Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) can infect a broad range of animal species and has been associated with severe disease in some taxa. Few studies have evaluated optimal strategies to mitigate the risk to susceptible zoo animals. This study evaluated the safety and immunogenicity of a protein-based veterinary SARS-CoV-2 vaccine (SpikeVet™) in zoo animals. Two to three doses of SpikeVet™ were administered intramuscularly or subcutaneously 3-4 weeks apart to 354 zoo animals representing 38 species. SpikeVet™ was very well tolerated across all species. Minor adverse effects were observed in 1.69% of animals vaccinated, or 1.04% of vaccine doses administered. Preliminary immunogenicity analyses in representative carnivores (meerkats, lions) and an artiodactylid (domestic goat) showed SpikeVet™-immunized animals developed serum antibodies able to neutralize a range of SARS-CoV-2 variants, including the vaccine-homologous Wuhan and Mu variants, as well as vaccine-heterologous Omicron BA.2 and XBB.1 strains. Prior to vaccination, all eight lions were seropositive for Wuhan strain by surrogate viral neutralization testing, suggesting past infection with SARS-CoV-2 or cross-reactive antibodies generated by another closely related coronavirus. These results from a range of zoo species support the ongoing development of SpikeVet™ as a safe and effective veterinary SARS-CoV-2 vaccine.

Zoos are increasingly implementing formalized animal welfare assessment programs to allow monitoring of welfare over time, as well as to aid in resource prioritization. These programs tend to rely on assessment tools that incorporate resource-based and observational animal-focused measures. A narrative review of the literature was conducted to bring together recent studies examining welfare assessment methods in zoo animals. A summary of these methods is provided, with advantages and limitations of the approaches presented. We then highlight practical considerations with respect to implementation of these tools into practice, for example scoring schemes, weighting of criteria, and innate animal factors for consideration. It is concluded that there would be value in standardizing guidelines for development of welfare assessment tools since zoo accreditation bodies rarely prescribe these. There is also a need to develop taxon or species-specific assessment tools to complement more generic processes and more directly inform welfare management.

In the 2019-2020 Australian bushfires, Kangaroo Island, South Australia, experienced catastrophic bushfires that burnt approximately half the island, with an estimated 80% of the koala population lost. During and after the event, rescued koalas were triaged at a designated facility and a range of initial data were recorded including rescue location and date, sex, estimation of age, body condition and hydration, and assessment of burn severity (n = 304 records available). Koalas were presented to the triage facility over a span of 10 weeks, with 50.2% during the first 14 days of the bushfire response, the majority of which were rescued from regions of lower fire severity. Burns were observed in 67.4% of koalas, with the majority (60.9%) classified as superficial burns, primarily affecting the limbs and face. Poor body condition was recorded in 74.6% of burnt koalas and dehydration in 77.1%. Negative final outcomes (death or euthanasia, at triage or at a later date) occurred in 45.6% of koalas and were significantly associated with higher mean burn score, maximum burn severity, number of body regions burnt, poor body condition score, and dehydration severity. The findings of this retrospective study may assist clinicians in the field with decision making when triaging koalas in future fire rescue efforts.

Macropod progressive periodontal disease (MPPD) is a necrotizing, polymicrobial, inflammatory disease commonly diagnosed in captive macropods. MPPD is characterized by gingivitis associated with dental plaque formation, which progresses to periodontitis and then to osteomyelitis of the mandible or maxilla. However, the underlying microbial causes of this disease remain poorly understood. In this study, we collected 27 oral plaque samples and associated clinical records from 22 captive Macropodidae and Potoroidae individuals that were undergoing clinical examination at Adelaide and Monarto Zoos in South Australia (15 healthy, 7 gingivitis and 5 periodontitis-osteomyelitis samples). The V3-V4 region of the 16S ribosomal RNA gene was sequenced using an Illumina Miseq to explore links between MPPD and oral bacteria in these animals. Compositional differences were detected between the microbiota of periodontitis-osteomyelitis cases compared to healthy samples (p-value with Bonferroni correction < 0.01), as well as gingivitis cases compared to healthy samples (p-value with Bonferroni correction < 0.05) using Permutational Multivariate Analysis of Variance (PERMANOVA). An overabundance of Porphyromonas, Fusobacterium, and Bacteroides taxa was also identified in animals with MPPD compared to healthy individuals using linear discriminant analysis effect size (LEfSe; p = < 0.05). An increased abundance of Desulfomicrobium also was detected in MPPD samples (LEfSe; p < 0.05), which could potentially reflect differences in disease progression. This is the first microbiota analysis of MPPD in captive macropods, and these results support a polymicrobial pathogenesis of MPPD, suggesting that the microbial interactions underpinning MPPD may be more complex than previously documented.

