BACKGROUND/OBJECTIVES: Acinar-to-ductal metaplasia (ADM) has been shown to contribute to the development of pancreatic ductal adenocarcinoma (PDAC) in genetically engineered mouse models, but little is known about whether acinar cell plasticity contributes to carcinogenesis in human PDAC. We aimed to assess whether cancer cells that stain positive for amylase and CK19 (ADM-like cancer cells) are present in human resected PDAC and to investigate their role in tumor progression. METHODS: We immunohistochemically investigated the presence of ADM-like cancer cells, and compared the clinical and histological parameters of PDAC patients with and without ADM-like cancer cells. RESULTS: ADM-like cancer cells were detected in 16 of 60 (26.7%) PDAC specimens. Positive staining for anterior gradient protein 2 (AGR2) was observed in 14 of 16 (87.5%) PDAC specimens with ADM-like cancer cells. On the other hand, the intensity of AGR2 expression (negative, low/moderate or high) was lower in PDAC with ADM-like cancer cells (9/7) than in PDAC without these cells (11/33) (P = 0.032). The presence of ADM-like cancer cells was significantly correlated with increased cell proliferation (P = 0.012) and tended to be associated with MUC1 expression (P = 0.067). CONCLUSIONS: These results indicated that acinar cells may act as the origin of human PDAC, and that their presence may be useful for the stratification of human PDAC to predict prognosis.

Camera traps are a powerful tool for wildlife surveys. However, camera traps may not always detect animals passing in front. This constraint may create a substantial bias in estimating critical parameters such as the density of unmarked populations. We proposed the 'double-observer approach' with camera traps to counter the constraint, which involves setting up a paired camera trap at a station and correcting imperfect detection with a reformulated hierarchical capture-recapture model for stratified populations. We performed simulations to evaluate this approach's reliability and determine how to obtain desirable data for this approach. We then applied it to 12 mammals in Japan and Cameroon. The results showed that the model assuming a beta-binomial distribution as detection processes could correct imperfect detection as long as paired camera traps detect animals nearly independently (Correlation coefficient ≤ 0.2). Camera traps should be installed to monitor a predefined small focal area from different directions to satisfy this requirement. The field surveys showed that camera trap could miss animals by 3-40%, suggesting that current density estimation models relying on perfect detection may underestimate animal density by the same order of magnitude. We hope that our approach will be incorporated into existing density estimation models to improve their accuracy.

Movement is a fundamental characteristic of animals, but challenging to measure noninvasively. Noninvasive methods for measuring travel have different weaknesses, so multiple techniques need to be applied multiple techniques for reliable inferences. We used two methods, direct tracking and camera trapping, to examine the variation in time and seasonal differences in movement rates of mandrills (Mandrillus sphinx), an elusive primate that lives in large groups in central Africa. In a 400-km2 rainforest area in Moukalaba-Doudou National Park, Gabon, we tracked unidentified groups 46 times from 2009 to 2013. We systematically placed 157 terrestrial camera traps in the same area from 2012 to 2014 and recorded groups 309 times. Generalized additive mixed models (GAMMs) of the tracking data indicated that the group travel speed varied with time and season. In the fruiting season, the movement rate fluctuated with time in a bimodal pattern, whereas in the nonfruiting season, it increased monotonously with time. The predicted day range was longer in the fruiting season (6.98 km) than in the nonfruiting season (6.06 km). These seasonal differences suggest responses to changes in food resources and temperature. Camera-trap detection rates showed similar temporal and seasonal patterns to the tracking data, allowing us to generalize our findings to the population level. Moreover, cameras never detected mandrills at night, and we observed that they slept high in trees and hardly moved until the next morning, all suggesting their strict avoidance of nighttime movement. This study demonstrated the significance of the multiple-method approach in drawing robust conclusions on temporal patterns of animal movement.


