RESUMO

The remarkable radiation of South American (SA) canids produced 10 extant species distributed across diverse habitats, including disparate forms such as the short-legged, hypercarnivorous bush dog and the long-legged, largely frugivorous maned wolf. Despite considerable research spanning nearly two centuries, many aspects of their evolutionary history remain unknown. Here, we analyzed 31 whole genomes encompassing all extant SA canid species to assess phylogenetic relationships, interspecific hybridization, historical demography, current genetic diversity, and the molecular bases of adaptations in the bush dog and maned wolf. We found that SA canids originated from a single ancestor that colonized South America 3.9 to 3.5 Mya, followed by diversification east of the Andes and then a single colonization event and radiation of Lycalopex species west of the Andes. We detected extensive historical gene flow between recently diverged lineages and observed distinct patterns of genomic diversity and demographic history in SA canids, likely induced by past climatic cycles compounded by human-induced population declines. Genome-wide scans of selection showed that disparate limb proportions in the bush dog and maned wolf may derive from mutations in genes regulating chondrocyte proliferation and enlargement. Further, frugivory in the maned wolf may have been enabled by variants in genes associated with energy intake from short-chain fatty acids. In contrast, unique genetic variants detected in the bush dog may underlie interdigital webbing and dental adaptations for hypercarnivory. Our analyses shed light on the evolution of a unique carnivoran radiation and how it was shaped by South American topography and climate change.

This study evaluated ticks and tick-borne agents in 104 captures of the maned wolf Chrysocyon brachyurus (50 different individuals and 54 recaptures) in the Serra da Canastra National Park (SCNP), a Cerrado preserved area in southeastern Brazil, from 2005 to 2012. From the 104 capture events, a total of 1,206 ticks were collected on 94 occasions (90.4 %), and identified into five species: Amblyomma tigrinum (77.3 % of all collected ticks), Amblyomma sculptum (16.6 %), Amblyomma ovale (0.1 %), Amblyomma brasiliense (0.1 %), Rhipicephalus microplus (0.1 %), and Amblyomma spp. larvae (5.8 %). Molecular analyses of A. tigrinum adult ticks revealed the presence of 'Candidatus Rickettsia andeanae', Rickettsia parkeri sensu stricto, two different haplotypes of 'Ca. Midichloria sp.', and a Hepatozoon canis haplotype. Molecular analyses of maned wolf blood samples revealed two distinct haplotypes of Hepatozoon spp., one identical to the H. canis genotype that was detected in the A. tigrinum ticks, and a Hepatozoon americanum-like haplotype. None tick or blood samples yielded amplicons through PCR assays targeting the genera Ehrlichia, Anaplasma, Babesia, Rangelia, Cytauxzoon, and Theileria. Maned wolf serum samples were tested by immunofluorescence assay against antigens of five Rickettsia species (R. parkeri, R. rickettsii, R. amblyommatis, R. rhipicephali, and R. bellii) and Ehrlichia canis. Among 78 serum samples (45 captures plus 33 recaptures), 74 (95 %) were reactive to at least one Rickettsia species, with R. parkeri eliciting the highest endpoint titers. Some maned wolves that were recaptured during the study were shown to seroconvert to R. parkeri. Serum-reactiveness to E. canis was detected in 36 % (16/45) maned wolves. During the study, general clinical signs of tick-borne diseases were not found in any of the captured animals, indicating that they were under a good health status in the SCNP, despite of been exposed to ticks (mostly A. tigrinum) and some tick-borne agents (Rickettsia, Hepatozoon, Ehrlichia). The results of the present study might represent baseline data for the conservation of the maned wolf in its natural habitat, which should be used to interpret further studies about ticks and tick-borne diseases in maned wolves within human-modified landscapes.

