RESUMO

To investigate the effects of long-term fertilization on the accumulation and availability of heavy metals in reddish paddy soil and to analyze the major influencing factors, soil samples were collected after the later rice was harvested in 2018 from a long-term fertilization field experiment that began in 1984. Six treatments were selected, namely CK (control without fertilization), PK (P and K fertilizer), and NPK (N, P, and K fertilizer), and different proportions of organic fertilizer plus chemical fertilizer (M1NPK: 30%M+70%NPK; M2NPK: 50%M+50%NPK; and M3NPK: 70%M+30%NPK), soil chemical properties, total and available heavy metal contents, and the relationships between the available forms of heavy metals and soil chemical parameters and total heavy metals (THM) were analyzed. The results showed that ① long-term fertilization changed the soil chemical properties; compared with those of CK, PK significantly increased the contents of soil available phosphorus (AP) and available potassium (AK), NPK significantly increased the soil organic matter (SOM), cation exchange capacity (CEC), AP, and AK, and the organic fertilizer treatments significantly increased the contents of SOM, CEC, AP, AK, and nitrate (NO3--N). ② There were slight variations in the THM contents under the chemical fertilizer treatments (PK and NPK), whereas the organic fertilizer treatments significantly increased the total contents of Cu, Zn, and Cd. ③ The chemical fertilizer treatments significantly increased the available Cr and As, whereas the organic fertilizer treatments significantly increased the available Cu, Zn, Cr, Cd, As, and Fe. ④ There were significant positive correlations between the available Cu, Zn, Cr, Cd, As, and Fe and the SOM, CEC, AP, and NO3--N. In addition, the available Zn and Cd were significantly positively correlated with the soil pH, whereas the available Pb was significantly negatively correlated with soil pH, SOM, CEC, and NO3--N. ⑤ There were significant positive correlations between the available and total contents of Cu, Zn, and Cd, whereas there were significant negative correlations between the available and total contents of Cr and Fe. ⑥ Redundancy analysis showed that SOM and pH accounted for 80.7% and 5.5% of the variation in THM, whereas the soil CEC, AP, and pH accounted for 81.1%, 4.9%, and 3.3% of the variation in the available heavy metals, respectively. ⑦ The partial least squares path model analysis showed that the path coefficients of the THM, CEC, and AP on the available state of heavy metals were 0.459, 0.417, and 0.293, respectively. Long-term application of organic manure, such as pig manure, significantly improved the soil chemical properties and affected the availability of heavy metals, and soil CEC and AP may play key roles in regulation.

RESUMO

Rice cultivar "Weiyou916" (Oryza sativa L. ssp. Indica) were cultured with control (10 mM NO3-) and nitrate deficient solution (0 mM NO3-) for four weeks. Nitrogen (N) deficiency significantly decreased the content of N and P, dry weight (DW) of the shoots and roots, but increased the ratio of root to shoot in O. sativa. N deficiency decreased the photosynthesis rate and the maximum quantum yield of primary photochemistry (Fv/Fm), however, increased the intercellular CO2 concentration and primary fluorescence (Fo). N deficiency significantly increased the production of H2O2 and membrane lipid peroxidation revealed as increased MDA content in O. sativa leaves. N deficiency significantly increased the contents of starch, sucrose, fructose, and malate, but did not change that of glucose and total soluble protein in O. sativa leaves. The accumulated carbohydrates and H2O2 might further accelerate biosynthesis of lignin in O. sativa leaves under N limitation. A total of 1635 genes showed differential expression in response to N deficiency revealed by Illumina sequencing. Gene Ontology (GO) analysis showed that 195 DEGs were found to highly enrich in nine GO terms. Most of DEGs involved in photosynthesis, biosynthesis of ethylene and gibberellins were downregulated, whereas most of DEGs involved in cellular transport, lignin biosynthesis and flavonoid metabolism were upregulated by N deficiency in O. sativa leaves. Results of real-time quantitative PCR (RT-qPCR) further verified the RNA-Seq data. For the first time, DEGs involved oxygen-evolving complex, phosphorus response and lignin biosynthesis were identified in rice leaves. Our RNA-Seq data provided a global view of transcriptomic profile of principal processes implicated in the adaptation of N deficiency in O. sativa and shed light on the candidate direction in rice breeding for green and sustainable agriculture.

