RESUMO

Crucian carp (Carassius carassius) is an important aquatic economic animal, and the immune barrier function of its intestine has been a focus of research into oral vaccines and drugs. However, the histological structures of the intestinal barrier and its adjacent areas have not been clearly established, and little subcellular evidence is available to elucidate the spatial distribution of intracellular biological processes. In this study, the spatial distribution of autophagy and endosome formation in the intestinal epithelial cells (IECs) of crucian carp were analyzed. These two biological activities are closely related to intestinal homeostasis, immunity, and cell communication. Periodic acid-Schiff (PAS) and Masson's trichrome staining were employed to elucidate the distinctive histological framework of the Crucian carp's myoid cell network, which resides within the subepithelial layer and is characterized by gap junctions. Transmission electron microscopy (TEM), immunohistochemistry (IHC), and immunofluorescence (IF) were used to detect the structural and functional aspects of the IEC in different intestinal segments. TEM and immunohistochemical analyses captured the biogenesis and maturation of early and late endosomes as well as multivesicular bodies (MVBs), as well as the initiation and progression of autophagy, including macroautophagy and mitophagy. The endosome and MVBs-specific marker CD63 and autophagy-related protein LC3 were highly expressed in IECs and were correlated with autophagy and endosome biosynthesis in the apical and basal regions of individual cells, and differed between different intestinal segments. In summary, this study elucidated the ubiquity and morphological characteristics of autophagy and endosome formation across different intestinal segments of crucian carp. A unique myoid cell network beneath the intestinal epithelium in crucian carp was also identified, expanding the histological understanding of this animal's intestinal tract.

Ether-based electrolytes are considered as an ideal electrolyte system for sodium metal batteries (SMBs) due to their superior compatibility with the sodium metal anode (SMA). However, the selection principle of ether solvents and the impact on solid electrolyte interphase formation are still unclear. Herein, we systematically compare the chain ether-based electrolyte and understand the relationship between the solvation structure and the interphasial properties. The linear ether solvent molecules with different terminal group lengths demonstrate remarkably distinct solvation effects, thus leading to different electrochemical performance as well as deposition morphologies for SMBs. Computational calculations and comprehensive characterizations indicate that the terminal group length significantly regulates the electrolyte solvation structure and consequently influences the interfacial reaction mechanism of electrolytes on SMA. Cryogenic electron microscopy clearly reveals the difference in solid electrolyte interphase in various ether-based electrolytes. As a result, the 1,2-diethoxyethane-based electrolyte enables a high Coulombic efficiency of 99.9 %, which also realizes the stable cycling of Na||Na3 V2 (PO4 )3 full cell with a mass loading of ≈9 mg cm-2 over 500 cycles.

Allergic contact dermatitis (ACD) is an occupation-dependent skin disease that afflicts humans with recurrent, non-specific episodes. Telocyte (TC) is a novel interstitial cell discovered in recent years and, together with fibroblasts, constitutes the predominant interstitial cell population in the skin. The purpose of this study was to investigate the morphodynamic changes of interstitial cells, especially TCs, in the skin during the development and treatment of ACD by histological and microscopic scientific methods. Hematoxylin-eosin staining, Masson staining, immunohistochemistry (IHC), immunofluorescence (IF), scanning electron microscopy (SEM), and transmission electron microscopy (TEM) were used to track morphodynamic changes in interstitial cells during the development and treatment in the ACD-involved skin induced by 2,4-dinitrochlorobenzene (DNCB). The results demonstrated that TCs were mainly present around dermal collagen fibers, perivascular (except dermal papillary vascular loop), and skin appendages, which expressed CD34+, Vimentin+, PDGFR-α+, and α-SMA-. The absence of TCs during ACD development and after ACD recovery causes dermal interstitial cell dysregulation. The special anatomical relationships between TCs, immune cells, and follicular stem cells were also revealed, suggesting their potential dermatitis-regulating function. In a nutshell, our results provide morphodynamic evidence for the process of ACD development and recovery and offer potential cytological ideas for ACD treatment.

