RESUMO

Expanding the network of connected and resilient protected areas (PAs) for climate change adaptation can help species track suitable climate conditions and safeguard biodiversity. This is often overlooked when expanding PAs and quantifying their benefits, resulting in an underestimate of the benefits of expanding PAs. We expanded PAs through terrestrial mammalian species distribution hotspots, Key Biodiversity Areas (KBAs), and wilderness areas. Then, we constructed climate connectivity networks using a resistance-based approach and further quantified the network resilience to propose resilient climate response strategies in China. The results showed that existing PAs suffered from location biases with important biodiversity areas. The existing PAs represented about half of the KBAs and wilderness areas, yet only 12.08% of terrestrial mammalian species distribution hotspots were located within existing PAs. Compared with the existing PA network, the network efficiency and resilience of the expanded PAs' climate connectivity increased to 1.80 times and 1.78 times, respectively. With 56% of the nodes remaining, the network efficiency of the expanded PAs was equivalent to that of the existing PAs with all nodes. The network resilience of preferentially protecting and restoring low human footprint patches was approximately 1.5-2 times that of the random scenario. These findings highlighted that confronted with the unoptimistic situation of global warming, nature conservation based on existing PAs was no longer optimal. It was critical to construct a connected and resilient conservation network relying on both important biodiversity areas and low human footprint patches.

Replacing expensive silver with inexpensive copper for the metallization of silicon wafer solar cells can lead to significant reductions in material costs associated with cell production, but the susceptibility of the Cu material to oxidation remains a challenging issue to solve. In this study, we investigate copper metallization of Indium Tin Oxide surfaces to define copper grid electrodes for heterojunction cells. We propose a novel laser-induced selective metallization (LISM) method to fabricate large-scale copper electrodes for heterojunction solar cells at low cost. This study includes a comprehensive evaluation of the morphological characteristics and electrical properties of the electrodes. The effects of laser parameters on the morphology, composition, size, and conductivity of copper electrodes are investigated. The goal of establishing the process window is to obtain the optimal laser parameters for manufacturing highly conductive copper electrodes. These optimized parameters will then be employed to fabricate high-performance electrodes for solar cells. Furthermore, a detailed analysis of the mechanism underlying laser selective metallization is provided. The resulting Cu electrodes exhibit high conductivity and low resistivity of 1.98 × 10-5Ω.cm, demonstrating the potential of this method for efficient and cost-effective solar electrode production.

It is unclear what caused the Bangong Nujiang suture zone in the central Tibetan plateau to rise from less than 2 km in early Cenozoic to more than 4 km at present. The zircon U-Pb ages and trace elements of samples from the Niubao Formation in the Paleogene of the Nima basin were analyzed and tested. Combined with the isostasy theory, the surface uplift height of the Nima Basin during the Cenozoic period was calculated. The zircon U-Pb age results of the Niubao formation are consistent with the ages of the Lhasa terrane on the south side of the basin, the Qiangtang terrane on the north side, and the uplift in central. The zircon Eu/Eu* results show that the crust in central part of Tibetan plateau thickened by ∼20 km in Paleogene, resulting in ∼3 km surface uplift. Sediments created a total of about 1 km of surface uplift throughout the Paleogene, and the deposition rate began to slow down significantly at ∼40 Ma. Therefore, it is inferred that in the early Cenozoic, the uplift of the valley was mainly caused by sedimentation. With the continuous downward subduction of the Indian plate, at about 40 Ma, factors such as crustal shortening dominated the uplift of the central valley, and the uplift caused by deposition only accounted for a very small part. In general, the uplift of the Central Valley in the Paleogene was mainly affected by crustal shortening, but a quarter of the surface uplift was caused by the accumulation of sediments.

