RESUMO

Craniofacial growth and development have been shown to be influenced by various environmental factors that impact child development. This study aims to analyze the different patterns of feeding during early childhood, starting from birth, and assess the variability of nutrition during the first stage of childhood, along with the habits developed, to study their impact on jaw development. The study was conducted on a sample of twenty-five patients aged 3 to 5, following approval from the ethics committee of the Catholic University of Valencia. Informed consent was obtained from the fathers, mothers, and/or legal guardians, who were administered surveys on habits and diet. Cephalometric measurements within the parameters of ideal occlusion were subsequently taken. While previous studies examined this subject, the findings are challenging to evaluate. However, this study identified significant associations (p = 0.001) between clinical measurements and children's eating habits. The growth and development of the craniofacial cavity are influenced by multiple factors, including a child's diet and habits. Nonetheless, further research is required to determine whether diet can be considered a determining factor in proper jaw growth.

The objective of this study was to evaluate volumetric changes in the upper airway using Cone Beam Computed Tomography (CBCT) in orthodontic patients with maxillary transversal hypoplasia undergoing maxillary disjunction. The influence of factors such as sex, age, and growth pattern on airway volumetric changes was also assessed. The sample consisted of 50 growing patients from the dental clinic of Cardenal Herrera CEU University of Valencia. Airway volume was measured in mm3 before treatment (T0) and after palatal disjunction (T1). The final sample included 37 subjects in the treatment group and 13 in the control group. The volume gained exclusively from the disjunction treatment was determined to differentiate it from natural growth. The control group showed a mean volume increase from 10,911.3 ± 1,249.6 mm3 to 13,168.9 ± 1,789.7 mm3, representing a mean increase of 2,257.6 mm3 or + 20.9%. The treatment group exhibited an increase from 14,126.3 ± 4,399.8 mm3 at T0 to 18,064.1 ± 4,565.9 mm3 at T1, corresponding to a gain of 3,937.8 mm3 or + 31.8%. Significant differences in airway volume were observed after palatal disjunction compared to the control group. The expansion of the maxilla led to a significant increase in airway volume in the treated patients, estimated at 5,183 mm3 (+ 41.5%).

Hypoplastic or hypomineralized enamel defects represent a recurrent reason for consultation within the pediatric population, causing great discomfort due to their aesthetic appearance, as well as their functional limitations. Current conservative dentistry requires minimally invasive treatments in order to treat such defects and provide successful, definitive solutions. A systematic review of the literature has been carried out in accordance with the PRISMA recommendations. A search was carried out in the PubMed, Scopus, SciELO and Web of Science databases, completed with a manual search. The following variables were extracted from the selected studies: author, year, publication journal, type of study, sample, age of the participants and the materials used for its development. From the initial electronic search of the four databases, 282 articles were identified: 34 from PubMed, 240 from Scopus, 0 from SciELO and 8 from Web of Science. After eliminating duplicate articles, a total of 225 remained. After reading the title and abstract, 158 articles were eliminated, leaving 68. Upon reading the full text, the remaining studies were eliminated for not answering the research question or the inclusion criteria, leaving a total of 13 articles. Finally, 12 articles were used to carry out the systematic review. Treatments performed to date with the ICON™ system in pediatric patients have shown good results after their application. Since the variability of diagnostic methods has been observed, new diagnostic and assessment protocols should be created after treatment to objectify their effect on hypoplastic or hypomineralized enamel defects. In the same way, it has been described that treatment provides better results if combined with other opalustre-type or remineralizing materials. Growth-associated protein 43 plays a key role in neurite outgrowth through cytoskeleton remodeling. We have previously demonstrated that structural damage of peripheral nerves induces growth-associated protein 43 upregulation to promote growth cone formation. Conversely, the limited regenerative capacity of the central nervous system due to an inhibitory environment prevents major changes in neurite outgrowth and should be presumably associated with low levels of growth-associated protein 43 expression. However, central alterations due to peripheral nerve damage have never been assessed using the growth-associated protein 43 marker. In this study, we used the tubulization technique to repair 1 cm-long nerve gaps in the rat nerve injury/repair model and detected growth-associated protein 43 expression in the peripheral and central nervous systems. First, histological analysis of the regeneration process confirmed an active regeneration process of the nerve gaps through the conduit from 10 days onwards. The growth-associated protein 43 expression profile varied across regions and follow-up times, from a localized expression to an abundant and consistent expression throughout the regeneration tissue, confirming the presence of an active nerve regeneration process. Second, spinal cord changes were also histologically assessed, and no apparent changes in the structural and cellular organization were observed using routine staining methods. Surprisingly, remarkable differences and local changes appeared in growth-associated protein 43 expression at the spinal cord level, in particular at 20 days post-repair and beyond. Growth-associated protein 43 protein was first localized in the gracile fasciculus and was homogeneously distributed in the left posterior cord. These findings differed from the growth-associated protein 43 pattern observed in the healthy control, which did not express growth-associated protein 43 at these levels. Our results revealed a differential expression in growth-associated protein 43 protein not only in the regenerating nerve tissue but also in the spinal cord after peripheral nerve transection. These findings open the possibility of using this marker to monitor changes in the central nervous system after peripheral nerve injury.

