AIM: A study was undertaken to evaluate the optimum roughage to concentrate ratio in maize stover (MS) based complete diets for efficient microbial biomass production (EMBP) using in vitro gas production technique. MATERIALS AND METHODS: MS based complete diets with roughage to concentrate ratio of 100:0, 90:10, 80:20, 70:30, 60:40, 50:50, 40:60, and 30:70 were formulated, and 200 mg of oven-dried sample was incubated in water bath at 39°C along with media (rumen liquor [RL] - buffer) in in vitro gas syringes to evaluate the gas production. The gas produced was recorded at 8 and 24 h of incubation. In vitro organic matter digestibility (IVOMD), metabolizable energy (ME), truly digestible organic matter (TDOM), partitioning factor (PF), and EMBP were calculated using appropriate formulae. Ammonia nitrogen and total volatile fatty acids (TVFAs) production were analyzed in RL fluid-media mixture after 24 h of incubation. RESULTS: In vitro gas production (ml) at 24 h incubation, IVOMD, ME, TDOM, TVFA concentration, and ammonia nitrogen production were increased (p<0.01) in proportion to the increase in the level of concentrate in the diet. Significantly (p<0.01) higher PF and EMBP was noticed in total mixed ration with roughage to concentrate ratio of 60:40 and 50:50 followed by 70:30 and 40:60. CONCLUSION: Based on the results, it was concluded that the MS can be included in complete rations for ruminants at the level of 50-60% for better microbial biomass synthesis which in turn influences the performance of growing sheep.

AIM: This study was undertaken to assess the pharmacodynamic interaction of fenugreek, insulin and glimepiride on sero-biochemical parameters in streptozotocin-induced diabetic rats. MATERIALS AND METHODS: A total of 56 male Sprague-Dawley rats, randomly divided into seven Groups. Group 1: Non-diabetic control; Group 2: Streptozotocin induced diabetic control; Groups 3, 4 and 5 were treated with insulin, glimepiride and fenugreek seed powder, respectively; Groups 6 and 7: Insulin + fenugreek seed powder treatment and glimepiride + fenugreek seed powder treatment respectively, in diabetic rats. Body weights, blood glucose, lipids total cholesterol (TC), high-density lipoprotein (HDL), low-density lipoprotein (LDL), triglycerides (TG) and proteins (total protein, albumin, globulin, A/G ratios) were studied at different time intervals. Rats were sacrified at the end of 8 weeks, pancreas and aorta collected for histopathological study. RESULTS: The results of Group 2 showed significantly (p<0.05) higher concentration of glucose, TC, TG, LDL, globulin, A/G ratios and significantly (p<0.05) lower concentration of albumin, total protein, HDL and body weights when compared to Group 1 at the end of 4(th) and 8(th) weeks intervals with marked alteration in histopathology of pancreas and aorta. All the treatment Groups 3-7 showed significantly (p<0.05) improvement in the all the parameters and the Groups 6 and 7 showed highest decrease in the concentration blood glucose, TC, TG, LDL and increase in the albumin, total protein and body weights during 6(th) and 8(th) week, respectively. CONCLUSION: The treatment with fenugreek, insulin and glimepiride countered the alteration in the sero biochemical parameters in diabetic rats, and their combination was found a positive interaction in improving the sero biochemical status of diabetic rats.

