RESUMO

Four-dimensional magnetic resonance imaging (4D-MRI) is becoming increasingly important in radiotherapy treatment planning for its ability to simultaneously provide 3D structural information and temporal profiles of the examined tissues in a non-ionizing manner. However, the relatively long acquisition time and the resulting motion artifacts severely limit the further application of 4D-MRI. In this paper, we propose a novel motion-aligned reconstruction method based on higher degree total variation and locally low-rank regularization (maHDTV-LLR) to recover 4D MR images from the highly undersampled Fourier coefficients. Specifically, we propose a two-stage reconstruction framework alternating between a motion alignment step and a regularized optimization reconstruction step. Moreover, we incorporate the 3D-HDTV and the locally low-rank penalties into a unified framework to simultaneously exploit the spatial and temporal correlation of the 4D-MRI data. A fast alternating minimization algorithm based on variable splitting is utilized to solve the optimization problem efficiently. The performance of the proposed method is demonstrated in the context of 4D cardiac and abdominal MR images reconstruction with high undersampling factors. Numerical results show that the proposed method enables accelerated 4D-MRI with improved image quality and reduced artifacts.

Klebsiella pneumoniae is an opportunistic pathogen, and its hypervirulent variants cause serious invasive community-acquired infections. A genomic view of K. pneumoniae NTUH-2044 for the carbohydrate phosphotransferase system (PTS) found a putative fructose PTS, namely, the Frw PTS gene cluster. The deletion mutant and the complemented mutant of frwC (KP1_1992), which encodes the putative fructose-specific enzyme IIC, were constructed, and the phenotypes were characterized. This transmembrane PTS protein is responsible for fructose utilization. frwC deletion can enhance biofilm formation and capsular polysaccharide (CPS) biosynthesis but decreases the growth rate and lethality in mice. frwC expression was repressed in the cyclic AMP receptor protein (CRP) mutant. Electrophoretic mobility shift assay showed that CRP can directly bind to the promoter of frwC These results indicated that frwC expression is controlled by CRP directly and that such regulation contributes to bacterial growth, CPS synthesis, and the virulence of the Δcrp strain. The findings help elucidate fructose metabolism and the CRP regulatory mechanism in K. pneumoniae.

OBJECTIVE: To construct the KbvR gene of LuxR family deletion mutant and complementation strains from Klebsiella pneumoniae NTUH-K2044 and analyze the effect of KbvR on bacterial growth, biofilm formation and capsular synthesis. METHODS: A KbvR gene deletion mutant strain was constructed using the suicide vector pKO3-Km, and the gene fragment including KbvR coding region, promoter area and transcription termination area were amplified and cloned into pGEM-T-easy plasmid to construct KbvR complementation strain. The growth curves of the wild-type strain, KbvR gene deletion mutant strain and complementation strain were observed to assess the effect of KbvR on bacterial growth. Crystal violet staining method was used to measure the effect of KbvR on biofilm formation; the effect of KbvR on capsular synthesis was detected using string test, centrifugal test and RT-PCR. RESULTS: The KbvR deletion mutant and complementation strains were constructed successfully. KbvR gene did not affect the growth of the bacteria, but biofilm formation and capsular synthesis were attenuated in KbvR deletion mutant strain. CONCLUSION: As a transcription factor of the LuxR family orphans of the quorum sensing system, KbvR positively regulates bacterial biofilm formation by affecting capsular synthesis.
