BACKGROUND: The roles of IL-10, IL-11, COX-2, BCL6, ZEB1, and ZEB2 genes in the potential correlation between polycystic ovary syndrome (PCOS), inflammation, and cancer remain controversial. AIMS: This study aimed to compare serum levels of IL-10 and IL-11 and gene expression of IL-10, IL-11, COX-2, BCL6, ZEB1, and ZEB2 in PBMCs of women with PCOS and healthy controls. METHODS: A case-control study included 40 women with PCOS as the case group and 40 healthy women as controls. Group matching for age and BMI was performed. Serum levels of IL-10 and IL-11 were assessed using ELISA, while gene expression was measured using real-time PCR. Parameters were compared between groups, and correlations among gene expression and serum levels were explored. RESULTS: In comparison to healthy women, women with PCOS exhibited a significant decrease in the expression of COX-2 and IL-10 genes (p<0.001), alongside a significant increase in ZEB2 gene expression (p<0.001). There were no significant differences observed in the expression of IL-11, BCL6, and ZEB1 genes. Furthermore, the serum level of IL-10 was significantly lower in women with PCOS compared to the control group (p<0.001), while no significant difference was found in IL-11 levels. Additionally, no significant correlations were identified between gene expression and serum levels. CONCLUSION: In women with PCOS, reduced IL-10 gene expression may indicate inflammation and serve as a diagnostic biomarker. However, conflicting findings on COX-2 expression complicate understanding. Elevated ZEB2 expression in PCOS women may lead to infertility, epithelial-mesenchymal transition, and aggressive phenotypes.

BACKGROUND: The present study aimed to evaluate the flexural strength, degree of conversion, and demineralization-prevention ability of an experimental fissure sealant containing nano-calcium-phosphate compounds. METHODS: An experimental sealant was formulated using silica and nano hydroxyapatite filler particles. The control group consisted of the DENU Seal (n = 10, each group). The flexural bond strength was evaluated by UTM. DC was evaluated by FTIR. To evaluate the demineralization-prevention ability, Cl V cavities in 10 third molar teeth restored with two sealant products, followed by an acid challenge then the Vickers microhardness test was carried out. RESULTS: The mean flexural strength in the commercial group was higher than the experimental group. However, the mean flexural modulus was not significantly different between the two groups. In the experimental group, DC was significantly higher than the commercial group. Adjacent to the interface, the decrease in microhardness in the experimental group was significantly less than the commercial group. However, on the tooth surface, there were no significant differences between the two groups. In the experimental group, the decrease in microhardness at the interface was less than at the tooth surface, however the situation was opposite in the commercial group. CONCLUSIONS: Incorporating hydroxyapatite into the sealant structure might prevent demineralization, without adverse effects on flexural modulus and degree of conversion.

The aim of the present study was to assess the positive predictive value (PPV) of panoramic radiographic signs in the assessment of the relationship between impacted mandibular third molars (IMTMs) and the mandibular canal (MC). This cross-sectional study was conducted by reviewing 102 cone-beam computed tomography (CBCT) and panoramic radiographs of patients with IMTMs and radiographic signs of the contact of the IMTMs with the MC on panoramic radiographs (i.e., root apex darkening and interference with the white line). A positive relationship of the IMTM roots with the MC based on CBCT findings was recorded as the gold standard. The PPV of panoramic radiographic signs was calculated for the detection of the relationship of the IMTM root with the MC. The IMTMs were in contact with the MC on CBCT scans in 90.1% of the cases. The PPV of root apex darkening and the interference with the white line was found to be 89.09% (95% CI: (77.75, 95.88)) and 91.48% (95% CI: (79.62, 97.63)), respectively. The MC had a buccal position in 63.7%, and a lingual position in 35.2%, of the cases. The contact of IMTMs with the MC was more commonly seen in patients with a lingual position (100% of the samples). The IMTM root apex darkening and interference with the white line of the MC on panoramic radiographs had a high PPV for determination of the contact of IMTMs with the MC. Thus, presence of the above-mentioned risk factors indicates the need for subsequent 3D radiographic assessments.

