BACKGROUND: Papillary thyroid cancer is an inert malignant tumor with a good response to surgical treatment, low recurrence and metastasis rate, and good prognosis. Diffuse sclerosing thyroid cancer is an invasive subtype that is more common in young people, with a higher rate of lymph node metastasis and recurrence, and a relatively poor prognosis. PATIENT CONCERNS: A 13-year-old girl underwent radical surgery for diffuse sclerosing thyroid cancer. Eight years later, due to a large number of lymph node metastases, she underwent another radical surgery on her neck lymph nodes. METHODS: The patient thyroid ultrasound and neck enhanced CT indicated that the patient had multiple enlarged lymph nodes in the neck with irregular morphology and structure, and the possibility of metastatic lymph nodes was high. Subsequently, the patient underwent thyroid fine-needle aspiration and the results showed that cancer cells were detected in both cervical lymph nodes. DIAGNOSIS: The patient was diagnosed with bilateral cervical lymph node metastases after thyroid surgery. RESULTS: After the second surgery, the patient recovered well, and no residual or focal iodine uptake tissue was found on the enhanced CT examination. CONCLUSION: As diffuse sclerosing thyroid cancer is prone to lymph node and recurrent metastases, once it is diagnosed, radical treatment should be actively performed. Postoperative adjuvant radiation therapy should be administered according to the patient condition and regular follow-ups should be conducted to monitor neck lymph node metastasis.

Ecological divergence due to habitat difference plays a prominent role in the formation of new species, but the genetic architecture during ecological speciation and the mechanism underlying phenotypic divergence remain less understood. Two wild ancestors of rice (Oryza rufipogon and Oryza nivara) are a progenitor-derivative species pair with ecological divergence and provide a unique system for studying ecological adaptation/speciation. Here, we constructed a high-resolution linkage map and conducted a quantitative trait locus (QTL) analysis of 19 phenotypic traits using an F2 population generated from a cross between the two Oryza species. We identified 113 QTLs associated with interspecific divergence of 16 quantitative traits, with effect sizes ranging from 1.61% to 34.1% in terms of the percentage of variation explained (PVE). The distribution of effect sizes of QTLs followed a negative exponential, suggesting that a few genes of large effect and many genes of small effect were responsible for the phenotypic divergence. We observed 18 clusters of QTLs (QTL hotspots) on 11 chromosomes, significantly more than that expected by chance, demonstrating the importance of coinheritance of loci/genes in ecological adaptation/speciation. Analysis of effect direction and v-test statistics revealed that interspecific differentiation of most traits was driven by divergent natural selection, supporting the argument that ecological adaptation/speciation would proceed rapidly under coordinated selection on multiple traits. Our findings provide new insights into the understanding of genetic architecture of ecological adaptation and speciation in plants and help effective manipulation of specific genes or gene cluster in rice breeding.

Energy utilization rates have been largely improved thanks to the wide application of smart grids, thereby realizing the reliable, economic and efficient operation of the grids. However, such an application is also accompanied by many security issues. In response to the many problems within existing security schemes, such as not supporting the communication between heterogeneous cryptosystems, low security levels and a low data retrieval efficiency, a heterogeneous signcryption (HSC) scheme that supports a trusted multi-ciphertext equality test (MET) is proposed. The adoption of the HSC helps to identify secure communications from identity-based cryptosystems to certificateless cryptosystem, eliminates the certificate management problems in the traditional public key cryptography scheme, and ensures the confidentiality and authentication of power data. The introduction of the MET technology can avoid the high cost of equality test calculations after grouping ciphertexts in pairs. Using blockchain and smart contract technologies ensure the credibility of test results and eliminates the reliance on trusted cloud servers. Under the random oracle model, on the basis of the bilinear Diffie-Hellman, the computational Diffie-Hellman and the q-strong Diffie-Hellman problems, this paper proves that the scheme proposed herein meets the requirements of indistinguishability and one-way security under adaptive choice ciphertext attacks, and the unforgeability under the adaptive choice message attack. From the findings of the analysis, it has been shown that the proposed scheme satisfies more security attributes and requires lower computational overhead compared to similar schemes.

