RESUMO

We configured a long-distance ranging apparatus to test the principle of dual-comb time-of-flight measurement using ultrashort lasers. Emphasis was given to the evaluation of open-air performance quantitatively in terms of the measurement resolution and stability. The test results revealed that our dual-comb asynchronous optical pulse sampling permits micrometer-resolved ranging with a repeatability of 2.05 µm over a 648 m distance in dry weather conditions. Further atmospheric effects were evaluated in three different weather conditions with corresponding Allan deviations. Finally, the capability of simultaneous determination of multiple targets was verified with the potential of advanced industrial applications, such as manufacturing, surveying, metrology, and geodesy.

High-damping rubber composites were prepared by mixing ethylene propylene diene monomer rubber (EPDM) with slide-ring (SR) materials using a two-roll mill, followed by a compression molding technique. SR material has a novel supramolecular structure with unique softness and slidable crosslink junctions. The mechanical strength, thermal stability, compression set property, and damping performance of the composites were investigated. The use of the high damping SR phase dispersed in the EPDM matrix displayed improved physical properties and damping performance compared to those of virgin rubber. As SR content increases in the composites, the damping factor of SR/EPDM blends becomes higher at room temperature. In addition to this, the SR composites showed excellent improvements in the compression set properties. The composites showed a compression set improvement of 35-38% compared to virgin EPDM. These improvements are due to the "pulley effect" of slide-ring materials. Therefore, these materials present a robust platform for making novel elastomer composites for high-performance damping and sealing applications.

The development of smart elastomeric materials with inherent self-repairing abilities after mechanical damage has important technological and scientific implications, particularly in regard to the durability and life cycle of rubber products. The interest in self-healing materials for automotive applications is rapidly growing along with the increasing importance of vehicle scratch quality and quantity. The creation of a reversible network by noncovalent ionic cross-linking in elastomer/rubber blends is an effective approach to generate the self-healing phenomenon, with reprocessing and recycling properties. In this work, thermoplastic vulcanizates (TPVs) were prepared using ethylene-propylene-diene (EPDM) polymers and high-acid-containing thermoplastic ionomers. Along with the general EPDM, maleic anhydride grafted EPDM (EPDM-g-MAH) was also used for the preparation of the TPVs. The strategy was based on a simple ionic crosslinking reaction between the carboxyl groups present in the ionomer and zinc oxide (ZnO), where the formation of reversible Zn2+ salt bondings exhibits the self-healing behavior. The heterogeneous blending of EPDM and ionomers was also used to investigate the thermal and mechanical properties of the TPVs. The experimental findings were further supported by the surface morphology of the fracture surfaces viewed using microscopy. The self-healing behavior of the TPVs has been identified by scratch resistance testing, where the EPDM-g-MAH TPVs showed excellent healing efficiency of the scratch surface. Therefore, this work provides an efficient approach to fabricate new ionically cross-linked thermoplastic vulcanizates with excellent mechanical and self-repairing properties for the skins of automotive interior door trims and instrument panel applications.

Although prime editing is a promising genome editing method, the efficiency of prime editor 2 (PE2) is often insufficient. Here we generate a more efficient variant of PE2, named hyPE2, by adding the Rad51 DNA-binding domain. When tested at endogenous sites, hyPE2 shows a median of 1.5- or 1.4- fold (range, 0.99- to 2.6-fold) higher efficiencies than PE2; furthermore, at sites where PE2-induced prime editing is very inefficient (efficiency < 1%), hyPE2 enables prime editing with efficiencies ranging from 1.1% to 2.9% at up to 34% of target sequences, potentially facilitating prime editing applications.

Multilateration tracking systems (MLTSs) are used in industrial three-dimensional (3D) coordinate measuring applications. For high-precision measurement, system parameters must be calibrated properly in advance. For an MLTS using absolute distance measurement (ADM), the conventional self-calibration method significantly reduces estimation efficiency because all system parameters are estimated simultaneously using a complicated residual function. This paper presents a novel self-calibration method that optimizes ADM to reduce the number of system parameters via highly precise and separate estimations of dead paths. Therefore, the residual function to estimate the tracking station locations can be simplified. By applying a suitable mathematical procedure and solving the initial guess problem without the aid of an external device, estimation accuracy of the system parameters is significantly improved. In three self-calibration experiments, with ADM repeatability of approximately 3.4 µm, the maximum deviation of the system parameters estimated by the proposed self-calibration method was 68.6 µm, while the maximum deviation estimated by the conventional self-calibration method was 711.9 µm. Validation of 3D coordinate measurements in a 1000 mm × 1000 mm × 1000 mm volume showed good agreement between the proposed ADM-based MLTS and a commercial laser tracker, where the maximum difference based on the standard deviation was 17.7 µm. Conversely, the maximum difference was 98.8 µm using the conventional self-calibration method. These results confirmed the efficiency and feasibility of the proposed self-calibration method.

