ABSTACT: The microbiota of traditional food provides a rich reservoir of biodiversity to find new strains with interesting features for novel functional food formulation. Therefore, this study aimed to investigate the biofunctional potential of the lactic acid bacteria (LAB) strain Jb21-11 isolated from Jben, a traditional Algerian fresh cheese. This isolate was selected out of a collection of 154 LAB based on its exopolysaccharide (EPS) phenotype and was preliminarily identified by polyphasic characterization as Lactiplantibacillus plantarum (previously known as Lactobacillus plantarum) and its biofunctional properties were then assessed in vitro. The tested strain demonstrated good resistance to gastric juice, acidity around pH 2, and 2% (v/v) bile salts, which are important characteristics for potential biofunctional LAB candidates. It also showed a good production of ropy EPS with 674 mg/L on MRS medium. However, this ability appears to compromise the adhesion of the strain to Caco-2 cells (less than 1%), which according to our results, seems not to be related to autoaggregation and hydrophobicity (44.88 ± 0.028% and 16.59 ± 0.012%). Furthermore, promising antimicrobial activity against three pathogenic bacteria (Escherichia coli, Staphylococcus aureus, and Salmonella) was detected probably due to antimicrobial metabolites excreted during fermentation process into the medium. Moreover, the strain L. plantarum Jb21-11 displayed a therapeutic functionality with both anti-inflammatory and immunomodulatory action using RAW 264.7 cells. The chemical features of the novel ropy Jb21-11-EPS were also investigated revealing the presence of three monosaccharides, namely, mannose, galactose, and glucose, with a molar ratio of 5.42:1.00:4.52 linked together by α- and ß-glycosidic bonds, presenting a relatively high molecular weight of 1.08 × 105 Da of interest for a texturing potential. Therefore, the new producing EPS strain Jb21-11 is a promising candidate for use as an adjunct culture for improving the texture of functional food.

AIM: Compare the laparoscopic treatment (LT) and the posterior sagittal anorectoplasty treatment (ST) of the rectovaginal fistulas (RvaF) in a single center. We have previously reported feasibility and results of LT in this rare variety of anorectal malformations (ARM) [1-3]. MATERIAL AND METHODS: 19 patients were treated between February 2000 and November 2020. Nine underwent a LT and 10 a ST. Both surgical techniques were previously described. [2][4][5] The distal posterior wall of the vagina was kept intact in the LT. A technical change was introduced in the ST for that purpose. The fistula was treated from the inside of the rectum, avoiding the opening of the distal vagina as described for the treatment of a urethra rectal bulbar fistula in males. Age at operation, associated anomalies, sacral ratio index (SR), complications, urinary continence, presence of spontaneous intestinal movements, constipation, soiling and requirements of bowel management program (BMP) were analyzed. RESULTS: Associated anomalies occurred in 17 patients (89.5 %), 63 % of which were urological. Five (26 %) had a SR below 0.4; 4 in the LT group and 1 in the ST group. The mean age at the time of operation was 23.2 (8-59) in ST and 17.6 months (4-32) in LT. Average operative time was 190.4 min for ST (120-334) and 195.8 min (90-270) for LT (p 0.13). One patient in the LT group presented a mild rectal prolapse and 2 a partial wound dehiscence after the ST. Only 15 patients were evaluable for functional results (8 in ST and 7 in LT). Mean follow up was 83 months (12-197). All patients are clean with a bowel management program. Five of the 7 patients undergoing a LT had a bad prognosis (SR < 0,4). Three (43 %) are clean with diet or any treatment, 3 (43 %) using laxatives or enemas and 1 (14 %) with a trans anal irrigation system. Only 1 of the 8 patients in the STgroup had a bad prognosis. Six (40 %) needed a diet; 4 (50 %) laxatives or enemas and 1 (10 %) a cecostomy button for antegrade enemas. CONCLUSIONS: Patients with RvaF had a high index of associated anomalies. The difference of operative time was not statistically significative. No differences in functional results between both groups were observed. LT is a valid option to treat RvaF.