The need to perform fast, effective and efficient pulpectomies has led to the development of numerous valid rotary systems. Its technical features allow the clinician to obtain good results in less working time. The objective of this study is to compare the characteristics of the different current rotary systems to favor a correct diagnosis and subsequent treatment. A systematic review of the literature has been carried out in accordance with the PRISMA recommendations. A search was carried out in PubMed, Embase Scopus, Cochrane and Web of Science databases, and was completed with a manual search. The following variables were extracted from the selected studies: author, year, sample, rotary systems used (length, diameter, taper, speed), obturation material and irrigant. From the initial electronic search of the five databases, 315 articles were identified. Once the duplicate articles were eliminated, a total of 233 remained. After reading both title and abstract, 200 articles were eliminated, leaving 33. On account of reading the full text, 22 were eliminated for not answering the research question or the inclusion criteria, leaving a total of 11 articles for the systematic review. Rotary systems which are able to adapt to the root anatomy of primary teeth and allow rapid and simple instrumentation, without producing excessive extrusion of debris at the root apex, will be the ones that provide the best results to the pediatric dentist during the performance of pulp treatment in primary teeth. Clinical success will only be achieved through proper prior diagnosis.

Drug-loaded nanocarriers (NCs) are new systems that can greatly improve the delivery and targeting of drugs to specific tissues and organs. In our work, a PPAR-γ agonist loaded into polymeric NCs was prepared, stabilized by spray-drying, and tested in vitro, ex vivo, and in vivo (animal models) to provide a safe formulation for optical anti-inflammatory treatments. The NCs were shown to be well tolerated, and no signs of irritancy or alterations of the eye properties were detected by the in vitro HET-CAM test and in vivo Draize test. Furthermore, no signs of cytotoxicity were found in the NC formulations on retinoblastoma cells (Y-79) analyzed using the alamarBlue assay, and the transmittance experiments evidenced good corneal transparency with the formulations tested. The ocular anti-inflammatory study confirmed the significant prevention efficacy using the NCs, and these systems did not affect the corneal tissue structure. Moreover, the animal corneal structure treated with the NCs was analyzed using X-ray diffraction using synchrotron light. Small-angle X-ray scattering (SAXS) analysis did not show a significant difference in corneal collagen interfibrillar spacing after the treatment with freshly prepared NCs or NCs after the drying process compared to the corresponding negative control when inflammation was induced. Considering these results, the PPAR-γ agonist NCs could be a safe and effective alternative for the treatment of inflammatory ocular processes.

Children who show better eating practices are less likely to suffer from severe caries than those who eat a diet rich in sugars. In the present study, we aimed to establish the relationship between the severity of dental caries and adherence to the Mediterranean diet. A cross-sectional study was carried out in which 263 children aged 2 to 14 years old were examined intraorally to analyze the presence and severity of caries. Children's parents/caregivers completed the KIDMED questionnaire to determine their degree of adherence to the Mediterranean diet. The results showed that the prevalence of caries is greater than 80% in children with medium or low adherence to the Mediterranean diet, and remains significant at 67% in the high adherence group (p = 0.010). A statistically significant negative correlation of weak magnitude (r = −0.29; p < 0.001) was found between adherence and the number of carious teeth. Caries severity in the first molars is also influenced by adherence to the diet in a statistically significant way. In conclusion, there is an association between adherence to the Mediterranean diet and the prevalence, extension, and severity of caries in the pediatric population.

The diagnosis of transverse growth deficit of the maxilla in daily clinical practice is carried out mainly through the experience of a well-trained clinician, which implies a lack of objective criteria applicable in a protocolized manner. The objective of this study was to establish a mathematical method to diagnose maxillary compression in relation to the dimensions of the skull and mandible. Methods: Records of 97 cases with an overall mean age of 9.8 ± 2.6 years were analyzed by three experienced orthodontists. The group of transverse compression was comprised of 62 cases and the control group of 35 cases. The main measurements of the widths were made on a frontal teleradiography of the skull (cranial, zygomatic, orbital, maxillary, bigonial and biantegonial width) and a lateral teleradiography of the skull (facial axis, mandibular plane, SNA, SNB, ANB and Wits). It was established that from the cranial width it is possible to predict the group to which each subject studied belongs-the compression group or the control group. A mathematical formula was obtained in the form of logistic regression that allows for the diagnosis of the presence of maxillary compression based on the cranial, maxillary and orbital widths with a sensitivity of 88.7% and a specificity of 77.1%.

