RESUMO

Encoded microparticles have great potential in small-volume multiplexed assays. It is important to link the micro-level assays to the macro-level by indexing and manipulating the microparticles to enhance their versatility. There are technologies to actively manipulate the encoded microparticles, but none is capable of directly manipulating the encoded microparticles with homogeneous physical properties. Here, we report the image-based laser-induced forward transfer system for active manipulation of the graphically encoded microparticles. By demonstrating the direct retrieval of the microparticles of interest, we show that this system has the potential to expand the usage of encoded microparticles.

Since the discovery of antibiotics, the emergence of antibiotic resistance has become a global issue that is threatening society. In the era of antibiotic resistance, finding the proper antibiotics through antibiotic susceptibility testing (AST) is crucial in clinical settings. However, the current clinical process of AST based on the broth microdilution test has limitations on scalability to expand the number of antibiotics that are tested with various concentrations. Here, we used color-coded droplets to expand the multiplexing of AST regarding the kind and concentration of antibiotics. Color type and density differentiate the kind of antibiotics and concentration, respectively. Microscopic images of a large view field contain numbers of droplets with different testing conditions. Image processing analysis detects each droplet, decodes color codes, and measures the bacterial growth in the droplet. Testing E. coli ATCC 25922 with ampicillin, gentamicin, and tetracycline shows that the system can provide a robust and scalable platform for multiplexed AST. Furthermore, the system can be applied to various drug testing systems, which require several different testing conditions.

Wireless monitoring of the physio-biochemical information is becoming increasingly important for healthcare. In this work, we present a proof-of-concept hydrogel-based wireless biochemical sensing scheme utilizing ultrasound. The sensing system utilizes silica-nanoparticle embedded hydrogel deposited on a thin glass substrate, which presents two prominent interfaces for ultrasonic backscattering (tissue/glass and hydrogel/glass). To overcome the effect of the varying acoustic properties of the intervening biological tissues between the sensor and the external transducer, we implemented a differential mode of ultrasonic back-scattering. Here, we demonstrate a wireless pH measurement with a resolution of 0.2 pH level change and a wireless sensing range around 10 cm in a water tank.

In this work, we present a proof-of-concept hydrogel-based sensor system capable of wireless biochemical sensing through measuring backscattered ultrasound. The system consists of silica-nanoparticle embedded hydrogel deposited on a thin glass substrate, presenting two interfaces for backscattering (tissue/hydrogel and hydrogel/glass), which allows for system output to be invariant under the change in acoustic properties (e.g. attenuation, reflection) of the intervening biological tissue. We characterize the effect of silica nanoparticles (acoustic contrast agents) loading on the hydrogel's swelling ratio and its ultrasonic backscattering properties. We demonstrate a wireless pH measurement using dual modes of interrogations, reflection ratio and time delay. The ultrasonic hydrogel pH sensor is demonstrated with a sensing resolution of 0.2 pH level change with a wireless sensing distance around 10 cm.

Stimuli-responsive hydrogels that exhibit reversible volume changes in response to stimulus cues such as heat, pH, and light have been utilized in soft robotics, microfluidics, electronics, and biomedical surgical tools. While the development of the soft robotics has widely expanded, most external triggering systems still have limited utilities due to the low selectivity. We present a hybrid gripper capable of undergoing preprogrammed shape transformation utilizing ultrasound energy on-off processes as the external triggering system, which can be utilized in invisible and nonselective environments. Furthermore, we describe the magnetic locomotion of the soft gripper enabled by the introduction of iron oxide (Fe2O3) ferrogel. By integrating these dual ultrasonic and magnetic control systems, we demonstrate the soft gripper could actively and safely perform pick-and-place tasks on a biological salmon roe in the aqueous maze environment. We expect that this study can provide the groundwork for the further important advances to the creation of ultrasound-responsive shape programmable and multifunctional smart soft robots.
