RESUMO

This study examined the relationships of sociocultural body ideal internalization, difficulties in emotion regulation, and their interaction with body image concerns in a group of 423 Canadian adolescent boys. Participants were 12 to 17 years old and completed multiple self-reported questionnaires. Regression analyses and latent moderation structural equations were performed. Both variables were positively correlated with body dissatisfaction, drive for muscularity, restrained eating, and emotional eating. The internalization of men's sociocultural body ideal moderated the relationships between multiple difficulties in emotion regulation and some body image concerns. These results are in line with findings of other studies among adult men and women, and dominant theories of body image concerns, which have been derived especially from women populations.

The Step Pyramid of Djoser in Saqqara, Egypt, is considered the oldest of the seven monumental pyramids built about 4,500 years ago. From transdisciplinary analysis, it was discovered that a hydraulic lift may have been used to build the pyramid. Based on our mapping of the nearby watersheds, we show that one of the unexplained massive Saqqara structures, the Gisr el-Mudir enclosure, has the features of a check dam with the intent to trap sediment and water. The topography beyond the dam suggests a possible ephemeral lake west of the Djoser complex and water flow inside the 'Dry Moat' surrounding it. In the southern section of the moat, we show that the monumental linear rock-cut structure consisting of successive, deep compartments combines the technical requirements of a water treatment facility: a settling basin, a retention basin, and a purification system. Together, the Gisr el-Mudir and the Dry Moat's inner south section work as a unified hydraulic system that improves water quality and regulates flow for practical purposes and human needs. Finally, we identified that the Step Pyramid's internal architecture is consistent with a hydraulic elevation mechanism never reported before. The ancient architects may have raised the stones from the pyramid centre in a volcano fashion using the sediment-free water from the Dry Moat's south section. Ancient Egyptians are famous for their pioneering and mastery of hydraulics through canals for irrigation purposes and barges to transport huge stones. This work opens a new line of research: the use of hydraulic force to erect the massive structures built by Pharaohs.

Amino acid (AA) transporters (AAT) control AA cellular fluxes across membranes, contributing to maintain cellular homeostasis. In this study, we took advantage of rainbow trout metabolic feature, which highly relies on dietary AA, to explore the cellular and physiological consequences of unbalanced diets on AAT dysregulations with a particular focus on cationic AAs (CAA), frequently underrepresented in plant-based diets. Results evidenced that 24 different CAAT are expressed in various trout tissues, part of which being subjected to AA- and CAA-dependent regulations, with y+LAT2 exchanger being prone to the strongest dysregulations. Moreover, CAA were shown to control two major AA-dependent activation pathways (namely mTOR and GCN2) but at different strength according to the CAA considered. A new feed formulation strategy has been put forward to improve specifically the CAA supplemented absorption in fish together with their growth performance. Such "precision formulation" strategy reveals high potential for nutrition practices, especially in aquaculture.

Despite numerous studies on Himalayan erosion, it is not known how the very high Himalayan peaks erode. Although valley floors are efficiently eroded by glaciers, the intensity of periglacial processes, which erode the headwalls extending from glacial cirques to crest lines, seems to decrease sharply with altitude1,2. This contrast suggests that erosion is muted and much lower than regional rock uplift rates for the highest Himalayan peaks, raising questions about their long-term evolution3,4. Here we report geological evidence for a giant rockslide that occurred around 1190 AD in the Annapurna massif (central Nepal), involving a total rock volume of about 23 km3. This event collapsed a palaeo-summit, probably culminating above 8,000 m in altitude. Our data suggest that a mode of high-altitude erosion could be mega-rockslides, leading to the sudden reduction of ridge-crest elevation by several hundred metres and ultimately preventing the disproportionate growth of the Himalayan peaks. This erosion mode, associated with steep slopes and high relief, arises from a greater mechanical strength of the peak substratum, probably because of the presence of permafrost at high altitude. Giant rockslides also have implications for landscape evolution and natural hazards: the massive supply of finely crushed sediments can fill valleys more than 150 km farther downstream and overwhelm the sediment load in Himalayan rivers for a century or more.

