RESUMO

Prednisolone in therapeutic concentrations blocks Ca(2+) channels of lymphocyte plasma membranes and prevents arachidonic acid-induced Ca(2+) entry into the cells. Glucocorticoid virtually did not modulate arachidonic acid-stimulated release of Ca(2+) from intracellular stores. No appreciable effect of the hormone on mitogen-induced changes in the intracellular content of cAMP was detected.

RESUMO

We studied the effect of polyvinylpyrrolidone-immobilized cortisol on the distribution of glucocorticoid-receptor complexes in hepatocytes. Immobilized cortisol 1.2-1.5-fold increased the number of glucocorticoid-receptor complexes associated with the nuclei in a dose-dependent manner. The transport of glucocorticoid-receptor complexes into hepatocyte nuclei induced by immobilized cortisol is associated with their activation. Moreover, the ratio between nuclear pools of glucocorticoid-receptor complexes resistant to 0.2 and 0.4 M KCl extraction was changed. It was concluded that glucocorticoid binding to specific plasma membrane sites induced further stages of interaction between the hormone and competent cell.

RESUMO

Glucocorticoid-sensitive alkaline proteases are localized in the nuclear and mitochondrial fractions of rat thymocytes. Dexamethasone load increased alkaline protease activity in the nuclear fraction and decreased or even eliminated it in the mitochondrial fraction. The inhibitory analysis showed that SH groups and disulfide bonds play a crucial role in the functioning of glucocorticoid-activated nuclear alkaline protease. Hence, this enzyme can be assigned to as cysteine proteases. Mitochondrial alkaline protease is a serine hydrolase, although it does not belong to the class of trypsin- or chymotrypsin-like enzymes. The role of alkaline proteases in apoptotic death of thymocytes is discussed.

By means of the method of potential-sensitive fluorescent probes is established that under dexametazone-induced apoptosis (0.1-10 microM) the reduction of transmembrane potentials (TMP, delta phi) on plasmatic (delta phi p) and mitochondrial (delta phi m) membranes of hormone-sensitive thymocytes is observed. Dexametazone does not changes transmembrane potentials of glucocorticoid-resistant thymocytes. Ca-ionophore A23187 realistically reduces (at the average on 26%) value of plasmatic TMP both in glucocorticoid-sensitive and glucocorticoid-resistant thymocytes. Change of delta phi is one of the mechanisms to realization an apoptotic process in rats' thymocytes. Dynamics and nature of TMP changes depend on the agent-inductor of apoptosis.

RESUMO

The preliminary characterization of glucocorticoid-sensitive alkaline proteinases of rats thymus and liver was carried out. The role of active serine and cysteine residues in proteolysis was estimated. The influence of Ca++, Mg++, Fe++, Fe , Cu++, EDTA and EGTA on enzymes activity was studied. It is shown that the alkaline proteinases of thymus and liver have similar properties, but are not completely identical.

RESUMO

The effect of hydrocortisone on the thymus and liver proteinases activity in the alkaline pH range was studied on rats. It is shown that the alkaline proteinases are activated in the thymus and inhibited in the liver of hormone-treated animals. Possible relationship between the effect of glucocorticoids on protein metabolism and alkaline proteinases activity is suggested.

RESUMO

The effect of single and chronic administration of hydrocortisone on the thymus proteinase activity in the alkaline pH range was studied on rats. The presence in the thymus of the glucocorticoid-sensitive alkaline proteinase system induced by hydrocortisone was shown. A decrease of the activity of the system in the adrenalectomized animals was found. The possible relationship between the thymolytic effects of hydrocortisone and the increased activity of the alkaline proteinase systems of the thymus is concluded.

The effect of adenosine (10(-6) M) on the cyclic AMP production in lymphocytes of peripheral blood of patients with bronchial asthma and healthy donors was studied. Cells response to adenosine-induced stimulation was significantly decreased for asthmatic patients as compared to normal subjects. Cortisol (3 x 10(-6) M) was shown to be able to normalize suppressed function of cells of the patients. Adenosine (10(-6) M) changed the distribution of hormone-receptor complexes in lymphocytes of the healthy donors to cause increment of the number of 3H cortisol-receptor complexes bound by nucleus. The treatment of lymphocytes of patients with adenosine didn't give any change in specific nuclear 3H cortisol binding. The obtained results may be used for a rational choice of hormonal therapy.

To evaluate the potentialities of the spin probe method in examination of the blood plasma state, a study was made into the blood plasma bound nitroxyl radical 2,2,4,4-tetramethyl-1,2,3,4-tetrahydro-5,6-benzo-gamma-carbolin-3-oxyl in healthy rabbits and those with pronounced experimental atherosclerosis. The probe absorption by high molecular plasma components was assayed in order to determine the binding constants and concentration of the binding centers, mobility of adsorbed probes and polarity of their surroundings. In the course of experimental atherosclerosis development these parameters underwent changes correlating with those of blood plasma cholesterol level. The data obtained suggest that the spin probe method may be used for assessement of the plasma state for diagnostic and research purposes.

The data obtained from the study of interaction of two steroids with serum albumin shows that D-ring of steroid molecule has more hidrophobic environment then A-ring in the complexes of steroid with protein. The plane of steroid is perpendicular to the long axis of protein molecule.

RESUMO

Binding of 17beta-estradiol with histones of calf thymus and the influence of the hormone on heart denaturing of partially reconstructed nucleohistones were investigated. By the degree of the hormone binding histone fractions could be arranged in the following order: F2a greater than F3 greater than F1 greater than or equal F2b. Addition of 17beta-estradiol gave rise to increase of the first and a corresponding decrease of the second melting phase of partially reconstructed nucleohistones F2a, F3 and F2b. Addition of the hormone to the partially reconstructed nucleohistone F1 gave rise to the opposite changes in the heat denaturing profile.
