RESUMO

After the Fukushima Daiichi Nuclear Power Plant (FDNPP) accident in 2011, the wild boar (Sus scrofa) population within the Fukushima Evacuation Zone (FEZ) increased substantially in size and distribution. This growing population and their potential dispersal from the FEZ, where they are exposed to high levels of radionuclides, into the surrounding landscape underscores the need to better understand boar movement patterns in order to establish policies for managing shipping restrictions for boar meat and develop management strategies. In this study, we quantified the genetic population structure of boar in and around Fukushima prefecture using sequence data of the mitochondrial DNA control region and MIG-seq analysis using 348 boar samples to clarify boar dispersal patterns. Among boar samples, seven Asian haplotypes and one European haplotype were detected. The European haplotype originated from hybridization between domestic pigs and native boar in the evacuation zone after the accident and was detected in 15 samples across a broad geographic area. Our MIG-seq analysis revealed genetic structure of boar was significantly different between boar inhabiting the eastern (including FEZ. i.e., East clade) and western (i.e., West clade) regions in Fukushima prefecture. In addition, we investigated the relationships between boar dispersal and Cesium (Cs)-137 activity concentrations in boar muscle using MIG-seq genetic data in Nihonmatsu city, located in the central-northern region of Fukushima. High Cs-137 activity concentrations, exceeding 1000 Bq/kg, in boar muscle had a significantly high probability of belonging to the East clade within localized regions. Thus, our results provide evidence of the spatial scale of dispersal of individuals or offspring of boar from the FEZ. Results of this research also indicate that dispersal of individuals between areas with different Cs-137 contamination levels is one of the biggest factors contributing to variation in Cs-137 activity concentration in boar muscle within localized regions.

Following the Fukushima Daiichi Nuclear Power Plant accident in 2011, tissue samples from wild boar (Sus scrofa) outside the evacuation zone (difficult-to-return zone, DRZ) tended to show high activity concentrations of cesium-137 (137Cs). Understanding the 137Cs dynamics of wild boar populations inside the DRZ is necessary because they affect 137Cs dynamics and wild boar management in areas outside the DRZ. Since few detailed, long-term studies have been conducted inside the DRZ, we measured 137Cs activity concentrations in 221 wild boar muscle samples obtained from wild boar caught inside the DRZ and surrounding areas over a 5-year period. Our results showed that the 137Cs activity concentration in wild boar from inside the DRZ were higher than those in wild boar outside this zone. No significant difference was observed between muscle and soil 137Cs levels, but significant correlations were observed between muscle 137Cs activity concentrations and body length and body weight in the low-activity-concentration season, but not between all seasons and the high-activity-concentration seasons. It is considered that the size effects observed during the low-activity-concentration season may be due to factors related to metabolism and changes in food habit. This is the first long-term survey of 137Cs in wild boar inside the DRZ.

We aimed to reveal the dispersal and gene flow of the local wild boar (Sus scrofa) population and find their genetic boundary in Fukushima Prefecture. After the nuclear incident in 2011, the land was considered a difficult-to-return zone, and the increase in the number of wild boars was pronounced. To provide an effective management strategy for the wild boar population, we used multiplexed inter-simple sequence repeat genotyping by sequencing (MIG-seq) and clarified the genetic structure of wild boars. We obtained 328 single-nucleotide polymorphisms from 179 samples. STRUCTURE analysis showed that the most likely number of population cluster was K = 2. Molecular analysis of variance showed significant genetic differences between groups of wild boars inhabiting in the east and west across the Abukuma River. The migration rate from the eastern population to the western population is higher than in the reverse case based on BayesAss analysis. Our study indicates that both the Abukuma River and anthropogenic urbanization along the river may affect the migration of wild boars and the population in western was established mainly by the migration from other neighboring prefectures.

Self-assembly properties and diversity in higher-order structures of DNA enable programmable tools to be used to construct algorithms at the molecular level. However, the utility of DNA-based programmable tools is hampered by the low orthogonality to natural nucleic acids, especially in complex molecular systems. To address this challenge, we report here the orthogonal regulation of DNA self-assembly by using an unnatural base pair (UBP) formation. Our newly designed UBP AnN:SyN is formed in combination with anti and unusual syn glycosidic conformation with high thermal stability and selectivity. Furthermore, AnC worked as a pH-sensitive artificial nucleobase, which forms a strong base pair with cytosine under a weak acidic condition (pH 6.0). The orthogonal AnN:SyN base pair functioned as a trigger for hybridization chain reaction to provide long nicked double-stranded DNA (ca. 1000 base pairs). This work represents the first example of the orthogonal DNA self-assembly that is nonreactive to natural four-letter alphabets DNA trigger and expands the types of programmable tools that work in a complex environment.