The dynamics of polymer nanocomposites varies depending on the physics and chemistry at the polymer-nanoparticle interface. The physical aging of the nanocomposites is accelerated or retarded based on interfacial interactions and the state of polymer adsorption at the interfaces. In this study, we investigated the aging kinetics of silica-polystyrene nanocomposites using differential scanning calorimetry, focusing on the effect of local conformations of chains adsorbed on the nanofiller surface. The results show that the temperature dependence of the aging rate follows a Vogel-Fulcher-Tammann relationship at high temperatures, whereas it exhibits an Arrhenius-like behavior below a characteristic temperature (Tc). Notably, at T < Tc, the aging rate decreases with increasing loop height of the chains adsorbed on the filler surface, but the activation energy remains unchanged. We proposed that the suppression of the aging rate at T < Tc is likely related to an increase in the length scale over which the slow interfacial dynamics can propagate due to the increased topological interactions between the chain loops of a larger size and the free chains in the matrix. The increased packing frustration occurring at the filler surface occupied by the larger loops might also contribute to the decreased aging rate.

Natural rubber (NR) is extensively utilized in numerous industries, such as aerospace, military, and transportation, because of its exceptional elasticity and all-around mechanical qualities. However, commercial NR made using various techniques typically has distinct mechanical characteristics. For instance, whole field latex rubber (SCR-WF) cured with accelerator 2-Mercaptobenzothiazole exhibits poor mechanical properties. This work attempts to enhance the mechanical property of SCR-WF via the addition of lanthanum stearate (LaSt). The influence of LaSt on strain-induced crystallization (SIC) and the mechanical properties of SCR-WF were investigated. The results of crosslinking density measured by the equilibrium swelling method demonstrate that the presence of LaSt significantly increases the crosslinking density of SCR-WF with lower loading of LaSt. The results of the mechanical properties show that the introduction of LaSt can enhance the tensile strength and fracture toughness of SCR-WF. To reveal the mechanism of LaSt improving the mechanical properties of SCR-WF, synchrotron radiation wide-angle X-ray diffraction (WAXD) experiments were used to investigate the SIC behaviors of SCR-WF. We found that the LaSt leads to higher crystallinity of SIC for the strain higher than 3.5. The tube model indicates the contribution of LaSt in both crosslinking and topological constraints. This work may provide an instruction for developing SCR-WF with superior mechanical properties.

Riboswitches are regulatory elements found in the untranslated regions (UTRs) of certain mRNA molecules. They typically comprise two distinct domains: an aptamer domain that can bind to specific small molecules, and an expression platform that controls gene expression. Riboswitches work by undergoing a conformational change upon binding to their specific ligand, thus activating or repressing the genes downstream. This mechanism allows gene expression regulation in response to metabolites or small molecules. To systematically summarise riboswitch structures and their related ligand binding functions, we present Ribocentre-switch, a comprehensive database of riboswitches, including the information as follows: sequences, structures, functions, ligand binding pockets and biological applications. It encompasses 56 riboswitches and 26 orphan riboswitches from over 430 references, with a total of 89 591 sequences. It serves as a good resource for comparing different riboswitches and facilitating the identification of potential riboswitch candidates. Therefore, it may facilitate the understanding of RNA structural conformational changes in response to ligand signaling. The database is publicly available at https://riboswitch.ribocentre.org.

Bovine enterovirus (BEV) is a highly infectious pathogen that may cause respiratory and gastrointestinal disease outbreaks in cattle. This study aimed to investigate the prevalence and genetic characteristics of BEVs in Guangxi Province, China. A total of 1,168 fecal samples from 97 different bovine farms were collected between October 2021 and July 2022 in Guangxi Province, China. BEV was confirmed using a reverse transcription-PCR (RT-PCR) method targeting the 5' untranslated region (UTR), and isolates were genotyped by sequencing their genomes. The nearly complete genome sequences of eight BEV strains showing cytopathic effects in MDBK cells were determined and analyzed. In total, 125 (10.7%) of 1,168 fecal samples were positive for BEV. BEV infection was significantly associated with farming patterns and clinical symptoms (P < 0.05; odds ratio [OR] > 1). Molecular characterization indicated that five BEV strains from this study belonged to EV-E2 and one strain to EV-E4. Two BEV strains (GXNN2204 and GXGL2215) could not be assigned to a known type. Strain GXGL2215 showed the closest genetic relationship with GX1901 (GenBank accession number MN607030; China) in its VP1 (67.5%) and P1 (74.7%) and with NGR2017 (MH719217; Nigeria) in its polyprotein (72.0%). It was also close to the EV-E4 strain GXYL2213 from this study when the complete genome (81.7%) was compared. Strain GXNN2204 showed the closest genetic relationship with Ho12 (LC150008; Japan) in the VP1 (66.5%), P1 (71.6%), and polyprotein (73.2%). Genome sequence analysis suggested that strains GXNN2204 and GXGL2215 originated from the genomic recombination of EV-E4 and EV-F3 and EV-E2 and EV-E4, respectively. This study reports the cocirculation of multiple BEV types and the identification of two novel BEV strains in Guangxi, China, and it will provide further insights into the epidemiology and evolution of BEV in China. IMPORTANCE Bovine enterovirus (BEV) is a pathogen that causes intestinal, respiratory, and reproductive disease infections in cattle. This study reports on the widespread prevalence and biological characteristics of the different BEV types which currently exist in Guangxi Province, China. It also provides a reference for the study of the prevalence of BEV in China.