OBJECTIVE: Heart failure (HF) is a complex clinical syndrome that impairs the ability to achieve proper filling or ejection, in which patients have typical symptoms and signs. It is a major Public Health problem with a high incidence and prevalence associated with high morbidity and mortality. The management of the patient with HF is complex, requiring in its treatment the work of specialized multidisciplinary teams in which the management of cardiac-healthy habits and self-care will play a leading role. Knowing the health literacy (HL) level of patients is a fundamental piece that will help us to provide a holistic attention, based on individual needs, promoting in this way the empowerment of the patient. Our aim will be to evaluate the effectiveness of an HL intervention for improving quality of life and decreasing morbidity/mortality. METHODS: A two-arm randomized controlled clinical trial will be conducted, with concealment of randomization. Patients with diagnosis of HF attended in cardiology and internal medicine consultations of 5 hospitals in Spain will be included.


OBJETIVO: La insuficiencia cardiaca (IC) es un síndrome clínico complejo con una sintomatología bien definida que constituye un problema de Salud Pública por su impacto en la morbi-mortalidad. El manejo del paciente con IC requiere el trabajo de equipos especializados multidisciplinares que enfaticen en los hábitos cardiovasculares y el autocuidado. Conocer el grado alfabetización en salud (AeS) de estos pacientes es una buena herramienta para prestarles una atención holística, basada en necesidades individuales, así como para fomentar su empoderamiento. Se ha diseñado un protocolo con el objetivo de evaluar la eficacia de una intervención en AeS en la mejora de su calidad de vida, así como en la disminución de la morbi/mortalidad. METODOS: Se llevará a cabo un estudio clínico controlado aleatorio a dos brazos multicéntrico, con ocultación del reparto aleatorio. Se incluirán pacientes con diagnóstico de IC atendidos en consultas de cardiología y medicina interna de cinco hospitales de España.

BACKGROUND: Pornography consumption is higher in men, but we do not know if this association can be modified by different variables, such as sexual attraction and place of origin. Given the impact pornography has on minors, there are limited studies that analyze the use of pornography in representative samples of the adult population. The aim was analyze the prevalence and factors associated with using pornography in young adult men and women, living in Spain, with different sexual attractions and different places of birth. METHODS: Cross-sectional study with an online survey conducted with 2515 men and women aged between 18 and 35 years of age. The prevalence of pornography consumption is described and analyzed in the total sample and stratified by sex, according to socio-demographic and sexual attraction variables. The association between covariates and pornography consumption at some point in life was estimated with prevalence ratios (PR) obtained with the Poisson models of robust variance. DEPENDENT VARIABLE: voluntarily using pornography at some point in life. Socio-demographic variables were included in the analysis: age, sex, level of education, place of birth. Sexual attraction was also analyzed. RESULTS: In Spain, 94.7% of men between 18 and 34 years and 74.6% of women have voluntarily used pornography at some point in their life. The mean age to start using it is earlier in men [Mean:14.2; Standard Deviation (SD):2.3]. Bisexual/homosexual attraction (reference: heterosexual) increases the probability of using pornography in women [(PR (95%CI): 1.30 (1.22; 1.38)]. Yet this is not observed in men. In both sexes, the probability of using pornography increases with age [(PR (95%CI): 1.01(1.00; 1.01)] and coming from abroad (reference: native), being the effect of country of birth significantly higher in women [(PR (95%CI): 1.17 (1.09; 1.26)] than in men [(PR (95%CI): 1.04 (1.01; 1.07). CONCLUSIONS: Public health programmes aimed at improving affective-sexual health should consider the high use of pornography among young adults in Spain, as well as those variables that increase its use.