When treatment is required for a herd/flock health concern, a range of factors must be considered to determine the preferred treatment strategy. If a group treatment strategy is pursued, considerations to optimize the likelihood of safe and effective group treatment include taxon-specific pharmacokinetics/pharmacodynamics, the formulation of prescribed medication, the type and amount of food, the number and type of water sources, enclosure design, size and demography of the group, weather conditions, and health status of individuals in the group. In addition, antimicrobial stewardship principles and relevant legislation/regulation must be at the forefront of decision making.

There is an increasing focus on evidence-based welfare assessment by animal care staff in zoos, along with a strong interest in animal welfare by the zoo-visiting public, to the extent that this can influence their choice of institutions to visit. Regulatory oversight of animal welfare standards continues to strengthen across many jurisdictions. Zoos are increasingly formalizing their practices with the development and refinement of evidence-based welfare assessment tools. There has been a drive for welfare assessment tools to comprise both resource-based and animal-based measures. However, animal-based indicators are not always well characterized, in terms of their nature and whether they infer a positive or negative affective state. This is especially so for reptiles, which are often considered behaviorally inexpressive and are under-researched. In this study, a Delphi consultation approach was used to gather expert opinion on the suitability of potential animal-based indicators of welfare for inclusion in a welfare assessment tool across four families of reptiles: Agamidae, Chelidae, Pythonidae, and Testudinidae. Two rounds of online surveys were conducted eliciting responses from a global group of professionals who work with reptiles. In the first survey, respondents were provided with an author-derived list of potential animal-based indicators for consideration of their validity and practicality as welfare indicators. The indicators were refined for the second survey including only those indicators that were considered valid or practical on the first survey (≥4 on a 5-point Likert scale), and that achieved ≥70% consensus amongst experts. In the second survey, respondents were asked to re-evaluate the reliability and practicality of the indicators and to rank them on these facets. Eight to ten assessment indicators for each family of reptiles were identified from Survey 2. These indicators were often health related, for example, presence of oculo-nasal discharge or wounds. However, some true behavioral indicators were identified, such as showing species-specific interest and alertness. These indicators should now be incorporated into taxon-tailored welfare assessment tools for trial and validation in captive reptile populations. This study provides a next step towards developing reptile-specific animal welfare assessment tools for these often-overlooked animals.

Pneumonia has been reported in both free-ranging and captive koalas and a number of causative agents have been described. Between 2016 and 2019, 16 free-ranging and 1 captive koala (Phascolarctos cinereus) from the Mount Lofty Ranges of South Australia were identified with pyogranulomatous lobar pneumonia, which involved the left caudal lobe in 14/17 (82%) cases. Within lesions, numerous gram-positive or gram-variable, non-acid-fast filamentous bacteria were observed in association with Splendore-Hoeppli phenomenon. Culture yielded growth of anaerobic bacteria, which were unidentifiable by MALDI-TOF-MS (matrix-assisted laser desorption ionization-time of flight mass spectrometry) analysis in 5/5 cases. Sequencing of the bacterial 16S rRNA gene identified a novel Actinomyces species in 4 samples, confirming a diagnosis of pulmonary actinomycosis. Concurrent examination of resin lung casts from healthy koalas suggested greater laminar flow of air to the left caudal lung lobe in koalas. Actinomyces spp. have been reported as commensals of the oral microbiome in other species, and an association with similar pulmonary lesions in other species. Considering the predilection for involvement of the left caudal lung lobe, aspiration is suggested as the likely cause in some cases of pulmonary actinomycosis in koalas. Pulmonary actinomycosis has not been previously described in koalas and further work needs to be undertaken in order to classify this organism within the Actinomyces genus.

The pharmacokinetics of fluoroquinolones in chelonians are well described but this does not extend to pradofloxacin, a broad-spectrum veterinary fluoroquinolone available as an oral suspension for cats and dogs. The aim of this study was to investigate the single-dose pharmacokinetic profile of pradofloxacin oral suspension at 7.5 mg/kg in eastern long-necked turtles (Chelodina longicollis). Eight treated turtles were sampled at multiple time points up to 168 hr. Plasma concentrations were measured using high-performance liquid chromatography. Pradofloxacin was quantifiable for up to 48 hr after drug administration. The Tmax (9.0 hr) and T½ to 48 hr (13.16 hr) were longer, and the Cmax (0.2 µg/ml) and AUC0-24 (2.2 hr*µg/ml) lower, than previously reported in cats and dogs. Pradofloxacin was measurable in tank water samples for up to 48 hr. No adverse effects were observed in six turtles administered 7.5 mg/kg sid for 7 days. Using mammalian MIC data, the AUC0-24 /MIC ratios for a range of bacterial isolates suggest that this dose of pradofloxacin in turtles is unlikely to be effective against many bacterial pathogens.

