Sixty patients with a chronic Opisthorchis felineus infection were administered one-day therapy with praziquantel in doses 25, 40, or 60-75 mg/kg b. m. The former two doses of the drug did not much improve the levels of the examined immunologic parameters. In patients treated with the highest dose of praziquantel a significant decrease of the total and specific IgE and CIC levels, reaching that in the reference groups, was observed in 6-8 months after the treatment, this indicating a 92% efficacy of the drug in this group of patients.

The effects of various metal alloys on the oral fluid levels of IgA, serum IgA and IgG, and IgE of 115 subjects aged 20 to 60 were under study. Mixed salivary IgG levels were measured before and in various periods after fixation of dentures made of different alloys. IgA levels failed to normalize even in a year after fixation of dentures made of steel and titanium nitrite-coated steel, though IgG levels were unchanged in the same patients.

During a remission and drug therapy, 71 children suffering from allergic diseases were vaccinated with DT-M toxoid containing different dose of diphtheria (1, 2.5, 5 LF) and tetanus (1, 2.5, 5 EC) antigens. It has been shown that all the children responded to tetanus antigen by adequate serological tests, whereas as regards diphtheria toxoid, the patients immunized with the least dose of DT-M preparation (1 LF + + 1 EC antigens) produced specific antibodies later and at a lower level. During the postvaccinal period, they also manifested an increase of the content of IgE and exacerbation of the underlying disease in 17.8% of cases. The use of DT-M toxoid with 2.5 LF + EC antigens was shown to hold promise in vaccination of children suffering from allergic diseases.

45 patients with Opisthorchis felineus infection, 8 in the acute and 37 in the chronic stage of the disease of moderate severity or mild course were treated with praziquantel dosed 25-60 mg per kilo for one-day course. Only in 4 out of 45 patients egg output was more than 1000 per gram of feces (Kato-Katz method). Blood hypereosinophilia, elevated serum IgE and CIC content (215 +/- 56 against 76 +/- 19 in control) in the acute and the tendency to elevation of IgE content with the chronic opisthorchiasis before treatment were found. Tolerance of the treatment was satisfactory. Six-eight months later in patients with acute opisthorchiasis the content of serum IgE was significantly lower but still three times higher than normal. The other parameters in both groups of patients were normal.

The determination of the parameters of cellular immunity in children with hepatitis A has demonstrated unbalance on the part of the cellular defence mechanisms which appeared more pronounced in patients suffering from hepatitis A combined with opisthorchiasis. This determines the clinical picture characteristics in patients with mixed infection.

The sensitivity and specificity of an enzyme immunoassay (EIA) system for the determination of total IgE in humans increases if polystyrene plates are sensitized with swine gamma-globulin and the conjugate is prepared with the use of sheep gamma-globulin obtained from the commercial preparation of sheep antiserum to human IgE. Such system becomes even more specific and sensitive if sheep antibodies to human IgE, purified by affinity chromatography, are used both for the sensitization of polystyrene plates and for the preparation of the conjugate. This conjugate is necessary for the development of EIA systems intended for the determination of specific human IgE-antibodies to various allergens.

A comparative study of local and humoral immunity in patients with increased sensitivity to ragweed pollen and in healthy persons has been carried out. In untreated patients a six-fold increase of the levels of total IgE and specific anti-ragweed IgE-antibodies in the blood sera and secretions has been registered, which corresponds to the 3rd and 4th grade reaction in the enzyme immunoassay; simultaneously, a decrease in the characteristics of immune responsiveness has been observed. As the levels of total IgE and specific IgE-antibodies are of particular diagnostic value in immediate-type allergic diseases, the authors have examined the patients with ragweed pollenosis at the periods of the absence of clinical manifestations and in the exacerbation of the disease during blossoming with pollen formation. During exacerbation the level of immunoglobulins of all classes, specific IgE-antibodies in the blood sera and secretions of patients, as well as some characteristics of immune responsiveness, are higher than in remission, which is indicative of the increased antigenic stimulation of the body and of the developing immune response.

An ELISA system for the detection of allergen-specific IgE antibodies to ragweed allergen has been developed. The system is highly sensitive and specific. Ragweed pollen allergen has been obtained by the dialysis of water-soluble extract through a kidney membrane. The high molecular fraction of ragweed allergen, showing the whole of the allergenic activity detected by skin tests in untreated patients, has been used for coating polystyrene assay plates. To detect IgE antibodies to ragweed allergen, the conjugate of sheep anti-IgE antibodies with horse-radish peroxidase has been used. The level of allergen-specific IgE antibodies has been determined on the basis of the data on the optical density of the samples in comparison with that of the normal sera. The correlation factor of the results obtained in the assay of specific IgE antibodies with the newly developed assay system and with the commercial kit Phadezyme RAST manufactured by Pharmacia AB (Sweden) has proved to be 0.82 at n = 39, p less than 0.01, while the variation factor in the reproduction of the assay results has proved to be 12% at n = 40.

[Development of a Soviet immunoenzyme test system for the quantitative determination of human immunoglobulin E]. / Razrabotka otechestvennoi immunofermentnoi test-sistemy dlia kolichestvennogo opredeleniia immunoglobulina E cheloveka.

An EIA system for the quantitative determination of human IgE was developed. As the solid phase, polystyrene microplates sensitized with the gamma globulin fraction of sheep antiserum to human IgE were used in this system. Peroxidase conjugate with IgE was prepared with the use of periodate technique. The optimum parameters for the sorption of antibodies on polystyrene were determined: protein concentration, the pH of the buffer, temperature, the time of fixation, the working dilution of the conjugate. As the substrate of peroxidase activity, ortho-phenilenediamine solution with hydrogen peroxide was used. The result was evaluated by means of a photometer, model Titertek Multiskan MC (Flow Laboratories Ltd., Great Britain), at the wavelength 429 nm. The new EIA system proved to be specific and detected IgE in the sera under investigation at a concentration of 10(-9) g/ml. The system permitted the determination of the normal level of IgE in a group of healthy adults, this level being, on the average, 76 +/- 9 kU/1.