The origin of domesticated Asian rice (Oryza sativa L.) has been controversial for more than half a century. The debates have focused on two leading hypotheses: a single domestication event in China or multiple domestication events in geographically separate areas. These two hypotheses differ in their predicted history of genes/alleles selected during domestication. Here we amassed a dataset of 1,578 resequenced genomes, including an expanded sample of wild rice from throughout its geographic range. We identified 993 selected genes that generated phylogenetic trees on which japonica and indica formed a monophyletic group, suggesting that the domestication alleles of these genes originated only once in either japonica or indica. Importantly, the domestication alleles of most selected genes (~80%) stemmed from wild rice in China, but the domestication alleles of a substantial minority of selected genes (~20%) originated from wild rice in South and Southeast Asia, demonstrating separate domestication events of Asian rice.

The Internet of Things (IoT), driven by wireless communication and other technologies, is gradually entering our lives and promoting the transformation of society from "informatization" to "intelligence". Certificateless signature (CLS) eliminates the characteristic of certificate management, making it an effective method for verifying large-scale data in the IoT environment. Nevertheless, hash functions are regarded as ideal random oracles in the security proofs of most CLS schemes, which cannot guarantee the security of CLS schemes in reality. In response to this problem, Shim devised a CLS scheme without random oracles in the standard model and declared it to be provably secure. Unfortunately, in this paper, we cryptanalyze Shim's CLS scheme and demonstrate that it is not resistant to public key replacement attacks from a Type Ⅰ attacker. Furthermore, to further improve the security of the Shim CLS scheme and avoid the single-point failure of the KGC and the signature forgery initiated, we propose a blockchain-based CLS scheme without a random oracle. Finally, we evaluate the comprehensive performance, and while maintaining the computational and communication performance of the Shim scheme, we resist both Type Ⅰ and Type Ⅱ attackers, as well as signature forgery initiated against public parameters.

Internet of Drones (IoD) is considered as a network and management architecture, which can enable unmanned aerial vehicles (UAVs) to collect data in controlled areas and conduct access control for UAVs. However, the current cloud-assisted IoD scheme cannot efficiently achieve secure communication between heterogeneous cryptosystems, and does not support multi-ciphertext equality tests. To improve the security and performance of traditional schemes, we propose a heterogeneous signcryption scheme (HSC-MET) that supports multi-ciphertext equality test. In this paper, we use a multi-ciphertext equality test technique to achieve multi-user simultaneous retrieval of multiple ciphertexts safely and efficiently. In addition, we adopt heterogeneous signcryption technology to realize secure data communication from public key infrastructure (PKI) to certificateless cryptography (CLC). At the same time, the proposed scheme based on the computation without bilinear pairing, which greatly reduces the computational cost. According to the security and performance analysis, under the random oracle model (ROM), the confidentiality, unforgeability and number security of HSC-MET are proved based on the computational Diffie-Hellman (CDH) problem.

It is of critical importance for our understanding of speciation process to determine the forms of reproductive isolation and their relative importance in species divergence. Oryza nivara and O. rufipogon are direct ancestors of Asian cultivated rice and a progenitor-daughter species pair. Investigating the reproductive isolation between them provides insights into plant speciation and helps understanding of the rice domestication. Here, we quantitatively measured the major components of reproductive isolation between the two species based on common garden and crossing experiments for three pairs of sympatric populations in Nepal, Cambodia and Laos. We revealed significant differences in the flowering times between species pairs, with O. nivara flowering much earlier than O. rufipogon. A very weak reduction in seed set but no reduction in F1 viability and fertility were detected for the crosses between species relative to those within species. Moreover, we detected asymmetrical compatibility between species and found that emasculation significantly decreased pollination success in O. nivara but not in O. rufipogon. Our study demonstrates that the divergence between O. nivara and O. rufipogon is maintained almost entirely by the difference in flowering times and suggests that differential flowering times contribute to both habitat preferences and reproductive isolation between species.

