RESUMO

Frequency and spectrum of chromosomal aberrations were studied in hemopoietic cells of Wistar rats myeloid leukaemia induced by the prolonged exposure to tritium oxide. Differences in quantitative and structural damages of chromosomes were revealed by the cytogenetic assessment of eighteen rats with acute myeloid leukaemia (AML) and six rats with chronic myeloid leukaemia (CML). The most characteristic chromosomal aberrations for AML appeared to be rearrangements in chromosomes 1-3. In CML translocations by one of chromosomes 15-20 coupled with defects in the groups of chromosomes 5-10 seemed to be nonrandom.

RESUMO

The studies were performed on Wistar rats with acute myeloblastic leukemia induced by a prolonged administration of tritiated water (370 MBq/kg/day-1; an absorbed dose--25.3 Gy) and by repeated pyrogenal injections. A reduction of the pool of polypotent hemopoietic cells (CFUs) with a simultaneous increase of their proliferative activity and a decline of their ability to self-supporting were observed in the leukemia-bearing animals. Populations of CFUdc in DC and of CFUc were reduced in all cases; their colony-forming ability was also found to decrease. All animals with leukemia showed a lowered amount of CFU-BL in the bone marrow and of CFU-TL in the thymus, although the content of these cells in the peripheral blood remained unchanged.

RESUMO

A large-dispersed plutonium 239 dioxide intravenously administered to rats in doses of 92.5, 46.3, 23.2 kBq/kg of body mass was shown to impair haemopoiesis which was manifested by a more profound injury to erythropoiesis, a stable increase in the number of bone marrow lymphocytes, a remote development of lymph node hypoplasia, and reduction of functional neutrophil and lymphocyte pools.

RESUMO

During chronic exposure to tritium oxide (a dose rate of 0.125 Gy/day-1, and a cumulative-absorbed dose of 22.1 Gy) different granulocytopoiesis compartments (i.e. polypotent CFUs, committed CFUc; proliferating, maturing, and functional pools) were differently damaged by radiation. In the course of the development of tritium oxide-induced affection granulocytopoiesis proceeds at an intense pace.

The data are submitted obtained on 10.5 thousands Wistar rats and 350 mongrel dogs exposed to gamma-radiation (LD40-LD100) in different seasons. The radiosensitivity of both animal species was the highest in autumn (August and September) and the lowest in winter (February and January). The seasonal DMF for rats and dogs with LD16, LD50 and LD84 was 1.24 and 1.31, 1.13 and 1.17 and 1.04 and 1.07, respectively. The curves for seasonal radiosensitivity of dogs with LD84, LD50 and LD16, exhibited a single--phase character throughout a year. For rats, the seasonal radiosensitivity curves with LD84 were single-phase, and the curve with LD50, and particularly LD16, exhibited the second peak of radioresistance in June. The increase in radioresistance of rats and dogs correlated with the increase in number of leukocytes in the peripheral blood of animals.

RESUMO

The kinetics of cell movement through the compartment of maturing, nondividing neutrophils of the Wistar rat bone marrow was studied by autoradiography using H3-thymidine. There were transit and reserved neutrophil populations in each cell compartment. The transit periods of the maturing nondividing cells, the mean time of cell renewal in the subcompartments of metamyelocytes, the band and the sementonuclear neutrophils were determined with consideration to the reserved cell population.
