While root canal anatomy in Middle Eastern populations is well-studied, research on isthmi in mandibular permanent teeth from this region is limited. This retrospective study used cone-beam computed tomography (CBCT) to examine isthmi prevalence and location, as well as root canal morphologies (per Vertucci's classification) in mandibular permanent teeth from a subpopulation in Ardabil, Iran. The study is aimed at enhancing our understanding of dental anatomical variations in Middle Eastern populations. A total of 3566 teeth from 384 CBCT scans were evaluated in this retrospective study. Mandibular teeth were evaluated on sagittal, coronal, and axial sections regarding the presence of isthmus and root morphology (Vertucci's classification). CBCT scans of 197 males (51.3%) and 187 females (48.7%) were evaluated, with a mean age of 41.1 ± 11.4 years. Isthmi were most prevalent in molar mesial roots, typically located in the middle third for anterior teeth and first premolars and cervical third for posterior teeth. While gender did not play a significant role, the presence of an isthmus in the mesial root of the left second molars was associated with a younger mean patient age (p < 0.05). Root morphology varied across tooth types. Central and lateral incisors predominantly showed Vertucci's Types I and III. Canines and premolars were mostly Type I, with some variation. Molar mesial roots frequently exhibited Types IV and II, while distal roots were predominantly Type I. Statistically significant differences were found between morphology and gender in the first left premolar (Type I more common in women; Type V in men; p < 0.001) and in the right canine (Type I more prevalent in men; Types III and V more prevalent in women; p < 0.001). The results revealed wide variations in root canal morphology and a relatively high prevalence of isthmi in the study population. Our findings suggest a potential difference in tooth anatomy based on sex and a relationship between age and the presence of isthmi.

The new Gentlefile (GF) system, made of stainless steel and developed by MedicNRG in Kibbutz Afikim, Israel, claims to have advantages over traditional nickel-titanium files. However, research has shown that nickel-titanium files are mechanically superior due to their increased flexibility, cutting efficiency, and ability to maintain canal anatomy with less risk of procedural errors. This study compared the amount of debris extrusion and the time required for root canal instrumentation using GF versus the nickel-titanium ProTaper Universal (PTU) system and a manual step-back (MSB) stainless steel technique. This in vitro experimental study utilized 66 extracted human single-canal mandibular premolars with mature apices and root curvature of less than 10 degrees. The teeth were randomly divided into three groups (n = 22) and standardized for working length before being placed in preweighed vials. Group 1 was instrumented with PTU, Group 2 with GF, and Group 3 with the MSB technique. Extruded debris was collected in the vials, dried in an incubator, and weighed using the same scale. The change in weight indicates the debris amount. Instrumentation time was recorded using a stopwatch. The normal distribution of data was assessed using the Kolmogorov-Smirnov test. The groups were then compared regarding the amount of extruded debris and instrumentation time using the Kruskal-Wallis test and one-way ANOVA, followed by the Games-Howell test, respectively (alpha = 0.05). No significant difference in apical debris extrusion was found among the three groups (P > 0.05). However, a significant difference in instrumentation time was detected between the groups (P < 0.05). MSB instrumentation took significantly longer than both the PTU (P = 0.001) and GF (P = 0.001) systems. The GF system did not demonstrate reduced apical debris extrusion or faster instrumentation time compared to PTU. MSB had the longest instrumentation time compared to the other techniques.

Background: This study compared apical extrusion of debris and instrumentation time following root canal instrumentation with Reciproc, Reciproc Blue, and Gentlefile (GF) rotary instruments versus the manual step-back technique. Methods: This in vitro study was conducted on 80 extracted mandibular premolars with mature apices and a root curvature of<10°. The teeth were randomly assigned to 4 groups (n=20), standardized regarding working length, and placed in pre-weighed vials. The root canals were instrumented with Reciproc, Reciproc Blue, and GF systems and the manual step-back technique in the four groups. The vials containing the collected debris were then dried and weighed. The instrumentation time was also recorded for each group. Data were analyzed with one-way ANOVA and post hoc Games-Howell test (α=0.05). Results: Minimum apical debris extrusion was noted in Reciproc, followed by Reciproc Blue, GF, and manual technique (P<0.05). Pairwise comparisons showed significantly lower apical extrusion of debris in the Reciproc compared with GF (P=0.015) and manual instrumentation (P=0.011) groups. The Reciproc system also had the shortest instrumentation time, followed by Reciproc Blue, GF, and manual instrumentation (P<0.05). Pairwise comparisons showed significant differences between all the systems (P<0.05) except between Reciproc and Reciproc Blue (P>0.05) in this respect. Conclusion: Although all systems caused apical extrusion of debris, manual instrumentation caused maximum extrusion of debris. In contrast, the Reciproc system was superior to others regarding minimal apical extrusion of debris and the shortest instrumentation time.