The stable cycling of Mg-metal anodes is limited by several problems, including sluggish electrochemical kinetics and passivation at the Mg surface. In this study, we present a high-entropy electrolyte composed of lithium triflate (LiOTf) and trimethyl phosphate (TMP) co-added to magnesium bis(trifluoromethane sulfonyl)imide (Mg(TFSI)2 /1,2-dimethoxyethane (DME) to significantly improve the electrochemical performance of Mg-metal anodes. The as-formed high-entropy Mg2+ -2DME-OTf- -Li+ -DME-TMP solvation structure effectively reduced the Mg2+ -DME interaction in comparison with that observed in traditional Mg(TFSI)2 /DME electrolytes, thereby preventing the formation of insulating components on the Mg-metal anode and promoting its electrochemical kinetics and cycling stability. Comprehensive characterization revealed that the high-entropy solvation structure brought OTf- and TMP to the surface of the Mg-metal anode and promoted the formation of a Mg3 (PO4 )2 -rich interfacial layer, which is beneficial for enhancing Mg2+ conductivity. Consequently, the Mg-metal anode achieved excellent reversibility with a high Coulombic efficiency of 98 % and low voltage hysteresis. This study provides new insights into the design of electrolytes for Mg-metal batteries.

Plant architecture is a crucial influencing factor of wheat yield and adaptation. In this study, we cloned and characterized TaSPL14, a homologous gene of the rice ideal plant architecture gene OsSPL14 in wheat. TaSPL14 homoeologs (TaSPL14-7A, TaSPL14-7B and TaSPL14-7D) exhibited similar expression patterns, and they were all preferentially expressed in stems at the elongation stage and in young spikes. Moreover, the expression level of TaSPL14-7A was higher than that of TaSPL14-7B and TaSPL14-7D. Overexpression of TaSPL14-7A in wheat resulted in significant changes in plant architecture and yield traits, including decreased tiller number and increased kernel size and weight. Three TaSPL14-7A haplotypes were identified in Chinese wheat core collection, and haplotype-based association analysis showed that TaSPL14-7A-Hap1/2 were significantly correlated with fewer tillers, larger kernels and higher kernel weights in modern cultivars. The haplotype effect resulted from a difference in TaSPL14-7A expression levels among genotypes, with TaSPL14-7A-Hap1/2 leading to higher expression levels than TaSPL14-7A-Hap3. As favorable haplotypes, TaSPL14-7A-Hap1/2 underwent positive selection during global wheat breeding over the last century. Together, the findings of our study provide insight into the function and genetic effects of TaSPL14 and provide a useful molecular marker for wheat breeding.

Selenium nanoparticles (SeNPs) are among the emerging selenium supplements because of their high bioactivity and low toxicity. However, bare SeNPs are prone to activity loss caused by aggregation and sedimentation. This study aims to stabilize SeNPs with curdlan (CUR), a polysaccharide, to maintain or even enhance their biological activity. Herein, the stable SeNPs were constructed via the unique conformational transition of CUR induced by alkali-neutralization (AN) pretreatment. The physicochemical properties and structures of the prepared SeNPs were characterized by dynamic light scattering (DLS), UV-visible spectroscopy, Fourier-transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), transmission electron microscopy (TEM), and free-radical-scavenging activity assays. The results show that most SeNPs are stabilized within the triple helix of CUR that has been pretreated with high-intensity AN treatment. These amorphous, small-sized (average size was 53.6 ± 17.7 nm), and stabilized SeNPs have significantly enhanced free-radical-scavenging ability compared to the control and can be well-stabilized for at least 240 days at 4 °C. This work indicates that CUR, as a food additive, can be used to well-stabilize SeNPs by AN pretreatment and provides a facile method to prepare and enhance the stability and bioactivity of SeNPs via triple-helix conformational transition.

RESUMO

Sodium metal batteries (SMBs) have been widely studied owing to their relatively high energy density and abundant resources. However, they still need systematic improvement to fulfill the harsh operating conditions for their commercialization. In this review, we summarize the recent progress in SMBs in terms of sodium anode modification, electrolyte exploration, and cathode design. Firstly, we give an overview of the current challenges facing Na metal anodes and the corresponding solutions. Then, the traditional liquid electrolytes and the prospective solid electrolytes for SMBs are summarized. In addition, insertion- and conversion-type cathode materials are introduced. Finally, an outlook for the future of practical SMBs is provided.

ABSTRACT: Background and aims: Genipin, an iridoid derived from geniposide by ß-glucosidase hydrolysis, has shown potential benefit in the treatment of heart function insufficiency despite its unclear therapeutic mechanism. This study aimed to investigate the primary cardioprotective mechanism of genipin. We hypothesized that genipin demonstrated the antiapoptosis and anti-inflammation for cardiac protection by inhibiting the cyclooxidase 2 (COX2)-prostaglandin D2 (PGD2) and murine double minute 2 (MDM2)-p53 pathways. Methods: The normal Sprague-Dawley rats were made into myocardial infarction models by conventional methods. Animals were treated with genipin for 5 weeks after myocardial infarction (MI). Morphometric and hemodynamic measurements were performed 5 weeks post-MI. Biological and molecular experiments were performed after the termination. Results: Both morphometry and hemodynamics in systole and diastole were significantly impaired in the model group but restored close to basal level after treatment with genipin. Genipin also restored the post-MI upregulated expressions of cytochrome c, p53, COX2, and PGD2 and downregulated expression of MDM2 to the approximate baseline. Genipin inhibited apoptotic and inflammatory pathways to prevent post-MI structure-function remodeling. Conclusions: This study showed the cardioprotective mechanism of genipin and implied its potential clinical application for the treatment of ischemic heart failure.