The occurrence of parallel speciation strongly implies the action of natural selection. However, it is unclear how general a phenomena parallel speciation is since it was only shown in a small number of animal species. In particular, the adaptive process and mechanisms underlying the process of parallel speciation remain elusive. Here, we used an integrative approach incorporating population genomics, common garden, and crossing experiments to investigate parallel speciation of the wild rice species Oryza nivara from O. rufipogon. We demonstrated that O. nivara originated multiple times from different O. rufipogon populations and revealed that different O. nivara populations have evolved similar phenotypes under divergent selection, a reflection of recurrent local adaptation of ancient O. rufipogon populations to dry habitats. Almost completed premating isolation was detected between O. nivara and O. rufipogon in the absence of any postmating barriers between and within these species. These results suggest that flowering time is a "magic" trait that contributes to both local adaptation and reproductive isolation in the origin of wild rice species. Our study thus demonstrates a convincing case of parallel ecological speciation as a consequence of adaptation to new environments.

Mycobacterium tuberculosis (M. tuberculosis) regions of difference (RD) encode proteins which are potentially useful as diagnostic reagents for tuberculosis (TB). In this study, 75 genes from M. tuberculosis RD1-RD16 were successfully cloned from which 68 proteins were expressed and purified. Three serum pools from patients with pulmonary TB (PTB), extra-pulmonary tuberculosis (EPTB) and healthy controls (HC) were used to preliminarily screen individual RD proteins. The OD630 ratio of the PTB or EPTB to the HC group ≥ 2-fold was positive. As a result, 29 proteins were obtained. The serological response to the identified antigens was further verified using 58 PTB samples with 38 sera from smear-positive PTB (PTB-SP) patients and 20 sera from smear-negative PTB (PTB-SN) patients, 16 EPTB samples, 42 latent M. tuberculosis infection samples and 40 HCs by indirect ELISA. With respect to the PTB diagnosis, receiver operating characteristic analysis showed that Rv0222 [area under the curve (AUC), 0.8129; 95% confidence interval (CI), 0.7280-0.8979] and Rv3403c (AUC, 0.8537; 95% CI, 0.7779-0.9294) performed better than ESAT6/CFP10 (AUC, 0.7435; 95% CI, 0.6465-0.8406). Rv0222 and Rv3403c demonstrated the highest diagnostic ability in the PTB-SP group (sensitivity, 86.8%; specificity, 80%), while Rv3403c demonstrated the highest diagnostic ability in the PTB-SN group (sensitivity, 70%; specificity, 80%). With respect to the EPTB diagnosis, Rv0222 exhibited the highest diagnostic value (AUC, 0.7523; sensitivity, 68.8%; specificity, 87.5%). In addition, the combination of Rv0222 and Rv3403c improved the test for PTB-SN. These results indicate that Rv0222 and Rv3403c would be potential diagnostic biomarkers for active TB serodiagnosis. Mouse experiments demonstrated that Rv0222 and Rv3403c elicited specific cellular and humoral responses which were characterized by production of IFN-γ, IgG1, and IgG2a, but a higher level of IgG1 than IgG2a.

