We examined the agreement between heart rate deflection point (HRDP) variables with maximal lactate steady state (MLSS) in a sample of young males categorized to different body mass statuses using body mass index (BMI) cut-off points. One hundred and eighteen young males (19.9 ± 4.4 years) underwent a standard running incremental protocol with individualized speed increment between 0.3 and 1.0 km/h for HRDP determination. HRDP was determined using the modified Dmax method called S.Dmax. MLSS was determined using 2-5 series of constant-speed treadmill runs. Heart rate (HR) and blood lactate concentration (La) were measured in all tests. MLSS was defined as the maximal running speed yielding a La increase of less than 1 mmol/L during the last 20 min. Good agreement was observed between HRDP and MLSS for HR for all participants (±1.96; 95% CI = -11.5 to +9.2 b/min, ICC = 0.88; P < 0.001). Good agreement was observed between HRDP and MLSS for speed for all participants (±1.96; 95% CI = -0.40 to +0.42 km/h, ICC = 0.98; P < 0.001). The same findings were observed when participants were categorized in different body mass groups. In conclusion, HRDP can be used as a simple, non-invasive and time-efficient method to objectively determine submaximal aerobic performance in nonathletic young adult men with varying body mass status, according to the chosen standards for HRDP determination.

Evaluation of spinal range of motions (RoMs) and movement coordination between its segments (thorax, lumbar, and pelvis) has clinical and biomechanical implications. Previous studies have not recorded three-dimensional primary/coupled motions of all spinal segments simultaneously. Moreover, magnitude/direction of the coupled motions of the thorax/pelvis in standing posture and lumbopelvic rhythms in the frontal/transverse planes have not been investigated. This study, hence, used an inertial tracking device to measure T1, T5, T12, total (T1-T12) thoracic, lower (T5-T12) and upper (T1-T5) thoracic, lumbar (T12-S1), and pelvis primary and coupled RoMs as well as their movement coordination in all anatomical planes/directions in twenty-two healthy individuals. RoMs were statistically compared between the anatomical planes and spinal segments as well as with available data in the literature. The spine had different primary RoMs in different planes/directions (flexion: lumbar: 55.4 ±â€¯12.4°, pelvis: 42.8 ±â€¯21.6°, and T1-T12 thoracic: 19.9 ±â€¯6.4°, extension: lumbar: 23.4 ±â€¯10.1°, thoracic: 11.7 ±â€¯3.4°, and pelvis: 10.2 ±â€¯6.4°, left/right lateral bending: thoracic: 24.5 ±â€¯7.4°/26.5 ±â€¯6.1°, lumbar: 16.4 ±â€¯7.2°/18.3 ±â€¯5.7°, and pelvis: 11.0 ±â€¯4.4°/9.3 ±â€¯6.2°, and left/right axial rotation: thoracic: 33.5 ±â€¯10.0°/37.1 ±â€¯11.7°, pelvis: 31.6 ±â€¯12.5°/27.2 ±â€¯12.0° and lumbar: 7.5 ±â€¯4.5°/9.2 ±â€¯7.3°). Pelvis, lumbar and thoracic spine had different/varying contributions/rhythms to generate total trunk (T1) movement, both within and between planes. Pattern of the coupled motions was inconsistent between subjects but side bending was generally associated with twisting to the same side at the thoracic spine and to the opposite side at the lumbar spine.

BACKGROUND: Natural killer (NK) cells play important roles in the immune defense against tumors such as colorectal cancer. In humans, NKG2D is an activating immune receptor constitutively expressed in most cytotoxic lymphocytes including NK and CD8+ T cells. In this study, the expression of NKG2D molecule was investigated in peripheral blood NK cells from colorectal cancer patients and compared with healthy subjects. METHODS: We studied 21 non-metastatic (low-grade), 17 non-metastatic (high-grade), 16 metastatic colorectal cancer patients, and 24 healthy controls. Peripheral blood samples were obtained to isolate peripheral blood mononuclear cells (PBMCs) and the percentage of peripheral blood NKG2D+CD3-CD56+ NK cells was analyzed by flow cytometry. The expression of NKG2D at mRNA level was also measured by real-time PCR in both, patients and control subjects. RESULTS: The results showed a significant reduction in the percentage of NKG2D+NK cells as well as NKG2D mRNA expression in peripheral blood of metastatic colon cancer patients. CONCLUSION: This result suggests that decreased expression of activating NKG2D receptor in metastatic colorectal cancer might compromise NK cell function and allow tumor to evade immunity (Tab. 3, Fig. 4, Ref. 33).

BACKGROUND: Carbon nanotubes (CNTs) have a large variety of applications in tissue engineering and biomedical devices. The biocompatibility and cytotoxicity of CNTs have been studied widely, however, up until now; there was uncertainty on how nanosized materials behave in the human body and stem cells. The current study describes the functionalized carbon nanotubes on adipose-derived stem cells (ADSCs) for viability and proliferation purposes in vitro. MATERIALS AND METHODS: After chemical modification of the CNTs, the ADSCs were cultured in Dulbecco's Modified Eagle's. Medium (DMEM) having doses of 0.1, 1, 10, 20, 50, and 100 µg/ml of CNTs. On the third and seventh days of the experiment, the cellular viability, proliferation, and stemness were determined, using the MTT, trypan Blue, and flow cytometry assays in variable CNTs dosage. RESULTS: In doses of 0.1 and 1 µg/ml, the expression of the surface markers were similar to the control groups on day three, but decreased in higher dosages on day seven. The viability of both groups was the same on day three, but in comparison to the control groups, was found to decrease in the higher dosages on day seven. CONCLUSION: The effect of CNTs on the viability and proliferation of ADSCs is a function of time and the doses used. Through further investigation by using these particles, we expect that we should be able to increase the viability and proliferation of ADSCs.