AIM: Present experiment was conducted to investigate the effect of selenium (Se) supplementation on performance, carcass characteristics, meat composition, shelflife of meat and biochemical profile in Nellore ram lambs. MATERIALS AND METHODS: 24 male Nellore ram lambs (15.75±0.47 kg) were randomly divided into four dietary groups with six lambs in each and reared under uniform management conditions for 120 days. Basal diet was not supplemented with Se and consisted of green fodder (Se 0.09 mg/kg dry matter [DM]), dry roughage (Se 0.11 mg/kg DM) and concentrate mixture (Se 0.019 mg/kg DM) and fed individually. Dietary treatments were prepared by adding graded levels Se (0, 0.45, 0.9, and 1.8 ppm) to concentrate mixture (1% body weight [BW]) from sodium selenite. Feed offered and refusal measured daily; and BWs were measured at fortnight interval to find out average daily gain (g), feed conversion ratio (FCR), cost economics and plane of nutrition. Serum biochemical profile (concentration of glucose, total protein, albumin, globulin, cholesterol, and hemoglobin) was assessed on 0, 60(th), and 120(th) day. At the end of experiment, the carcass characteristics (dressing percentage, cut-up parts, meat to bone ratio) and meat chemical composition were evaluated. Meat keeping (thiobarbituric acid reactive substances) quality from different groups was evaluated on day 0, 3, and 6 post-slaughter. RESULTS: Dietary Se supplementation did not show any effect on weight gain, FCR, cost economics, plane of nutrition, and serum biochemical profile in Nellore ram lambs. However, Se supplemented lambs had numerically higher weight gain than the unsupplemented lambs. Similarly, carcass characteristics and keeping quality were comparable among the four treatments. However, numerical increase in post-slaughter keeping quality with increasing Se supplementation was observed. CONCLUSION: It can be concluded that supplementation of Se in the form of sodium selenite (inorganic source) at different levels did not influence animal performance in growing Nellore ram lambs had no effect on lamb performance, cost economics, carcass characteristics, and serum biochemical profile.

The present study was conducted to assess the effect of fenugreek, insulin and glimepiride alone and their combination in diabetic rat liver. Fifty six male Sprague dawley rats of uniform age were randomly divided into seven groups. Group 1: Non-diabetic control; Group 2: Streptozotocin (40 mg/Kg i/p single dose)-induced diabetic control; Group 3: Insulin (4 U/kg once daily for 8 weeks) treatment in diabetic rats; Group 4: Glimepiride (4 mg/Kg orally once daily for 8 weeks) treatment in diabetic rats; Group 5: Fenugreek seed powder treatment (1 g/kg orally once daily for 8 weeks) in diabetic rats; Group 6: Insulin + Fenugreek seed powder treatment (once daily for 8 weeks) in diabetic rats; Group 7: Glimepiride + Fenugreek seed powder treatment (once daily for 8 weeks) in diabetic rats. Livers were collected at the end of experiment for histopathology and estimation of reduced glutathione (GSH), thiobarbituric acid reacting substances (TBARS), protein carbonyls, glutathione S-transferase (GST), glucose-6-phosphate dehydrogenase (G6PD), Na(+)/K(+) ATPase and Mg(2)+ ATPase, cytochrome P450 (CYP) and glycogen. There was an increase in the concentration of TBARS and protein carbonyls, and decrease in the concentration of GSH and glycogen, and the activity of GST, G6PD, Na(+)/K(+) ATPase and Mg(2)+ ATPase in diabetic livers, while treatment groups showed significant (P < 0.05) increase in the above parameters. The histology of liver revealed marked changes in diabetic rats and mild changes in combination treatment groups. The treatment with fenugreek, insulin and glimepiride improved the liver parameters in diabetic rats and their combination showed a beneficial effect on liver.

An experimental study was conducted to evaluate the toxicological effects, if any, due to feeding of Bt (Bacillus thuringiensis) cotton plants to sheep. A total of 32 sheep of one year of age belonging to Deccani breed were randomly divided into four groups, consisting of eight sheep in each group. Group 1 was maintained on basal diet (concentrate feed at the rate of 300 g + green fodder at the rate of 3 kg/sheep/day), group 2 on non-Bt cotton plant at the rate of 1.5 kg + green fodder at the rate of 1.5 kg + concentrate feed at the rate of 300 g/sheep/day, group 3 on Bt cotton plants (50%) at the rate of 1.5 kg + green fodder at the rate of 1.5 + concentrate feed at the rate of 300 g/sheep/day, and group 4 on Bt cotton plants ad libitum + concentrate feed at the rate of 300 g/sheep/day. All the groups of sheep were maintained for three months and various hemato-biochemical parameters were studied at monthly intervals. The activity of aspartate transaminase, gamma glutamyltransferase, and creatine kinase in sera samples, and the concentration of blood urea nitrogen and creatinine did not differ significantly among different groups at different time intervals. The histological examination of liver and kidney did not reveal any significant changes in Bt and non-Bt cotton-fed groups. In conclusion, the results of the present investigation enunciated that feeding of genetically modified (Bt) cotton plants to sheep was without detrimental effects in the biological system of sheep.