BACKGROUND: Hegemonic masculinity has been recognized as contributing to the perpetration of different forms of gender-based violence (GBV). Abandoning hegemonic masculinities and promoting positive masculinities are both strategies used by interventions that foreground a "gender-transformative approach." Preventing GBV among young people could be strengthened by engaging young men. In this article, we aim to systematically review the primary characteristics, methodological quality, and results of published evaluation studies of educational interventions that aim to prevent different forms of GBV through addressing hegemonic masculinities among young people. MAIN BODY: We conducted a systematic review of available literature (2008-2019) using Medline (PubMed), Scopus, Web of Science, PsycInfo, the CINAHL Complete Database, and ERIC as well as Google scholar. The Template for Intervention Description and Replication was used for data extraction, and the quality of the selected studies was analyzed using the Mixed Method Appraisal Tool. More than half of the studies were conducted in Africa (n = 10/15) and many were randomized controlled trials (n = 8/15). Most of the studies with quantitative and qualitative methodologies (n = 12/15) reported a decrease in physical GBV and/or sexual violence perpetration/victimization (n = 6/15). Longitudinal studies reported consistent results over time. CONCLUSIONS: Our results highlight the importance of using a gender-transformative approach in educational interventions to engage young people in critical thinking about hegemonic masculinity and to prevent GBV.

OBJECTIVE: Despite the efforts to reduce gender inequality, sexism persists. There are still few studies on this matter that consider variables such as sexual desire or migration background. The aim of this study is to analyse the social circumstances (individual and from the immediate context) associated with sexism as well as its relationship with intimate partner violence perpetration amongst adolescents. METHOD: Cross-sectional study with 1421 students aged 13-17 from Alicante and Terrassa (2019-2021). Sexism was measured with the Ambivalent Sexism Inventory, which assesses ambivalent, hostile and benevolent sexism. Generalized linear models were carried out and stratified by sex to identify variables associated with sexism. A logistic regression model was used to assess the association between sexism and intimate partner violence. RESULTS: The likelihood of a high score in ambivalent sexism was higher in migrants (girls and boys: p<0.01) and adolescents with migrant parents (girls: p<0.01; boys: p<0.05). Non-heterosexual desire was negatively associated with sexism in girls (benevolent: p<0.01) and boys (ambivalent and hostile: p<0.01; benevolent: p<0.05). Hostile sexism showed association with intimate partner violence perpetration, independently of the rest of the adjustment variables included in the model, for both girls (odds ratio [OR]: 1.07; 95% confidence interval [95% CI]: 1.00-1.14) and boys (OR: 1.11; 95% CI: 1.03-1.20). CONCLUSIONS: Sexism is present amongst teenagers and its relationship with intimate partner violence is confirmed. Sexual desire and migration background should be taken into account for intimate partner violence prevention.

Study of the neurodevelopment of children is vital to promote good quality of life during childhood. Few batteries showing adequate reliability and validity indices are available to evaluate the different neuropsychological domains. The objective of this study was to obtain initial evidence on the validity of the Computerized Battery for Neuropsychological Evaluation of Children (BENCI) in a Spanish population. To assess the validity of the BENCI battery and other measures of task switching, abstract reasoning, linguistic abilities, processing speed, and attention were used. The sample was composed of a total of 73 children aged 9, 10, and 11 years. Significant differences among age groups were observed in the domains of sustained attention, memory, and executive function. In addition, the BENCI subtests showed statistically significant correlations with the other neuropsychological tools. Further research is warranted on the relationship of the BENCI with other tests in wider age groups and to assess the factorial structure of the scale and the reliability values of the subtests. In conclusion, this study seems to indicate that the Spanish version of the BENCI has promising validity to be used for evaluating the main neuropsychological domains in children.