Pancreatic carcinoma is currently considered as a rapidly progressive and fatal disease, and is typically diagnosed late in its natural course. It is characterized by a poor diagnosis and lack of response to conventional therapy. Recent studies have suggested that disulfiram (DSF), a member of the dithiocarbamate family, may have antitumor activity. This study aimed to evaluate the in vitro effect of DSF on apoptosis in human pancreatic cancerous cell line (PANC-1). PANC-1 cells were cultured and treated with DSF at doses of 5, 10, 13 µM for 24 h and apoptosis was measured. Methylation specific PCR (MS-PCR) and real-time quantitative PCR were carried out to detect the methylation pattern and to estimate the mRNA expression levels of RASSF1A, p21 and Bax. MS-PCR analysis demonstrated that no unmethylated band was apeared in PANC-1 cell line after DSF treatments. The real-time quantitative PCR results showed no significant mRNA expression for RASSF1A (p>0.05); whereas p21 and Bax expression were significantly (p<0.01) enhanced after treatment with DSF. The results of the current study indicated that DSF can induce appoptosis in PANC-1 through p21 and Bax pathway but not through RASSF1A.

BACKGROUND: In this study the level of IL-23 and IL-27 produced by macrophages derived from peripheral blood mononuclear cell culture collected from patients with healing or non-healing form of cutaneous leishmaniasis lesion were compared before and after treatment with live Leishmania to explore whether IL-23 or IL-27 plays any role in healing process of cutaneous lesions induced by L. major. METHODS: Twenty patients resident in Isfahan Province, with healing or non-healing form of cutaneous leishmaniasis lesion caused by Leishmania major participated in this study. In vitro productions of IL-23 and IL-27 by peripheral blood derived macrophages, before and after stimulation with live L. major (MRHO/IR/75/ER) promastigotes were evaluated using ELISA method. Patient with healing form of lesion received no treatment and patient with non-healing form of lesion received at least 2 courses of glucantime. RESULTS: The mean production of IL-23 and IL-27 from macrophages of patients with healing form of lesion was significantly higher than patients with non-healing form of lesion. The levels of IL-23 and IL-27 in culture supernatants before and after stimulation in healing form of CL was significantly higher than non- healing form of CL (P < 0.001). CONCLUSION: IL-23 and IL-27 might play a role in human leishmaniasis and further studies are needed to understand the role of IL-23 and IL-27 in leishmaniasis.

For any informed decision making about community-oriented nursing, it is necessary to map out the community regarding the psychological factors. This study is an attempt to estimate the mental disorders' rate among the age group of 15-65 in the urban areas of Ardabil province. A sample of 1430 individuals was selected through random multistage cluster sampling. The instruments employed in this study included Personal Data Questionnaire and the 28-item General Health Questionnaire (GHQ). The findings revealed that, on the whole, 27.83% of the subjects suffer from different mental disorders. The prevalence rate of mental disorders appeared to vary from 13.3% to 44.4% based on the place of residence, and the highest prevalence rate was found to be social dysfunction and anxiety disorders. According to GHQ test, the prevalence rate of social dysfunction, anxiety, somatization and depression disorders were 32%, 22.30%, 20% and 14%, respectively. The study also showed that the prevalence rate of mental disorders was higher among women as compared with men (34.2% vs. 21.1%). The findings of this study is hoped to contribute to community-oriented nursing practice and education.

BACKGROUND: Based on the efficacy of paromomycin ointment and recent ongoing clinical trials of combination of paromomycin and gentamicin, a new physical form of films of the paromomycin and gentamicin was prepared and anti-Leishmania activities of the prepared films were assessed in vitro and in vivo. METHODS: Paromomycin 15% and gentamicin 0.5% was incorporated in a film using ethyl cellulose and HPMC (Hydroxyl Propyl Methyl Cellulose). In order to assess the drug release and anti-Leishmania activities of the preparation, a clone L. major parasite was established using a set of modified NNN medium without overlay liquid layer. Therapeutic effects of the films were evaluated using Balb/c mice model. The mice were inoculated with 2×10(6)L. major promastigotes (MRHO/IR/75/ER) and then when the lesions developed the mice were randomly divided in 3 groups, 10 mice per group, and treated with either perpetrated films or placebo for 28 days or left untreated. RESULTS: Growth inhibition of cloned promastigotes showed that the films have enough releasing capacity and in vivo system, the films containing paromomycin and gentamicin was able to reduce the lesion size and induced complete cure in 80% of the mice but relapse was seen in 60% of the cured mice and overall 50% cure rate was seen during 20 weeks period of the study. CONCLUSION: It seems that the prepared films might be further used in human clinical trials.