A juvenile yellow-tailed black cockatoo (Calyptorhynchus funereus) was presented with paresis of the right wing, ptosis, and miosis of the right eye; feather erection of the right side of the head and neck; and a penetrating injury over the right pectoral muscle. Temporary reversal of ptosis, miosis, and feather erection after administration of phenylephrine drops confirmed a diagnosis of Horner syndrome. Computed tomographic imaging revealed a fractured rib, traumatic lung lesions, and subcutaneous emphysema. The right-sided Horner syndrome and wing paresis were attributed to a sympathetic nerve trauma of the eye and feathers and to the brachial plexus, respectively. This report describes the diagnosis and resolution of ptosis and miosis within 8 weeks and recovery of feather symmetry and wing function within 11 weeks of the cockatoo's initial presentation with a conservative-management treatment plan.

Psittacid Adenovirus-2 (PsAdv-2) was identified in captive orange-bellied parrots (Neophema chrysogastor) during a multifactorial cluster of mortalities at the Adelaide Zoo, South Australia, and an outbreak of Pseudomonas aeruginosa septicaemia at the Tasmanian Department of Primary Industries, Parks, Water and Environment captive breeding facility, Taroona, Tasmania. This was the first time that an adenovirus had been identified in orange-bellied parrots and is the first report of PsAdv-2 in Australia. To investigate the status of PsAdv-2 in the captive population of orange-bellied parrots, 102 healthy birds from five breeding facilities were examined for the presence of PsAdv-2 DNA in droppings and/or cloacal swabs using a nested polymerase chain reaction assay. Additionally, eight birds released to the wild for the 2016 breeding season were similarly tested when they were recaptured prior to migration to be held in captivity for the winter. PsAdv-2 was identified in all breeding facilities as well as the birds recaptured from the wild. Prevalence of shedding ranged from 29.7 to 76.5%, demonstrating that PsAdv-2 is endemic in the captive population of orange-bellied parrots and that wild parrots may have been exposed to the virus. PsAdv-2 DNA was detected in both cloacal swabs and faeces of the orange-bellied parrots, but testing both samples from the same birds suggested that testing faeces would be more sensitive than cloacal swabs. PsAdv-2 was not found in other psittacine species housed in nearby aviaries at the Adelaide Zoo. The source of the infection in the orange-bellied parrots remains undetermined. In this study, PsAdv-2 prevalence of shedding was higher in adult birds as compared to birds less than one year old. Preliminary data also suggested a correlation between adenovirus shedding prevalence within the breeding collection and chick survival.

Reptiles are held at wildlife parks and zoos for display and conservation breeding programs and are increasingly being kept as pets. Reliable indicators of welfare for reptiles need to be identified. Current guidelines for the captive management of reptiles utilize resource-based, rather than animal-based indicators; the latter being a more direct reflection of affective state. In this paper we review the literature on welfare assessment methods in reptiles with a focus on animal-based measures. We conclude that, whilst a number of physiological and behavioral indicators of welfare have been applied in reptiles, there is need for further validation of these methods across the diversity of species within the Class. Methods of positive welfare state assessment are comparatively understudied and need elucidation. Finally, we examine some widely-used welfare assessment tools in mammals and explore the application of the Welfare Quality® Protocol to the endangered pygmy blue-tongue skink, Tiliqua adelaidensis. We propose that this framework can form the basis for the development of taxon-specific tools with consideration of species-specific biology.

A 47-day-old orphaned Goodfellow's tree kangaroo (Dendrolagus goodfellowi) joey was successfully cross-fostered onto a yellow-footed rock wallaby (Petrogale xanthopus). The joey was subsequently taken for hand-rearing at age 5 months. This is the first report of the cross-fostering technique, well-established in other macropods, being applied to a Dendrolagus sp. This technique can be considered as a viable option to raise young orphaned tree kangaroos, and as a tool to accelerate breeding in captive breeding programs of Dendrolagus spp.

In 2009, an outbreak of white nodular cutaneous lesions was detected in one of only two known breeding colonies of the critically endangered southern bentwing bat (Miniopterus schreibersii bassanii), at Naracoorte, South Australia. Necropsies were conducted on 10 euthanized bats in September 2009. In October 2009, 123 bats were examined under anesthesia, with skin biopsies collected from 18 affected bats. Prevalence of skin lesions was 45.2%. The prevalence among males was three times greater than among females. The majority of lesions examined histologically were granulomas, typically centered on a nematode. A single lesion had epidermal hyperplasia with intracytoplasmic inclusions consistent with a pox virus; pox virions were identified on electron microscopy. Nematodes dissected from frozen lesions were identified morphologically as Riouxgolvania beveridgei, previously described in the eastern bentwing bat (Miniopterus schreibersii oceanensis). The factors contributing to this apparent disease emergence and outbreak remain undetermined. Lesions consistent with white nose syndrome were not identified.

