RESUMO

This research was designed to find out the effect of different factors such as influence of lunar cycle, harvesting interval, rope type and seeding gap on the production of G. tenuistipitata in coast of Cox's Bazar. Duration of these experiments were sixty days and all the parameters were recorded fortnightly. Monitoring of water quality parameters indicated that salinity, temperature, transparency, pH and DO were suitable for seaweed cultivation. In determining lunar cycle effect, results envisaged that fresh yield was 14.43% increased when seeding and harvesting time was selected considering the moon cycle. Regarding the selection of harvesting interval, it was found that T3 (30 days interval) was the best to harvest the seaweed whereas T4 (40 days interval) showed decreasing trend in production. Our study also found that semi floating single line showed better yield performance compared to semi floating double line system. In case of influence on seeding gap, it has been found that 20 cm gap between two seed showed the highest yields followed by 10 cm, 30 cm and 40 cm, respectively. Overall, it can be concluded that yield of G. tenuistipitata in coast of Cox's Bazar could be improved considering those factors.

RESUMO

Seaweeds are now recognized as a treasure of bioactive compounds. However, the bioactivity of seaweed originating in Bangladesh is still unexplored. So, this study was designed to explore the secondary metabolites and antioxidant activities of solvent extracts of Padina tetrastromatica and Gracilaria tenuistipitata. Phytochemical screening and FTIR spectra confirm the diverse type of bioactive compounds. Antioxidant activity of extracts were evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2, 2-Azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), reducing power (RP), phosphomolybdenum, hydrogen peroxide and nitric oxide (NO) scavenging assays. Here, methanolic extract of P. tetrastromatica showed highest amount of total phenolic content (85.61 mg of GA/g), total flavonoid content (41.77 mg of quercetin/g), DPPH (77.07%), ABTS (77.65%), RP (53.24 mg AAE/g), phosphomolybdenum (31.58 mg AAE/g), hydrogen peroxide (67.89%) and NO (70.64%) assays compared to its methanolic extracts of G. tenuistipitata. This study concluded that methanol as a solvent extract of brown seaweed (P. tetrastromatica) exhibited bioactivity and antioxidant potentiality which will be useful for pharmacological as well as in functional food application.

RESUMO

The biological and therapeutic properties of seaweeds have already been well known. Several studies showed that among the various natural marine sources of antioxidants, seaweeds have become a potential source of antioxidants because of their bioactive compounds. Most of the metabolic diseases are caused by oxidative stress. It is very well known that antioxidants have a pivotal role in the treatment of those diseases. Recent researches have revealed the potential activity of seaweeds as complementary medicine, which have therapeutic properties for health and disease management. Among the seaweeds, brown seaweeds (Phaeophyta) and their derived bioactive substances showed excellent antioxidant properties than other seaweeds. This review focuses on brown seaweeds and their derived major bioactive compounds such as sulfated polysaccharide, polyphenol, carotenoid, and sterol antioxidant effects and molecular mechanisms in the case of the oxidative stress-originated disease. Antioxidants have a potential role in the modification of stress-induced signaling pathways along with the activation of the oxidative defensive pathways. This review would help to provide the basis for further studies to researchers on the potential antioxidant role in the field of medical health care and future drug development.

RESUMO

Seminal composition and semen quality are the important determinants in assessing the reproductive performance of different fishes. This study was carried out to evaluate the seminal composition and sperm quality of Barbonymus gonionotus. The seminal plasma contained 17.2 ± 0.34 mmol/l, 20.9 ± 0.48 mmol/l, 0.72 ± 0.04 mmol/l, 3.8 ± 0.2 mmol/l, and 1.49 ± 0.02 g/dl of Na+, K+, Ca++, Mg++, and total protein, respectively. The physical spermatological parameters, such as sperm volume, sperm motility, motility duration, sperm density, osmolality, and pH values were 1.55 ± 0.15 ml, 89 ± 2%, 391.9 ± 8.5 s, 2.8 ± 0.2 × 1010 /ml, 400.6 ± 5.1 mmol/kg, and 8.75 ± 0.10, respectively. In correlation matrix, the K+ (R2 = 0.39, P < 0.01) and Ca++ (R2 = 0.27, P < 0.05) ions and osmolality (R2 = 0.29, P < 0.05) showed significant positive correlations with sperm motility. Similarly, fertilization rate significantly influenced by sperm motility (R2 = 0.26, P < 0.05) and K+ (R2 = 0.30, P < 0.05) and Ca++ (R2 = 0.26, P < 0.05) ions. Also, osmolality significantly and negatively correlated with Mg++ (R2 = 0.33, P < 0.05) and sperm motility duration (R2 = 0.28, P < 0.05). Therefore, based on this results, it can be concluded that seminal plasma ions, K+ and Ca++ and osmolality are the key factors for the determination of sperm quality of silver barb, and these parameters could be considered during standardization of artificial fertilization or cryopreservation technique of silver barb spermatozoa.

RESUMO

The present study was performed to investigate the chemical composition and antioxidant activity of glycoprotein purified from Undaria pinnatifida Harvey (UPGP). On SDS-PAGE, UPGP migrated as a single band with a molecular weight of approximately 10 kDa and confirmed by staining with Schiff's reagent as glycoprotein. It consists of a carbohydrate component (42.53%) and protein component (57.47%). Amino acid profile, FT-IR spectrum and enzymatic glycosylation analysis suggested that protein is linked with carbohydrate by O-glycosylation. UPGP showed dose-dependent antioxidant activities as detected by different assays before and after in vitro digestion. The IC50 values of undigested UPGP were 0.25 ± 0.03, 0.08 ± 0.005, 0.69 ± 0.12, and 0.25 ± 0.08 mg/mL for DPPH, ABTS, FRAP, and NO, respectively. Following in vitro digestion, the antioxidant activities of UPGP were decreased during the gastric phase compared to those of undigested UPGP, with an increase occurring during the duodenal phase in all assays. However, the reducing power was unchanged after in vitro digestion. Furthermore, UPGP showed protective activity against oxidative DNA damage both undigested, after saliva and duodenal phase of digestion. These results indicate that the antioxidant and DNA protection activities of UPGP may be pH-dependent and assay specific.