BACKGROUND: Machismo and acceptance of violence (AV) against women are part of the social construction of hegemonic masculinity and are related to the risk of dating violence. This study aims to analyze the effectiveness of the Lights4Violence program in reducing machismo and AV in secondary school students from different European cities. METHODS: Quasi-experimental longitudinal study using a convenience sample of 1,146 high school students from different European cities (12-17 years old) including 575 intervention group students (59.1% girls) and 571 control group students (62.7% girls). We performed linear regression models to identify the effect of the intervention, modelling the difference in means in machismo and AV (dependent variables) between wave-2 and wave-1. RESULTS: An interaction was identified between the group variable and the empathy variable. In wave-2, girls with high empathy at baseline in the intervention group obtained lower mean AV scores (ß: -0.131; p = 0.004). However, the boys in the intervention group (reference: control group) with low empathy at baseline registered a significant increase in the mean values of machismo (ß: 0.247; p < 0.001). CONCLUSION: The importance of empathy is shown in the effectiveness of interventions to reduce machismo and AV in adolescents. While the Lights4Violence program focuses on promoting healthy relationships, there were some controversial results. It is possible that some children, especially those with less empathy, may have felt "challenged" during the intervention and/or assessment. This suggests the need for the development of interventions that also consider psychological processes and integrate the promotion of positive expressions of masculinity.

BACKGROUND: Dating Violence (DV) is a public health problem that is on the rise. In this paper, we aim to analyse different factors associated with DV victimization among female and male adolescents in Spain, considering socioeconomic circumstances, sexual orientation and the presence of different attitudes and experiences related to violence. METHODS: Cross-sectional data from a convenience sample of 640 ever-partnered adolescents aged 13 to 17 at schools in the cities of Alicante (n = 359, 50.1% girls) and Terrassa (n = 281, 51.9%) in the context of an educational intervention to promote healthy relationships. We calculated the prevalence of different forms of DV (physical, sexual and control and fear) and carried out multivariate regression models by sex. RESULTS: 5.5% of girls and 8.7% of boys declared having suffered lifetime physical and/or sexual violence, while 22% of girls and 20.5% of boys reported control and/or fear victimization. The likelihood of DV was higher among migrants and those with foreign-born parents (aPR girls = 2.1 CI95%: 1.1-3.9; aPR boys = 1.9: CI95%: 1.0-3.6); prior experiences of abuse (aPR girls = 1.6; CI95%: 1.0-2.6; aPR boys = 1.7; CI95%: 1.1-2.6); and those who showed higher levels of machismo (aPR girls = 1.0; CI95%: 1.0-1.1; aPR boys = 1.0; CI95%: 1.0-1.1). In girls, DV increased among those who reported lesbian/bisexual orientation and poor relationship with teachers. CONCLUSIONS: DV is socially patterned and increases among LGB adolescents (especially in the case of girls), migrants, and those with foreign-born parents, and adolescents who reported prior experiences of violence in childhood. Future DV prevention programs should consider social inequalities in the likelihood of DV and by reinforcing adolescents' abilities to recognize social support sources and reject machismo and violence.

In the letter, Urro et al. performed a search on the sucrose, fructose and sorbitol content in the approved Sars-Cov-2 vaccines and they concluded that these vaccines can be safely administered in adults affected by Hereditary fructose intolerance.The Pfizer-BioNTech COVID-19 Vaccine is currently approved for use in adolescents ≥ 12 years and the Moderna COVID-19 vaccine is close to approval for use in children over 12 years of age. Furthermore, both vaccines have initiated clinical trials that will include infant as young as 6 months. Therefore, we considerate important to analyze the safely administration of this two vaccines in children with Hereditary fructose intolerance.

The Maudsley Violence Questionnaire (MVQ) is an instrument specifically developed to evaluate violent thinking through two subscales examining macho attitudes and the acceptance of violence. This study analyzed the psychometric properties and factor structure of the Spanish version of the MVQ questionnaire in a large sample of 1933 Spanish adolescents. An online questionnaire was used to collect variables, such as sociodemographic and sexism data. The factor structure showed good fit indices in Spanish adolescents, which were similar to the original scale. The exploratory analysis yielded a first factor that explained 11.3% of the total variance and a second factor that explained 10.8% of the total variance. The Goodness of Fit Index (GFI) (0.902), Adjusted Goodness of Fit Index (AGFI) (0.90), Normed Fit Index (NNFI) (0.85), and the Comparative Fit Index (CFI) (0.86) suggested that the model fit the data adequately (with values ≥ 0.90) and the Root Mean Square Error of Approximation (RMSEA) (≤0.10) values indicative of an adequate fit. This study contributes a Spanish-language validated tool to measure machismo and the acceptance of violence among adolescents.

