RESUMO

Zoonotic viral infections continue to pose significant threats to global public health, as highlighted by the COVID-19 pandemic caused by the SARS-CoV-2 virus. The emergence of SARS-CoV-2 served as a stark reminder of the potential for zoonotic transmission of viruses from animals to humans. Understanding the origins and dynamics of zoonotic viruses is critical for early detection, prevention, and effective management of future outbreaks. Metagenomics has emerged as a powerful tool for investigating the virome of diverse ecosystems, shedding light on the diversity of viral populations, their hosts, and potential zoonotic spillover events. We provide an in-depth examination of metagenomic approaches, including, NGS metagenomics, shotgun metagenomics, viral metagenomics, and single-virus metagenomics, highlighting their strengths and limitations in identifying and characterizing zoonotic viral pathogens. This review underscores the pivotal role of metagenomics in enhancing our ability to detect, monitor, and mitigate zoonotic viral infections, using SARS-CoV-2 analogues as a case study. We emphasize the need for continued interdisciplinary collaboration among virologists, ecologists, and bioinformaticians to harness the full potential of metagenomic approaches in safeguarding public health against emerging zoonotic threats.

RESUMO

The objective of this study was to evaluate the effects of adapting Nellore and ½ Angus/Nellore (AN) feedlot cattle over periods of 9 and 14 days to high-concentrate diets on performance, feeding behaviour, carcass traits and rumen morphometrics. Seventy-two yearling bulls (313.5 kg ± 24.5), 36 Nellore and 36 AN, were randomly allocated in 24 pens (3 animals/pen; 24 m2 and 2.0 m of bunk space/animal) according to a randomized complete block design with a 2 × 2 factorial arrangement of treatments as follows: Nellore adapted for 9 days, Nellore adapted for 14 days, AN adapted for 9 days, and AN adapted for 14 days. Each treatment was composed by 6 pens (considered the experimental unit in this study). The adaptation lasted either 9 or 14 days and consisted of 3 step-up diets. Therefore, yearling bulls received the finishing diet containing 86% concentrate either on day 10 or 15 of the study, which lasted 89 days taking into account adaptation and finishing periods. Cattle were slaughtered in a commercial abattoir, and two 1-cm2 -rumen fragments, one from cranial and another from ventral sac, were collected. The AN cattle outperformed Nellore in terms of average daily gain (1.71 kg/day vs. 1.27 kg/day, p < 0.01), gain:feed ratio (0.137 kg/kg vs. 0.127 kg/kg, p = 0.02) and hot carcass weight (243.64 kg vs. 228.98 kg, p < 0.01). No main effect of the adaptation period was observed for any of the feedlot performance and carcass traits variables evaluated. Compared to feedlot cattle adapted for 9 days, feedlot cattle adapted for 14 days sorted against long (0.68 vs. 0.91, p < 0.01) and for fine particles (1.04 vs. 1.00, p = 0.01). An interaction (p < 0.01) of genotype and adaptation period was observed for rumenitis, where Nellore bulls adapted for 14 days presented the highest scores. In conclusion, there was no evidence that either Nellore or AN cattle benefit from an adaptation period shorter than 14 days.

RESUMO

Every year, a considerable volume of medications is consumed. Because these medications are not entirely eliminated in the sewage treatment plants and impact the surface waterways, the environmental pollution problem arises. This study objective was to evaluate the possibility of using an absorbent material made with of polyethylene terephthalate and sugarcane bagasse ash functionalized with iron oxide (PETSCA/Fe3+) in the removal of naproxen from water. The feasibility of having viable features in becoming an efficient adsorbent was first determined. The batch test was performed, allowing the dose effect, adsorption kinetics, and isotherm models to be evaluated. The determination of naproxen (NAP) concentration in water was analyzed on a high-performance liquid chromatograph and Langmuir method best represented the adsorption isotherm model. PETSCA/Fe3+ adsorbent material demonstrated potential in the naproxen removal at a low cost. The batching process was satisfactory, with 0.30 g of composite being the optimum fit for the system. The adsorption kinetics was determined and described by the pseudo second order model, with an average correlation coefficient (R2) of 0.974. The adsorption system model was best represented by the Langmuir isotherm curve. Moreover, adsorption in the presence of H2O2 had a positive impact on the process, removing 81.9% of NAP, whereas the process without H2O2 did not remove more than 62.0% of NAP. Therefore, because of its good qualities for NAP removal, PETSCA/Fe3+ is recommended as adsorbent material, primarily in small-volume water filtration systems.