Ultrashort pulse lasers are emerging as an advanced tool of distance measurement, with their unique temporal and spectral characteristics being extended to diverse principles of absolute ranging and instrumentation. Here, a systematic methodology is presented for absolute ranging by means of the time-of-flight measurement of ultrashort light pulses using dual-comb asynchronous optical sampling. Based on an elaborate uncertainty analysis, influencing system parameters such as the pulse duration, repetition rate, and averaging time are optimized to achieve a sub-µm measurement accuracy. The absolute ranging system developed in this study demonstrates a combined standard uncertainty of 0.986 µm for a 0.5 ms averaging over a distance range of 3.0 m, with a further reduction to 0.056 µm when the averaging time is increased to 0.5 s. The outstanding performance leads to unprecedented multitarget applications: machine feed control with thermal error compensation in real time as well as the nondestructive inspection of multilens assembly in a production line.

In this study, we have investigated the recycling of flexible polyurethane foam (PUF) using an ultrasonic reactor. Waste PUF of automotive seat cushions were subjected to ultrasonic horn oscillation of various amplitudes at different flow rate. Successful decrosslinking of ultrasonically treated waste PUF was confirmed by thorough characterizations such as gel fraction, crosslink density, and thermal property. Decrosslinked PUFs were blended with the virgin PUF in various proportions to prepare the recrosslinked PUFs. Automotive seat cushions made of the blended PUFs exhibit enhanced comfort than that of the virgin PUFs by lowering the hardness and hysteresis loss and increasing the sag factor.

Inspired by the epidermis-dermis composition of human skin, here we have simply developed a lightweight, robust, flexible, and biocompatible single-electrode triboelectric nanogenerator (S-TENG)-based prototype of bilayer artificial skin, by attaching one induction electrode with unfoamed skin layer of microcellular thermoplastic polyurethane (TPU) foam, which shows high-performance object manipulation [by responding differently toward different objects, viz., aluminum foil, balloon, cotton glove, human finger, glass, rubber glove, artificial leather, polyimide, poly(tetrafluoroethylene) (PTFE), paper, and wood], due to electrification and electrostatic induction during contact with the objects having different chemical functionalities. Comparative foaming behavior of ecofriendly supercritical fluids, viz., CO2 over N2 under variable temperatures (e.g., 130 and 150 °C) and constant pressure (15 MPa), have been examined here to pursue the soft and flexible triboelectric TPU foam. The foam derived by CO2 foaming at 150 °C has been prioritized for development of S-TENG. Foam derived by CO2 foaming at 130 °C did not respond as well due to the smaller cell size, higher hardness, and thicker skin. Inflexible N2-derived foam was not considered for S-TENG fabrication. Object manipulation performance has been visualized by principal component analysis (PCA), which shows good discrimination among responses to different objects.

Growth impairment (GI) is one of the adverse effects of dexamethasone (DXM), and growth hormone (GH) has been used clinically to improve GI. The present study aimed to evaluate the manner in which DXM disturbs the growth rate of longitudinal bones, and the recovery effects of GH on DXM-induced GI in the longitudinal bones of adolescent male rats. In the first experiment, DXM (0, 0.5, 1, 2 and 5 mg/kg) was administered subcutaneously to identify a potential dose-dependent activity and calculate the median effective dose (ED50) of DXM-induced GI. The ED50 was identified to be 1.15 mg/kg. In the second experiment, GH (0, 2.5, 5 and 10 mg/kg) with 1.15 mg/kg DXM was injected subcutaneously to assess the recovery effects of GH on DXM-induced GI. The growth rates of the longitudinal bones, total height of the growth plate, local mRNA expressions of insulin-like growth factor 1 (IGF-1), GH receptor (GHR) and IGF-1 receptor (IGF-1R), and local protein expression of IGF-1 were measured to evaluate the recovery effects of GH on DXM-induced GI. The local expressions of IGF-1, GHR and IGF-1R mRNA, and IGF-1 protein were measured using quantitative polymerase chain reaction following laser microdissection and antigen-specific immunohistochemistry, respectively. GH administration partially recovered DXM-induced GI in the longitudinal bones and growth plate. GH significantly increased the levels of IGF-1, GHR and IGF-1R mRNA in the proliferative zone of the control group (P<0.05), whereas it failed to increase them in the proliferative zone of the DXM-treated group. Furthermore, GH increased the levels of IGF-1, GHR and IGF-1R mRNA in the hypertrophic zone of both the vehicle and DXM-treated groups (P<0.05). Immunohistochemical analysis of IGF-1 protein expression revealed a similar pattern to that of IGF-1 mRNA. These results suggest that increased GH insensitivity in the proliferative zone of the growth plate, induced by DXM, leads to GI in longitudinal bones. Thus, combined administration of GH with GH insensitivity-alleviating medications may be more effective in the treatment of DXM-induced GI.