Patients with Obsessive-Compulsive Disorder (OCD) present neuropsychological deficits across different cognitive domains, especially in executive functioning and information processing speed. Some studies have even suggested that speed deficits may underlie poor neuropsychological performance. However, this hypothesis remains unanswered in both OCD general population and OCD refractory subgroup. In addition, it is not clear whether such deficits are secondary to the clinical symptoms or may constitute a primary deficit. The aim of this study was to explore the speed of processing hypothesis in treatment-refractory OCD patients, and to clarify to what extent slowness is related to psychopathological symptoms. Both clinical and neuropsychological examination was conducted to assess 39 OCD refractory patients candidates for neurosurgery and 39 healthy matched individuals. Principal component analysis revealed a three-component structure in the neuropsychological battery being used, including a speed of processing, working memory, and conflict monitoring components. Group comparisons revealed that OCD patients performed significantly worse than healthy individuals in speed measures, but no differences were found in executive tests not influenced by time. Correlation analyses revealed a lack of association between neuropsychological and clinical measures. The results suggest that treatment-refractory OCD patients exhibit a primary deficit in information processing speed independent of clinical symptoms.

Resumen: la presencia de fístulas como complicaciones de aneurismas de aorta abdominal (AAA) es una condición conocida, aunque poco frecuente. Se ha reportado en la literatura una incidencia para fístulas aortocavas (FAC) del 1 % o, incluso, se encuentran valores inferiores en algunas series de casos. El objetivo de este estudio es realizar una revisión de la literatura basada en un caso clínico para dar a conocer el abordaje diagnóstico y los hallazgos por imagen de las FAC con el fin de familiarizar a los lectores con las características más comunes de esta entidad mediante tomografía computarizada. Presentamos el caso de una paciente de 59 años de edad con un cuadro clínico de cuatro días de evolución de dolor abdominal, sin antecedentes de importancia. En el examen físico abdominal se identificó la presencia de una masa pulsátil asociada a soplo abdominal como hallazgos principales. Se procedió a evaluación mediante angioTC abdominal en donde se identificó dilatación aneurismática fusiforme de la aorta abdominal con fístula hacia la vena cava inferior (VCI) y compromiso de la arteria renal derecha con hipoperfusión renal ipsilateral.


Abstract: the presence of fistulas as complications of abdominal aortic aneurysms (AAA) is a known condition, however, infrequent. An incidence of 1 % for aortocaval fistulas (AFC) has been reported in the literature, or even lower values are found in some case series. The aim of this study is to review the literature based on a clinical case to present the diagnostic approach and imaging findings of AFC in order to familiarize readers with the most common characteristics of this entity by computerized tomography. We present the case of a 59-year-old female patient with a clinical picture of four-day evolution abdominal pain, with no relevant history. The abdominal physical examination identified the presence of a pulsatile mass associated with abdominal murmur as the main findings. Abdominal TC angiography assessment identified fusiform aneurysmal dilatation of the abdominal aorta with fistula to the inferior vena cava (VCI), and involvement of the right renal artery with ipsilateral renal hypoperfusion.


