RESUMO

The activity and specificity of monoclonal antibodies (MCA) anti-A and anti-B, which were presented for testing at the 4th International Workshop on Monoclonal Antibodies to Red Blood Cells and Related Antigens, Paris, 2001, were studied. First, the activity of antibodies with erythrocytes with good expression of antigens A and B (controls) was tested. Then, the activity of MCA with erythrocytes with a weak expression of erythrocyte antigens was tested and the reaction was, after that, estimated in percent versus the controls. According to the results, the activity of interaction of anti-B antibodies with the phenotype A2 erythrocytes was lower versus such activity with the phenotype A1 erythrocytes. It is noteworthy, that all samples of antibodies, except for series 5, 6 and 10, were more active with the phenotype A2 erythrocytes versus the phenotype A2B erythrocytes. All tested samples of anti-B-antibodies possessed a high activity (within a range of 85-100%) and were effective in the detection of antigen B. The activity of MCA anti-AB with erythrocytes A2 was lower versus such activity with erythrocytes A and B and made 45-85%. MCAs of anti-A and of anti-B significantly differed by their ability to interact with erythrocytes A2. This may generate problems in using the above antibodies for making the typing reagents to be applied in routine blood group tests.

RESUMO

Prevalence of Lewis antigens and phenotypes has been investigated in 60 healthy donors and 74 patients with ischemic heart disease. It was found that phenotype Le(a-b-) marks high risk and phenotype Le(a-b+) is a resistance marker for development of ischemic heart disease. It would be valid to elucidate relationships between Lewis system and pathogenesis of internal diseases.

RESUMO

Effectiveness has been shown of human blood serum fractionation with polyethylene glycol for isolation of anti-A and anti-B isohemagglutinins from stocks with low antibody activity, inadequate for the production of AB0-typing standards. An optimal diluent of fractions containing antibodies of the AB0 system has been selected. The reagents prepared did not differ, by their activity and specificity, from the standard ones produced from the whole sera.