Tri-training expands the training set by adding pseudo-labels to unlabeled data, which effectively improves the generalization ability of the classifier, but it is easy to mislabel unlabeled data into training noise, which damages the learning efficiency of the classifier, and the explicit decision mechanism tends to make the training noise degrade the accuracy of the classification model in the prediction stage. This study proposes the Tri-training algorithm for adaptive nearest neighbor density editing and cross-entropy evaluation (TTADEC), which is used to reduce the training noise formed during the classifier iteration and to solve the problem of inaccurate prediction by explicit decision mechanism. First, the TTADEC algorithm uses the nearest neighbor editing to label high-confidence samples. Then, combined with the relative nearest neighbor to define the local density of samples to screen the pre-training samples, and then dynamically expand the training set by adaptive technique. Finally, the decision process uses cross-entropy to evaluate the completed base classifier of training and assign appropriate weights to it to construct a decision function. The effectiveness of the TTADEC algorithm is verified on the UCI dataset, and the experimental results show that compared with the standard Tri-training algorithm and its improvement algorithm, the TTADEC algorithm has better classification performance and can effectively deal with the semi-supervised classification problem where the training set is insufficient.

This study developed a novel, ultrasensitive sandwich-type electrochemical immunosensor for detecting the porcine epidemic diarrhea virus (PEDV). By electrochemical co-deposition of graphene and Prussian blue, a Prussian blue-reduced graphene oxide-modified glassy carbon electrode was made, further modified with PEDV-monoclonal antibodies (mAbs) to create a new PEDV immunosensor using the double antibody sandwich technique. The electrochemical characteristics of several modified electrodes were investigated using cyclic voltammetry (CV). We optimized the pH levels and scan rate. Additionally, we examined specificity, reproducibility, repeatability, accuracy, and stability. The study indicates that the immunosensor has good performance in the concentration range of 1 × 101.88 to 1 × 105.38 TCID50/mL of PEDV, with a detection limit of 1 × 101.93 TCID50/mL at a signal-to-noise ratio of 3σ. The composite membranes produced via co-deposition of graphene and Prussian blue effectively increased electron transport to the glassy carbon electrode, boosted response signals, and increased the sensitivity, specificity, and stability of the immunosensor. The immunosensor could accurately detect PEDV, with results comparable to real-time quantitative PCR. This technique was applied to PEDV detection and served as a model for developing additional immunosensors for detecting hazardous chemicals and pathogenic microbes.

Constructing an ecological security pattern by matching the supply and demand of ecosystem services is of great significance for Shenzhen, China. To determine the strategic positioning of sustainable development pioneers and benchmarks for human well-being in a pilot demonstration area of socialism with Chinese characteristics, we selected water yield, carbon sequestration, and recreational services to identifiy the integrated supply and demand status of ecosystem services in Shenzhen using the InVEST model, ecological supply-demand ratio, cell phone signalling data analysis, and circuit theory. An ecological security pattern based on the matching of supply and demand of ecosystem services was constructed. We further examined the impacts of different delimitation boundaries on the area and number of ecological sources. The results showed that the comprehensive ecological supply-demand ratio in Shenzhen generally showed a distribution pattern of abundant in the east and scarce in the west. Among them, water yield services met the demand in terms of quantity and spatial distribution, but carbon sequestration services can not. Recreational services met the demand in terms of quantity, but their spatial distribution was extremely uneven. We identified 25 ecological sources covering a total area of 347.62 km2 and 34 ecological corridors with a total length of 346.06 km. The ecological source area was large in the east and small in the west, while the length of the corridors showed the opposite distribution pattern. Different delineation boundaries had different effects on the area and number of ecological sources. Primary, secondary, and tertiary ecological source areas cover 347.62, 520.84, and 557.58 km2, respectively, showing a gradual increasing trend, the numbers of ecological sources were 25, 35, and 32, showing an increasing then decreasing trend. In Shenzhen, there were important ecological patches that were not included in the ecological control line, and some patches within the ecological control line had poor ecological sustainability. Targeted protection and management policies could be formulated based on such pattern. The results could provide spatial guidance for the delimitation of ecological management and control units in Shenzhen.