The health effects associated with chronic low-dose, low-dose rate (LD-LDR) exposures to environmental radiation are uncertain. All dose-effect studies conducted outside controlled laboratory conditions are challenged by inherent complexities of ecological systems and difficulties quantifying dose to free-ranging organisms in natural environments. Consequently, the effects of chronic LD-LDR radiation exposures on wildlife health remain poorly understood and much debated. Here, samples from wild boar (Sus scrofa leucomystax) and rat snakes (Elaphe spp.) were collected between 2016 and 2018 across a gradient of radiation exposures in Fukushima, Japan. In vivo biomarkers of DNA damage and stress were evaluated as a function of multiple measurements of radiation dose. Specifically, we assessed frequencies of dicentric chromosomes (Telomere-Centromere Fluorescence in situ Hybridization: TC-FISH), telomere length (Telo-FISH, qPCR), and cortisol hormone levels (Enzyme Immunoassay: EIA) in wild boar, and telomere length (qPCR) in snakes. These biological parameters were then correlated to robust calculations of radiation dose rate at the time of capture and plausible upper bound lifetime dose, both of which incorporated internal and external dose. No significant relationships were observed between dicentric chromosome frequencies or telomere length and dose rate at capture or lifetime dose (p value range: 0.20-0.97). Radiation exposure significantly associated only with cortisol, where lower concentrations were associated with higher dose rates (r2 = 0.58; p < 0.0001), a relationship that was likely due to other (unmeasured) factors. Our results suggest that wild boar and snakes chronically exposed to LD-LDR radiation sufficient to prohibit human occupancy were not experiencing significant adverse health effects as assessed by biomarkers of DNA damage and stress.

Testosterone is an important regulator of male reproduction in terms of spermatogenesis and physiological changes in the accessory reproductive organs. Despite the important role of the testosterone level in male reproduction, testosterone levels vary among male bears even during the breeding season, and the causative underlying factors remain unknown. We postulated that testosterone levels are higher during the breeding season than during other seasons in free-ranging male bears, and that testosterone levels increase with advancing age, larger body size, and better nutritional status. We assessed potential factors (season, time of sampling, body condition index, head circumference, and age) associated with the testosterone level in 80 blood samples collected from free-ranging Asian black bears (Ursus thibetanus) captured in barrel traps in the Ashio-Nikko Mountains and Okutama Mountains in central Honshu Island, Japan. The plasma testosterone level was higher during the breeding season (May-July) than during the non-breeding season (August-November). The body condition index was significantly and positively associated with the plasma testosterone level. None of the other factors were significantly associated with the plasma testosterone level. Therefore, the body condition index may be essential for maintaining high plasma testosterone levels. These findings imply that testosterone secretion might be activated in free-ranging males during the breeding season, and animals with good nutritional status may be able to maintain high plasma testosterone levels. This study uncovered an association between nutritional conditions and reproductive activity in male bears.

After the Tokyo Electric Power Company Fukushima Dai-ichi Nuclear Power Station accident, wild boar was found to have greater radiocesium (137Cs) activity concentrations in their bodies than other wild animals in Japan; however, the reason for this remains unknown. To understand the mechanism of 137Cs transfer from the environment to wild boar, and the factors that affect variation in 137Cs contamination in wild boar, we sampled muscle and stomach contents from wild boar captured in Fukushima Prefecture and analyzed the relationships among 137Cs concentrations in muscle tissue and in the stomach contents, 137Cs ground deposition at capture sites, and wild boar food habits. Significant positive relationships were observed among 137Cs activity concentrations in muscle and stomach contents, as well as 137Cs deposition density at capture sites. These results suggest that 137Cs is transferred from the environment to plant and animal materials consumed by wild boar, and then from these foods to the bodies of wild boar through digestion. However, no correlation was observed between 137Cs concentrations in stomach contents and the presence of any particular food item in stomachs of wild boar, including mushrooms. These findings suggest mushrooms and underground food items, which were found to affect 137Cs concentrations in wild animals in Europe, were not important contributors to high levels of 137Cs contamination in Japanese wild animals.

