Introduction: Maxillary reconstruction is often a challenging task for the surgeons because of the complex anatomy. However, with the advances in virtual surgical planning (VSP) and 3D printing technology there is a new avenue for the surgeons which offers a suitable alternative to conventional flap-based reconstructions. Patients and Methods: In this article, we have described 4 case scenarios which were managed with the help of VSP and additive manufacturing technology for complex maxillary reconstruction procedures. Use of the technologies aided the clinician in achieving optimal outcomes with regards to form, function and esthetics. Discussion: Virtual surgical planning (VSP) has gained a lot of impetus in past 1 decade. These aides the surgeon in determining the extent of disease and also carry out the treatment planning. In addition to VSP, the concept of additive manufacturing provides a viable alternative to the conventional reconstruction modalities for maxillary defect rehabilitation. Increased accuracy, rehabilitation of normal anatomical configuration, appropriate dental rehabilitation, decreased intra-operative time and post-operative complications are some of the advantages. In addition, patient-specific implants eliminate the need for a separate donor site. Apart from the treatment of pathologies, they also can be used for reconstruction of post-traumatic defect, where endosteal implant placement is not possible. Conclusion: These modalities show promising results for reconstruction of complex maxillary defects.

L-Tyrosine (L-Tyr), a critical amino acid whose aberrant levels impact melanin and dopamine levels in human body while also increasing insulin resistance thereby increasing the risk of type 2 diabetes. The objective of this study was to detect the amount of L-Tyr in human fluids by tailored electrochemical synthesis of well adhered, homogenous and thin molecularly imprinted polymers (MIPs) by the electro-polymerization of pyrrole on glassy carbon electrode modified functionalized multi-walled carbon nanotubes. The key benefits of this procedure over previous imprinting techniques were the elimination of expensive materials like Au and tedious multi-step synthesis, for L-Tyr detection using a handheld potentiostat. The developed particles were characterized using Fourier Transform Infrared Spectroscopy, Scanning Electron Microscope, Chronoamperometry, and Cyclic Voltammetry. With strong reproducibility and stability, this optimized approach provides a rapid and effective method of preparing and sensing MIPs for the target analyte with a broad linear range of [Formula: see text] to [Formula: see text]. The Limit of Detection and Limit of Quantification were [Formula: see text] and [Formula: see text], respectively. The engineered sensor was validated for quantifying the concentrations of L-Tyr in human blood and serum samples, yielding satisfactory recovery and can be expanded in future to detect analytes simultaneous.

The authors describe a simple technique to overcome the task of stabilizing the down-fractured maxilla.

Chronic kidney disease is a universal topic gravitating towards various aspects of widespread illness, impacting the overall well-being of human beings. Patients with longstanding renal complaints under dialysis encounter challenges correlated with physical, intuitive, and socio-economic conditions to a greater extent in their daily existence. These portions may include changes in the appearance of a person, restricted physique movements, curbed diet, duration of surgical protocols, travelling time during the period of prevention, financial load, role reversal in the family followed by ruining their livelihood, deprived social rank, difficulty in relational, cordial relationships, and so on. Excluding these details, the sick may be profoundly influenced by sorrow, health anxiety, despair, itching, the impoverished essence of vitality, dysfunction in sexual intimacy, impaired cognition, disturbances in disposition, sleeping fluctuations, frequent panic attacks, delirium, brain-afflicted degeneration disabilities, etcetera. Our analysis focuses on exploring a few unidentified intrinsic factors that distinguish these views over combined elements due to the existing disorder.