INTRODUCTION: Introduction: dental caries is the most common contagious infectious disease of childhood. Much has been studied about the effect of a healthy diet on oral health, but little attention has been paid to the importance of eating patterns. Objectives: to establish the relationship between dental caries and eating behavior pattern using the Child Eating Behavior Questionnaire (CEBQ) in a sample of children. Materials and methods: a cross-sectional and descriptive study was carried out by selecting any boy or girl between 3 and 9 years who attended a dental clinic in the province of Alicante (Spain). An oral examination was performed to diagnose caries in children whose parents signed an informed consent. These parents/caregivers were also given the CEBQ to fill out. Results: a total of 276 children with a mean age of 86.5 months were examined. A higher average score was observed in the "food-avoidant" dimension in: "demand for food" and in the "response to satiety". A statistically significant difference was obtained in the mean values of the variables "slow eating" (p = 0.016) and "response to satiety" (p = 0.001) of the groups with and without caries. That is, the time a person spends eating influences the development of caries. Likewise, the ability of a person to be satisfied is also related to caries. Conclusions: the eating behavior pattern of children is a risk factor associated with infant caries.


INTRODUCCIÓN: Introducción: la caries dental es la enfermedad infecciosa contagiosa más común de la infancia. Se ha estudiado mucho sobre el efecto que tiene una dieta saludable en la salud oral, pero no se ha prestado mucha atención a la importancia que tiene el patrón de conducta alimentaria. Objetivos: establecer la relación entre la caries dental y el patrón de conducta alimentaria mediante el cuestionario Child Eating Behaviour Questionnaire (CEBQ) en una muestra de niños. Materiales y métodos: se realizó un estudio transversal y descriptivo seleccionando a cualquier niño o niña de entre 3 y 9 años que acudió a una clínica odontológica de la provincia de Alicante. Se hizo una exploración oral para diagnosticar la caries a los niños cuyos padres firmaron un consentimiento informado. También se les dio a estos padres/cuidadores el CEBQ para cumplimentar. Resultados: en total se examinaron 276 niños con una edad media de 86,5 meses. Se observó una mayor puntuación promedio en la dimensión Antiingesta en: "exigencia frente a los alimentos" y "respuesta a la saciedad". Se obtuvo una diferencia estadísticamente significativa en los valores medios de la variables "lentitud para comer" (p = 0,016) y "respuesta a la saciedad" (p = 0,001) de los grupos con y sin caries. Es decir, el tiempo que dedica una persona a comer influye en la aparición de caries. Asimismo, la capacidad que tiene una persona de saciarse también está relacionada con el desarrollo de caries. Conclusiones: el patrón de conducta alimentaria en los niños es un factor de riesgo asociado a la caries infantil.

Pioglitazone-loaded PLGA-PEG nanoparticles (NPs) were stabilized by the spray drying technique as an alternative to the treatment of ocular inflammatory disorders. Pioglitazone-NPs were developed and characterized physiochemically. Interaction studies, biopharmaceutical behavior, ex vivo corneal and scleral permeation, and in vivo bioavailability evaluations were conducted. Fibrillar diameter and interfibrillar corneal spacing of collagen was analyzed by synchrotron X-ray scattering techniques and stability studies at 4 °C and was carried out before and after the spray drying process. NPs showed physicochemical characteristics suitable for ocular administration. The release was sustained up to 46 h after drying; ex vivo corneal and scleral permeation profiles of pioglitazone-NPs before and after drying demonstrated higher retention and permeation through cornea than sclera. These results were correlated with an in vivo bioavailability study. Small-angle X-ray scattering (SAXS) analysis did not show a significant difference in the organization of the corneal collagen after the treatment with pioglitazone-NPs before and after the drying process, regarding the negative control. The stabilization process by Nano Spray Dryer B-90 was shown to be useful in preserving the activity of pioglitazone inside the NPs, maintaining their physicochemical characteristics, in vivo bioavailability, and non-damage to corneal collagen function after SAXS analysis was observed.

We report the in vivo regulation of Inosine-5´-monophosphate dehydrogenase 1 (IMPDH1) in the retina. IMPDH1 catalyzes the rate-limiting step in the de novo synthesis of guanine nucleotides, impacting the cellular pools of GMP, GDP and GTP. Guanine nucleotide homeostasis is central to photoreceptor cells, where cGMP is the signal transducing molecule in the light response. Mutations in IMPDH1 lead to inherited blindness. We unveil a light-dependent phosphorylation of retinal IMPDH1 at Thr159/Ser160 in the Bateman domain that desensitizes the enzyme to allosteric inhibition by GDP/GTP. When exposed to bright light, living mice increase the rate of GTP and ATP synthesis in their retinas; concomitant with IMPDH1 aggregate formation at the outer segment layer. Inhibiting IMPDH activity in living mice delays rod mass recovery. We unveil a novel mechanism of regulation of IMPDH1 in vivo, important for understanding GTP homeostasis in the retina and the pathogenesis of adRP10 IMPDH1 mutations.