BACKGROUND: Endometrial receptivity is one of the important factors in assisted reproductive technology (ART) success. In the luteal phase of an ART cycle, serum estradiol (E2) and progesterone are often placed in low levels. Supporting the luteal phase with progesterone is a usual method. OBJECTIVE: To evaluate the effects of E2 supplementation plus progesterone on the luteal phase support in the antagonist protocol who have undergone intracytoplasmic sperm injection-embryo transfer cycles. MATERIALS AND METHODS: In this cross-sectional study, 200 patients with antagonist stimulation protocol, who had undergone intracytoplasmic sperm injection treatment, were divided into two groups based on the use of E2 supplementation. In both groups, 400 mg progesterone suppositories (CyclogestⓇ), twice a day/vaginally, was administered starting from the day of oocyte collection until the fetal heart activity. However, in the E2 group, in addition to progesterone, 4 mg tablet of E2 was received daily. Beta hCG was checked 14 days after the embryo transfer, and the clinical pregnancy rate was the main endpoint. RESULTS: The patients' characteristics were matched, and insignificant differences were observed, except for endometrial thickness. The clinical outcomes showed the rate of pregnancy was higher in the E2 group compared to the control group; nonetheless, statistically, there was no noticeable difference. CONCLUSION: E2 supplementation had no beneficial effect in the luteal phase support of IVF cycles. Nevertheless, more studies are required to confirm the supportive role of E2 supplementation for embryo implantation and to improve the outcomes in ART cycles.

Trachyspermum ammi is an important medicinal plant that contains a bioactive compound namely thymol. In the study, T. ammi was transformed by Agrobacterium rhizogenes strains. Seedling stem explants were inoculated with A. rhizogenes strains A4, LBA 9402, ATCC 15834, and the effect of different co-cultivation media along with incorporation of acetosyringone (100 µM) was evaluated comparatively on the frequency of hairy root induction. The polymerase chain reaction using rolB and virD specific primers was served to confirm the putative transformed hairy roots. All strains established hairy root with various frequencies, among which strain ATCC 15834 was significantly the most efficient strain for hairy root induction (84.3%). Half-strength B5 medium and incorporation of acetosiryngone (100 µM) were also significantly optimal for hairy root induction. Hairy roots culture induced by ATCC 15834 using half-strength B5 liquid medium supplemented with 30 g L-1 sucrose indicated the highest accumulation of biomass (99.05 g L-1 FW and 10.95 g L-1 DW) and thymol content (11.30 mg g-1 DM) at 20 days. Nearly 4.9-fold and 5.3-fold increment of biomass and thymol accumulation was observed, respectively, at 20 days in comparison with the untransformed control roots. The results showed the high potential of T. ammi hairy roots for the biosynthesis of thymol.

Cytokines are the main factors involved in the normal functions of the placenta and delivery process. The aim of this project was to compare serum levels of interleukin-8 (IL-8), IL-6, tumour necrosis factor α (TNF-α) and transforming growth factor ß (TGF-ß) in term and prolonged-pregnancy mothers and their neonates. This study was performed on 240 participants including 60 term and prolonged-pregnancy neonates and their corresponding mothers. Serum levels of IL-8, IL-6, TNF-α and TGF-ß were evaluated by the enzyme-linked immunosorbent assay technique. The results revealed that IL-8 serum levels were significantly lower in the prolonged-pregnancy mothers and their neonates when compared with term mothers and their neonates. Data analysis also revealed a negative correlation between TGF-ß and age of prolonged-pregnancy mothers. A poor positive correlation between IL-6 and head circumference of term neonates was also observed. IL-8 may play crucial roles in the process of on-time delivery and age may significantly affect TGF-ß production in prolonged-pregnancy mothers. Pro-inflammatory cytokines, such as IL-6, can also be considered as main factors involved in fetal growth.