Le mouvement est une caractéristique fondamentale des animaux, mais il est difficile de le mesurer de manière non invasive. Les méthodes non invasives de mesure des déplacements présentent des faiblesses différentes, de sorte que nous devons appliquer plusieurs techniques à un même sujet pour obtenir des déductions fiables. Nous avons utilisé deux méthodes, le suivi direct et le piégeage photographique, pour examiner les variations temporelles et la différence saisonnière des taux de déplacement des mandrills (Mandrillus sphinx), un primate insaisissable vivant en grands groupes. Dans une zone de forêt tropicale humide de 400 km2 du parc national de Moukalaba-Doudou au Gabon, nous avons suivi des groupes non identifiés 46 fois entre 2009 et 2013. Dans la même zone, nous avons systématiquement placé 157 pièges photographiques terrestres de 2012 à 2014 et enregistré des groupes 309 fois. Les modèles mixtes additifs généralisés (GAMM) des données de suivi ont indiqué que la vitesse de déplacement des groupes variait en fonction du temps et des saison. Pendant la saison de fructification, la vitesse de déplacement fluctuait dans le temps selon un schéma bimodal, tandis que pendant la saison de non-fructification, elle augmentait de façon monotone avec le temps. Les distances journalières prédites étaient plus longues pendant la saison de fructification (6,98 km) que pendant la saison de non-fructification (6,06 km). Ces différences saisonnières suggèrent des réponses aux changements de ressources alimentaires et de température. Les taux de détection des pièges photographiques ont montré des tendances temporelles et saisonnières similaires à celles des données de suivi, ce qui nous permet de généraliser nos conclusions au niveau de la population. De plus, les caméras n'ont jamais détecté les mandrills la nuit, et nous avons observé qu'ils dormaient haut dans les arbres et ne bougeaient pratiquement pas jusqu'au lendemain matin, ce qui suggère qu'ils évitent strictement de se déplacer la nuit. Cette étude a démontré l'importance de l'approche à méthodes multiples pour tirer des conclusions solides sur les modes temporels de déplacement des animaux.

Vehicle exhaust gases are important sources of nitrous acid (HONO). In this study, HONO in diesel vehicle exhaust was measured by incoherent broadband cavity-enhanced absorption spectroscopy using a chassis dynamometer system. The mean HONO concentrations in exhaust gases emitted by passenger cars and light-duty trucks were high when the after treatment devices were not fully working during the warming up period. The HONO/NOx ratio is a good index of HONO formation. The HONO/NOx ratios were 9.7 × 10-3-18.1 × 10-3, and were higher than what we found in a previous study. The estimated HONO emission factors were 7.71-64.70 mg (kg fuel)-1, and were lower than were found in previous studies. The results indicated that the frequency particulate matter is removed from a diesel particle filter affects the HONO concentration in the emitted gases and the HONO emission factor.

Analyses of life history and population dynamics are essential for effective population control of wild mammals. We developed a model for the simultaneous estimation of seasonal changes in three parameters-population density, habitat preference and trap catchability of target animals-based on camera-trapping data and harvest records. The random encounter and staying time model, with no need for individual recognition, is the core component of the model-by combining this model with the catch-effort model, we estimated density at broad spatial scales and catchability by traps. Here, the wild boar population in central Japan was evaluated as a target population. We found that the estimated population density increased after the birth period and then decreased until the next birth period, mainly due to harvesting. Habitat preference changed seasonally, but forests having abandoned fields nearby were generally preferred throughout the season. These patterns can be explained by patterns of food availability and resting or nesting sites. Catchability by traps also changed seasonally, with relatively high values in the winter, which probably reflected changes in the attractiveness of the trap bait due to activity changes in response to food scarcity. Based on these results, we proposed an effective trapping strategy for wild boars, and discussed the applicability of our model to more general conservation and management issues.

Missing hydroxyl radical (OH) reactivity from unknown/unmeasured trace species empirically accounts for 10%-30% of total OH reactivity and may cause significant uncertainty regarding estimation of photochemical ozone production. Thus, it is essential to unveil the missing OH reactivity for developing an effective ozone mitigation strategy. In this study, we conducted simultaneous observations of total OH reactivity and 54 reactive trace species in a suburban area as part of the Air QUAlity Study (AQUAS)-Tsukuba campaign for the summer of 2017 to gain in-depth insight into total OH reactivity in an area that experienced relatively high contributions of secondary pollutants. The campaign identified on average 35.3% of missing OH reactivity among total OH reactivity (12.9 s-1). In general, ozone-production potential estimation categorized ozone formation in this area as volatile organic compound (VOC)-limited conditions, and missing OH reactivity may increase ozone production potential 40% on average if considered. Our results suggest the importance of photochemical processes of both AVOCs and BVOCs for the production of missing OH reactivity and that we may underestimate the importance of reducing precursors in approach to suppressing ozone production if we ignore the contribution of their photochemical products.