RESUMO

Benzimidazoles are anthelmintics frequently used in sheep farming due to the high susceptibility of these animals to parasitic diseases. Sheep excreta are often disposed onto soils as a fertilizer, and they may contain benzimidazole residues that can contaminate soil and water. This work aimed to assess the leaching behavior of benzimidazole drugs (albendazole, fenbendazole, and thiabendazole) and their metabolites in two Brazilian soils of different textural classifications (sandy and clay), as well as sheep excreta-amended soils, following the OECD 312 Guidelines. Ewes received a single oral dose of 10 mg kg-1 b.w. of either albendazole or fenbendazole. The feces were collected at 24, 48, 72, 96, and 120 h post-dose, and the parent drugs and their metabolites extracted using the QuEChERS approach and quantified by UHPLC-MS/MS. For the leaching assays, a benzimidazole solution was directly applied onto the soil columns, or an amount of 5 g of the medicated sheep feces was distributed over the top of the soil columns. In soil samples, benzimidazoles were extracted by solid-liquid extraction and quantified by UHPLC-MS/MS. For the leaching studies, atrazine was used as a reference substance to determine the relative mobility factor of the analytes of interest. Benzimidazoles were considered slightly to moderately mobile in both soils tested, with a leaching distance of up to 25 cm in a 30-cm soil column. Approximately 3 to 6% of the benzimidazoles present in ewe feces were able to leach into the soil columns. This finding is of concern since benzimidazoles are persistent in soil and may pose a risk to soil biota and induce the development of resistant strains of parasites.

It was evaluated the effects of sunflower grains inclusion or protected fat in the diet and compensatory growth on the loin eye area, subcutaneous fat thickness, chemical composition, cholesterol concentration and fatty acid composition of intramuscular fat of Longissimus lumborum of lambs distributed in a completely randomized design in factorial scheme (3 diets x 2 feeding management). Intramuscular cholesterol from lambs fed with diet containing protected fat (28.5mg 100g-1) was lower (P<0.05) than that of the control (36.8mg 100g-1) and sunflower grains (38.1mg 100g-1). The restriction and refeeding didn't cause change (P>0.05) in the fatty acids profile, but diet containing sunflower grains increased polyunsaturated/saturated ratio due to(P<0.05) the superiority of 20% in the proportion of polyunsaturated, reflecting directly the increase in linoleic acid. Inclusion of lipid sources in the diet of feedlot sheep is a tool that helps to improve the quality of intramuscular fat of lambs, whereas the protected fat reduces cholesterol and sunflower grains elevates the concentration of polyunsaturated fatty acids. Feeding restriction and refeeding period does not alter the chemical composition and quality of intramuscular fat.

This study aimed to assess the influence of transport on the occurrence of carcass bruising of cattle slaughtered in the Pantanal, by quantifying the number and size of bruises, locating the frequency of occurrence of bruises in the main commercial cuts standardized for interne market. From a total of 121 carcasses assessed, 102 (84.3 percent) had one or more bruises, totaling 270 bruises, which resulted in the removal of 56.1kg meat. The frequency of carcass bruises of cattle showed significant difference, according to the conditions of transport. The highest proportions of bruises found in animals submitted to road transport for more than one hour in distances longer than 70km, a great part of which on dirty toads. The conclusion is that the animals submitted to the successive series of handling and transported on dirty roads for long distances caused the highest proportion of bruises, whereas the animals submitted to lot IV presented the highest number of bruises.