Imbalanced immune responses against fetus alloantigens can lead to abnormality in pregnancy. Interleukin-10 (IL-10) plays key roles in regulation of immune responses against self and foreign antigens to induce tolerance to these antigens. Therefore, alteration in expression of IL-10 during pregnancy may result in several pathologic conditions such as preterm labor. IL-10 leads to a normal pregnancy via several molecular mechanisms including development of tolerogenic dendritic cells, T regulatory lymphocytes and activation of the JAK1/STAT3 pathway in the target cells. This review has collected recent data regarding the status of IL-10 expression during term and preterm deliveries and also its molecular mechanisms that lead to a normal pregnancy.

BACKGROUND: Gestational diabetes mellitus (GDM) is common during pregnancy. This survey was designed based on the frequency of GDM among an urban population according to the American Diabetes Association (ADA) and the International Association of Diabetes and Pregnancy Study Groups (IADPSG) criteria. METHODS: We included all pregnant women who were admitted to a gynecology clinic from September 2012 until May 2013. The fasting blood sugar (FBS) was measured. Those having FBS≥ 126 mg/dl were excluded from the study. All women underwent a standard OGTT (oral glucose tolerance test) by ingesting 75g of glucose in the 24th to 32nd week of their pregnancy. RESULTS: Two hundred ninety pregnant women with a mean±SD age of 27.72±5.091 years were included in the study. The mean±SD FBS, blood glucose one hour and two hours after ingesting 75g of glucose were 82.48±9.41, 146.86±34.22 and 114.21±27.79 mg/ dl, respectively. Based on the criteria of the ADA, 9.3% (n= 27) of the admitted patients suffered from GDM. For the IADPSG and the WHO, those numbers were 31% (n= 90) and 15.2% (n= 44), respectively. CONCLUSION: The prevalence of GDM was 1.5-times and 3 times higher when the IADPSG based data were compared to those of the WHO or the ADA.

The kinetics of the reaction of ozone with hydrogen sulfide was studied theoretically. High-level ab initio calculations were carried out to build the potential energy surface. The mechanism of the title reaction was found to be much more complicated than what is reported in the literature to date. According to our results, six different chemically activated intermediates are involved along the proposed mechanism on its lowest singlet potential energy surface that play an important role in the kinetics of this system. Multichannel RRKM-TST and CVT calculations have been carried out to compute the temperature dependence of the individual rate constants for different channels and also the overall rate constant for the consumption of the reactants. The major products are sulfur dioxide and water at lower temperatures, in good agreement with experimental reports, while at higher temperatures, formation of the other products like O2, H2SO, and radicals like cis/trans-HOSO, SH, HO3, and OH also become important.

The objective was to investigate the effect two modes of labor (vaginal delivery and elective cesarean section) on thiobarbituric reactive substances (TBARS) as markers of lipid peroxidation, total antioxidant power (ferric reducing ability, TAP), and total thiol molecules (TTM) in blood of mothers and their newborns. One hundred and twenty women with normal pregnancy and normal blood biochemical parameters were divided into groups of vaginal delivery (VD) and elective cesarean surgery (ECS). Blood samples were obtained firstly in the time 37-40 weeks of pregnancy and secondly during labor phase for VD or during ECS. Blood samples from umbilical cord arterial of newborns were also obtained at birth after separation of cord. Plasma levels of TBARS, TAP, and TTM were determined. There was no significant differences between VD and ECS mothers before labor in plasma levels of TBARS, TTM, and TAP. Mothers in the VD group showed a significant increase in TBARS (p < 0.05) after delivery. And TTM level showed a significant increase in ECS group (p < 0.05) as compared to pre-delivery levels. Comparing oxidative stress variables between VD and ECS groups after labor, plasma levels of TBARS, and TTM significantly increased (p < 0.05) in VD mothers. TAP was not significantly different between VD and ECS groups. Newborn of VD mothers showed a significant increase in TBARS (p < 0.05), and TAP (p < 0.05) as compared to newborn of ECS. TTM was not significantly different between two groups of newborns. The results indicate that mothers in VD and their newborns are in more oxidative stress than those who underwent ECS for delivery. Linking oxidative stress to severe neonatal diseases, it may be reasonable to assess whether antioxidant supplementation during pregnancy may reduce the frequency of neonatal diseases.