BACKGROUND: Peficitinib (ASP015K), a novel oral Janus kinase inhibitor, has demonstrated efficacy and safety for the treatment of rheumatoid arthritis (RA) in randomized, controlled trials of up to 52 weeks' duration. However, safety and effectiveness after long-term treatment have not been assessed. METHODS: This was an interim analysis of an ongoing open-label, multicenter extension study in RA patients who completed phase 2b (RAJ1; 12 weeks) and phase 3 (RAJ3 and RAJ4; 52 weeks) peficitinib studies in Asia (mainly Japan). Eligible patients (n = 843) received oral peficitinib once daily (100 mg, or 50 mg for patients transferring from RAJ1). The peficitinib dose could be increased (up to 150 mg) or reduced (to 50 mg) at the discretion of the investigator. Efficacy variables assessed included American College of Rheumatology (ACR) response rates, ACR components, and disease activity score in 28 joints based on C-reactive protein (DAS28-CRP). RESULTS: Results up to May 2018 are summarized. Mean peficitinib duration of exposure was 22.7 months and the maximum dose was 100 mg in most (66.5%) patients. ACR responses were maintained during the extension study, with ACR20/50/70 response rates of 71.6%, 52.1%, and 34.7% at week 0 and 78.9%, 61.4%, and 42.7% at end of treatment, respectively. ACR components and DAS28-CRP showed improvements from baselines of the preceding studies and continued to show improvements during the extension study. Treatment-emergent adverse events (TEAEs) were reported in 757/843 (89.8%) patients, the most common being nasopharyngitis (39.7%) and herpes zoster (11.7%). The majority of TEAEs were severity grade 1/2. Drug-related TEAEs leading to permanent study drug discontinuation occurred in 55/843 (6.5%) patients. Regarding AEs of special interest, the incidence per 100 patient-years of serious infections was 2.3 (95% CI 1.6 - 3.1), herpes zoster-related disease 6.8 (95% CI, 5.6 - 8.3), and malignancies 1.1 (95% CI, 0.7 - 1.8). One death from diffuse large B cell lymphoma during the study and one death from uterine sarcoma after the study were considered probably and possibly related to study drug, respectively. CONCLUSIONS: The effectiveness of peficitinib was maintained or improved during long-term administration and treatment up to 6 years was well tolerated in Asian patients with RA. Water-soluble organic nitrogen (WSON) affects the formation, chemical transformations, hygroscopicity, and acidity of organic aerosols as well as biogeochemical cycles of nitrogen. However, large uncertainties exist in the origins and formation processes of WSON. Submicrometer aerosol particles were collected at a suburban forest site in Tokyo in summer 2015 to investigate the relative impacts of anthropogenic and biogenic sources on WSON formations and their linkages with aerosol liquid water (ALW). The concentrations of WSON (ave. 225 ± 100 ngN m-3) and ALW exhibited peaks during nighttime, which showed a significant positive correlation, suggesting that ALW significantly contributed to WSON formation. Further, the thermodynamic predictions by ISORROPIA-II suggest that ALW was primarily driven by anthropogenic sulfate. Our analysis, including positive matrix factorization, suggests that aqueous-phase reactions of ammonium and reactive nitrogen with biogenic volatile organic compounds (VOCs) play a key role in WSON formation in submicrometer particles, which is particularly significant in nighttime, at the suburban forest site. The formation of WSON associated with biogenic VOCs and ALW was partly supported by the molecular characterization of WSON. The overall result suggests that ALW is an important driver for the formation of aerosol WSON through a combination of anthropogenic and biogenic sources.