The regenerative capability of peripheral nerves is very limited, and several strategies have been proposed to increase nerve regeneration. In the present work, we have analyzed the in vivo usefulness of a novel nanostructured fibrin-agarose bio-artificial nerve substitute (Nano) used alone or in combination with NeuraGen® collagen type I conduits (Coll-Nano) in laboratory rats with a 10-mm sciatic nerve defect. Control animals were subjected to the gold-standard autograft technique (Auto). Results first demonstrated that the percentage of self-amputations was lower in Nano and Coll-Nano groups as compared to the Auto group. Neurotrophic ulcers were more abundant in the Auto group (60%, with 66.6% of them being >2-mm) than Nano and Coll-Nano groups (0%) at 4 weeks, although Nano showed more ulcers after 12 weeks. Foot length was significantly altered in Auto animals due to neurogenic retraction, but not in Nano and Coll-Nano groups after 12 weeks. At the functional level, all animals showed a partial sensory recovery as determined by the pinch test, especially in Nano and Auto groups, but did not reach the levels of native animals. Toe-spread test revealed a partial motor function recovery only in Nano animals at 4 weeks and Auto and Nano at 12 weeks. Electromyography showed clear denervation signs in all experimental groups, with few differences between Auto and Nano animals. After 12 weeks, an important denervation decrease and an increase of the reinnervation process was found in Auto and Nano groups, with no differences between these groups. Histological analyses demonstrated an active peripheral nerve regeneration process with newly formed peripheral nerve fascicles showing S-100, GAP-43 and myelin in all experimental groups. The peripheral nerve regeneration process was more abundant in Auto group, followed by Nano group, and both were better than Coll-Nano group. Muscle histology confirmed the electromyography results and showed some atrophy and fibrosis signs and an important weight and volume loss in all groups, especially in the Coll-Nano group (56.8% weight and 60.4% volume loss). All these results suggest that the novel Nano substitutes used in in vivo were able to contribute to bridge a 10-mm peripheral nerve defect in rats.

A simple and accurate reversed phase high-performance liquid chromatography coupled with diode array detector (HPLC-DAD) method for simultaneously determining and quantifying the antioxidants carotenes, xanthophylls, and retinol in human plasma is presented in this paper. Compounds were extracted with hexane, a C30 column, and a mobile phase of methanol, methyl tert-butyl ether, and water were used for the separation of the compounds. A total of 8 carotenoids, 3 Z-ß-carotene isomers, and 1 fat-soluble vitamin (retinol) were resolved within 72 min at a flow rate of 0.6 mL/min. Detection was achieved at 450 nm for carotenoids and 330 nm for retinol. To evaluate the effectiveness of themethod, it has been applied to an intervention study conducted on eight volunteers. Results. Limits of detection were between 0.1 µg/mL for lycopene and astaxanthin and 1.3 µg/mL for 15-Z-ß-carotene. Recoveries were ranged between 89% and 113% for α-carotene and astaxanthin, respectively. Accuracy was between 90.7% and 112.2% and precision was between 1% and 15% RSD. In human plasma samples compounds studied were identified besides three lycopene isomers, demonstrated to be suitable for application in dietary intervention studies. Conclusions. Due to its accuracy, precision, selectivity, and reproducibility, this method is suitable to dietary habits and/or antioxidants status studies.

BACKGROUND: Carotenoids are important antioxidant compounds in the human diet. Owing to their constrained polyene structure, carotenoids can exist in cis and trans isomeric forms that can be difficult to differentiate in natural samples. In ketchups and gazpachos, all-trans isomeric forms are predominant. However, during thermal processing, cis isomers are formed. Therefore the determination of carotenoid isomers is required for the assessment of the nutritional value of foods. RESULTS: The main carotenoid found in ketchups was trans-lycopene, whereas in gazpachos a higher contribution to the total carotenoid content was made by other carotenoids such as 5-, 9- and 13-cis-lycopene, lutein, ß-carotene and α-carotene. Ketchups exhibited the highest lipophilic antioxidant content owing to their higher content of trans-lycopene, the main carotenoid in tomatoes. CONCLUSION: Direct analysis of carotenoids by electrospray ionisation mass spectrometry (ESI-MS) often produces poor results requiring offline time- and sample-consuming derivatisation techniques. Therefore in this work a simple ESI-MS approach is described for the direct analysis of carotenoids in ketchups and gazpachos using the post-column addition of lithium chloride to promote the cationisation of carotenoids. To the best of the authors' knowledge, this paper presents for the first time the identification of 5-, 9- and 13-cis-lycopene in ketchups and gazpachos.