PURPOSE: Mucinous histology is generally considered as a risk factor of prognosis in stage II colon cancer, but there is no appropriate model for prognostic evaluation and treatment decision in patients with stage II colon mucinous adenocarcinoma (C-MAC) Thus, it is urgent to develop a comprehensive, individualized evaluation tool to reflect the heterogeneity of stage II C-MAC. METHODS: Patients with stage II C-MAC who underwent surgical treatment in the Surveillance, Epidemiology, and End Results Program were enrolled and randomly divided into training cohort (70%) and internal validation cohort (30%). Prognostic predictors which were determined by univariate and multivariate analysis in the training cohort were included in the nomogram. The calibration curves, decision curve analysis, X-tile analysis, and Kaplan-Meier curve of the nomogram were validated in the internal validation cohort. RESULTS: Three thousand seven hundred sixty-two patients of stage II C-MAC were enrolled. The age, pathological T (pT) stage, tumor number, serum carcinoembryonic antigen (CEA), and perineural invasion (PNI) were independent predictors of overall survival (OS), which were used to establish a nomogram. Calibration curves of the nomogram indicated good consistency between nomogram prediction and actual survival for 1-, 3- and 5-year OS. Besides, patients with stage II C-MAC could be divided into high-, middle-, and low-risk subgroups by the nomogram. Further subgroup analysis indicated that patients in the high-risk group could have a survival benefit from chemotherapy after surgical treatment. CONCLUSIONS: We established the first nomogram to accurately predict the survival of stage II C-MAC patients who underwent surgical treatment. In addition, the nomogram identified low-, middle-, and high-risk subgroups of patients and found chemotherapy might improve survival in the high-risk subgroup of stage II C-MAC patients.

Background: Icariin presents protective effect in several kidney diseases. However, the role of icariin in contrast-induced acute kidney injury (CIAKI) is still unclear. This study aimed to investigate the effect of icariin in CIAKI, as well as exploring the underlying mechanism from the aspect of interaction between protein-coding genes and non-coding RNAs. Methods: The effect of icariin was evaluated in both in vivo and in vitro CIAKI models. Rat kidneys were collected for genome-wide sequencing. The differentially expressed genes (DEGs) were screened and visualized by R software. The function annotation of DEGs was analyzed by Metascape. By Cytoscape software, the competing endogenous RNA (ceRNA) network was constructed, and hub genes were selected. Expressions of hub genes were validated by PCR. Association of hub genes in the ceRNA network and renal function was also examined. Results: Icariin protected against CIAKI in both in vivo and in vitro models. Based on DEGs in icariin pretreated CIAKI rats, lncRNA- and circRNA-associated ceRNA networks were constructed, respectively. Function annotation showed the ceRNA networks were enriched in ERK1 and ERK2 cascade, MAPK signaling and NF-κB signaling. Further, two circRNAs, six lncRNAs, four miRNAs and nine mRNAs were selected as hub genes of the ceRNA network. Among them, eight mRNAs (Acot1, Cbwd1, Ly6i, Map3k14, Mettl2b, Nyap1, Set and Utp20) were negatively correlated with renal function, while one mRNA (Tmem44) was positively correlated with renal function. Conclusion: Icariin presented a protective effect against CIAKI. The ceRNA network, involving Acot1, Cbwd1, Ly6i, Map3k14, Mettl2, Nyap1, Set, Tmem44 and Utp20, might partially contribute to the underlying mechanism of icariin protection by regulation of ERK1 and ERK2 cascade, MAPK signaling and NF-κB signaling.