The change in the ozone production rate on reducing its precursors, namely, ozone production sensitivity, is important information for developing a strategy to reduce ozone. We expanded a conventional sensitivity analysis theory by including peroxy radical loss by uptake onto particle surfaces in the aim of examining their potential impact. We also propose a new concept of absolute sensitivity that enables us to evaluate the quantitative effectiveness of precursor reduction toward mitigating ozone production over a given period and area. This study applies the theory to observations in Tsukuba, a city in Japan. The relative sensitivity analysis shows that ozone production was more sensitive to volatile organic compounds (VOCs) in the morning and evening, and it became more sensitive to NOx in the afternoon. NO depletion was a main trigger in this sensitivity regime transition. The absolute sensitivity analysis indicates that the VOC-sensitive period in the morning determines the total ozone production sensitivity in a day. While particles did not have significant impact on regime classification in Tsukuba, they have a potential to decrease the mitigating effect of VOC reduction on ozone production and to moderate the enhancement effect of NOx reduction depending upon uptake coefficients. A further study will benefit from a combination with an observation-constrained box model simulation or chemical transport modeling system, which may provide sensitivity analysis over a large spatial and temporal range.

BACKGROUND: A multicenter, randomized, open-label, parallel group, pilot, 52-week study in Asian countries that assessed the renal function, efficacy, and safety of reduced-exposure versus standard-exposure prolonged-release tacrolimus (PR-T) in adult kidney transplant recipients (KTRs). METHODS: Posttransplantation, KTRs received PR-T from weeks 0 to 4 (initial dose, 0.2-0.3 mg/kg; target trough level, 6-10 ng/mL). At week 4, KTRs were randomized (1:1) to receive reduced-exposure PR-T (target 4-6 ng/mL, weeks 4-12; 3-5 ng/mL, weeks 12-52) or standard-exposure PR-T (target: 6-10 ng/mL, weeks 4-52). Primary end point: estimated glomerular filtration rate (eGFR) over 52 weeks. Secondary end points (week 52) included creatinine clearance, serum creatinine, graft/patient survival, biopsy-confirmed acute rejection (AR), composite of graft loss/patient death/biopsy-confirmed AR, and steroid-resistant AR. Treatment-emergent adverse events were recorded. RESULTS: Sixty-six KTRs received PR-T (reduced-exposure, n = 32; standard-exposure, n = 34) and were analyzed. After per-protocol dose adjustment, mean ± standard deviation tacrolimus trough level was lower with reduced- versus standard-exposure PR-T (week 52, 4.5 ± 1.1 ng/mL vs 8.0 ± 2.2 ng/mL). In the reduced- versus standard-exposure group, eGFR was similar at weeks 8 to 52 (overall least-square mean difference, -2.82; 95% confidence interval, -7.91 to 2.27; P = 0.272). At week 52, there was no significant difference in creatinine clearance (P = 0.375) or serum creatinine (P = 0.547) between groups. All grafts/patients survived, no steroid-resistant AR was reported, and 4 and 3 patients had AR in reduced- and standard-exposure groups, respectively. Drug-related treatment-emergent adverse events were reported in 34.4% and 38.2% of patients, respectively. CONCLUSIONS: Reducing exposure to PR-T resulted in a clinically acceptable short-term safety profile and was generally as effective as standard tacrolimus exposure for Asian patients.

High resolution Fourier transform emission spectroscopy of the A∼2Πi-X∼2Πi band of the OCS+ ion was performed in the UV region to observe the ν1 (CO stretch) progression bands (υ1 = 0 → 2-5) for both the Ω=3/2 and 1/2 spin components. Accurate molecular constants including the rotational constants, B0 = 0.194 765(13) and 0.187 106(13) cm-1, and the spin-orbit interaction constants, A0 = -381.0(56) and -126.5(56) cm-1, were determined for the X∼2Π and A∼2Π states, respectively, by the simultaneous analysis of the observed progression bands. The CO bond length (rCO = 1.2810 Å) for the A∼2Π state, derived from the rotational constant B0 and Franck-Condon factors, is longer by 0.1756 Å than that (1.1054 Å) for the X∼2Π state, while the CS bond length for the A∼2Π state is shorter by 0.0905 Å than that for the X∼2Π state. Pure rotational transition frequencies in the ground X∼2Π state are predicted, as well as transition frequencies of the ν1 fundamental band, with the present molecular constants.