Background: Cleaning and shaping represent a vital step in the endodontic procedure. In routine endodontic therapy, the fracture of nickel-titanium (NiTi) instruments is a procedural problem creating a major obstacle to therapy. Aim: This study examines the life span of one Neoendo Flex and ProTaper Next (PTN) rotary file using reciprocating motion and compares the time required by both file systems for canal preparation. Materials and Methods: One hundred maxillary permanent central incisors were selected. In that, fifty teeth are utilized in a reciprocating motion (RM) with the PTN file (n = 25) and Neoendo Flex file (n = 25). To check fractured resistance and the overall root canal preparation time, fifty more teeth are employed in continuous motion (CM) with canals prepared using PTN (n = 25) and Neoendo Flex (n = 25). All canals are prepared following the sequence of respective files. Later, the fracture mechanism of the files was inspected using a scanning electron microscope. Results: In an RM motion, both PTN and Neoendo Flex files can be used in a minimum of 25 canals in single-rooted teeth. Statistical analysis using the Mann-Whitney U-test showed no significant difference in total time taken by PTN and Neoendo Flex in both motions. Conclusion: Within the limitation of this study, the RM was found to be better than CM with less incidence of instrument fracture.

Marker-assisted backcross breeding enables selective insertion of targeted traits into the genome to improve yield, quality, and stress resistance in wheat. In the current investigation, we transferred four drought tolerance quantitative trait loci (QTLs) controlling traits, viz canopy temperature, normalized difference vegetative index, chlorophyll content, and grain yield from the drought-tolerant donor line, C306, into a popular high-yielding, drought-sensitive variety, HD2733. Marker-assisted selection coupled with stringent phenotypic screening was used to advance each generation. This study resulted in 23 improved lines carrying combinations of four drought tolerance QTLs with a range of 85.35%-95.79% background recovery. The backcross-derived lines gave a higher yield under moisture-deficit stress conditions compared with the recipient parent. They also showed higher phenotypic mean values for physiological traits and stability characteristics of HD2733. A promising genotype, HD3411, derived from this cross was identified for release after national multi-location coordinating trials under the All India Coordinated Wheat Improvement Project. Our study is a prime example of the advantages of precision breeding using integrating markers and phenotypic selection to develop new cultivars with desirable traits like drought tolerance.

Schizophrenia is a chronic mental disorder that impairs a person's thinking capacity, feelings and emotions, behavioural traits, etc., Emotional distortions, delusions, hallucinations, and incoherent speech are all some of the symptoms of schizophrenia, and cause disruption of routine activities. Computer-assisted diagnosis of schizophrenia is significantly needed to give its patients a higher quality of life. Hence, an improved adaptive neuro-fuzzy inference system based on the Hybrid Grey Wolf-Bat Algorithm for accurate prediction of schizophrenia from multi-channel EEG signals is presented in this study. The EEG signals are pre-processed using a Butterworth band pass filter and wICA initially, from which statistical, time-domain, frequency-domain, and spectral features are extracted. Discriminating features are selected using the ReliefF algorithm and are then forwarded to ANFIS for classification into either schizophrenic or normal. ANFIS is optimized by the Hybrid Grey Wolf-Bat Algorithm (HWBO) for better efficiency. The method is experimented on two separate EEG datasets-1 and 2, demonstrating an accuracy of 99.54% and 99.35%, respectively, with appreciable F1-score and MCC. Further experiments reveal the efficiency of the Hybrid Wolf-Bat algorithm in optimizing the ANFIS parameters when compared with traditional ANFIS model and other proven algorithms like genetic algorithm-ANFIS, particle optimization-ANFIS, crow search optimization algorithm-ANFIS and ant colony optimization algorithm-ANFIS, showing high R2 value and low RSME value. To provide a bias free classification, tenfold cross validation is performed which produced an accuracy of 97.8% and 98.5% on the two datasets respectively. Experimental outcomes demonstrate the superiority of the Hybrid Grey Wolf-Bat Algorithm over the similar techniques in predicting schizophrenia.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) transmission in India in 2020-2022 was driven predominantly by Wild (Wuhan-Hu-1 and D614G), Delta, and Omicron variants. The aim of this study was to examine the effect of infections on the humoral immune response and cross-reactivity to spike proteins of Wuhan-Hu-1, Delta, C.1.2., and Omicron. Residual archival sera (N = 81) received between January 2020 and March 2022 were included. Infection status was inferred by a positive SARS-CoV-2 RT-PCR and/or serology (anti-N and anti-S antibodies) and sequencing of contemporaneous samples (N = 18) to infer lineage. We estimated the levels and cross-reactivity of infection-induced sera including Wild, Delta, Omicron as well as vaccine breakthrough infections (Delta and Omicron). We found an approximately two-fold increase in spike-specific IgG antibody binding in post-Omicron infection compared with the pre-Omicron period, whilst the change in pre- and post-Delta infections were similar. Further investigation of Omicron-specific humoral responses revealed primary Omicron infection as an inducer of cross-reactive antibodies against predecessor variants, in spite of the weaker degree of humoral response compared to Wuhan-Hu-1 and Delta infection. Intriguingly, Omicron vaccine-breakthrough infections when compared with primary infections, exhibited increased humoral responses against RBD (7.7-fold) and Trimeric S (Trimeric form of spike protein) (34.6-fold) in addition to increased binding of IgGs towards previously circulating variants (4.2 - 6.5-fold). Despite Delta breakthrough infections showing a higher level of humoral response against RBD (2.9-fold) and Trimeric S (5.7-fold) compared to primary Delta sera, a demonstrably reduced binding (36%-49%) was observed to Omicron spike protein. Omicron vaccine breakthrough infection results in increased intensity of humoral response and wider breadth of IgG binding to spike proteins of antigenically-distinct, predecessor variants.

