agreement with the full wave simulation results. We show that the NPM sensor based on a microring resonator possesses higher sensitivity than the traditional microring sensor since with the evanescent wave amplification and the increase of NPM layer thickness, the sensitivity will be greatly increased. This may open a door for designing sensors with specified sensitivity.

OBJECTIVE: To investigate the potential cause of an outbreak of eosinophilic meningitis in Dali City in March, 2011. METHODS: A retrospective investigation was performed among 18 consumers who had meal together. The clinical documents of inpatients were analyzed and the diagnoses were checked. The outpatients were investigated through a case survey. Pomacea snails on sale were examined for the infection status of Angiostrongylus cantonensis. RESULTS: Of 18 persons, 16 consumed Pomacea snail food and 9 were infected with an infection rate of 56.3%. The two persons who did not consumed snail food were not infected. Serum samples of seven patients were examined for antibody to Angiostrongylus cantonensis in peripheral blood and 3 were proved positive (42.9%). The snails consumed by the patients came from the market, and the surveillance data showed that the infection rate of Angiostrongylus cantonensis of Pomacea snails was 7.3%. CONCLUSION: According to the epidemiological survey, clinical manifestations, and laboratory examinations, the outbreak of eosinophilic meningitis is due to the consumption of Pomacea snails infected with Angiostrongylus cantonensis.