Incoherent broadband cavity-enhanced absorption spectroscopy (IBBCEAS) is a useful technique for measuring trace gaseous species in the atmosphere. Recently, IBBCEAS was used to measure concentrations of nitrous acid (HONO) in the troposphere to resolve controversies related to its formation and loss. Here, measurements of HONO and a mixture of HONO and NO2 using IBBCEAS were validated by comparing them with those obtained with a NOx analyzer. Good agreement was found between these methods, given their respective experimental uncertainties. The detection limit of our IBBCEAS instrument was 0.2 ppbv, with a signal-to-noise ratio of 1, and a 5-min integration time.

BACKGROUND: Previous studies have indicated that ramosetron, a 5-hydroxytryptamine-3 receptor antagonist, achieves global improvement in irritable bowel syndrome (IBS) symptoms in male patients with IBS with diarrhea (IBS-D). However, in addition to global assessment it was deemed important to assess "clinically meaningful improvements, focusing on the patient's chief complaint and the severity of major IBS symptoms". We performed a randomized, placebo-controlled, phase IV pilot study to explore and examine efficacy variables that allow such evaluation of ramosetron in male patients with IBS-D. METHODS: We performed a prospective study of 115 male outpatients with IBS-D (according to the Rome III criteria), from June 2009 to December 2009 at 25 centers in Japan. After a one-week baseline period, subjects received either 5 µg of ramosetron (n = 47) or placebo (n = 51) once daily for 12 weeks. To evaluate "clinically meaningful improvements focusing on the severity of major IBS symptoms," the Japanese version of the IBS severity index (IBSSI-J) was used. RESULTS: Change in IBSSI-J overall score from baseline was -133.5 ± 110.72 in the ramosetron 5 µg group and -108.2 ± 94.44 in the placebo group (P = 0.228) at the last evaluation point. Differences in responder rates for at least a 50% reduction from baseline in IBSSI-J between the ramosetron 5 µg group and the placebo group were over 10%, except Month 1. The monthly responder rate for global assessment of relief of overall IBS symptoms in the ramosetron 5 µg group showed a statistically significant improvement compared to placebo at the second month (44.4% vs 18.4%, P = 0.012). The proportion of patients who had a ≥ 50% reduction in IBSSI-J overall score was 24/37 (64.9%) in the responder group on global assessment and 18/54 (33.3%) in the non-responder group at Week 12. CONCLUSIONS: Further examination will be needed before IBSSI-J can be used in clinical trials of agents for IBS-D. However, this study revealed that response on global assessment was correlated with improvement in the IBSSI-J, suggesting that global assessment reflects improvement of the symptom severity of patients with IBS-D. BACKGROUND: Global assessment allows patients to assess improvement in multiple irritable bowel syndrome (IBS) symptoms. However, it was deemed important to assess "clinically meaningful improvements, focusing on the patient's chief complaint and the severity of major IBS symptoms" in addition to global assessment to show how ramosetron is effective for individual IBS symptoms. This is a pilot study to explore clinical endpoints focusing on the chief complaint of patients with IBS with diarrhea (IBS-D). METHODS: The same database was used in a previously reported post-marketing phase IV, randomized placebo-controlled pilot trial in male patients with IBS-D. The hypothesis is completely different from that of the other study. Patients with IBS-D diagnosed according to Rome III criteria were given either 5 µg of ramosetron (n = 47) or placebo (n = 51) once daily for 12 weeks after a one-week baseline period. To explore and examine endpoints that allow evaluation of "clinically meaningful improvements focusing on the patient's chief complaint," the chief complaint and its relief by this study drug were assessed in this exploratory study. RESULTS: Rates of patients with abdominal pain/discomfort, stool form and stool frequency which patients had as a chief complaint before administration were 34.0, 19.1 and 25.5%, respectively, in the ramosetron 5 µg group and 42.0, 18.0, and 20.0% in the placebo group. Responder rates for improvement in symptoms of the chief complaint that patients had before administration were 53.2% in the ramosetron 5 µg group and 42.0% in the placebo group at the last point. The greatest symptomatic improvement in the chief complaint in the ramosetron 5 µg group compared to the placebo group was shown with respect to stool consistency. Bristol Stool Form Scale (BSFS) scores were significantly lower in the ramosetron group than in the placebo group (4.36 ± 1.195 vs 4.85 ± 0.890 at the last point, P = 0.027) throughout the treatment period, except at week 6. CONCLUSIONS: Ramosetron acted most effectively on stool consistency. Nitrous acid (HONO) plays an important role in the formation of OH radicals, which are involved in photochemical oxidation. HONO concentrations in ambient air at urban sites have previously been measured, but very few studies have been performed in central Tokyo. In this study, HONO concentrations in ambient air in southeast central Tokyo (near Tokyo Bay) in winter were determined by incoherent cavity enhanced absorption spectroscopy. The O3, NO, NO2, and SO2 concentrations were simultaneously determined. The NO concentrations were used to classify the parts of the study period into types I (high pollution), II (medium pollution), and III (low pollution). The maximum HONO concentrations in the type I, II, and III periods were 7.1, 4.5, and 3.0ppbv, respectively. These concentrations were comparable to concentrations previously found in other Asian megacities. The mean HONO concentration varied diurnally, and HONO was depleted between 00:00 and 03:00 each day. The sampling site is surrounded by roads with high traffic loads, but vehicular emissions were estimated to contribute <10% of the HONO concentrations. Two positive and negative relative humidity dependences of the HONO to NO2 ratio were confirmed, implying the existence of the two different secondary formation process of HONO. The NO2 to HONO conversion rates at night in the type I, II, and III periods were 6.3×10-3, 7.6×10-3, and 4.2×10-3h-1, respectively.

