RESUMO

The present work investigates the influence of isothermal annealing on the microstructure and oxidation behavior of nanocomposite coatings. AlTiSiN/TiSiN coatings with TiSiN adhesive layer were deposited onto a high-speed steel substrate via physical vapor deposition. The coatings were investigated in the as-deposited state as well as after annealing in air at 700, 800, 900 and 1000 °C, respectively. The microstructure and morphology of the coatings were observed using scanning electron microscopy and transmission electron microscopy. The chemical composition and presence of oxidation products were studied by energy-dispersive X-ray spectroscopy. The phase identification was performed by means of X-ray diffraction. In the microstructure of the as-deposited coating, the (Ti1-xAlx)N particles were embedded in an amorphous Si3N4 matrix. TiO2 and SiO2 were found at all annealing temperatures, and Al2O3 was additionally identified at 1000 °C. It was found that, with increasing annealing temperature, the thickness of the oxide layer increased, and its morphology and chemical composition changed. At 700 and 800 °C, a Ti-Si-rich surface oxide layer was formed. At 900 and 1000 °C, an oxidized part of the coating was observed in addition to the surface oxide layer. Compared to the as-deposited sample, the oxidized samples exhibited considerably worse mechanical properties.

Development of new types of antimicrobial coatings is of utmost importance due to increasing problems with pathogen transmission from various infectious surfaces to human beings. In this study, new types of highly potent antimicrobial polyurethane composite films encapsulated by hydrophobic riboflavin-based carbon polymer dots are presented. Detailed structural, optical, antimicrobial, and cytotoxic investigations of these composites were conducted. Low-power blue light triggered the composites to eradicate Escherichia coli in 30 min, whereas the same effect toward Staphylococcus aureus was reached after 60 min. These composites also show low toxicity against MRC-5 cells. In this way, RF-CPD composites can be used for sterilization of highly touched objects in the healthcare industry.

Iron, an essential element for most living organism, participates in a wide variety of physiological processes. Disturbance in iron homeostasis has been associated with numerous pathologies, particularly in the heart and brain, which are the most susceptible organs. Under iron-overload conditions, the generation of reactive oxygen species leads to impairment in Ca2+ signaling, fundamentally implicated in cardiac and neuronal physiology. Since iron excess is accompanied by increased expression of iron-storage protein, ferritin, we examined whether ferritin has an effect on the ryanodine receptor - isoform 2 (RYR2), which is one of the major components of Ca2+ signaling. Using the method of planar lipid membranes, we show that ferritin induced an abrupt, permanent blockage of the RYR2 channel. The ferritin effect was strongly voltage dependent and competitively antagonized by cytosolic TEA+, an impermeant RYR2 blocker. Our results collectively indicate that monomeric ferritin highly likely blocks the RYR2 channel by a direct electrostatic interaction within the wider region of the channel permeation pathway.

Diclofenac sodium salt (DSS)-loaded electrospun nanofiber mats on the base of poly(ε-caprolactone) (PCL) were investigated as biocompatible nanofibrous mats for medical applications with the ability to inhibit bacterial infections. The paper presents the characteristics of fibrous mats made by electrospinning and determines the effect of medicament on the fiber morphology, chemical, mechanical and thermal properties, as well as wettability. PCL and DSS-loaded PCL nanofibrous mats were characterized using scanning electron microscopy, transmission electron microscopy, attenuated total reflectance-Fourier transform infrared spectrometry, dynamic mechanical analysis, and contact angle measurements. Electron paramagnetic resonance measurements confirmed the lifetime of DSS before and after application of high voltage during the electrospinning process. In vitro biocompatibility was studied, and it was proved to be of good viability with ~92% of the diploid human cells culture line composed of lung fibroblast (MRC 5) after 48 h of incubation. Moreover, the significant activity of DSS-loaded nanofibers against cancer cells, Ca Ski and HeLa, was established as well. It was shown that 12.5% (m/V) is the minimal concentration for antibacterial activity when more than 99% of Escherichia coli (Gram-negative) and 99% of Staphylococcus aureus (Gram-positive) have been exterminated.

