Newcastle disease virus (NDV) strains, while falling under a single serotype, are classified into distinct genotypes. Genotype VII virulent NDVs pose a significant threat to poultry due to their association with high mortality rates and economic losses. This study aimed to evaluate the efficacy of three commercial live vaccines based on genotype II against genotype VII virulent NDV (vNDV) in specific pathogen-free (SPF) chickens. Forty one-day-old chickens were randomly divided into four groups (n = 10) and inoculated with one dose of each ND pneumotropic vaccine-B1, Clone.12IR, and La Sota-or received phosphate-buffered saline (PBS) as a control at eight days of age via eye drop. At 28 days of age (20th post-vaccination days), chickens were intramuscularly challenged with genotype VII virulent NDV (≥ 105 LD50). Serum samples were collected at 28 days of age (challenge day), 7 and 14 post-challenge days to measure NDV antibodies via the hemagglutination inhibition (HI) test. Cloacal and oropharyngeal swabs were taken on the 3rd, 5th, 7th, and 10th post-challenge days to evaluate virus shedding. Vaccinated groups exhibited significantly higher antibody titers and greater protection levels compared to the control group (P≤ 0.001). While HI antibody titer was not different at 28 and 35 days of age between vaccinated chickens, the Clone.12IR groups showed higher HI antibody titer compared to B1 at day 42 of age (9.43 vs. 7.42; P≤ 0.002). La Sota and Clone.12IR vaccines demonstrated superior protection against mortality compared to the B1 vaccine (90 %, 80% vs. 60 %, respectively) with 6.0 and 2.67 odds ratio of survivability. All three mismatched vaccines effectively curbed the shedding of virulent genotype VII NDV, with 0 % to 11 % positive cloacal samples up to the 3rd post-challenge day. These findings demonstrate that in the experimental setting, the administration of mismatched ND vaccines, particularly La Sota and Clone.12IR, confer protection against genotype VII virulent NDV and control viral shedding, which can help to develop effective vaccination strategies to mitigate the impact of vNDV outbreaks in the poultry farms.

BACKGROUND: The presence of antimicrobial drugs residues in animal products at levels higher than the maximum residue level (MRL) may have adverse effects on consumer health such as allergic reactions and resistance development. Therefore, it is necessary to monitor animal products for the presence of antimicrobial residues. OBJECTIVES: The aim of this study was to evaluate the detection limit of microbial inhibition assay (MIA) in microplate by using of Bacillus licheniformis as indicator microorganism for two antibiotics, enrofloxacin (ENR) and sulfamethazine (SMT), in broiler chicken's kidney, liver and muscle tissue samples. METHODS: Spiked tissues samples for the two antibiotics were analysed separately by this method. The results of the assay were evaluated by the determination of the absorbance after mean 3.47 h of incubation at 45°C. RESULTS: Results showed that the detection limits of MIA for ENR and SMT in kidney (124.03 and 23.21 µg/kg, respectively) and liver (90.02 and 62.03 µg/kg, respectively) as well as SMT in muscle (46.95 µg/kg) were lower than EU (European Union) - MRL, whereas the detection limit for ENR in muscle was slightly higher than MRL (136.3 µg/kg compared to 100 µg/kg MRL). Furthermore, the MIA in the current study was found to be more sensitive to SMT than ENR (92% and 88% sensitivity rate, respectively). No false-positive was observed in the assay. CONCLUSIONS: Based on the results, the MIA investigated in this study had the potential to detect ENR and SMT residues in broiler chicken kidney, liver and muscle tissues at levels below or close to EU - MRL but offered lower capability for the detection of ENR compared with SMT in kidney and muscle tissue samples.

BACKGROUND: Evaluating the prevalence of vancomycin resistance genes (van genes) in enterococcal isolates from food-producing animals is an important public health issue because of the possibility of resistance genes spread to human. OBJECTIVES: The present study aimed to determine the occurrence of vancomycin resistance genes among Enterococcus species obtained from ostrich faecal samples. METHODS: One hundred and twenty-five faecal samples of apparently healthy ostriches from five different farms were investigated. Genes encoding vancomycin resistance were studied by multiplex-PCR, and susceptibility to six antibiotics was evaluated by disk-diffusion method. RESULTS: In total, 107 Enterococcus spp. isolates were obtained and confirmed by biochemical and molecular tests. Enterococcus faecium was the prevailing species (56 isolates of 107; 52.3%), followed by E. hirae (24 isolates; 22.4%) and E. gallinarum (12 isolates; 11.2%). Of the 107 recovered isolates, 44% harboured at least a type of van genes. vanA, vanC2/3 and vanC1 were identified in 34 (31.7%), 13 isolates (12.1%) and 4 (3.7%) isolates respectively. Additionally, four isolates (E. gallinarum, E. rafinosus) co-harboured the the vanA and vanC1 or vanA and vanC2/3. Enterococcus faecium and Enterococcus hirae strains with the vanA genotype were the most frequent van-carrying enterococci from ostrich faecal samples. Among van-carrying enterococcal isolates, 23.4% were phenotypically resistant to vancomycin. This study revealed a relatively high prevalence (44%) of van-carrying enterococci in ostrich faecal samples. CONCLUSIONS: Results of the present study suggest that ostrich faeces could be considered as a reservoir of vancomycin resistance genes, especially vanA containing enterococci that could be potentially transferred to human through the food chain.

OBJECTIVE: Spirulina has been recognized formerly as a filamentous spiral-shaped blue-green algae but more recently as a genus of photosynthetic bacteria (Arthrospira). This microorganism is considered as a rich source of essential nutrients for human and animals. The present study was conducted to determine potential application of Spirulina for heat-exposed broilers. METHODS: Two hundred and fifty Cobb 500 chicks with male to female in equal ratio with average initial weight of 615.6 g at 17 days of age were divided into 5 treatments with 5 replicates of 10 chicks. Treatment groups were as follows: positive and negative controls with 0% Spirulina supplement and three Spirulina receiving groups with 5 g/kg (0.5%), 10 g/kg (1%), and 20 g/kg (2%) supplementation. Spirulina receiving groups as well as positive control were exposed to high ambient temperature at 36°C for 6 h/d from 38 to 44 days of age. Biochemical variables were measured in serum samples at 35, 38, 42, and 45 days of broiler chickens age. RESULTS: The results showed that supplementation of the diet with Spirulina decreased concentration of stress hormone and some serum lipid parameters while enhanced humoral immunity response and elevated antioxidant status whereas it didn't meaningfully affect performance characteristics. Nevertheless, feed conversion ratio was improved numerically but not statistically in broilers fed with 1% Spirulina under high ambient temperature. CONCLUSION: Overall, the present study suggests that alleviation of adverse impacts due to high ambient temperature at biochemical level including impaired enzymatic antioxidant system, elevated stress hormone and lipid profile can be approached in broiler chickens through supplementation of the diet with Spirulina platensis.