In recent years, hunniviruses have been reported in a variety of animal species from many countries. Here, hunnivirus was detected in fecal samples from water buffaloes and named as BufHuV-GX-2106. The samples were inoculated into cultures of MDBK cells supplemented with TPCK trypsin and the BufHuV-GX-2106 strain was stably passaged and replicated. Electron microscopic analysis showed the BufHuV-GX-2106 virus particles were spherical and 20~30 nm in diameter. The complete genome of a plaque purified sample of BufHuV-GX-2106 was determined and analyzed. Genomic analysis revealed that the whole sequence of BufHuV-GX-2106 was ~7,601 nucleotides (nt) in length and consisted of a large open reading frame of 6,759nt, a 5'UTR, a 3'UTR and a poly(A) tail. The complete genome sequence of BufHuV-GX-2106 shares 68-85% nucleotide identities with other known hunnivirus strains, indicating high genetic heterogeneity among these viruses. Phylogenetic analysis showed that BufHuV-GX-2106 belonged to the Hunnivirus A species and was more closely related to ovine hunnivirus than other known viruses of this type. This study describes the first isolation and complete genome sequence of a hunnivirus strain from water buffaloes. In addition, this study will help to understand the mechanisms involved in the pathogenesis of Hunnivirus A among different animal species.

Urban deciduous forests are an important ecological resource and seasonal landscape in the urban environment. However, in the abundant literature on how urban green space promotes human health and well-being, research on urban seasonal deciduous forests is limited. This study aimed to investigate the physiological and psychological recovery potential provided of urban deciduous forest space for youths and the spatial preferences of youths regarding such spaces. We recruited 120 participants to study the restorative potential of two typical urban deciduous forest landscape spaces (experimental groups) and one urban road environment (control group). The results showed that after 15 min of observation, the blood pressure (especially the diastolic blood pressure (p < 0.01)) and pulse of the deciduous forest trail setting (DFTS) group effectively decreased, and the restorative mood significantly increased. Regarding change in emotional parameters, the DFTS group scored higher on "interest" and significantly higher than the other two groups on positive emotion. The correlation results show that density and level are the key factors affecting spatial preferences regarding complex deciduous forests. An increase in density reduces the mood of re-laxation, and an increase in level decreases fatigue and interest. We suggest (1) constructing foot-paths in urban deciduous forests to reduce their spatial density as to improve the relaxation effect and (2) increasing landscape diversity according to the forest space to facilitate user participation and interest. This study provides a scientific basis for the environmental restoration of deciduous landscapes and for urban forestry management decision-makers based on space type construction.

Heat-stable antifungal factor (HSAF) isolated from Lysobacter enzymogenes has shown a broad-spectrum of antifungal activities. However, little is known about its mode of action. In this study, we used the model filamentous fungus Neurospora crassa to investigate the antifungal mechanism of HSAF. We first used HSAF to treat the N. crassa strain at different time points. Spore germination, growth phenotype and differential gene expression analysis were conducted by utilizing global transcriptional profiling combined with genetic and physiological analyses. Our data showed that HSAF could significantly inhibit the germination and aerial hyphae growth of N. crassa. RNA-seq analysis showed that a group of genes, associated with cell wall formation and remodeling, were highly activated. Screening of N. crassa gene deletion mutants combined with scanning electron microscopic observation revealed that three fungal cell wall integrity-related genes played an important role in the interaction between N. crassa and L. enzymogens. In addition, Weighted Gene Co-Expression Network Analysis (WGCNA), accompanied by confocal microscopy observation revealed that HSAF could trigger autophagy-mediated degradation and eventually result in cell death in N. crassa. The findings of this work provided new insights into the interactions between the predatory Lysobacter and its fungal prey.

