Rabies is a lethal zoonotic disease that threatens human health. As the only viral surface protein, the rabies virus (RABV) glycoprotein (G) induces main neutralizing antibody (Nab) responses; however, Nab titre is closely correlated with the conformation of G. Virus-like particles (VLP) formed by the co-expression of RABV G and matrix protein (M) improve retention and antigen presentation, inducing broad, durable immune responses. RABV nucleoprotein (N) can elicit humoral and cellular immune responses. Hence, we developed a series of nucleoside-modified RABV mRNA vaccines encoding wild-type G, soluble trimeric RABV G formed by an artificial trimer motif (tG-MTQ), membrane-anchored prefusion-stabilized G (preG). Furthermore, we also developed RABV VLP mRNA vaccine co-expressing preG and M to generate VLPs, and VLP/N mRNA vaccine co-expressing preG, M, and N. The RABV mRNA vaccines induced higher humoral and cellular responses than inactivated rabies vaccine, and completely protected mice against intracerebral challenge. Additionally, the IgG and Nab titres in RABV preG, VLP and VLP/N mRNA groups were significantly higher than those in G and tG-MTQ groups. A single administration of VLP or VLP/N mRNA vaccines elicited protective Nab responses, the Nab titres were significantly higher than that in inactivated rabies vaccine group at day 7. Moreover, RABV VLP and VLP/N mRNA vaccines showed superior capacities to elicit potent germinal centre, long-lived plasma cell and memory B cell responses, which linked to high titre and durable Nab responses. In summary, our data demonstrated that RABV VLP and VLP/N mRNA vaccines could be promising candidates against rabies.

Pyrolysis is an effective process for disposing of municipal sewage sludge (SS). Plastics can affect the SS pyrolysis behaviour and pyrolysis products due to their low ash and high hydrocarbon ratio. The secondary pollutants from the pyrolysis process may also be affected. Therefore, polyethylene terephthalate (PET), a typical plastic, was chosen to investigate the release characteristics of pollutants containing nitrogen, sulphur, and chlorine via SS pyrolysis, and the changes of biochar to adsorb two typical heavy metals, Pb and Cu. The pyrolysis of PET plastics facilitates the migration of N toward solid and liquid-phase products, S and Cl to the gas-phase products via pyrolysis. Oxygenated compounds of pyrolytic volatiles decreased from 38.18% to 28.43%, concurrently promoting the formation of phenolic compounds. The co-pyrolysis improved the quality of biochar and the ability to adsorb Pb and Cu. This systematic study can provide some support for the further improvement of SS pyrolysis technology, and will also be beneficial for subsequent applications.

For portable devices with limited resources, it is often difficult to deploy deep networks due to the prohibitive computational overhead. Numerous approaches have been proposed to quantize weights and/or activations to speed up the inference. Loss-aware quantization has been proposed to directly formulate the impact of weight quantization on the model's final loss. However, we discover that, under certain circumstances, such a method may not converge and end up oscillating. To tackle this issue, we introduce a novel loss-aware quantization algorithm to efficiently compress deep networks with low bit-width model weights. We provide a more accurate estimation of gradients by leveraging the Taylor expansion to compensate for the quantization error, which leads to better convergence behavior. Our theoretical analysis indicates that the gradient mismatch issue can be fixed by the newly introduced quantization error compensation term. Experimental results for both linear models and convolutional networks verify the effectiveness of our proposed method.

In this study, a novel class of multifunctional responsive nanoparticles is designed and fabricated as drug nanocarriers for synergetic chemo-photothermal therapy of tumors. The proposed nanoparticles are composed of a thermo-/pH-responsive poly(N-isopropylacrylamide-co-acrylic acid) (PNA) nanogel core, a polydopamine (PDA) layer for photothermal conversion, and an outer folic acid (FA) layer as a targeting agent for the folate receptors on tumor cells. The fabricated nanoparticles show good biocompatibility and outstanding photothermal conversion efficiency. The proposed nanoparticles loaded with doxorubicin (DOX) drug molecules are stable under physiological conditions with low leakage of drugs, while rapidly release drugs in environments with low pH conditions and at high temperature. The experimental results show that the drug release process is mainly governed by Fickian diffusion. In vitro cell experimental results demonstrate that the PNA-DOX@PDA-FA nanoparticles can be phagocytized by 4T1 tumor cells and release drugs in tumor cell acidic environments, and confirm that the combined chemo and photothermal therapeutic efficacy of PNA-DOX@PDA-FA nanoparticles is higher than the photothermal therapeutic efficacy or the chemotherapeutic efficacy alone. The proposed multifunctional responsive nanoparticles in this study provide a novel class of drug nanocarriers as a promising tool for synergetic chemo-photothermal therapy of tumors.

A simple device, which is equipped with a non-woven fabric filter medium immobilized with ion-recognizable smart hollow microgels, is developed for facile detection of trace lead(II) ions (Pb2+ ). The ion-recognizable smart microgels are made of poly(N-isopropylacrylamide-co-benzo-18-crown-6-acrylamide) (PNB), in which the 18-crown-6 groups act as the sensors of Pb2+ and the N-isopropylacrylamide groups act as the actuators. The PNB hollow microgels can isothermally change from a shrunk state to a swollen state in response to recognizing Pb2+ in the aqueous environment due to the electrostatic repulsion among the charged 18-crown-6/Pb2+ complex groups and the enhancement of hydrophilicity of the microgels. Due to the hollow structures, the PNB microgels show remarkable isothermal swelling ratio. Thus, the flux of solution pass through the non-woven fabric filter medium decreases significantly because of the remarkable reduction in the space for liquid flowing upon recognizing Pb2+ . The Pb2+ concentration can be detected quantitatively by simply and easily measuring the change of solution flux using the proposed device, which is operated without external power supply or spectroscopic measurements. The strategy proposed in this study provides a promising method for facile detection of trace Pb2+ in aqueous environments.