Thermococcales, a major order of hyperthermophilic archaea inhabiting iron- and sulfur-rich anaerobic parts of hydrothermal deep-sea vents, are known to induce the formation of iron phosphates, greigite (Fe3S4) and abundant quantities of pyrite (FeS2), including pyrite spherules. In the present study, we report the characterization of the sulfide and phosphate minerals produced in the presence of Thermococcales using X-ray diffraction, synchrotron-based X ray absorption spectroscopy and scanning and transmission electron microscopies. Mixed valence Fe(II)-Fe(III) phosphates are interpreted as resulting from the activity of Thermococcales controlling phosphorus-iron-sulfur dynamics. The pyrite spherules (absent in abiotic control) consist of an assemblage of ultra-small nanocrystals of a few ten nanometers in size, showing coherently diffracting domain sizes of few nanometers. The production of these spherules occurs via a sulfur redox swing from S0 to S-2 and then to S-1, involving a comproportionation of (-II) and (0) oxidation states of sulfur, as supported by S-XANES data. Importantly, these pyrite spherules sequester biogenic organic compounds in small but detectable quantities, possibly making them good biosignatures to be searched for in extreme environments.

Microbial processes can be involved in the remobilization of uranium (U) from reduced sediments under O2 reoxidation events such as water table fluctuations. Such reactions could be typically encountered after U-bearing sediment dredging operations. Solid U(IV) species may thus reoxidize into U(VI) that can be released in pore waters in the form of aqueous complexes with organic and inorganic ligands. Non-uraninite U(IV) species may be especially sensitive to reoxidation and remobilization processes. Nevertheless, little is known regarding the effect of microbially mediated processes on the behaviour of U under these conditions.

Coastal sediments downstream of ultramafic catchments can show Ni and Cr concentration well above sediment quality guidelines. Despite their potential ecological impact, the bioavailability of these trace metals in such sedimentary settings has been poorly investigated. In this study, we tried to fill this gap by performing kinetic EDTA-extractions across a shore-to-reef gradient in lagoon sediments downstream of an ultramafic catchment in New Caledonia and interpreting the results in regard of synchrotron-derived speciation. Measured bioavailability ranged from very low for Cr (below 1% of total Cr) to medium for Ni (below 5% of total Ni). Both trace metals showed a decreasing shore-to-reef bioavailability gradient reflecting the larger deposition of ultramafic sediments close to the shore. According to synchrotron-derived speciation data, the very low bioavailability of Cr is attributed to its major occurrence as Cr(III)-bearing Fe-(oxyhydr)oxides and phyllosilicates, with no evidence of Cr(VI). Considering the low occurrence of Fe-sulfides, the medium bioavailability of Ni is considered to arise mainly from the reductive dissolution of Ni-bearing Fe-(oxyhydr)oxides during early diagenesis. This reaction also explains the medium bioavailability of Fe (up to 15% of total Fe) and the positive correlation observed with Total Organic Carbon (TOC). In this regard, this latter parameter appears as a major driver of Ni and Fe bioavailability in coastal sediments downstream of ultramafic catchments. On the opposite, in the absence of Mn-oxides, TOC has no influence on Mn bioavailability (up to 30% of total Mn) that appears more likely driven by sediment sources. From an ecological point of view, considering the Australian and New-Zealand High Interim Sediment Quality Guidelines (ANZ-ISQG-H), Cr should not represent a significant risk towards benthic communities in coastal sediments downstream of ultramafic catchments. On the opposite, Ni, Fe and Mn might represent an ecological risk that should be further investigated in such sedimentary settings.