Avian hepatitis E virus (HEV) causes liver diseases and multiple extrahepatic disorders in chickens. However, the mechanisms involved in avian HEV entry remain elusive. Herein, we identified the RAS-related protein 1b (Rap1b) as a potential HEV-ORF2 protein interacting candidate. Experimental infection of chickens and cells with an avian HEV isolate from China (CaHEV) led to upregulated expression and activation of Rap1b both in vivo and in vitro. By using CaHEV capsid as mimic of virion to treat cell in vitro, it appears that the interaction between the viral capsid and Rap1b promoted cell membrane recruitment of the downstream effector Rap1-interacting molecule (RIAM). In turn, RIAM further enhanced Talin-1 membrane recruitment and retention, which led to the activation of integrin α5/ß1, as well as integrin-associated membrane protein kinases, including focal adhesion kinase (FAK). Meanwhile, FAK activation triggered activation of downstream signaling molecules, such as Ras-related C3 botulinum toxin substrate 1 RAC1 cell division cycle 42 (CDC42), p21-activated kinase 1 (PAK1), and LIM domain kinase 1 (LIMK1). Finally, F-actin rearrangement induced by Cofilin led to the formation of lamellipodia, filopodia, and stress fibers, contributes to plasma membrane remodeling, and might enhance CaHEV virion internalization. In conclusion, our data suggested that Rap1b activation was triggered during CaHEV infection and appeared to require interaction between CaHEV-ORF2 and Rap1b, thereby further inducing membrane recruitment of Talin-1. Membrane-bound Talin-1 then activates key Integrin-FAK-Cofilin cascades involved in modulation of actin kinetics, and finally leads to F-actin rearrangement and membrane remodeling to potentially facilitate internalization of CaHEV virions into permissive cells. IMPORTANCE Rap1b is a multifunctional protein that is responsible for cell adhesion, growth, and differentiation. The inactive form of Rap1b is phosphorylated and distributed in the cytoplasm, while active Rap1b is prenylated and loaded with GTP to the cell membrane. In this study, the activation of Rap1b was induced during the early stage of avian HEV infection under the regulation of PKA and SmgGDS. Continuously activated Rap1b recruited its effector RIAM to the membrane, thereby inducing the membrane recruitment of Talin-1 that led to the activation of membrane α5/ß1 integrins. The triggering of the signaling pathway-associated Integrin α5/ß1-FAK-CDC42&RAC1-PAK1-LIMK1-Cofilin culminated in F-actin polymerization and membrane remodeling that might promote avian HEV virion internalization. These findings suggested a novel mechanism that is potentially utilized by avian HEV to invade susceptible cells.

Previous studies on the supply and demand of ecosystem services (ES) mainly focused on inter-annual changes, and no studies have explored the impact of demographic change on the ES supply and demand on fine-grained time scales. Thus, taking Shenzhen as an example, the status of ES supply and demand, as well as diurnal population changes and their impacts on cultural services were analyzed at different time periods using mobile phone signaling data, ecological supply-demand ratio (ESDR), Geo-Informatic Tupu, InVEST model and buffer zone. The results showed that the population declines successively on workdays, weekends and holidays, and that the daytime population is greater than the nighttime. Water yield services can basically meet the demand in terms of quantity and spatial distribution, however, carbon sequestration and cultural services showed the opposite results. The main type of ESDR changes in cultural services are the mutual conversion of deficit and balance, and these are concentrated in areas with high forest coverage and small populations, but frequent population changes. In addition, when the fixed population is too large, the use of time-varying population data will conceal the impact of demographic changes on ES supply and demand, and other data are needed for auxiliary analysis. Overall, this study provides a new research perspective for the ES supply and demand and can provide a theoretical basis for refined sustainable urban management.

