RESUMO

Effects of various doses of semicarbazide on Lewis carcinoma metastasing and peroxidation processes in C57B1 mice have been investigated. In the animals with inoculated Lewis carcinoma, the semicarbazide in the dose of 1/120 LD50 had practical influence on the tumour growth and inhibited the metastasing into mice lungs (P < 0.05). Importantly, this dose significantly inhibited the formation of free radicals and active forms of oxygen against the background of decrease of the aldehydes level that was related to the acceptor properties of the drug.

RESUMO

Experiments were carried out on rats with lathyrism, which was induced by adding semicarbazide (0.075%) into drinking water for 45 days. The data obtained show a 30% reduction in the body weight and an increase in.organ weight coefficients. Semicarbazide intake led to the pelvic limb paralysis, scoliosis, bone tissue degradation, cartilage growth, 46% decrease of the calcium level in the femur. It has been detected essential structural changes in extracellular matrix based on the collagen cross-links reduction. The activity of lysyl oxidase, a key enzyme for the collagen development, showed 5-fold decrease in the aorta tissues. The level of formaldehyde, a nonenzymic cross-links developer, has been measured in the liver tissue by the aldehyde trap (5,5-dimethyleyclohexane-1,3-dione) administration and then fluorimetric determination of formaldimedone. Under semicarbazide load, the formaldehyde level in the liver tissue was reduced by 47%. Therefore, semicarbazide influences not only the enzymic development of aldehyde groups in collagen, but the level of other aldehydes, which can cause cross-links. This experimental model of lathyrism is appropriate for investigation of the lysyl oxidase inhibitors effect on extracellular matrix.

RESUMO

Subchronic administration of semicarbazide in the experiment with the rats was used to reduce the formaldehyde level in the organism in order to reveal the interaction between formaldehyde metabolism and biochemical parameters, which define the oxidant-antioxidant system condition and NO metabolism. It has been found that under semicarbazide impact the generation of free radicals, ROS, nitrite and nitrate were enhanced while aldehydes level was reduced that resulted from not only semicarbazide effect like the aldehydes acceptor, but the formaldehyde synthesis slowdown and acceleration of its transformation into format as well. We suppose that formaldehyde plays certain role in the development of connective tissue pathology.

RESUMO

It is shown that the intensity of ammonium genesis, purines biosynthesis and redox processes increases in the organism of chicken with visceral gout. Sodium bicarbonate-correction of the acid-alkaline state of chickens is performed in order to normalize disturbances observed under gout.

RESUMO

Experiments on chickens of different age groups (one day--1 months) from large poultry plants of the Kiev Region established that metabolic acidosis develops in chickens at the age of 10-30 days. Compensated alkalosis is observed in them at the age of two months and further. In chickens whose HCO3- level in blood is 15-18 mM/l the content of uric acid in the liver and blood increases as compared to the control (HCO3- --23 mM/l); they also differ in a high intensity of the growth. A rise of the HCO3- concentration in chicken blood up to 33 mM/l favours a still greater decrease in the level of uric acid in tissues and an increase in the chicken growth intensity as compared to the control. Studies in the state of the blood acid-base balance in pigs of the commercial complex "Kalityansky" showed that compensated alkalosis develops in the whole pig stock.

RESUMO

Under conditions of an increase in the carbonic acid concentration in blood of rabbits (experimental group) utilization of ammonia nitrogen administered to the organism intensifies and simultaneously the level of keto-acids in blood as well as of free amino acids and urea in the liver tissue increases. In these animals as compared to the control ones the content of glutamine in blood and in the liver tissue decreases; the level of alpha-glutarate and glutamate in the liver tissue increases and that of pyruvate lowers. An increase is also observed in reducing power of the liver and kidney cell cytoplasm.