The replacement of fishmeal by plant proteins in aquafeeds imposes the use of synthetic methionine (MET) sources to balance the amino acid composition of alternative diets and so to meet the metabolic needs of fish of agronomic interest such as rainbow trout (RT-Oncorhynchus mykiss). Nonetheless, debates still exist to determine if one MET source is more efficiently used than another by fish. To address this question, the use of fish cell lines appeared a convenient strategy, since it allowed to perfectly control cell growing conditions notably by fully depleting MET from the media and studying which MET source is capable to restore cell growth/proliferation and metabolism when supplemented back. Thus, results of cell proliferation assays, Western blots, RT-qPCR and liquid chromatography analyses from two RT liver-derived cell lines revealed a better absorption and metabolization of DL-MET than DL-Methionine Hydroxy Analog (MHA) with the activation of the mechanistic Target Of Rapamycin (mTOR) pathway for DL-MET and the activation of integrated stress response (ISR) pathway for MHA. Altogether, the results clearly allow to conclude that both synthetic MET sources are not biologically equivalent, suggesting similar in vivo effects in RT liver and, therefore, questioning the MHA efficiencies in other RT tissues.

BACKGROUND: Most very premature newborns (<32 weeks of gestation) receive parenteral nutrition (PN) that is inherently contaminated with peroxides. Oxidative stress induced by PN is associated with bronchopulmonary dysplasia, a main pathological complication in these infants who have weak antioxidant capacity to detoxify peroxides because of their glutathione deficiency. In animals, glutathione supplementation of PN prevented oxidative stress and alveolar loss (the main characteristic of bronchopulmonary dysplasia). Of its two forms-oxidized glutathione (GSSG) and reduced glutathione (GSH)-GSSG was used because of its better stability. However, a 30% loss of GSSG in PN is observed. The potentially high therapeutic benefits of GSSG supplementation on the health of very premature infants make the study of its stability highly important. METHODS: GSSG was incubated in combination with the following components of PN: dextrose, multivitamins, Primene, and Travasol, and with cysteine, cystine, and peroxides, for 24 h. Total glutathione in these solutions was measured 0-24 h after the addition of GSSG. RESULTS: The combination of cysteine and multivitamins caused the maximum loss of glutathione. The stability of GSSG was not affected by multivitamins. The cysteine was responsible for ∼20% of the loss of GSSG; in the presence of multivitamins, the loss reached >70%. Removing the cysteine prevented the degradation of glutathione. CONCLUSION: GSSG reacts with cysteine to form cysteine-glutathione mixed disulfide, another suitable glutathione substrate for preterm neonates. The study confirms that GSSG added to PN can potentially provide a precursor to de novo synthesis of glutathione in vivo.

Uranium (U) isotopic signatures and concentration in sediments are widely used as paleo-redox proxies, as the behavior of U is often controlled by bottom water oxygenation. Here, we investigated the processes controlling U accumulation in the sediments of Lake Nègre (Mediterranean Alps, South-East France) over the past 9200 years. Exceptionally high natural U concentrations (350-1250 µg·g-1) allowed the measurement of U along with other elements by high-resolution X-Ray Fluorescence core-scanning. Weathering and erosion proxies (Ti content, Zr/Al and K/Ti ratios) indicate that sedimentary inputs were controlled by Holocene climatic variations. After a period of low erosion during the Holocene Climatic Optimum, a major regime shift was recorded at 4.2 kyr BP when terrigenous fluxes consistently increased until present with high sensitivity to centennial-scale climatic events. Sedimentary organic matter (OM) inputs were dominated by terrigenous OM from the catchment soils until 2.4 kyr BP, as attested by carbon to nitrogen (C/N) and bromine to organic carbon (Br/TOC) ratios. From 2.4 kyr BP to present, lake primary production and soils equally contributed to sedimentary OM. Uranium fluxes to the sediments were well correlated to terrigenous OM fluxes from 7 kyr BP to present, showing that U supply to the lake was controlled by U scavenging in the soils of the watershed followed by transport of U bound to detrital organic particles. Higher U/OM ratios before 7 kyr BP likely reflect the development of the upstream wetland. The fluctuations of U sedimentary inputs appear to be independent of bottom water oxygenation, as estimated from constant Fe/Mn ratios and δ238U isotopic signatures, and rather controlled by the production, erosion and sedimentation of terrigenous OM. This finding confirms that the use of U (and potentially other metals with high affinity to OM) concentrations alone should be used with caution for paleo-redox reconstructions.