After the accident at the TEPCO Fukushima Daiichi Nuclear Power Plant in 2011, it became important to study radiation dynamics, assess internal radiation exposure and specify factors affecting radionuclide variation in wildlife. Therefore, it is necessary to investigate which physicochemical fractions of radiocaesium (137Cs) are absorbed from ingested material in species with high activity concentrations of 137Cs, such as wild boar. This study analysed the physicochemical fractions of 137Cs in the stomach contents of wild boar to evaluate the transfer from ingested food to muscle. The 137Cs activity concentration in muscle showed a significantly positive relationship with the 137Cs activity concentration in the exchangeable fraction, and the sum of the 137Cs activity concentrations in the exchangeable and bound to organic matter fractions. Seasonal variations were also found in the 137Cs activity concentration in the exchangeable fraction, and the sum of the 137Cs activity concentrations in the exchangeable and bound to organic matter fractions. These findings suggest that the proportions of the physicochemical fractions of 137Cs in the exchangeable and bound to organic matter fractions in the stomach contents are important factors affecting the increases and seasonal dynamics of the activity concentrations of 137Cs in wild boar muscle.

This study was conducted to assess radiocesium accumulation in birds after the accident at Tokyo Electric Power Company's Fukushima Daiichi Nuclear Power Station in 2011, with a particular focus on 137Cs, which has a long physical half-life. Results of 137Cs monitoring in four game bird species including two pheasant species and two duck species (copper pheasant, green pheasant, spot-billed duck and mallard) were assessed in Fukushima Prefecture. We also obtained samples from rescued raptors that died during treatment or rehabilitation at the Wildlife Symbiosis Centre in Fukushima Prefecture because of severe injury. We measured the muscle concentrations of 137Cs in four of these raptor species (black kite, northern goshawk, peregrine falcon and ural owl). Comparison of the two pheasant species showed that the copper pheasants inhabiting forested areas had higher 137Cs concentrations in muscle (Bq/kg, fresh mass) than the green pheasants inhabiting mountainous areas near human habitation (i.e., Satoyama). No clear tendencies were observed in 137Cs concentration in muscle of copper pheasants over time, but a tendency to decrease was observed in green pheasants over time. The difference in tendencies between species may be attributable to differences in their food habits and its 137Cs concentration, and also differences in the situation of 137Cs accumulation in their habitat. No significant differences were observed in 137Cs concentration in muscle between the resident spot-billed duck and migratory mallard because of the comparatively short biological of effective half-life of radiocesium. Analysis of 137Cs concentration in muscle of raptor revealed that the concentration was similar to, or lower than, those of pheasants and ducks.

To elucidate and reduce the risk of radionuclide contamination in wildlife caused by the Tokyo Electric Power Company Fukushima Dai-ichi Nuclear Power Station accident, it is important to understand radionuclide variations in the wild animal population. Here, we used environmental monitoring data and muscle samples collected from Asian black bear (Ursus thibetanus) and wild boar (Sus scrofa) from May 2011 to March 2016 to examine seasonal variation in radiocesium (137Cs) concentrations in muscle tissues (hereafter, muscle 137Cs) of these important game species in Fukushima Prefecture. We measured muscle 137Cs of bears and wild boars killed by hunters or in animal control culls. First, using a linear mixed model (LMM), we tested for a positive relationship between muscle 137Cs and 137Cs in the soil at the site of capture (hereafter, soil 137Cs) estimated from a soil 137Cs deposition map produced by the Japan Atomic Energy Agency. In the LMM, muscle 137Cs was positively related to estimated soil 137Cs, which corroborates the results of previous studies. The LMM regression coefficients differed between the two species, with wild boar muscle 137Cs being higher than that of bears sampled at the same locations. We then employed a generalized additive mixed model (GAMM) to estimate seasonal variation in the muscle 137Cs of the target species. GAMM showed that muscle 137Cs varied seasonally and that this seasonal variation also differed between the two species. In bears, muscle 137Cs decreased from spring to early autumn, before increasing in winter. Wild boar muscle 137Cs remained low during spring and summer and was high during autumn and early spring. These patterns are likely influenced by differences in diet, habitat use, and physiology between these two species.

The present study aimed to investigate the nutritional aspects of the bear diet quantitatively, in order to understand plant food selection in spring. Bears were observed directly from April to July in 2013 and 2014, to visually recognize plant species consumed by bears, and to describe the foraging period in the Ashio-Nikko Mountains, central Japan. Leaves were collected from eight dominant tree species, regardless of whether bears fed on them in spring, and their key nutritional components analyzed: crude protein (CP), neutral detergent fiber (NDF), and total energy. Bears tended to consume fresh leaves of specific species in May, and nutritional analysis revealed that these leaves had higher CP and lower NDF than other non-food leaves. However, CP in consumed leaves gradually decreased, and NDF increased from May to July, when the bears' food item preference changed from plant materials to ants. Bears may consume tree leaves with high CP and low NDF after hibernation to rebuild muscle mass.