The field of movement ecology has rapidly grown during the last decade, with important advancements in tracking devices and analytical tools that have provided unprecedented insights into where, when, and why species move across a landscape. Although there has been an increasing emphasis on making animal movement data publicly available, there has also been a conspicuous dearth in the availability of such data on large carnivores. Globally, large predators are of conservation concern. However, due to their secretive behavior and low densities, obtaining movement data on apex predators is expensive and logistically challenging. Consequently, the relatively small sample sizes typical of large carnivore movement studies may limit insights into the ecology and behavior of these elusive predators. The aim of this initiative is to make available to the conservation-scientific community a dataset of 134,690 locations of jaguars (Panthera onca) collected from 117 individuals (54 males and 63 females) tracked by GPS technology. Individual jaguars were monitored in five different range countries representing a large portion of the species' distribution. This dataset may be used to answer a variety of ecological questions including but not limited to: improved models of connectivity from local to continental scales; the use of natural or human-modified landscapes by jaguars; movement behavior of jaguars in regions not represented in this dataset; intraspecific interactions; and predator-prey interactions. In making our dataset publicly available, we hope to motivate other research groups to do the same in the near future. Specifically, we aim to help inform a better understanding of jaguar movement ecology with applications towards effective decision making and maximizing long-term conservation efforts for this ecologically important species. There are no costs, copyright, or proprietary restrictions associated with this data set. When using this data set, please cite this article to recognize the effort involved in gathering and collating the data and the willingness of the authors to make it publicly available.

The Pantanal is the largest inland wetland in the world and is under increasing anthropogenic threats, including long-term regionally intensive gold mining practices. Gold mining activities are known to cause the release of harmful pollutants such as mercury (Hg) to the surrounding environment. Jaguars (Panthera onca (Linnaeus, 1758)) are apex predators, and therefore show great potential to accumulate Hg by biomagnification. We hypothesize that total Hg content in the fur of jaguars from two sites within the Brazilian Pantanal would be significantly different as a function of distance from active gold mining operations. The Hg content was determined by fluorescence spectrometry. The mean ± SD Hg content in jaguars from the study site influenced by gold mining (SB) was compared to jaguars sampled in the area free of gold mining activities (CA) using a one-way ANOVA. The mean Hg content in jaguars from SB (673.0 ± 916.8 µg g-1) is significantly different from jaguars sampled in CA (29.7 ± 23.3 µg g-1), p = 0.03. The maximum recorded content of Hg was 2,010.4 ± 150.5 µg g-1, highest level ever recorded in a wild animal. The data indicate that Hg is an important threat to jaguars within at-risk regions of the Pantanal.

Accurately estimating home range and understanding movement behavior can provide important information on ecological processes. Advances in data collection and analysis have improved our ability to estimate home range and movement parameters, both of which have the potential to impact species conservation. Fitting continuous-time movement model to data and incorporating the autocorrelated kernel density estimator (AKDE), we investigated range residency of forty-four jaguars fit with GPS collars across five biomes in Brazil and Argentina. We assessed home range and movement parameters of range resident animals and compared AKDE estimates with kernel density estimates (KDE). We accounted for differential space use and movement among individuals, sex, region, and habitat quality. Thirty-three (80%) of collared jaguars were range resident. Home range estimates using AKDE were 1.02 to 4.80 times larger than KDE estimates that did not consider autocorrelation. Males exhibited larger home ranges, more directional movement paths, and a trend towards larger distances traveled per day. Jaguars with the largest home ranges occupied the Atlantic Forest, a biome with high levels of deforestation and high human population density. Our results fill a gap in the knowledge of the species' ecology with an aim towards better conservation of this endangered/critically endangered carnivore-the top predator in the Neotropics.