Objectives: The complex anatomy of C-shaped canals poses challenges for clinicians compared to teeth with normal root canal anatomy. This study is aimed at evaluating the frequency and morphology of C-shaped canals in the mandibular first and second molars among an Iranian population using cone-beam computed tomography (CBCT). Materials and Methods: This cross-sectional study evaluated 369 CBCT scans from the archives of a radiology clinic in Ardabil, Iran. The sample included 248 mandibular first molars and 478 mandibular second molars. The presence of C-shaped canals and their classification, according to Fan et al., were evaluated at four levels: orifice, coronal, middle, and apical. Prevalence based on gender and tooth type was also assessed. Results: A total of 199 (53.9%) males and 170 (46.1%) females were evaluated. C-shaped canals were found in 11 (8 males/3 females) out of 248 (4.4%) first molars and in 20 (11 males/9 females) out of 478 (3.7%) second molars. The C2 configuration was most prevalent in the orifice, coronal, and middle levels of both molar types, while C3 and C4 morphologies were most commonly found apically in the second and first molars, respectively. A significant difference in configuration was found only at the coronal level between molar types. A significant association between gender and configuration was observed only at the orifice level of the second molars. No other significant differences were found (P > 0.05). Conclusion: C-shaped canal configurations should be expected in 4.4% of mandibular first molars and 3.7% of the second molars in this Iranian population, with a predominance of the C2 configuration in the orifice, coronal, and middle levels.

Objectives: This study assessed the quality of endodontic treatment and the prevalence of procedural errors in permanent mandibular molars using cone-beam computed tomography (CBCT). Materials and Methods: This cross-sectional study was conducted on 328 CBCT scans (182 females and 146 males) of endodontically treated mandibular molars retrieved from the archives of two radiology centers in Ardabil city, Iran, in 2019. Mandibular molars were evaluated on sagittal, coronal, and axial sections regarding obturation length, obturation density (voids), missed canals, broken instruments, apical perforation, strip perforation, ledge formation, transportation, root fracture, root resorption, and periapical lesions by a senior dental student under the supervision of an oral and maxillofacial radiologist and an endodontist. Differences between the frequency of procedural errors and tooth type and gender were analyzed by the chi-square test. Results: The frequency of underfilling, missed canals, overfilling, voids, apical perforation, transportation, ledge formation, broken instruments, root fracture, strip perforation, root resorption, and periapical lesions was 34.8%, 17.4%, 16.8%, 14.3%, 7.3%, 6.1%, 4.3%, 3%, 1.2%, 0.6%, 5.5%, and 46%, respectively. The frequency of root fracture was significantly higher in females than in males (P < 0.05). The prevalence of underfilling was the highest in right second molars (47.2%), followed by right first molars, left second molars, and left first molars (P < 0.005). The frequency of transportation was maximum in right first molars (10%), followed by right second molars, left first molars, and left second molars (P < 0.04). Conclusion: Underfilling, missed canals, and overfilling were the most prevalent procedural errors in mandibular molars in our study population.