The long-term fate of uranium-contaminated sediments, especially downstream former mining areas, is a widespread environmental challenge. Essential for their management is the proper understanding of uranium (U) immobilization mechanisms in reducing environments. In particular, the long-term behavior of noncrystalline U(IV) species and their possible evolution to more stable phases in subsurface conditions is poorly documented, which limits our ability to predict U long-term geochemical reactivity. Here, we report direct evidence for the evolution of U speciation over 3,300 y in naturally highly U-enriched sediments (350-760 µg ⋅ g-1 U) from Lake Nègre (Mercantour Massif, Mediterranean Alps, France) by combining U isotopic data (δ238U and (234U/238U)) with U L3 -edge X-ray absorption fine structure spectroscopy. Constant isotopic ratios over the entire sediment core indicate stable U sources and accumulation modes, allowing for determination of the impact of aging on U speciation. We demonstrate that, after sediment deposition, mononuclear U(IV) species associated with organic matter transformed into authigenic polymeric U(IV)-silica species that might have partially converted to a nanocrystalline coffinite (UIVSiO4·nH2O)-like phase. This diagenetic transformation occurred in less than 700 y and is consistent with the high silica availability of sediments in which diatoms are abundant. It also yields consistency with laboratory studies that proposed the formation of colloidal polynuclear U(IV)-silica species, as precursors for coffinite formation. However, the incomplete transformation observed here only slightly reduces the potential lability of U, which could have important implications to evaluate the long-term management of U-contaminated sediments and, by extension, of U-bearing wastes in silica-rich subsurface environments.

Radium is a naturally occurring radioactive element commonly found at low levels in natural systems such as lacustrine or marine sediments. Anthropogenic activities including former uranium mining activities can lead to the dissemination of radium isotopes having high radiological toxicities, which potentially threaten the safety of nearby environments. Although radium mobility in oxidized environments is known to be largely governed by sorption/desorption onto Fe and Mn oxyhydroxides and coprecipitation with sulfate minerals (e.g. barite), little is known regarding its behavior under reducing conditions, which are the conditions typically encountered in organic-rich systems such as wetlands and lake sediments. The present study aims at understanding the behavior of long-lived radium isotopes (226Ra and 228Ra), during early diagenesis of lake sediments contaminated by former uranium mining activities. Solid and pore water concentrations of 226Ra and 228Ra were determined using ultra low background gamma spectrometry, which allowed improvement of detection limits and measurement accuracy. This study shows that the downcore distribution of radium isotopes is closely related to the reductive dissolution of iron and manganese oxyhydroxides below the sediment-water interface. The resulting diffusive fluxes of 226Ra and 228Ra (4.1 10-25 and 4.7 10-28 mol cm-2.s-1) are however significantly lower than other radium-impacted environments, such as uranium mill tailings pond and phosphate industry-impacted sediments, and are similar to those reported for natural marine environments. Hence, in the reduced lake sediments of Saint-Clement, the major fraction of radium is trapped by the solid phase, while early diagenesis only induces a slight mobility of this radioelement.

Nowadays, aquaculture provides more than 50% of fish consumed worldwide but faces new issues that challenge its sustainability. One of them relies on the replacement of fish meal (FM) in aquaculture feeds by other protein sources without deeply affecting the whole organism's homeostasis. Multiple strategies have already been tested using in vivo approaches, but they hardly managed to cope with the multifactorial problems related to the complexities of fish biology together with new feed formulations. In this context, rainbow trout (RT) is particularly concerned by these problems, since, as a carnivorous fish, dietary proteins provide the amino acids required to supply most of its energetic metabolism. Surprisingly, we noticed that in vitro approaches considering RT cell lines as models to study RT amino acid metabolism were never previously used. Therefore, we decided to investigate if, and how, three major pathways described, in other species, to be regulated by amino acid and to control cellular homeostasis were functional in a RT cell line called RTH-149-namely, the mechanistic Target Of Rapamycin (mTOR), autophagy and the general control nonderepressible 2 (GCN2) pathways. Our results not only demonstrated that these three pathways were functional in RTH-149 cells, but they also highlighted some RT specificities with respect to the time response, amino acid dependencies and the activation levels of their downstream targets. Altogether, this article demonstrated, for the first time, that RT cell lines could represent an interesting alternative of in vivo experimentations for the study of fish nutrition-related questions.