This study identified urinary biomarkers for tuberculosis (TB) diagnosis. The urine proteomic profiles of 45 pulmonary tuberculosis patients prior to anti-TB treatment and 45 healthy controls were analyzed and compared using two-dimensional electrophoresis with matrix-assisted laser desorption/ionization time of flight mass spectrometry. Nineteen differentially expressed proteins were identified preliminarily, and western blotting and qRT-PCR were performed to confirm these changes at the translational and transcriptional levels, respectively, using samples from 122 additional pulmonary tuberculosis patients and 73 additional healthy controls. Two proteins, mannose-binding lectin 2 and a 35-kDa fragment of inter-α-trypsin inhibitor H4, exhibited the highest differential expression. We constructed a protein-microRNA interaction network that primarily involved complement and inflammatory responses. Eleven microRNAs from microRNA-target protein interactions were screened and validated using qRT-PCR with some of the above samples, including 97 pulmonary tuberculosis patients and 48 healthy controls. Only miR-625-3p exhibited significant differential expression (p < 0.05). miR-625-3p was increased to a greater extent in samples of smear-positive than smear-negative patients. miR-625-3p was predicted to target mannose-binding lectin 2 protein. A binary logistic regression model based on miR-625-3p, mannose-binding lectin 2, and inter-α-trypsin inhibitor H4 was further established. This three-biomarker combination exhibited better performance for tuberculosis diagnosis than individual biomarkers or any two-biomarker combination and generated a diagnostic sensitivity of 85.87% and a specificity of 87.50%. These novel urine biomarkers may significantly improve tuberculosis diagnosis.

Purpose: With the development of digital X-ray imaging and processing methods, the categorization and analysis of massive digital radiographic images need to be automatically finished. What is crucial in this processing is the automatic retrieval and recognition of radiographic position. To address these concerns, we developed an automatic method to identify a patient's position and body region using only frequency curve classification and gray matching. Methods: Our new method is combined with frequency analysis and gray image matching. The radiographic position was determined from frequency similarity and amplitude classification. The body region recognition was performed by image matching in the whole-body phantom image with prior knowledge of templates. The whole-body phantom image was stitched by radiological images of different parts. Results: The proposed method can automatically retrieve and recognize the radiographic position and body region using frequency and intensity information. It replaces 2D image retrieval with 1D frequency curve classification, with higher speed and accuracy up to 93.78%. Conclusion: The proposed method is able to outperform the digital X-ray image's position recognition with a limited time cost and a simple algorithm. The frequency information of radiography can make image classification quicker and more accurate.

Objective: Designed a method that can automatically distinguish the X-ray radiological positions based on image information.For any selected input clinical X-ray digital images, we extract the image outline in the edge detection method using optimal threshold selection, then process binarization. After compared with the whole body phantom X-ray images, then process gray level similarity traverse detection.By optimizing the decision, the best matching region of the same input film was obtained, and the imaging part of the patient is determined based on prior knowledge.The algorithm is fast and has a high degree of robust.

To construct a universal vaccine against mastitis induced by either Streptococcus agalactiae or Staphylococcus aureus, the B cell epitopes of the surface immunogenic protein (Sip) from S. agalactiae and clumping factor A (ClfA) from S. aureus were analyzed and predicted. sip-clfA, a novel chimeric B cell epitope-based gene, was obtained by overlap PCR, and then the recombinant Sip-ClfA (rSip-ClfA) was expressed and purified. rSip-ClfA and inactivated S. agalactiae and S. aureus were formulated into different vaccines with mineral oil as the adjuvant and evaluated in mouse models. The rSip-ClfA vaccination induced immunoglobulin G (IgG) titers higher than those seen in groups immunized with inactivated bacteria. Furthermore, the response to rSip-ClfA immunization was characterized as having a dominant IgG1 subtype, whereas both bacterial immunizations produced similar levels of IgG1 and IgG2a. The antiserum capacities for opsonizing adhesion and phagocytosis were significantly greater in the rSip-ClfA immunization group than in the killed-bacterium immunization groups (P < 0.05). The immunized lactating mice were challenged with either S. agalactiae or S. aureus via the intramammary route. At 24 h postinfection, the numbers of bacteria recovered from the mammary glands in the rSip-ClfA group were >5-fold lower than those in both inactivated-bacterium groups (P < 0.01). Histopathological examination of the mammary glands showed that rSip-ClfA immunization provided better protection of mammary gland tissue integrity against both S. agalactiae and S. aureus challenges. Thus, the recombinant protein rSip-ClfA would be a promising vaccine candidate against mastitis induced by either S. agalactiae or S. aureus.