Resumo: a presença de fístulas como complicações dos aneurismas da aorta abdominal (AAA) é urna condição conhecida, porém rara. Na literatura, os relatos da incidencia de fístulas aorto-cava (FAC) é de apenas 1 % (valores ainda menores são encontrados em algumas séries de casos). O objetivo deste estudo é realizar uma revisão da literatura baseada em um caso clínico para informar a abordagem diagnóstica e os achados de imagem da FAC, a fim de familiarizar os leitores com as características mais comuns dessa entidade por meio da tomografia computadorizada. Apresentamos o caso de um paciente de 59 anos com quadro clínico de quatro dias de evolução de dor abdominal, sem história significativa. No exame físico abdominal, a presença de massa pulsante associada a sopro abdominal foi identificada como principais achados. Foi realizada uma avaliação com angioTC abdominal, na qual foi identificada dilatação aneurismática fusiforme da aorta abdominal com fístula na veia cava inferior (VCI) e acometimento da artéria renal direita com hipoperfusão renal ipsilateral.

The most important approach for the management of hereditary fructose intolerance is a strict avoidance of fructose, sucrose and sorbitol from the diet and medications. A safe threshold of 2.4 mg/kg/dose was recently established by the Instituto Superiore di Sanità of Italy for both oral and parenteral routes, thus shouldering a safe administration of a majority of vaccines in these patients. This would not include, Rotarix® pre-established oral suspension and Rotateq® vaccines, which are indeed contraindicated. Moreover, Rotarix® white powder and solvent for oral suspension would only be safely administered at a weight above 9.3 kg.Overall, these recommendations to avoid rotavirus vaccination are difficult to implement because these vaccines are given during exclusive breastfeeding, prior to fructose-containing food introduction.

Dating violence (DV) among adolescents is a public health issue because of its negative health consequences. In this study, we aimed to analyse the prevalence and the psychosocial and socioeconomic risk and protective factors associated DV among male and female adolescents in Europe. It was performed a cross-sectional study based on a non-probabilistic sample of 1555 students aged 13-16 years (2018-2019). The global prevalence of DV victimization was significantly greater among girls than boys (girls: 34.1%, boys: 26.7%; p = 0.012). The prevalence of DV in both girls and boys was greater for those over age 15 (girls: 48.5% p < 0.001; boys: 35.9%; p = 0.019). There was an increased likelihood of DV victimization among girls whose fathers did not have paid employment (p = 0.024), who suffered abuse in childhood, and reported higher Benevolent Sexism [PR (CI 95%): 1.01 (1.00-1.03)] and machismo [1.02 (1.00-1.05)]. In the case of boys, the likelihood of DV increased with abuse in childhood (p = 0.018), lower parental support [0.97 (0.96-0.99)], high hostile sexism scores (p = 0.019), lower acceptance of violence (p = 0.009) and high machismo (p < 0.001). Abuse in childhood was shown to be the main factor associated with being a victim of DV in both population groups, as well as sexism and machismo attitudes. These results may contribute to future DV prevention school programs for both, teenagers and children of elementary school ages.

PURPOSE: To analyze the potential association between social support, experiences of violence, and sociodemographic characteristics of adolescents and the likelihood of acceptance of violence and machismo in different European countries. METHODS: Cross-sectional design. We recruited 1,555 participants ages 13-16 from secondary schools in Alicante (Spain), Rome (Italy), Iasi (Romania), Matosinhos (Portugal), Poznan (Poland), and Cardiff (UK). We used linear regression models to identify how social support from teachers and parents, experiences of violence-dating violence, bullying, cyberbullying, abuse in childhood-and sociodemographic characteristics were associated with violent thinking, specifically: machismo and acceptance of violence. The analysis was stratified by sex. RESULTS: Acceptance of violence was higher for those who had lower perceived social support from parents (ßgirls = -154, p < .001; ßboys = -.114, p = .019) for both sexes. Perpetration of bullying and/or cyberbullying was associated with higher scores for machismo and acceptance of violence for both sexes (ßgirls = .067, p = .035; ßboys = .225, p < .001; (ßgirls = .118, p < .001; ßboys = .210, p < .001). Being the victim of dating violence, having suffered physical and/or sexual abuse in childhood, and lower perceived social support from teachers were associated with higher scores for both machismo and acceptance of violence. These associations differed between girls and boys. CONCLUSIONS: Machismo and acceptance of violence are widely present amongst adolescents in different European countries. Our results suggest the importance of providing educational/psycho-educational interventions with boys and girls to prevent these attitudes and, in turn, prevent interpersonal violence, including bullying and dating violence.