The deformation behaviour of bimodal sized Al2 O3 /Al nanocomposites were investigated by hot compression tests conducted in the temperature range 350-500°C and strain rates of 0.001, 0.01 and 0.1 s-1 . The dynamic recrystallisation behaviour of the nanocomposites strongly depended on the forming parameters. The bimodal sized Al2 O3 particles played a crucial role in the recrystallised microstructure. The addition of bimodal sized Al2 O3 particles led to a significant increase of activation energy of plastic deformation, corroborating the enhanced resistance of the nanocomposite to hot deformation. This was also reflected by the increased compressive yield strength in the nanocomposite due to both dislocation strengthening caused by n-Al2 O3 and preventing the grain growth due to the presence of µ-Al2 O3 at grain boundaries. It was found that with the decrease of Z values, local strain induced by deformation was released and the grain size of aluminium matrix gradually increased, indicating that the main softening mechanism of the bimodal sized Al2 O3 /Al nanocomposites was dynamic recrystallisation (DRX). The lower the Z value was, the easier the DRX occurred. The highly beneficial role of the bimodal sized Al2 O3 reinforcement in improving the high-temperature performance of aluminium matrix nanocomposite was discussed.

Eusociality in its various degrees represents an animal social system characterised by cooperative brood care, differentiation into castes and generational overlap. The fossil record indicates that eusociality is likely to have originated in hymenopterans and blattodeans during the Cretaceous. In this study, we present findings from surveys in Peruvian (Villa Carmen) and Ecuadorian (Rio Bigal, El Reventador) cloud forests revealing the first extant cockroach species living in complex, structured groups (n = 90-200 individuals, ˃ 20 adults). We observed and described behaviours that suggest the existence of cooperative care, nest guarding, nest chamber preparation within hardwood Casearia sp. (Salicaceae) and bamboo (Bambusoideae), multiple overlapping generations ('different stages of' instars), colony translocation, possibly a sole reproductive female (1.25 times larger white 'queen', but no potential 'king' observed), and morphologically diversified immature stages. In order to define the lineage where this type of sociality originated and occurs, the forms of Melyroidea magnifica Shelford, 1912, M. ecuadoriana sp. n., M. mimetica Shelford, 1912 and an undescribed species from Peru are also described in a separate section of this study. Blattoid morphological characteristics such as typical styli suggest categorisation within distinct Oulopterygidae (Rehn, 1951), outside Corydiidae Saussure 1864. Transitional advanced sociality or semisociality in related Aclavoidea socialis gen. et sp. n. is documented in a rotting stump (n = 80 individuals, few adults). Close phylogenetic relation between the genera, conserved morphology of numerous characters and their diverse feeding strategies generally lacking specialisation suggests a rather recent origin of a social way of life in this group. Eusociality in invertebrates and vertebrates can thus originate in various phylogenetical and ecological trajectories including predation, parasitism, care for herbs and the new one, documented through diet shift from detritivory to fungivory and algaevory. Interdisciplinary approaches reveal the low degree of knowledge of rainforest ecosystems, with fundamental groups remaining still systematically and also behaviourally undescribed.

Silk fibroin is a biocompatible, non-toxic, mechanically robust protein, and it is commonly used and studied as a material for biomedical applications. Silk fibroin also gained particular interest as a drug carrier vehicle, and numerous silk formats have been investigated for this purpose. Herein, we have prepared electrospun nanofibers from pure silk fibroin and blended silk fibroin/casein, followed by the incorporation of an anti-inflammatory drug, diclofenac. Casein serves as an excipient in pharmaceutical products and has a positive effect on the gradual release of drugs. The characteristics of the investigated composites were estimated by scanning electron microscope, transmission electron microscope, thermogravimetric analysis, and a lifetime of diclofenac by electron paramagnetic resonance analysis. The cumulative release in vitro of diclofenac sodium salt, together with the antiproliferative effect of diclofenac sodium salt-loaded silk nanofibers against the growth of two cancer cell lines, are presented and discussed.