Hepatitis E Virus (HEV) causes viral hepatitis in humans worldwide, while a subset of HEV species, avian HEV, causes hepatitis-splenomegaly syndrome in chickens. To date, there are few reports on the host proteins interacting with HEV and being involved in viral infection. Previous pull-down assay combining mass spectrometry indicated that cell division control protein 42 (CDC42), a member belonging to the Rho GTPase family, was pulled down by avian HEV capsid protein. We confirmed the direct interaction between CDC42 and avian and mammalian HEV capsid proteins. The interaction can increase the amount of active guanosine triphosphate binding CDC42 state (GTP-CDC42). Subsequently, we determined that the expression and activity of CDC42 were positively correlated with HEV infection in the host cells. Using the different inhibitors of CDC42 downstream signaling pathways, we found that CDC42-MRCK (a CDC42-binding kinase)-non-myosin IIA (NMIIA) pathway is involved in naked avian and mammalian HEV infection, CDC42-associated p21-activated kinase 1 (PAK1)-NMIIA/Cofilin pathway is involved in quasi-enveloped mammalian HEV infection and CDC42-neural Wiskott-Aldrich syndrome protein-actin-polymerizing protein Arp2/3 pathway (CDC42-(N-)WASP-Arp2/3) pathway participates in naked and quasi-enveloped mammalian HEV infection. Collectively, these results demonstrated for the first time that HEV capsid protein can directly bind to CDC42, and non- and quasi-enveloped HEV use different CDC42 downstream signaling pathways to participate in viral infection. The study provided some new insights to understand the life cycle of HEV in host cells and a new target of drug design for combating HEV infection.

Hepatitis E, a significant global public health issue in China, is caused by sporadic infections with regional hepatitis E virus (HEV) genotypes 1, 3, and 4. To date, most immunoassays currently used to test human sera for the presence of anti-HEV antibodies cannot identify HEV at the genotype level. However, such information would be useful for identifying the source of infecting virus. Therefore, here we describe the development of a competitive enzyme-linked immunosorbent assay (ELISA) for detecting anti-genotype 1 HEV antibodies in human sera. Using recombinant genotype 1 HEV ORF3 protein as immunogen, traditional hybridoma technology was employed to generate seven monoclonal antibodies (mAbs), of which two mAbs specifically reacted with the immunogen. One of these two mAbs, 1D2, was labeled with horseradish peroxidase (HRP) for use in competitive ELISA (cELISA). After cELISA optimization using a checkerboard assay design, the amount of ORF3SAR-55 as coating antigen (100 ng/well), HRP-1D2 mAb concentration (1 µg/mL), and test serum dilution (1:10) were selected and a result ≥ 19.5 was used as the cutoff for a positive result. Importantly, cross-genotype cELISA results indicated that the cELISA could not detect anti-genotype 3 rabbit and 4 swine HEV antibodies. Moreover, human sera confirmed as negative for anti-HEV antibodies using the commercial ELISA kit were all negative via cELISA. However, because the commercial ELISA kit detects anti-all genotypes HEV antibodies and the cELISA only detects anti-genotype 1 HEV antibodies, the consistence rate of two assays detecting positive sera is low. In summary, here a cELISA for detecting anti-genotype 1 HEV antibodies was developed for use in epidemiological investigations of genotype 1 HEV infections in humans. KEY POINTS: • Seven mAbs were produced using genotype 1 HEV ORF3 protein as immunogen. • One mAb that specifically bound to genotype 1 HEV ORF3 protein was selected and labeled for use in a cELISA to detect anti-genotype 1 HEV antibodies. • The competitive ELISA developed here will aid clinical diagnosis of HEV infections and will be useful for large-scale serological testing of genotype 1 HEV infections in humans.

Since the implementation of the Chinese economic reforms. The habitat quality of coastal has gradually deteriorated with economic development, but the concept of "ecological construction" has slowed the negative trend. For quantitative analysis of the correlation between the Pearl River Delta urban expansion and changes in habitat quality under the influence of the policy, we first analyzed the habitat quality change based on the InVEST model and then measured the impact of construction land expansion on the habitat quality through habitat quality change index (HQCI) and contribution index (CI) indicators. Finally, the correlation between urbanization level and habitat quality was evaluated using geographically weighted regression (GWR) and the Self-organizing feature mapping neural network (SOFM). The results indicated that: (1) during the study period from 2000 to 2020, habitat quality declined due to urban sprawl, indicating a deterioration of ecological structure and function, and the decrease was most significant from 2000 to 2010. (2) The urbanization index had a negative effect on the habitat quality, but the negative effect have improved after 2000, reflecting the positive effect of policies such as "ecological civilization construction" (3) The implementation degree of ecological civilization varies greatly among cities in the study area: Shenzhen, Dongguan, Foshan, and Zhongshan have the best level of green development. These results reflect the positive role of policies in the prevention of damage to habitat quality caused by economic development and provide a reference for the formulation of sustainable urban development policies with spatial differences.