Background and Aims: Diagnostic delay (DD) is especially relevant in children with inflammatory bowel disease, leading to potential complications. We examined the intervals and factors for DD in the pediatric population of Spain. Methods: We conducted a multicentric prospective study, including 149 pediatric inflammatory bowel disease patients, obtaining clinical, anthropometric, and biochemical data. Time to diagnosis (TD) was divided into several intervals to identify those where the DD was longer and find the variables that prolonged those intervals. Missed opportunities for diagnosis (MODs) were also identified. Results: Overall TD was 4.4 months (interquartile range [IQR] 2.6-10.4), being significantly higher in Crohn's disease (CD) than in ulcerative colitis (UC) (6.3 [IQR 3.3-12.3] vs. 3 [IQR 1.6-5.6] months, p = 0.0001). Time from the visit to the first physician until referral to a pediatric gastroenterologist was the main contributor to TD (2.4 months [IQR 1.03-7.17] in CD vs. 0.83 months [IQR 0.30-2.50] in UC, p = 0.0001). One hundred and ten patients (78.3%) visited more than one physician (29.9% to 4 or more), and 16.3% visited the same physician more than six times before being assessed by the pediatric gastroenterologist. The number of MODs was significantly higher in CD than that in UC patients: 4 MODs (IQR 2-7) vs. 2 MODs ([IQR 1-5], p = 0.003). Referral by pediatricians from hospital care allowed earlier IBD diagnosis (odds ratio 3.2 [95% confidence interval 1.1-8.9], p = 0.025). Conclusions: TD and DD were significantly higher in CD than those in UC. IBD patients (especially those with CD) undergo a large number of medical visits until the final diagnosis.

BACKGROUND: CD6 is a lymphocyte surface co-receptor physically associated with the T-cell receptor (TCR)/CD3 complex at the center of the immunological synapse. There, CD6 assists in cell-to-cell contact stabilization and modulation of activation/differentiation events through interaction with CD166/ALCAM (activated leukocyte cell adhesion molecule), its main reported ligand. While accumulating evidence is attracting new interest on targeting CD6 for therapeutic purposes in autoimmune disorders, little is known on its potential in cancer. In an attempt to elucidate the in vivo relevance of blocking CD6-mediated interactions in health and disease, we explored the consequences of expressing high circulating levels of a soluble form CD6 (sCD6) as a decoy receptor. METHODS: High sCD6 serum levels were achieved by using transgenic C57BL/6 mice expressing human sCD6 under the control of lymphoid-specific transcriptional elements (shCD6LckEµTg) or wild type either transduced with hepatotropic adeno-associated virus coding for mouse sCD6 or undergoing repeated infusions of recombinant human sCD6 protein. Characterization of sCD6-induced changes was performed by ex vivo flow cytometry and functional analyses of mouse lymphoid organ cells. The in vivo relevance of those changes was explored by challenging mice with subcutaneous or metastatic tumors induced by syngeneic cancer cells of different lineage origins. RESULTS: Through a combination of in vitro and in vivo studies, we show that circulating sCD6 expression induces defective regulatory T cell (Treg) generation and function, decreased CD166/ALCAM-mediated tumor cell proliferation/migration and impaired galectin-induced T-cell apoptosis, supporting the fact that sCD6 modulates antitumor lymphocyte effector function and tumorigenesis. Accordingly, sCD6 expression in vivo resulted in delayed subcutaneous tumor growth and/or reduced metastasis on challenge of mice with syngeneic cancer cells. CONCLUSIONS: Evidence is provided for the disruption of CD6 receptor-ligand interactions as a feasible immunomodulatory approach in cancer.
