RESUMO

In a bid to explore the on-site biorefinery approach for conversion of forestry residues, lignocellulosic biomass into value-added products was studied. The bark white pine wood was subjected to the microwave technique of fast and slow hydrolysis under varying acid and biomass concentrations to produce levulinic acid (LA). The HCl (2% v/v) and plant biomass (1% w/v) were identified as the optimum conditions for fast wood hydrolysis (270 ºC for 12 sec), which led to maximum LA yield of 446.68 g/kgPB. The proposed sustainable approach is mild, quick, and utilized a very low concentration of the HCl for the production of LA. The hydrolysate was used as a medium for Kluyveromyces marxianus growth to produce 2-phenylethanol (2-PE). K. marxianus used 74-95% of furfural from hydrolysate as a co-substrate to grow. The proposed model of the integrated biorefinery is an affordable on-site approach of using forest waste into localized solutions to produce LA and 2-PE.

RESUMO

An I2-mediated approach for selective C-H functionalization of unprotected aniline derivatives for synthesizing benzils and quinoxaline derivatives from sulfoxonium ylides has been described. Aniline derivatives and sulfoxonium ylides ornamented with different functional groups showed good compatibility. They afforded the corresponding products with moderate to high yields via a mild and simple procedure. Finally, we validated the practicality of this method by scaling up the reaction and further conversion of the synthesized derivatives into other valuable molecules.

BACKGROUND: The etiology of atypical hemolytic uremic syndrome (aHUS) is unknown in 30-40% of patients. Anti-factor B (FB) antibodies are reported in C3 glomerulopathy (C3G) and immune-complex membranoproliferative glomerulonephritis (IC-MPGN), though not in aHUS. METHODS: We screened patients < 18-year-old from cohorts of aHUS and C3G/idiopathic IC-MPGN. Anti-FB IgG antibodies were measured by ELISA and confirmed by Western blot. Normative levels were based on antibody levels in 103 healthy blood donors. RESULTS: Prevalence of anti-FB antibodies was 9.7% (95% CI 6.1-14.5%; n = 21) in 216 patients with aHUS, including 11.5% (95% CI 6.4-18.5%; n = 14) in anti-FH associated aHUS and 11.8% (95% CI 4.4-23.9%; n = 6) in patients without a definitive genetic or autoimmune etiology. Patients with significant genetic variants did not show anti-FB antibodies. In patients with concomitant anti-FB and anti-FH antibodies, median anti-FH titers were higher (11,312 AU/mL vs. 4920 AU/mL; P = 0.04). Anti-FB antibody titer correlated with disease severity (hemoglobin and platelets; P < 0.05), declined following plasma exchange and increased during relapse. While 4/64 patients with C3G (6.3%) and 1/17 with IC-MPGN showed anti-FB antibodies, titers were higher in aHUS (544.8 AU/mL vs. 1028.8 AU/mL; P = 0.003). CONCLUSION: Anti-FB antibodies are present in 6-10% of patients with aHUS and C3G/IC-MPGN, with higher titers in the former. The diagnostic and therapeutic implication of anti-FB antibodies in aHUS needs confirmation and further studies. The study shows propensity for autoantibody generation and co-existence of multiple risk factors for aHUS in Indian children.