Syngnathidae is the family of fish that includes sea horses, pipefish, and sea dragons. To date, only a single publication has described neoplasia in syngnathids, a fibrosarcoma of the brood pouch in an aquarium-reared lined sea horse (Hippocampus erectus). From 1998 until 2010, the Toronto Zoo submitted 172 syngnathids for postmortem; species included the spotted or yellow sea horse (Hippocampus kuda), the pot-bellied sea horse (Hippocampus abdominalis) and the weedy sea dragon (Phyllopteryx taeniolatus). Seven neoplasms and two neoplastic-like lesions were identified from these cases. Under light microscopy, the neoplasms had morphological characteristics of a cardiac rhabdomyosarcoma, renal adenocarcinoma, renal adenoma, renal round cell tumors, which were likely lymphomas, exocrine pancreatic carcinoma, and intestinal carcinoma. Of these neoplasms, four had clear evidence of metastasis: the pancreatic and intestinal carcinomas and both round cell tumors. As syngnathids are highly fastidious animals, they can be difficult to maintain in captivity. In order to improve their husbandry, preventative and palliative care, as well as treatment, it is important to investigate and document the types of diseases affecting syngnathids.

The study of secretive snakes, such as rattlesnakes, has benefited from the use of radiotelemetry. However, the principal assumption in telemetry studies is that the transmitter has no significant effect on the study animal. To test the validity of this assumption, the physiologic and pathologic effects of intracoelomic implants were examined in a group of 24 eastern massasauga rattlesnakes (Sistrurus catenatus catenatus) in a laboratory setting over a period of 58 wk between March 2005 and April 2006. Inflammation and infection were evaluated using gross examination, histopathology, bacteriology, hematology, and plasma protein electrophoresis. Inflammation and infection occurred despite careful surgical procedures and advanced veterinary care. Four of 12 (33%) snakes developed extensive inflammatory response to the transmitter and associated anaerobic and gram-negative bacterial infections. Another four (33%) snakes showed mild inflammatory responses without infection. Reaction to the transmitters was reflected in changes in values for heterophils, monocytes, alpha-1, and beta globulin levels. Some conclusions reached in field studies using implanted radiotransmitters in snakes may be invalid if the implant influences the behavior or survival of the subject. Advances in attachment methods and transmitter coating technology may prevent some of the adverse effects associated with surgically implanted transmitters.

Five tammar wallabies (Macropus eugenii) were injected intramuscularly with 10 mg/kg amoxicillin trihydrate. Serial blood samples were collected through to 26 hr postinjection. Plasma amoxicillin concentrations were measured using high-performance liquid chromatography. Pharmacokinetic parameters were estimated using noncompartmental analysis. The terminal half-life (1.77 +/- 0.40 hr) was comparable to that previously reported in domestic small ruminants. Without intravenous kinetic data, it is unclear whether the terminal phase is elimination- or absorption-dependent; both scenarios have been reported in domestic species. Plasma concentrations of amoxicillin remained above a reported minimum inhibitory concentration (MIC) breakpoint for staphylococci and streptococci for at least 8 hr; the MIC breakpoint for enterobacteria and enterococci was never attained.

A sarcomatoid carcinoma was diagnosed in the lung of a 10-year-old captive Egyptian fruit bat (Rousettus aegyptiacus). Both carcinomatous and sarcomatous cytologic phenotypes were identified histologically. Cells of both types stained positive for pancytokeratin and S-100. Stromal cells stained positively for muscle actin. No staining for vimentin was noted in either neoplastic or normal internal control tissues. To the authors' knowledge, this is the first report of a pulmonary sarcomatoid carcinoma in a bat, and only the third report of sarcomatoid carcinoma outside the human literature.

Encephalomyocarditis virus (EMCV) is a picornavirus with a worldwide distribution, capable of infecting a wide range of species. Episodes of EMCV-associated mortality have been reported in zoos and national parks around the world, including sporadic cases at Taronga Zoo, Sydney. An inactivated EMCV vaccine was evaluated by inoculating Barbary sheep (Ammotragus lervia), Indian antelope (Antilope cervicapra), Eastern wallaroos (Macropus robustus), and chimpanzees (Pan troglodytes). A proportion of the vaccinated ungulates were administered a second vaccination 4 wk after the initial dose. Neutralizing antibody titers were monitored for a period of 12 mo. One month after vaccination, all vaccinated groups had developed significant antibody titers that persisted for at least 6 mo. Animals receiving two doses of vaccine had higher titers 3, 6, and 12 mo after the initial vaccination compared with animals receiving a single vaccine dose.