The incidence rate of cardiovascular diseases is increasing year by year. The demand for coronary artery bypass grafting has been very large. Acellular blood vessels have potential clinical application because of their natural vascular basis, but their biocompatibility and anticoagulant energy need to be improved. We decellularized the abdominal aorta of SD rats, and then modified with bivalirudin via polydopamine. The mechanical properties, blood compatibility, cytocompatibility, immune response, and anticoagulant properties were evaluated, and then the bivalirudin-modified acellular blood vessels were implanted into rats for remodeling evaluation in vivo. The results we got show that the bivalirudin-modified acellular blood vessels showed good cytocompatibility and blood compatibility, and its anti-inflammatory trend was dominant in the immune response. After 3 months of transplantation, the bivalirudin-modified acellular blood vessels did not easily form thrombus. It was not easy to form calcification and could make the host cells grow better. Through vascular stimulation and immunofluorescence test, we found that vascular smooth muscle cells and endothelial cells proliferated well in the bivalirudin group. Bivalirudin-modified acellular blood vessels provided new idea for small diameter tissue engineering blood vessels, and may become a potential clinical substitute for small-diameter vascular grafts.

Triple-helical aggregates (THAs) have been proven to affect the biological activities and functional properties of triple-helix polysaccharides. Thus, it's urgent to seek a method to reduce the size of THAs while preserving independent triple helices (ITHs). In this study, the effects of alkali-neutralization (AN) treatment on THAs and ITHs of curdlan were studied. The positive values of the Congo red test data (R2>0.99) fitted using a Logistic model indicated that AN treatment (CNaOH/HCl>0.28 mol/L) facilitated the disaggregation of THAs. Congo red test, sedimentation test, and turbidity test showed that AN treatment (CNaOH/HCl = 1.0 mol/L) significantly reduced the size of THAs to approximately 1 µm while effectively increasing the relative amount of ITHs to approximately 199 %. Fourier transform infrared spectroscopy and X-ray diffraction analysis showed that AN treatment basically unchanged the primary structure of curdlan chains, but affected the crystalline structure and the intermolecular and intramolecular hydrogen bonding of curdlan.

BACKGROUND: Ichthyosis with confetti (IWC) is an extremely rare autosomal-dominant genodermatosis characterized by erythroderma with numerous confetti-like pale spots. IWC is caused by mutations in KRT10 (IWC-I) or KRT1 (IWC-II) which affect their tail domains. In IWC-I, the mutations lead to replacement of glycine/serine-rich keratin 10 (K10) tail with arginine- or alanine-rich frameshift motifs, causing K10 mis-localization which might trigger loss of the mutant KRT10 allele via mitotic recombination, leading to genetic reversion. OBJECTIVE: To investigate mutations in five IWC-I patients and their functional consequences. METHODS: We performed Sanger sequencing of KRT1 and KRT10 in peripheral blood samples of five patients, with highly polymorphic KRT10 SNPs genotyped to confirm loss-of-heterozygosity in the epidermis of pale spots. K10 expression pattern was examined in both patient skin biopsies and HaCaT cells overexpressing mutant KRT10-enhanced green fluorescence protein fusion. RESULTS: Four novel and one recurrent KRT10 mutations were identified in patient peripheral blood samples but not in the corresponding pale spot epidermis. Two of the mutations, c.1696_1699dupCACA and c.1676dupG, affected residues close to K10 carboxyl terminus and encoded only 3 and 6 arginine residues, which were far fewer than reported previously. Interestingly, imaging analyses for K10 in HaCaT cells overexpressing either of these two mutations and in the corresponding patients' affected skin, showed a remarkably lower level of K10 mis-localization compared to that of other mutations reported in this study. CONCLUSIONS: Our findings suggest that the number of arginine residues in the mutant tail may correlate with the level of K10 mis-localization in IWC-I keratinocytes. These results expand the genotypic and phenotypic spectrum of IWC-I.