BACKGROUND: Plasma exchanges (PEX) and immunosuppression are the cornerstone of management of anti-factor H (FH) antibody-associated atypical hemolytic uremic syndrome (aHUS), particularly if access to eculizumab is limited. The duration of therapy with PEX for anti-FH aHUS is empirical. METHODS: We compared the efficacy of abbreviated PEX protocol (10-12 sessions) in a prospective cohort of patients diagnosed with anti-FH aHUS (2020-2022), to standard PEX protocol (20-22 sessions) in a historical cohort (2016-2019; n = 65). Efficacy was defined as 70% decline in anti-FH titers or fall to ≤ 1300 AU/ml at 4 weeks. Patients in both cohorts received similar immunosuppression with oral prednisolone, IV cyclophosphamide (5 doses) and mycophenolate mofetil. Outcomes included efficacy, rates of hematological remission and adverse kidney outcomes at 1, 3 and 6 months. RESULTS: Of 23 patients, 8.2 ± 2.1 years old enrolled prospectively, two were excluded for significant protocol deviation. PEX was abbreviated in 18/21 (86%) patients to 11.5 ± 3.3 sessions. Abbreviation failed for lack of hematological remission by day 14 (n = 2) and persistent neurological manifestations (n = 1). All patients in whom PEX was abbreviated achieved > 70% reduction in anti-FH titers at day 28. The percentage fall in anti-FH titers was similar for the abbreviated vs. standard PEX protocols at 1, 3 and 6 months. At last follow-up, at median 50 months and 25 months for standard and abbreviated cohorts, the estimated GFR was similar at 104.8 ± 29.1 vs. 93.7 ± 53.4, respectively (P = 0.42). CONCLUSION: Abbreviation of the duration of PEX is feasible and efficacious in reducing anti-FH titers. Short-term outcomes were comparable in patients managed by abbreviated and standard PEX protocols.

Rhodosporidium toruloides, an oleaginous yeast, is a potential feedstock for biodiesel production due to its ability to utilize lignocellulosic biomass-derived hydrolysate with a considerably high lipid titer of 50-70 % w/w. Hence, for the first-time environmental assessment of large-scale R. toruloides-based biodiesel production from wood hydrolysate and crude glycerol was conducted. The global warming potential was observed to be 0.67 kg CO2 eq./MJ along with terrestrial ecotoxicity of 1.37 kg 1,4-DCB eq./MJ and fossil depletion of 0.13 kg oil eq./MJ. The highest impacts for global warming (∼45 %) and fossil depletion (∼37 %) are attributed to the use of chloroform for lipid extraction while fuel consumption for transportation contributed more than 50 % to terrestrial ecotoxicity. Further, sensitivity analysis revealed that maximizing biodiesel yield by increasing lipid yield and solid loading could contribute to reduced environmental impacts. In nutshell, this investigation reveals that environmental impact varies with the type of chemical utilized.

Fusarium wilt caused by Fusarium oxysporum f. sp. pisi (Fop) is an important disease and major obstacle to pea production, causing huge losses to growers. The focus of this study was on isolation followed by morphological, molecular characterization and analyzing the growth of the casual agent under variable temperature, pH and Nitrogen levels. The morphological features of radial growth, sporulation, pigmentation and mycelial characterization were examined and the variability of all isolates was presented. Molecular characterization of the fungus by ITS rDNA sequencing revealed that all 13 isolates belong to Fusarium oxysporum species. Six isolates were tested for temperature, pH and nitrogen dosage optimization studies. Seven different temperatures, viz., 21, 23, 25, 27, 29, 31, 33°C and pH values, having 3, 4, 5, 6, 7, 8, and 9 pH, as well as nitrogen dosage levels of 0 g, 3 g, 5 g, 7 g, 9 g, 11 g, and 13 g were tested against all six isolates, respectively. The results showed that all isolates exhibited the highest growth at a temperature of 25°C and the optimal temperature range for growth of Fusarium oxysporum was 23-27°C. All isolates showed the highest growth at pH5. Change in the nitrogen doses of the base ended in formation of thick, dense, fluffy mycelium of the casual agent. Six isolates were used for combination studies with seven different levels of temperatures, pH levels and nitrogen dosages. The density plots revealed the variations in the growth of the isolates with changes in temperature, pH and nitrogen levels, which can lead to mutations or genetic changes in the pathogens that could potentially introduce new threats to pea cultivation.