Reducing conditions and high organic carbon content make wetlands favorable to uranium (U) sequestration. However, such environments are subjected to water-table fluctuations that could impact the redox behavior of U and its mobility. Our previous study on U speciation in a contaminated wetland has suggested a major role of water-table redox fluctuations in the redistribution of U from U(IV)-phosphate minerals to organic U(VI) and U(IV) mononuclear species. Here, we investigate the mechanisms of these putative processes by mimicking drying or flooding periods via laboratory incubations of wetland samples. LCF-XANES and EXAFS analyses show the total oxidation/reduction of U(IV)/U(VI)-mononuclear species after 20 days of oxic/anoxic incubation, whereas U-phosphate minerals are partly oxidized/reduced. SEM-EDXS combined with µ-XRF and µ-XANES analyses suggest that autunite Ca(UO2)2(PO4)2⋅11H2O is reduced into lermontovite U(PO4)(OH)⋅H2O, whereas oxidized ningyoite CaU(PO4)2⋅2H2O is locally dissolved. The release of U from this latter process is observed to be limited by U(VI) adsorption to the soil matrix and further re-reduction into mononuclear U(IV) upon anoxic cycling. Analysis of incubation waters show, however, that dissolved organic carbon enhances U solubilization even under anoxic conditions. This study brings important information that help to assess the long-term stability of U in seasonally saturated organic-rich contaminated environments.

Stable Pb isotope ratios were measured and compared to U distributions in three soil cores located in a wetland highly impacted by water discharge of a former U-mine. Pb isotope ratios showed notable alignments in binary mixing plots, demonstrating the dissemination of radioactive-enriched material from the U-mine. Thanks to these alignments and to the measurement of the 204Pb isotope, a precise characterization of the Pb isotope composition of the U-ore was performed without the use of U-ore samples. The well-defined end-members with the help of a reevaluated isotope mixing model allowed the accurate determination of the radiogenic Pb percentages in the cores that were overall found to be >50%. Noncorrelated distributions of radiogenic 206Pb and U are observed in several of the wetland soil samples. They reveal postdepositional U redistribution explained by major U speciation changes due to redox cycling in the wetland. On the contrary, the radiogenic 206Pb showed no or little postdepositional mobility and thus can be considered to be a memory tracer of the dissemination of U-rich radioactive material: even after an important U loss, the radiogenic 206Pb is able to reveal past contamination by U-rich materials.

Peroxides contaminating parenteral nutrition (PN) limit the use of methionine as a precursor of cysteine. Thus, PN causes a cysteine deficiency, characterized by low levels of glutathione, the main molecule used in peroxide detoxification, and limited growth in individuals receiving long-term PN compared to the average population. We hypothesize that glutathione supplementation in PN can be used as a pro-cysteine that improves glutathione levels and protein synthesis and reduces oxidative stress caused by PN. One-month-old guinea pigs (7-8 per group) were used to compare glutathione-enriched to a non-enriched PN, animals on enteral nutrition were used as a reference. PN: Dextrose, amino acids (Primene), lipid emulsion (Intralipid), multivitamins, electrolytes; five-day infusion. Glutathione (GSH, GSSG, redox potential) and the incorporation of radioactive leucine into the protein fraction (protein synthesis index) were measured in the blood, lungs, liver, and gastrocnemius muscle. Data were analysed by ANOVA; p < 0.05 was considered significant. The addition of glutathione to PN prevented the PN-induced oxidative stress in the lungs and muscles and supported protein synthesis in liver and muscles. The results potentially support the recommendation to add glutathione to the PN and demonstrate that glutathione could act as a biologically available cysteine precursor.