The rapid urbanization of China and associated demand for land resources necessitates remediation, redevelopment, and reclamation of contaminated soil. Before these measures are taken, a basic investigation and inventory of heavy metal (HM) pollution levels in contaminated soil is necessary for establishing and implementing the redevelopment plan. In the present study, to identify the policy implications of inventorying and mapping HM pollution of soil in brownfields throughout China, the Bayan Obo giant rare earth element (REE)-Nb-Fe ore deposit of Baotou in Inner Mongolia, China, which is the largest REE mineral deposit in the world, was taken as a case study. Soil samples from 24 sites in Bayan Obo mining area (MA) and 76 sites in mine tailing area (TA) were collected for determining contents of soil HMs (Cr, Cd, Pb, Cu, and Zn). The results showed that the average concentrations of Cr, Cd, Pb, Cu, and Zn in both MA and TA were all higher than their corresponding background values for Inner Mongolia but lower than the Class II criteria of the National Soil Quality Standards of China (GB 15618-1995). Enrichment factor (EF) analysis of the soil samples indicated that the soil in the brownfield sites was highly enriched with Cr, Cd, Pb, Cu, and Zn compared to the corresponding background values. In MA, the EF for Cd was the highest among the studied elements, while in TA, the EF for Cr (3.45) was the highest, closely followed by the EF for Cd (3.34). The potential ecological risk index (RI) indicated a moderate potential ecological risk from the studied HMs in MA and a low potential ecological risk in TA, and the results of RI also suggested that the soil was most heavily polluted by Cd. According to the spatial distribution maps of HM, contamination hot-spots were primarily located near mining-related high-pollution plants. Based on the results, policy recommendations are proposed related to brownfield management in urban planning.

Mining activities including mineral excavation and ore transportation produce and release large amounts of pollutants to the surrounding environment, raising concerns regarding the effects of environmental exposure to pollutants on human health. The concentration of elements in hair can be used as a biomarker of exposure to chemical elements. In the present study, hair samples from 89 relatively healthy volunteers aged 11-77 years old (57 living near Bayan Obo giant REE-Nb-Fe ore deposit and 32 in non-mining areas) from Baotou (Inner Mongolia, China) were analyzed to determine the contents of 25 chemical elements (Ag, Al, As, Ba, Bi, Ca, Cd, Co, Cr, Cs, Cu, Fe, Ga, Li, Mg, Mn, Mo, Ni, Pb, Rb, Se, Tl, U, V and Zn) with the aim of gaining insights about the potential exposure to chemical elements of the population living within the Bayan Obo opencast mining area. The effects of gender and living area on element concentrations in hair were also investigated. The results showed that the mean concentrations of the target elements (Ag, Al, Ba, Cd, Co, Li, Mn, Ni, Pb, Tl, V, Cs, Ga, Rb, U, Bi, Mo, As, Ca, Fe, Mg and Zn) in human hair from the mining area (MA) were primarily higher than that in non-mining areas (NMA). The area of residence had an influence on the presence of select trace elements (As, Cd, Mg, Pb, Tl, U and Zn) in human hair. In addition, the hair samples from MA showed statistically significant differences between males and females for a larger number of elements than those in NMA in which only Ag, Bi and Se were significantly different. This result implies a potential influence of mining activities on residents living in the vicinity. These findings confirm the need for competent authorities to act as early as possible and to implement strategies aimed to protect exposed populations and the entire ecosystem.

In the current study, we hypothesize that mine (metallicolous) populations of metallophytes form a trade-off between the roots and shoots when under copper (Cu) stress to adapt themselves to heavy metal contaminated habitats, and thus, differ from normal (non-metallicolous) populations in biomass allocation. To test the hypothesis, two populations of the metallophyte Kummerowia stipulacea, one from an ancient Cu mine (MP) and the other from a non-contaminated site (NMP), were treated with Cu(2+) in hydroponic conditions. The results showed that MP plants had higher root/shoot biomass allocation and more complicated root system architecture compared to those of the NMP plants when under Cu stress. The net photosynthetic capacity was more inhibited in the NMP plants than in the MP plants when under Cu stress. The sugar (sucrose and hexose) contents and acid invertase activities of MP plants were elevated while those in NMP plants were inhibited after Cu treatment. The neutral/alkaline invertase activities and sucrose synthase level showed no significant differences between the two populations when under Cu stress. The results showed that acid invertase played an important role in biomass allocation and that the physiological responses were beneficial for the high root/shoot biomass allocation, which were advantageous during adaptive evolution to Cu-enriched mine soils.