RESUMO

Psoriasis is a usual immune-mediated inflammatory skin disease with undefined pathogenesis. Aromatic-turmerone (ATM) is a mainly constituent of essential oil from Curcuma longa L. It has been shown to exhibit strong anti-oxidant, anti-tumor activities and anti-inflammatory effects. In this study, we investigated the effects of ATM on imiquimod (IMQ)-induced psoriasis-like BALB/c mice and its molecular mechanisms for anti-inflammatory effect. ATM showed inhibition of the transfer of CD8+ T cells in epidermis, and reduced expression of NF-κB and COX-2 as well as phosphorylation of p38 MAPK. It also decreased the level of TNF-α and IL-6, and down-regulates IL-17 IL-22 and IL-23 mRNA synthesis. Notably, we demonstrated that topically applied ATM alleviated skin inflammation in IMQ-induced mice. These results indicate that ATM, a natural active compound exhibits anti-inflammatory activity and is a promising candidate molecule to treat inflammatory skin diseases, such as psoriasis.

RESUMO

Aspirin eugenol ester (AEE) is a promising drug candidate which is used for the treatment of inflammation, pain, fever, and the prevention of cardiovascular diseases. This study focuses on the effect of AEE on five proteins which are related to inflammation and thrombosis, including cyclooxygenase-1 (COX-1), cyclooxygenase-2 (COX-2), C-reactive protein (CRP), prothrombin (FII) and arachidonate 5-lipoxygenase (ALOX5). Meanwhile, the study was administrated to compare the drug effect between AEE and its precursor from the view of chemical-protein interactions. Healthy rats were given AEE, aspirin, eugenol and integration of aspirin and eugenol. Carboxyl methyl cellulose sodium (CMC-Na) was used as control. After drugs were administered intragastrically for seven days, the blood samples were collected to measure the proteins concentration by enzyme linked immuno-sorbent assay (ELISA). The results showed that the concentrations of key endogenic bioactive enzymes were significantly reduced in AEE groups when compared with CMC-Na and aspirin groups (P < 0.01). Drug effects of AEE on five proteins were stronger than aspirin and eugenol. From the view of chemical-protein interactions, AEE had positive effects on anti-inflammation and anti-thrombosis and showed stronger effects than aspirin and eugenol.

RESUMO

Mast cells and basophils are multifunctional effector cells that contain abundant secretory granules in their cytoplasm. Both cell types are involved in a variety of inflammatory and immune events, producing an array of inflammatory mediators, such as cytokines. The aim of the study was to examine whether isoquercitrin modulates allergic and inflammatory reactions in the human basophilic KU812 cells and to elucidate its influence on the phosphorylation of mitogen-activated protein kinase (MAPK) and nuclear factor (NF)-κB activation. The KU812 cells were stimulated with phorbol-12-myristate 13-acetate plus the calcium ionophore A23187 (PMACI). The inhibitory effects of isoquercitrin on the productions of histamine and pro-inflammatory cytokines in the stimulated KU812 cells were measured using cytokine-specific enzyme-linked immunosorbent (ELISA) assays. Western blotting analysis was used to assess the effects of isoquercitrin on the MAPKs and NF-κB protein levels. Our results indicated that the isoquercitrin treatment of PMACI-stimulated KU812 cells significantly reduced the production of histamine and the pro-inflammatory cytokines, such as interleukin (IL)-6, IL-8, IL-1ß, and tumor necrosis factor (TNF)-α. The treated cells exhibited decreased phosphorylation of extracellular signal-regulated kinase (ERK), revealing the role of ERK MAPK in isoquercitrin-mediated allergy inhibition. Furthermore, isoquercitrin suppressed the PMACI-mediated activation of NF-κB in the human basophil cells. In conclusion, the results from the present study provide insights into the potential therapeutic use of isoquercitrin for the treatment of inflammatory and allergic reactions.

RESUMO

Based on the prodrug principle, aspirin eugenol ester (AEE) was synthesized, which can reduce the side effects of aspirin and eugenol. As a good candidate for new antithrombotic and anti-inflammatory medicine, it is essential to evaluate its preventive effect on thrombosis. Preventive effect of AEE was investigated in κ-carrageenan-induced rat tail thrombosis model. AEE suspension liquids were prepared in 0.5% sodium carboxymethyl cellulose (CMC-Na). AEE was administrated at the dosage of 18, 36 and 72 mg/kg. Aspirin (20 mg/kg), eugenol (18 mg/kg) and 0.5% CMC-Na (30 mg/kg) were used as control drug. In order to compare the effects between AEE and its precursor, integration of aspirin and eugenol group (molar ratio 1:1) was also designed in the experiment. After drugs were administrated intragastrically for seven days, each rat was injected intraperitoneally with 20 mg/kg BW κ-carrageen dissolved in physiological saline to induce thrombosis. The length of tail-thrombosis was measured at 24 and 48 hours. The blank group just was given physiological saline for seven days without κ-carrageenan administrated. The results indicated that AEE significantly not only reduced the average length of thrombus, PT values and FIB concentration, but also reduced the red blood cell (RBC), hemoglobin (HGB), hematocrit (HCT) and platelet (PLT). The effects of AEE on platelet aggregation and anticoagulant in vitro showed that AEE could inhibit adenosine diphosphate (ADP)-induced platelet aggregation as dose-dependence but no notable effect on blood clotting. From these results, it was concluded that AEE possessed positive effect on thrombosis prevention in vivo through the reduction of FIB, PLT, inhibition of platelet aggregation and the change of TT and PT values.