Manipulation of flowering time for adaptation through natural or genetic approaches may combat heat-stress damage that occurs at the reproductive stages in production conditions. HD2733, a popular wheat variety of the eastern plains of India, is largely sensitive to heat stress. Therefore, the current study aims to improve heat tolerance of HD2733 by introgression of QTLs associated with early anthesis and high kernel weight linked to markers Xbarc186 and Xgwm190, respectively, through marker-assisted backcross breeding (MABB) from a tolerant donor, WH730. A total of 124 simple sequence repeat (SSR) markers distributed evenly across the genome were used for the background selection. The alleles of Xbarc186 and Xgwm190 were fixed in BC2F1 and BC1F2 generations by selecting individual plants heterozygous for both marker loci and backcrossed with HD2733 and simultaneously selfed to generate BC2F1 and BC1F2 populations, respectively. Furthermore, the selected BC1F2 were selfed to generate the BC1F4 population. By background screening, a total of 39 BC2F3 and 21 BC1F4 families homozygous for the targeted QTLs with 90.9-97.9% and 86.8-88.3% RPG recoveries were selected. The best performing 17 BC2F3 and 10 BC1F4 lines were evaluated for various morpho-physiological traits. Phenotypic evaluation and multi-location trials of the introgressed lines under late sown conditions led to the selection of three promising lines with early anthesis and higher grain yield. The improved lines will serve as an excellent genetic material for functional genomics and expression studies to understand the molecular mechanisms and pathways underlying the stress tolerance.

The present study was aimed to develop silybin phytosome (SIBP) and evaluate its effectiveness against cerebral ischemia-reperfusion (CIR) injury in rats. Initially, SIBP was prepared and characterized with Fourier transform-infrared spectroscopy, differential scanning calorimetry, and scanning electron microscopy. Drug loading and entrapment efficiency of SIBP were also calculated. High-performance liquid chromatography was used to carry out bioavailability studies of SIBP. Adult Wistar rats were divided randomly into five groups. The CIR injury was induced after 14 days of pretreatment by occlusion of bilateral common carotid arteries for 30 min followed by 4 h of reperfusion. Biochemical estimation, histopathological studies, and in silico studies were carried out. Bioavailability studies revealed that SIB concentration was increased to twofolds in SIBP-treated rats. SIBP treatment significantly increases superoxide dismutase and glutathione levels while it decreases monoaldehyde, tumor necrosis factor-α (TNF-α), and interleukin 6 (IL-6) levels in both the hippocampus and cortex of the SIBP-treated CIR-injured rats. Histopathological studies reveal SIBP treatment alleviates cortex cell death and arrangement of CA1 neurons in CIR-injured rats. In silico studies against proteins (TNF-α and IL-6) involved in cerebral ischemia revealed that silybin (SIB) exhibits strong binding interaction with the target proteins when compared to thalidomide which was used as the positive control. Phytosome increase SIB bioavailability and SIBP treatment showed promising results when compared to treatment with SIB only. Based on our study, we conclude that phytosome is a suitable drug delivery agent to the brain for SIB as SIBP treatment was able to provide neuroprotective action against CIR injury.