The present research is targeted at E. cardamomum-derived TiO2-photocatalyst synthesis, reporting for the first time. The structural properties observed from the XRD pattern reveal that EC:TiO2 has an anatase phase and crystallite size is assessed by Debye-Scherrer's method (3.56 nm), WH-method (3.30 nm), and Modified-Debye-Scherrer's method (3.27 nm). An optical study by the UV-Vis spectrum shows strong absorption at 313 nm, and the corresponding band gap value is 3.28 eV. The topographical and morphological properties revealed by SEM and HRTEM images, elucidate the formation of multi-shaped particles of nano-size. Further, the phytochemicals on the EC:TiO2 NPs' surface are confirmed by the FTIR spectrum. The photocatalytic activity is well studied under UV light towards Congo Red dye, along with an effect of the dose of catalyst. EC:TiO2 (20 mg) has exhibited high photocatalytic efficiency up to 97% for 150 min of exposure due to the morphological, structural, and optical properties. CR degradation reaction exhibits pseudo-first-order kinetics, displaying a rate constant value of 0.01320 min-1. Reusability investigations reveal that after four photocatalysis cycles, EC:TiO2 has an effective efficiency of >85%. Additionally, EC:TiO2 NPs have been assessed for antibacterial activity and show potential against two bacterial species (S. aureus and P. aeruginosa). Therefore, these research outcomes from the eco-friendly and low-cost synthesis, are promising for the use of EC:TiO2 as a talented photocatalyst towards the removal of crystal violet dye as well as an antibacterial agent against bacterial pathogens.

Uncontrolled amyloid aggregation is a frequent cause of neurodegenerative disorders such as prions and Alzheimer's disease (AD). As a result, many drug development approaches focus on evaluating novel molecules that can alter self-recognition pathways. Herein, we designed and synthesized the cyclometallated pyrene (Pd-1 and Pd-3) and anthracene (Pd-2) based palladium complexes ([Pd((L1)Cl] Pd-1, [Pd(L2)Cl](Pd-2), and [Pd(L3)Cl] (Pd-3)). This study explores the effect of these complexes on the aggregation, fibrillation, and amyloid formation of bovine serum albumin (BSA) and Aß1-42 peptide. Several spectroscopic methods were used to characterize all the Pd-complexes, and the molecular structure of Pd-3 was determined by X-ray crystallography. The secondary structures were studied using circular dichroism (CD) and transmission electron microscopy (TEM), while amyloid aggregation and inhibitory activities were investigated using the Thioflavin-T (ThT) fluorescence assay. Molecular docking of the Pd-complex (Pd-3) was done using fibril (PDB: 2BEG) and monomeric (PDB: 1IYT) peptides using Auto-dock Vina. As a result, the hydrogen bonding and hydrophobic interaction between the aromatic rings of the Pd-complexes and the amino acids of amyloid-ß peptides significantly reduced the production of ordered ß-sheets of amyloid fibrils and protein aggregation in the presence of Pd-2 and Pd-3 complexes.

The present study portrays a facile, cost-effective, and environmentally benign way for the preparation of TiO2 nanoparticles utilizing C. limon extract, which possesses phytochemicals as reducing and stabilizing agents. Structural characterization by XRD reveals that C. limon/TiO2 NPs exhibit anatase-type tetragonal crystallinity. An average crystallite-size is calculated using Debye Scherrer's method (3.79 nm), Williamson-Hall plot (3.60 nm), and Modified Debye Scherrer plot (3.68 nm), which are very closely intercorrelated. The absorption peak at 274 nm (UV-visible spectrum) corresponds to the bandgap (Eg) value of 3.8 eV. The existence of different phytochemicals containing organic groups like N-H, C=O, and O-H, has been elucidated from FTIR along with Ti-O bond stretching at wavenumber 780 cm-1. Micro-structural investigations of TiO2 NPs using FESEM and TEM display different geometrical configurations involving spherical, pentagonal, hexagonal, heptagonal and capsule-like structures. BET and BJH analysis show mesoporous characteristics of synthesized nanoparticles with specific surface-area (97.6 m2 g-1), pore-volume (0.018322 cm3 g-1), and mean pore-diameter (∼7.5 nm) values. In adsorption studies, the influence of reaction parameters, i.e., catalyst dosage and contact-time, for removal of Reactive Green dye is explored along with Langmuir and Freundlich models. The highest adsorption capability is ∼219 mg g-1 for green dye. TiO2 displays an excellent photocatalytic efficiency of ∼96% towards the degradation of reactive green dye within 180 min and excellent reusable performance. C. limon/TiO2 is found to have an outstanding performance with a quantum yield value of 4.68 × 10-5 molecules photon-1 for Reactive Green dye degradation. Additionally, synthesized nanoparticles have exhibited antimicrobial activity against gram-positive Staphylococcus aureus (S. aureus) and gram-negative Pseudomonas aeruginosa (P. aeruginosa) bacteria.