Nitrous acid (HONO) is a well-known source of hydroxyl radicals in the troposphere. Vehicle exhaust is considered to be one of the primary emission sources of HONO. In this study, measurements of HONO in gasoline vehicle exhaust were carried out using a chassis dynamometer combined with incoherent broadband cavity-enhanced absorption spectroscopy. When catalysts were warm, concentrations of HONO were higher than those prior to catalysts warming. Other species, such as CO, and total hydrocarbons (THCs), showed the opposite pattern. There were no correlations evident between HONO and other trace species concentrations immediately after emission. The HONO/NOx ratio, a good proxy for the formation of HONO in atmosphere, ranged from 1.1 to 6.8×10-3, which was consistent with previous studies. HONO emission factors (EFs) were calculated to be 0.01-3.6mgkg-1 fuel, which was different from the vehicle's specifications and those reported under different driving cycles. Annual HONO emissions in Japan were estimated using the calculated EFs and other statistical data.

BACKGROUND: The long-term safety of administration of ramosetron in female patients with irritable bowel syndrome with diarrhea (IBS-D) is unknown. The aim of this study was to assess the long-term safety, tolerability, and outcomes with the use of ramosetron in female patients with IBS-D. METHODS: This was a phase III, open-label, uncontrolled, long-term safety trial of the treatment of female Japanese patients with IBS-D, diagnosed according to the Rome III criteria. A total of 151 patients were given 2.5 µg of ramosetron for 4 weeks, and responders continued the same dose for another 48 weeks. Non-responders at 4 weeks were given 5 µg of ramosetron for 48 weeks. At the end of week 52, 106 patients receiving 2.5 µg and 17 patients receiving 5 µg had completed the study. Safety and efficacy including symptoms and quality of life (QOL) were evaluated. RESULTS: Concerning safety, no serious adverse event related to ramosetron, specifically ischemic colitis, was observed in patients with either dose of ramosetron. However, constipation occurred in 19.7 % of patients given 2.5 µg and 10.5 % of patients given 5 µg of ramosetron. Ramosetron-treated patients showed high rates of global improvement. Stool consistency, abdominal pain and discomfort, and IBS-QOL were also improved at the last evaluation point. CONCLUSIONS: The results provide evidence of the long-term safety and efficacy of treatment with 2.5 and 5 µg of ramosetron in female patients with IBS-D. Clinicians should be aware that one-fifth of women with IBS-D receiving ramosetron may suffer from constipation during treatment (ClinicalTrials.gov ID: NCT01736423).