The present investigation was framed to understand the genetics and development of conspicuous purple coloured corolla tip flower and multicapsules at axil in sesame (Sesamum indicum L.) from the cross between genotypes IC-205776 (♀) 9 EC-118591 (♂). The conspicuous corolla lip colour is recessive in expression and under digenic control, differing from the earlier reports. The ratio at F2 generation was best fit for 13:3 indicating inhibitory gene action for purple corolla lip colour. Among two genes, one acts as an inhibitory gene at recessive condition to produce conspicuous purple corolla lip colour. Multicapsules/axil is dominant in expression, controlled by more than one gene. The ratio of multiple capsules/axil and single capsules/axil at F2 generation was the best fit for the ratio 11:5 indicating dominance modification of duplicate genes for a number of capsules per axil. Single capsule/axil results due to dominance modification of duplicate genes where the homozygous condition of one gene reverses the dominance relation of another gene in heterozygous condition. Joint segregation analysis indicated independent segregation of corolla lip colour and capsule number per axil.

BACKGROUND: To emphasize the role of odontometric parameters which may be used as a reliable forensic tool especially in cases with partial remains of the individual. AIM: To assess the reliability of odontometric parameters in stature analysis. MATERIALS AND METHODS: The study was conducted on 100 patients (50 male and 50 Female). Mesiodistal width of anterior teeth, inter-canine width, Inter-premolar width and maxillary arch length were the parameters included. The results were tabulated with a linear regression formula obtained for each parameter. STATISTICAL ANALYSIS: The data collected was statistically analysed using SPSS version 20 and a linear regression formula was obtained thereafter. RESULTS: Mesio-distal widths of individual maxillary canines, total mesiodistal width of maxillary anteriors and arch length showed a significant co-relation with stature. The combined linear regression formula was obtained for all parameters. CONCLUSION: Odontometric parameters of mesio - distal widths of individual maxillary canines, total mesiodistal width of maxillary anteriors and arch length can be used as reliable parameters for stature analysis.

Malnutrition is considered as major public health concern and is emerging challenge to food and nutrition security particularly in developing countries. Rice is the staple food and consumed by the half of the world's population which is the source of daily requirement of the nutrients. Attempts are being made to fortify rice with micronutrients, but the loss or retention of these micronutrients in different cooking methods is not well studied and documented especially in fortified rice. In the present study, paddy seeds of six Indian varieties were fortified with iron and zinc by parboiling process. Consequently, fortified polished rice had higher micronutrient contents (Fe, 106.31 ± 12.56; Zn, 97.72 ± 9.75) than non-fortified polished rice (Fe, 7.44 ± 1.05; Zn, 14.74 ± 2.94) expressed in ppm. Polished rice of both fortified and non-fortified were cooked under five different cooking conditions and analyzed for remaining iron and zinc content. Cooking rice in rice cooker without prior washing (NRC) retained highest concentration of Fe and Zinc in both fortified and non-fortified rice varieties. It also showed that fortified rice suffered higher percentage loss of micronutrient, than the non-fortified rice. But the average retained micronutrient amount measured in ppm, was higher in fortified rice (Fe, 43.54 ± 6.88; Zn, 36.7 ± 3.12) than in non-fortified rice (Fe, 4.24 ± 0.87; Zn, 9.3 ± 2.11). Hence, adopting appropriate cooking method, higher amount of micronutrients will be retained in the cooked food which will in turn help in combating the malnutrition and improve health.