Coordination complexes offer great potential as cellular imaging probes, which allow to examine specific cell organelle structures in their physiological conditions to better understand the biological system. Understanding the heterogeneous nature of the cell membrane could unveil details of their functionality. Here, we have developed a new anthracene conjugated fluorescent palladium(II) cyclometallate [PdL1Cl] where L1H = [2-(2- (anthracen-9-ylmethylene)-1-phenylhydrazineyl)pyridine] (H stands for dissociable proton), which not only specifically stains the cell membrane, but could be utilized to visualise the membrane by the confocal and fluorescence lifetime imaging microscopy (FLIM). This probe is unable to enter inside the cell as it did not pass through the cell membrane via diffusion or various organic and metal transporters. However, the great lipophilicity of fluorescein improves the interaction of the probe with the peptidoglycan layer of the cell membrane. Probable dissociation of chloride ion and formation of positively charged palladium complex resulted in staining the negatively charged cell membrane. The 3D confocal imaging clearly expressed sole membrane staining by the probe. The probe efficiently stains both cancer cells (HeLa and MCF-7 cell lines) and normal cell (HEK 293 T), confirming the universality of the probe in membrane staining.

Wood hydrolysate has been regarded as sustainable and renewable substrate to produce microbial lipids, a potential feedstock for the biodiesel industry. Moreover, the major by-product of biofuel industries is crude glycerol but its implementation as a carbon source is still constrained due to the presence of impurities resulting in low biomass production and low lipid titer. Thus, this study investigates the effect of different carbon ratios of hydrolysate and crude glycerol on R. toruloides-1588. Hydrolysate to crude glycerol ratio of 60:40 resulted in maximum lipid accumulation of 49% (w/w), more than 90% of sugars and glycerol consumption. Further, scale-up to bench-scale fermenter resulted in 12% higher lipid accumulation (56.3% w/w, 0.15 g/L∙h) in 50% less time than flask fermentation. Hence, the ability of R. toruloides-1588 to flourish on different carbohydrates and accumulate high lipid content will be beneficial for the further development of biorefinery industries.

The presence of furfural in the hydrolysates obtained from lignocellulosic biomass sources represents an enormous challenge during their fermentation because furfural is a toxic compound for different microorganisms. Rhodosporidium toruloides-1588 can grow and accumulate lipids using wood hydrolysate as a substrate containing up to 1 g/L of furfural. In this study, the capacity of R. toruloides-1588 to grow and accumulate lipids using furfural without glucose in the media has been observed. R. toruloides-1588 degraded up to 3 g/L of furfural into furfuryl alcohol (1.8 g/L) and 2-furoic acid (0.9 g/L). Furthermore, R. toruloides-1588 accumulated 52% and 30% of its dry weight into lipids using YM media and YM media without glucose, respectively. Fatty acids such as palmitic, stearic and oleic were the most abundant. Finally, R. toruloides-1588 could potentially utilize furfural as a carbon source.

Plastics use is growing due to its applications in the economy, human health and aesthetics. The major plastic particles in the form of microplastics (MPs) released into the environment are made up of polyethylene (PE), polypropylene (PP), polyvinylchloride (PVC), and polyethylene terephthalate (PET). Tremendous usage and continuous accumulation of MPs in the environment pose a global threat to ecosystems and human health. The current knowledge of biotechnological, aerobic and aerobic biodegradation approaches emphasizes the microbial culture's potential towards MPs removal. This review selectively provides recent biotechnological advances such as biostimulation, bioaugmentation and enzymatic biodegradation that can be applied for MPs removal by biodegradation and bioaccumulation. This review summarizes the knowledge and the research exploration on the biodegradation of synthetic organic MPs with different biodegradability. However, further research is still needed to understand the underlying mechanism of MPs biodegradation in soil and water systems, leading to the development of an effective method for MPs removal.