BACKGROUND: Periodicity in activity level (rest/activity cycles) is ubiquitous in nature, but whether and how these periodicities translate into periodic patterns of space use by animals is much less documented. Here we introduce an analytical protocol based on the Lomb-Scargle periodogram (LSP) to facilitate exploration of animal tracking datasets for periodic patterns. The LSP accommodates missing observations and variation in the sampling intervals of the location time series. RESULTS: We describe a new, fast algorithm to compute the LSP. The gain in speed compared to other R implementations of the LSP makes it tractable to analyze long datasets (>10(6) records). We also give a detailed primer on periodicity analysis, focusing on the specificities of movement data. In particular, we warn against the risk of flawed inference when the sampling schedule creates artefactual periodicities and we introduce a new statistical test of periodicity that accommodates temporally autocorrelated background noise. Applying our LSP-based analytical protocol to tracking data from three species revealed that an ungulate exhibited periodicity in its movement speed but not in its locations, that a central place-foraging seabird tracked moon phase, and that the movements of a range-resident canid included a daily patrolling component that was initially masked by the stochasticity of the movements. CONCLUSION: The new, fast algorithm tailored for movement data analysis and now available in the R-package ctmm makes the LSP a convenient exploratory tool to detect periodic patterns in animal movement data.

A major question in global environmental policy is whether schemes to reduce carbon pollution through forest management, such as Reducing Emissions from Deforestation and Degradation (REDD+), can also benefit biodiversity conservation in tropical countries. We identified municipalities in Brazil that are priorities for reducing rates of deforestation and thus preserving carbon stocks that are also conservation targets for the endangered jaguar (Panthera onca) and biodiversity in general. Preliminary statistical analysis showed that municipalities with high biodiversity were positively associated with high forest carbon stocks. We used a multicriteria decision analysis to identify municipalities that offered the best opportunities for the conservation of forest carbon stocks and biodiversity conservation under a range of scenarios with different rates of deforestation and carbon values. We further categorized these areas by their representativeness of the entire country (through measures such as percent forest cover) and an indirect measure of cost (number of municipalities). The municipalities that offered optimal co-benefits for forest carbon stocks and conservation were termed REDDspots (n = 159), and their spatial distribution was compared with the distribution of current and proposed REDD projects (n = 135). We defined REDDspots as the municipalities that offer the best opportunities for co-benefits between the conservation of forest carbon stocks, jaguars, and other wildlife. These areas coincided in 25% (n = 40) of municipalities. We identified a further 95 municipalities that may have the greatest potential to develop additional REDD+ projects while also targeting biodiversity conservation. We concluded that REDD+ strategies could be an efficient tool for biodiversity conservation in key locations, especially in Amazonian and Atlantic Forest biomes.

In this study we measured excreted fecal corticoid metabolites (FCM) in maned wolves (Chrysocyon brachyurus) living within a protected reserve, on farmlands or in a boundary zone between the two habitats, and determined the impacts of season and reproductive status on adrenal activity. Feces were collected within a national park (n=191 samples), a park boundary zone (n=39) and on nearby farmlands (n=27), processed and analyzed by enzyme immunoassay. FCM amounts from samples collected on farmlands were higher (P<0.05) than in those collected inside the reserve and from the boundary zone. In relation to seasonality, FCM were elevated (P<0.05) in spring (September-November) when wolf pairs were raising young. We then divided the samples collected during breeding season (March-August) into cycling females and male/non-cycling females based on fecal progesterone: fecal testosterone ratio. FCM concentrations of the former collected inside the park were higher than (P<0.05) than the latter group. However, there were no differences in FCM levels between the two groups for samples collected in the boundary zone and on farmlands. Furthermore, FCM concentrations of male/non-cycling females samples collected on farmlands were 2- to 5-fold higher (P<0.05) than in counterparts collected inside the park. The consistently high FCM concentrations in samples collected on farmlands indicate that, in addition to seasonality, gender and reproductive status, anthropogenic pressures also contribute to elevating adrenal steroid for individuals living in altered habitat.

We tested 47 tetranucleotide microsatellite loci developed for the domestic dog in four species of Neotropical canids, aiming to produce a standardized set that could be successfully used even in noninvasive samples across this group. We identified 13 suitable loci, nine of which constitute a standardized set for all species. Considering only the ideal panel of nine loci, the mean expected heterozygosity (averaged across species) per locus ranged from 0.58 to 0.92 (overall mean 0.76), and the maximum probability of identity value was 1.3 × 10(-9) . This set of loci has a great potential for application in evolutionary, ecological and conservation studies.