INTRODUCTION: Disease transmission patterns of COVID-19 have shown that masking, social distancing, contact tracing and quarantine measures are important strategies for reducing transmission. The effective implementation of quarantine is determined by the commitment of the people and monitoring by the State. The aim of the study was to find out the effectiveness of home quarantine practises and its role in determining SARS CoV2 transmission. METHODS: Record-based retrospective cohort study was conducted among expatriates of Kerala who were on quarantine at their homes and later tested positive for SARS -CoV-2. Quarantine practises were categorised as strict room quarantine, incomplete room quarantine, home quarantine and no quarantine. Risk of transmission was assessed using risk ratios. Multiple logistic regression analysis was performed to find out the determinants of SARS CoV2 transmission. RESULTS: The median (IQR) age and duration of quarantine of 95 study participants were found to be 35 (29, 44) years and 7 (3,13) days, respectively. Majority of the participants practised strict room quarantine (57%), whereas 11.6%, 16.8% and 14.7% practiced incomplete room, home and no quarantine, respectively. Home quarantine without room quarantine had 24 times odds for transmitting disease [OR (95%CI)): 24.14 (4.87--119.75), P < 0.001] and not being in quarantine for any duration before being diagnosed was found to be 14 times riskier when compared with strict room quarantine [OR (95%CI)): 14.44 (2.42-86.17), P = 0.003]. DISCUSSION: Low-resource settings successful in the initial phases of COVID-19 pandemic should make periodic revisions in the quarantine guidelines while continually promoting physical distancing strategies.

Alveolar soft part sarcoma is a rare soft tissue tumor with uncertain histogenesis. It is a slow growing tumor with a high rate of metastasis. The tumor is not easily identified as clinical symptoms are not pronounced. The retroperitoneum is a rare location of tumor, with a few cases published in literature. Surgical excision is the mainstay of treatment. Here we describe a rare case of a large retroperitoneal Alveolar soft part sarcoma in a young female with radiological and histopathological findings.

The genus Lathyrus consists of more than 184 herbaceous annual and perennial species suitable for multifaceted sustainable food and feed production system in the arid and semi-arid regions of the world. The grasspea is a promising source of protein nutrition. However, its potential is not being utilized fully due to the presence of neurotoxin content (ß-N-oxalyl-l-α, ß diaminopropionic acid, ß-ODAP), a causal agent of non-reversible lower limbs paralysis. The high protein contents in seeds and leaves with ~90% digestibility make it sustainable super food to beat protein malnutrition in future. Therefore, it is desired to breed new grasspea cultivars with low ß-ODAP contents. Limited research has been carried out to date about this feature. A draft genome sequence of grasspea has been recently published that is expected to play a vital role in breeding and identifying the genes responsible for biosynthesis pathway of ß-ODAP contents in grasspea. Efforts to increase awareness about the importance of genus Lathyrus and detoxify ß-ODAP in grasspea are desired and are in progress. Presently, in South Asia, systematic and dedicated efforts to support the farmers in the grasspea growing regions by disseminating low ß-ODAP varieties has resulted in a considerable improvement in reducing the incidence of neurolathyrism. It is expected that the situation will improve further by mainstreaming grasspea cultivation by implementing different approaches such as the development and use of low ß-ODAP varieties, strengthening government policies and improved detox methods. The present review provides insight into the multifaceted characteristics of sustainable nutritious grasspea in the global and Indian perspective.