RESUMO

Renewable biofuel has a great potential in replacing the conventional transportation fuels as well as aiding the current issue of climate change and global warming. In the present scenario, tremendous initiatives have been implemented to encourage large-scale biofuel production and reduce greenhouse gas emissions. However, the information on the current biofuel status specifically in Canada and where it lacks in biofuel production, tax rebate and policies in comparison with other countries is limited. In this sense, the current work focuses on the liquid biofuel status, recent advancements and evaluation of programs aimed at reducing greenhouse gas emissions in coming years. Additionally, the role of private and government programs in scaling up the projects is elaborated using several examples of successful as well as failed attempts to commercialize biofuels. Moreover, the Canadian government regulations and policies for greenhouse gas mitigation, and biofuel blending policies are also briefly described. In summary, future aspects and suggestions to further increase biofuel production are portrayed in this review.

BACKGROUND: Inguinal hernia containing ovary and fallopian tube can be found in paediatric population and is a rare finding in women of reproductive age group. Most of the cases are associated with congenital abnormalities of the female genital tract. CASE PRESENTATION: A 20 year old female presented with right reducible inguinal hernia, primary amenorrhea and normal secondary sexual characteristics. Clinical examination revealed scoliosis with convexity towards left side, prominence of left rib cage with Sprengel deformity and right sided heart sounds. Ultrasound of the inguinal swelling revealed right ovary within the hernial sac, Chest X-ray revealed right lung collapse and dextrocardia. Further Magnetic resonance imaging (MRI) of pelvis revealed inguinal hernia with right ovary as its content, normal left ovary and absent uterus. Computed tomography (CT) revealed complete collapse of right lung with compensatory left lung hyperinflation and absent right kidney. Karyotyping of the patient was normal, 46XX. A diagnosis of MURCS syndrome with right ovarian hernia was made. The hernia was surgically managed with repositioning of ovary and fallopian tube into the pelvis. DISCUSSION: Ovary in inguinal hernia is rare in women of reproductive age group. MRKH syndrome, a mullerian duct anomaly, is the congenital aplasia of uterus and upper two-thirds of vagina in a female with normal ovaries, fallopian tube, secondary sexual characteristics and 46XX karyotype. MURCS is a subtype of MRKH type 2 having mullerian duct agenesis with renal, cardiac, muscular & vertebral defects. General physical examination and primary investigations if yields abnormal findings; the patient must undergo an array of investigations to rule out MRKH/MURCS, or other congenital abnormality. Early diagnosis is essential to prevent its incarceration or torsion. The primary treatment of ovary in inguinal hernia is repositioning the ovary and fallopian tube back to pelvis to preserve fertility and repair of inguinal hernia. A multidisciplinary team is required to deal with various abnormalities present in a patient with MURCS.

Utilizing the undetoxified wood hydrolysate to accumulate maximum lipids in Rhodosporidium toruloides under optimum conditions has been regarded as a renewable and cost-effective strategy. The current investigation aims to identify the best carbon to nitrogen (C/N 20, 70, and 120) ratio for maximum lipid accumulation in R. toruloides-1588 using wood hydrolysate. Additionally, a fed-batch-like condition was employed, where C/N ratios were maintained during the fermentation that inherently decreases in batch fermentation. The C/N ratio 70 has been identified as the best condition with 3 times higher lipid accumulation (43% w/w) than the control. Additionally, >95% and 70% of glucose and xylose consumption were observed, respectively. Moreover, 50% increase in polyunsaturated fatty acids compared to the control media reinforced the potential of R. toruloides-1588 to thrive on undetoxified hydrolysate, high lipid productivity (3.8 mg/g of dry weight per hour) and produce high value monosaturated and polyunsaturated fatty acids.