In the tropics, continental weathering and erosion are major sources of trace metals towards estuaries and lagoons, where early diagenesis of sediments may influence their mobility and bioavailability. Determining trace metals speciation in tropical sedimentary settings is thus needed to assess their long-term fate and potential threat to fragile coastal ecosystems. In this study, we determined Fe, Ni and S speciation across a shore-to-reef gradient in sediments from the New Caledonia lagoon that receive mixed contribution from lateritic (iron-oxyhydroxides and clay minerals), volcano-sedimentary (silicates) and marine (carbonate) sources. Sulfur K-edge XANES data indicated a major contribution of pyrite (FeS2) to S speciation close to the shore. However, this contribution was found to dramatically decrease across the shore-to-reef gradient, S mainly occurring as sulfate close to the coral reef. In contrast, Fe and Ni K-edge XANES and EXAFS data indicated a minor contribution of pyrite to Fe and Ni speciation, and this contribution could be evidenced only close to the shore. The major fractions of Fe and Ni across the shore-to-reef gradient were found to occur as Ni- and Fe-bearing clay minerals consisting of smectite (~nontronite), glauconite and two types of serpentines (chrysotile and greenalite/berthierine). Among these clay minerals, greenalite/berthierine, glauconite and possibly smectite, were considered as authigenic. The low contribution of pyrite to trace metals speciation compared to clay minerals is interpreted as a result of (1) a reduced formation rate due to the low amounts of organic carbon compared to the Fe pool and (2) repeated re-oxidation events upon re-suspension of the sediments top layers due to the specific context of shallow lagoon waters. This study thus suggests that green clay authigenesis could represent a key process in the biogeochemical cycling of trace metals that are delivered to lagoon ecosystems upon continental erosion and weathering.

Peat layers within alluvial sediments are considered effective arsenic (As) sinks under reducing conditions due to the binding of As(III) to thiol groups in natural organic matter (NOM) and the formation of As-bearing sulfide phases. However, their possible role as sources of As for anoxic groundwaters remains unexplored. Here, we perform laboratory experiments to provide evidence for the role of a sediment peat layer in releasing As. Our results show that the peat layer, deposited about 8,000 years ago in a paleomangrove environment in the nascent Mekong Delta, could be a source of As to porewater under reducing conditions. X-ray absorption spectroscopy (XAS) analysis of the peat confirmed that As was bound to NOM thiol groups and incorporated into pyrite. Nitrate was detected in peat layer porewater, and flow-through and batch experiments evidenced the release of As from NOM and pyrite in the presence of nitrate. Based on poisoning experiments, we propose that the microbially mediated oxidation of arsenic-rich pyrite and organic matter coupled to nitrate reduction releases arsenic from this peat. Although peat layers have been proposed as As sinks in earlier studies, we show here their potential to release depositional- and/or diagenetically-accumulated As.

Wetlands have been proposed to naturally attenuate U transfers in the environment via U complexation by organic matter and potential U reduction. However, U mobility may depend on the identity of particulate/dissolved uranium source materials and their redox sensitivity. Here, we examined the fate of uranium in a highly contaminated wetland (up to 4500 mg·kg-1 U) impacted by former mine water discharges. Bulk U LIII-EXAFS and (micro-)XANES combined with SEM-EDXS analyses of undisturbed soil cores show a sharp U redox boundary at the water table, together with a major U redistribution from U(IV)-minerals to U(VI)-organic matter complexes. Above the water table, U is fully oxidized into mono- and bidentate U(VI)-carboxyl and monodentate U(VI)-phosphoryl complexes. Minute amounts of U(VI)-phosphate minerals are also observed. Below the water table, U is fully reduced and is partitioned between U(IV)-phosphate minerals (i.e., ningyoite and a lermontovite-like phase), and bidentate U(IV)-phosphoryl and monodentate U(IV)-carboxyl complexes. Such a U redistribution from U-minerals inherited from mine water discharge deposits could result from redox cycling nearby the water table fluctuation zone. Oxidative dissolution of U(IV)-phosphate minerals could have led to U(VI)-organic matter complexation, followed by subsequent reduction into U(IV)-organic complexes. However, uranium(IV) minerals could have been preserved in permanently waterlogged soil.