RESUMO

To understand the potential of rice husk to fix Fe2+ and S2- ions, the sorption of Fe2+ and S2- by fermented rice husk was studied by using batch incubation experiments in the present study. The effects of adsorption time, Fe2+ and S2- concentration, pH, the temperature and ionic strength in adsorption reaction solution on the sorption were investigated. Therefore, the stability of Fe2+ and S2- adsorbed by fermented rice husk was further validated by desorption experiments performed under similar conditions as adsorption. The results showed that, the adsorption kinetics of Fe2+ (r = 0.912 1) and S2- (r = 0.901 1) by fermented rice husk fits the Elovich kinetics equation, and Freundlich isotherm model could simulate the isotherm adsorption processes of Fe2+ (R2 = 0.965 1) and S2- (R2 = 0.936 6) on fermented rice husk was better than other models. The adsorption processes on fermented rice husk were non- preferential adsorption for Fe2+ and S2, while the adsorption process of Fe2+ on fermented rice husk was spontaneous reaction and the adsorption process of S2- was non-spontaneous reaction. The adsorption processes of Fe2+ and S2- on fermented rice husk were endothermic process since high temperature could benefit to the adsorption. The adsorption mechanism of Fe2+ on fermented rice husk was mainly controlled by coordination adsorption, the adsorption mechanism of S2- on fermented rice husk was mainly controlled by ligand exchange adsorption. The adsorption processes of Fe2+ and S2- on fermented rice husk showed greater pH adaptability which ranged from 1.50 to 11.50. With the increasing of ionic strength, the amount of adsorbed Fe2+ on fermented rice husk wasincreased in some extent, the amount of adsorbed S2- on fermented rice husk was slightly decreased, which further proved the adsorption of Fe2+ was major in inner sphere complexation and the adsorption of S2- was major in outer complexation. The desorption rates of Fe2+ and S2- which was adsorbed by fermented rice husk were lower in different pH or ionic strength conditions, the desorption rates were all below 10 percentage which proved that the adsorption stabilities of Fe2+ and S2- on fermented rice husk were superior. The above results indicated that, the adsorption abilities to Fe2+ and S2- on fermented rice husk were better and had greater environmental adaptability. The Fe2+ and S2- adsorbed by fermented rice husk showed higher stability, and were not easy to release again.

RESUMO

The maximum absorption wavelengths (lambda(a-max)), absorption coefficient (epsilon), maximum emission wavelengths (lambda(e-max)), fluorescent quantum yields (phi(f)), and second-order nonlinear polarizations (beta(xxx)) of seventeen 4,4'-bis-(2-(substituted-styryl))biphenyl and three 1,4-bis-(2-(substituted-styryl))benzene were measured. The results showed that some of this series of compounds possess high fluorescent quantum yields in DMF, such as, 2 (0.801), 3 (0.680), 5 (0.565), 15 (0.538) 16 (0.848), 18 (2.175), 19 (1.314) and 20 (1.060), as compared with quinine-sulfuric acid. They could be used as fluorescent whiteners and fluorescent colorants. Some of these compounds were of a high beta(xxx) values, such as in DMSO, 2 (29.00/10(-30) m(5)c(-1)), 3 (25.29/10(-30) m(5)c(-1)), 8 (21.79/10(-30) m(5)c(-1)) and 9 (24.08/10(-30) m(5)c(-1)). Electron-withdrawing substituent NO(2), which is attached to the two terminal phenyl rings could cause lambda(a-max) obviously to be shorter, but it made lambda(e-max) change longer. Electron-donating substituent at two end benzene rings, such as OCH(3), N((CH(3))(2)), even Cl, could make lambda(a-max) and lambda(e-max) longer, and the larger the electron-donating ability of the substituent, the longer the lambda(a-max) and lambda(e-max). This influence of 4-position substituent on lambda(a-max) or lambda(e-max) is obviously larger than that of 2-position substituent, and the action of substituent on 2-position is larger than that of substituent on 3-position. The values of lambda(a-max) and lambda(e-max) of biphenyl compounds 2 or 3 were respectively close to these values of corresponding benzene compounds 18 or 19.