BACKGROUND: Filarial infections are common in most tropical and subtropical regions of the world. Lymphatic filariasis is caused by either Wuchereria bancrofti, Brugia malayi, or Brugia timori. Extralymphatic filariasis presenting as a primary retroperitoneal mass is very rare despite filariasis being endemic in many regions of India. On review of literature, only a few isolated case reports have been described. CASE PRESENTATION: We report a case of a huge retroperitoneal cystic mass in a 46-year-old patient who presented with a long-standing, painless progressive abdominal swelling. On examination, there was a large, non-tender, firm swelling of size around 20 × 15 cm occupying the left upper and lower quadrant. The computed tomography of the abdomen was suggestive of thin-walled hypodense cyst of size 25.7 × 15 × 14.3 cm. Laboratory investigations and cyst aspirate were inconclusive for a definite diagnosis. On exploration, a 3-kg cystic mass was removed. The diagnosis of filarial origin was confirmed by the demonstration of microfilaria in the cyst wall and immunochromatographic test (ICT) which was positive. CONCLUSION: Retroperitoneal lymphatic cyst of filarial origin is very unusual and requires a high index of suspicion if the patient is an inhabitant of an endemic area. The clinical dilemma cannot be resolved with imaging modalities alone, unless a disease-specific manifestation is there. The retroperitoneal cysts often pose a challenge in their diagnosis and management. Small cysts might respond to medical management, whereas large symptomatic cysts will require excision for the final diagnosis and treatment.

BACKGROUND: The growing use of neuromodulation techniques to treat neurological disorders has motivated efforts to improve on the safety and reliability of implantable nerve stimulators. NEW METHOD: The present study describes the ReStore system, a miniature, implantable wireless nerve stimulator system that has no battery or leads and is constructed using commercial components and processes. The implant can be programmed wirelessly to deliver charge-balanced, biphasic current pulses of varying amplitudes, pulse widths, frequencies, and train durations. Here, we describe bench and in vivo testing to evaluate the operational performance and efficacy of nerve recruitment. Additionally, we also provide results from a large-animal chronic active stimulation study assessing the long-term biocompatibility of the device. RESULTS: The results show that the system can reliably deliver accurate stimulation pulses through a range of different loads. Tests of nerve recruitment demonstrate that the implant can effectively activate peripheral nerves, even after accelerated aging and post-chronic implantation. Biocompatibility and hermeticity tests provide an initial indication that the implant will be safe for use in humans. COMPARISON WITH EXISTING METHOD(S): Most commercially available nerve stimulators include a battery and wire leads which often require subsequent surgeries to address failures in these components. Though miniaturized battery-less stimulators have been prototyped in academic labs, they are often constructed using custom components and processes that hinder clinical translation. CONCLUSIONS: The results from testing the performance and safety of the ReStore system establish its potential to advance the field of peripheral neuromodulation.

BACKGROUND: Muscle-derived interleukin-6 (IL-6) not only enhances glucose and fat metabolism but also has an anti-inflammatory effect that can prevent the development of cardiovascular disease (CVD) and metabolic syndrome. Dynamic resistance exercise comprises a muscle shortening and a lengthening phase known as concentric and eccentric contraction respectively. We compared an acute bout of concentric exercise (CE) with eccentric exercise (EE) for the serum IL-6 concentration, myocardial oxygen consumption (MVO2) and rating of perceived exertion (RPE). METHODS: The present non-randomized crossover study involved 24 physically inactive but apparently healthy males, aged 18-25 years with body mass index of 18.5-23 kg/m2. Each participant performed an acute bout of moderate intensity CE followed by an EE bout after two weeks of rest. Exercise bout lasted for about 45 minutes that involved brisk walking on a treadmill at ±21% gradient for approximately 30 minutes, followed by the dumbbell exercises for biceps, triceps, quadriceps and hamstring muscles. MVO2 and IL-6 were estimated by the rate pressure product (RPP) and ELISA (enzyme-linked immunosorbent assay) kit respectively. Participants told their RPE after the workout. Wilcoxon or paired-t test were applied appropriately to compare the means. P<0.05 was considered significant. RESULTS: RPP, RPE, and serum IL-6 levels were significantly lower after EE (164.4±16.3, 12, and 1.1±0.5, respectively) than CE (191.2±22.9, 13, and 1.2±0.6, respectively). A significant, strong and positive correlation of RPE was obtained with post-exercise heart-rate and RPP. CONCLUSIONS: Regular EE might be beneficial for individuals having a low tolerance to exertion or at risk of developing CVD.