We report a method of calculating the motion error of a linear motion bearing stage. The transfer function method, which exploits reaction forces of individual bearings, is effective for estimating motion errors; however, it requires the rail-form errors. This is not suitable for a linear motion bearing stage because obtaining the rail-form errors is not straightforward. In the method described here, we use the straightness errors of a bearing block to calculate the reaction forces on the bearing block. The reaction forces were compared with those of the transfer function method. Parallelism errors between two rails were considered, and the motion errors of the linear motion bearing stage were measured and compared with the results of the calculations, revealing good agreement.

In order to enhance the electron transport on the photoelectrodes of dye-sensitized solar cells, one-dimensional rutile nanorods were prepared using electrospun TiO2 nanofibers. The grain size of the nanorods increased with increasing temperature. Electrochemical impedance spectroscopy measurements revealed reduced interface resistance of the cells with the one-dimensional rutile nanorods due to the improved electron transport and the enhanced electrolyte penetration. Intensity-modulated photocurrent/photovoltage spectroscopy showed that the one-dimensional rutile nanorods provided the electrons with a moving pathway and suppressed the recombination of photogenerated electrons. However, an excessive quantity of rutile nanorods created an obstacle to the electrons moving in the TiO2 thin film. The photoelectrode with 7 wt.% rutile nanorods optimized the performance of the dye-sensitized solar cells.

In this study, we used a bacteriophage λQ⁻S⁻ mutant that increased the stability of recombinant Escherichia coli during continuous culture. The operation was conducted in two stages: the first stage was carried out to promote cell growth, and the second stage was performed for product formation. The productivity of recombinant proteins depends on the substrate concentration of the fresh medium supplied to the second stage (S3) and dilution rate of the second stage (D2). With the optimal value of S3 and D2, the first and second stages were stably maintained for 170 and 80 h, respectively. To further improve this process, a three-stage continuous process was conducted with an additional induction stage between the growth and production stages. Compared with the two-stage operation, the stable production period was extended by 1.7 fold, and the recombinant protein production increased by 1.3 fold.

We discuss two ways to use femtosecond pulsed lasers as a new interferometric light source for enhanced precision surface-profile metrology. First, a train of ultrafast laser pulses yields repeated low temporal coherence, which allows unequal-path scanning interferometry, which is not feasible with white light, to be performed. Second, the high spatial coherence of femtosecond pulsed lasers enables large-sized optics to be tested in nonsymmetric configurations with relatively small-sized reference surfaces. These two advantages are verified experimentally with Fizeau and Twyman-Green type scanning interferometers.

A two-stage continuous culture of Escherichia coli in combination with a bacteriophage lambda system was performed in order to overcome the intrinsic plasmid instability that is frequently observed in recombinant fermentation. A phage lambda vector with a Q(-) mutation was used to enhance the expression of the lambda system. The optimal values of the important operational variables such as the substrate concentration, the dilution rate, and the mean residence time on the expression of the cloned gene were determined in both batch and continuous cultures. For all culturing modes, the full induction of the cloned gene was observed 4 h after the temperature shift. In the two stage continuous culture, the overproduction reached their maxima at D=0.25 h(-1) with 1.5 S(0) of the medium supply. The maximum productivity of the total beta-galactosidase was 16.3x10(6) U l(-1) h(-1), which was approximately seven times higher than that in the single-copy lysogenic stage. The recombinant cells were stable in the lysogenic state for more than 260 h, while they were stable for 40 h in the lytic state. The instability that developed rapidly in the second tank is believed to be due to the accumulation of lysis proteins as a result of vector leakage during the operation.