Chitinase from the leaves of Simarouba glauca, a plant used in traditional anti-inflammatory therapy is purified and characterized. Peptide mass finger print analysis revealed the protein as an endo-chitinase which was further confirmed using chitin-agar assay. The enzyme exhibited significant anti-fungal efficacy against phyto-pathogens such as Macrophomina phaseolina, Fusarium oxysporum and Sclerotium rolfsii. Chitinolysis was also examined against insoluble chitin using SEM. Using X-ray diffraction data up to 1.66 Å, the structure was determined by Molecular Replacement using crystal structure of GH19 Chitinase-like protein from Hevea brasiliensis. During structure refinement, an extra domain could be traced and identified as hevein domain. To our knowledge, this is the first report of any chitinase with intact hevein domain. The GH19 chitinase and hevein domains though connected by a lengthy loop, are restricted to be close by disulfide bridges. These bridges connecting each domain with the loop may be important for proper chitin feeding into the active site. By considering reports on hevein and chitinase domains as well as the traditional use of the plant, this report of an intact hevein-chitinase protein and their relative orientation may add further insights for the usefulness of this protein.

AIMS AND OBJECTIVES: The aim of this study was to evaluate the effectiveness of three different caries excavation methods using micro-computed tomography (micro-CT). MATERIALS AND METHODS: Fifteen freshly extracted human molar teeth with occlusal dentinal caries were selected. The teeth were sectioned longitudinally into two halves and were randomly divided into three groups (n = 10) depending on the caries removal technique: Group 1: tungsten carbide (TC) bur, Group 2: cerabur (CB), and Group 3: Excavus (EX) tipEX). A preoperative micro-CT scan of all the samples was taken. The caries excavation procedures were carried out followed by postoperative micro-CT scan. The preoperative and postoperative scans of each tooth were analyzed for caries removal effectiveness (CRE), mineral density (MD), and minimal invasiveness potential (MIP) using Avizo 9.4 software (Thermo Fisher Scientific, Germany). Statistical analysis was conducted by applying three-way analysis of variance and independent sample t-test using the Statistical Package for the Social Sciences software, version 20.0 (IBM Corporation, USA). RESULTS: Among the three groups, the TC group had the smallest RC/IC (residual caries/initial caries) ratio, highest mean MD at the cavity floor, and highest MIP. The EX group significantly had the highest RC/IC, lowest mean MD, and lowest MIP. Both the CRE and MIP parameters of CB group were acceptable (RC/IC = 0.08, mean MD = 1.09g/cm3, and MIP = 1.09). CONCLUSION: As compared with the three excavation methods, CBs can be considered as an alternative to TC burs because of its MIP and complete removal of infected carious dentin.

Perivascular adipose tissue (PVAT), the fat surrounding peripheral blood vessels, is protective and reduces the contraction of blood vessels in health. PVAT is composed of adipocytes, stromal cells, and immune cells. Recent work supports eosinophils as one of the cell types key to the anti-contractile nature of PVAT in health. Hence, we hypothesized that there exists a basally activated immune cell community in healthy PVAT that is distinctly different from non-PVAT fats. PVATs were from around mesenteric resistance vessels (MRPVAT - white fat) and thoracic aorta (APVAT - brown fat). Non-PVATs included retroperitoneal (RP fat - white fat) and subscapular (SS fat - brown fat) while the spleen was a positive control. Tissues were harvested from adult male and female Sprague Dawley rats. Six primary immune cell types were identified in PVATs. T cells (CD4 and CD8), B cells, natural killer (NK) cells, macrophages, mast cells, and neutrophils in the stromal vascular fraction of each fat were identified using nine-color flow cytometry. PVATs contained a higher number of total immune cells vs. their respective non-PVAT fats in females. Females had a higher number of T cells in MRPVAT vs. males. Females also had a greater number of T cells and total immune cells in APVAT vs. males. Further, activation, differentiation, and/or polarization of various immune cell types were similarly determined by flow cytometry. PVATs were similar to their respective non-PVAT fats in density of recently activated B cells (B220+ CD25+). However, MRPVAT in females had a higher number of naïve CD4 T cells vs. MRPVAT in males and APVAT in females. MRPVAT also had denser naïve CD8 T cells vs. APVAT in females. Overall, this research for the first time has identified a community of discrete populations of immune cells (naive/recently activated/regulatory/memory) in healthy PVATs. Contrary to our hypothesis, PVATs are more similar than different in density to their respective non-PVAT fats.
