RESUMO

The current biocatalytic method of industrial Cytidine triphosphate (CTP) production suffers from reaction rate loss. It is caused by gradually increasing acetate salt concentration, which inhibits enzyme activities and decreases the final yield. This work gave a possible solution to this problem through computational aided design of CMP kinase (CMPK), an enzyme in the CTP production system, to increase its stability in solution with high acetate salt concentration. Enlightened by the features of natural halophilic enzymes, the basic and neutral surface residues were replaced with acidic amino acids. This protein design strategy effectively increased the activity of CMPK in the working condition (acetate concentration over 1200 mM). The halotolerant CMPK was applied in fed-batch production of CTP. The maximum titer was 201.4 ± 1.6 mM, and the productivity was 12.6 mM L-1 h-1, increased 26.4% and 27.8% from the process using wild-type CMPK, respectively.

RESUMO

Fermentation and chemical methods for industrial cytidine diphosphate choline (CDP-choline) catalytic production both suffer from several disadvantages such as relatively low efficiency and productivity. To overcome these problems, we applied the concept of synthetic biology to develop a new one-pot multi-enzyme system to produce CDP-choline from orotic acid. Enzymes from different sources were selected and optimized as building blocks of the system, and parameters such as oxygen supply were also optimized. This system shows a titer of 37.6 ± 1.1 mM and a reaction rate of 1.6 mM L-1 h-1, both increase 66 % from traditional processes. It also has an efficiency of energy of 25.4%, improves 2-folds. This new one-pot CDP-choline-producing system has a potential for industrial use, and the procedure to design one-pot multi-enzyme system can be applied to build other one-pot system producing energy-rich compounds.

RESUMO

We show that when an external magnetic field parallel to the boundary is applied, the Weyl anomaly gives rises to a new anomalous current in the vicinity of the boundary. The induced current is a magnetization current in origin: the movement of the virtual charges near the boundary give rise to a nonuniform magnetization of the vacuum and hence a magnetization current. Unlike other previously studied anomalous current phenomena such as the chiral magnetic effect or the chiral vortical effect, this induced current does not rely on the presence of a material system and can occur in vacuum. Similar to the Casimir effect, our discovered phenomenon arises from the effect of the boundary on the quantum fluctuations of the vacuum. However this induced current is purely quantum mechanical and has no classical limit. We briefly comment on how this induced current may be observed experimentally.

RESUMO

We investigate the Casimir effect at a finite temperature in the electromagnetic Rindler space, and we find that the Casimir energy is proportional to T(4)/d(2) in the high-temperature limit, where T ≈ 27 °C is the temperature and d ≈ 100 nm is a small cutoff. We propose to make metamaterials to mimic the Rindler space and measure the predicted Casimir effect. Because the parameters of metamaterials we proposed are quite simple, this experiment would be easily implemented in the laboratory.

RESUMO

We applied the transformation optics to mimic a black hole of Schwarzschild form. Similar properties of photon sphere were also found numerically for the metamaterial black hole. Several reduced versions of the black hole systems were proposed for easier implementations.

RESUMO

We estimate the dominating frequencies contributing to the Casimir energy in a cavity of meta- materials mimicking de Sitter space, by solving the eigenvalue problem of Maxwell equations. It turns out the dominating frequencies are the inverse of the size of the cavity, and the degeneracy of these frequencies also explains our previous result on the unusually large Casimir energy. Our result suggests that carrying out the experiment in laboratory is possible theoretically.