Introduction: This study examined the effect of 1.5% NaOCl, 17% EDTA, and curcumin on the proliferation, attachment, and differentiation of dental pulp stem cells (DPSCs) placed on the dentin specimens. Methods: MTT assay was performed to evaluate the proliferation of DPSCs on the dentin specimens treated with different concentrations of NaOCl, 17% EDTA, and curcumin (0.97-250 µM). Cell-adhering ability of DPSCs was tested via the LDH assay to calculate the attached DPSCs. In addition, the western blotting assay was performed to investigate the expression levels of fibronectin as a cell-adhesion marker and analyze the expressions level of differentiation markers, including DMP-1, OCN, ALP, and DSPP, to detect the odontogenic potential of hDPCs. Results: NaOCl had lower toxicity on DPSCs at lower concentrations (P < 0.001). The cytotoxicity of irrigants increased with increased dosage. The difference between the cell-adhesion ability of NaOCl and curcumin was not significant (∼4.4 MU/mL), whereas EDTA (∼3.8 MU/mL) exhibited the lowest release of LDH and less damage to hDPSCs. Regarding fibronectin expression, the pattern differed between irrigants in inducing cell adhesion. NaOCl increased fibronectin expression more than EDTA and curcumin. All the treated groups upregulated the expression of DSPP, DMP-1, OCN, and ALP compared to the control group, in which NaOCl showed a higher effect on the overexpression of differentiation markers. Conclusion: The results showed that all the tested irrigants could be used in regenerative endodontic treatment. However, as an herbal-based and biocompatible irrigant, curcumin exhibited fewer adverse effects than NaOCl and EDTA.

Background: Mineral trioxide aggregate (MTA) is a commonly used endodontic biomaterial. The physicochemical properties of MTA have a crucial role in designating clinical outcome, and different factors can affect these properties. Various methods have been used for mixing MTA, including manual, mechanical, and ultrasonic. The aim of this systematic review was to evaluate the effect of different mixing methods on the physicochemical properties of MTA. Materials and Methods: Electronic databases including PubMed, Embase, Web of Science, and Scopus were searched up to May 2022. In order to cover gray literature, the ProQuest and Google Scholar databases were also searched to detect theses and conference proceedings. For quality assessment of the included studies, we used a modified version of the Cochrane risk of bias tool for randomized controlled trials (RCTs). Experimental studies which had assessed at least one property of MTA and compared at least two different mixing methods of MTA were included in this study. All animal studies, reviews, case reports, and case series were excluded. Results: Fourteen studies were included. The results showed that the ultrasonic mixing method significantly improved some MTA characteristics, including microhardness, flowability, solubility, setting time, and porosity. However, the mechanical mixing method improved other properties including flowability, solubility, push-out bond strength, and hydration. The manual mixing method showed inferior results compared to other mixing methods in terms of microhardness, flowability, solubility, setting time, push-out bond strength, porosity, and hydration. Different mixing methods had a similar effect on compressive strength, sealing ability, pH and calcium ion release, volume change, film thickness, and flexural strength of MTA. Conclusion: Mechanical and ultrasonic mixing methods are superior to the manual mixing method in terms of improving physicochemical properties of MTA. No report of selection bias and varieties in methodologies were limitations of evidence.

Objectives: This study compared the obturation quality and push-out bond strength of single cone obturation (SCO) and cold lateral compaction (CLC) with AH-Plus and Sure Seal Root (SSR). Materials and Methods: This in vitro experimental study was conducted on 88 single-rootedsingle-canal teeth with straight roots that were randomly divided into four groups (n = 22). All teeth were decoronated and underwent cleaning and shaping. Obturation was performed with AH-Plus and SCO technique in group 1 (SAH), AH-Plus and CLC technique in group 2 (LAH), SSR and SCO technique in group 3 (SS), and SSR and CLC technique in group 4 (LS). The roots were then sectioned into 3-mm thick slices and underwent digital photography at x25 magnification to assess the quality of obturation in the coronal, middle, and apical thirds by Image J software. The PBS was measured by a universal testing machine. The mode of failure was also determined under a stereomicroscope. Results: The PBS was significantly higher in the LSS group than LAH and SAH groups, and also in the SSS group than the SAH group in all sections. The PBS in the LSS group was significantly higher than SSS in the coronal and middle thirds. Voids were significantly lower in LAH than in the SAH group in all sections. In LSS, voids in the coronal third were significantly lower than in LAH. In the middle third, voids in SSS were significantly lower than in SAH. The groups had no significant difference in the mode of failure (P > 0.05). The mean percentage of gutta-percha in the use of AH-Plus sealer was significantly higher than SSR (P < 0.05). The mean percentage of gutta-percha in the coronal third was lower than that in the middle and apical thirds (P < 0.05). Conclusion: SSR showed higher PBS and less voids than AH-Plus. High PBS of the CLC/SSR group showed that CLC should still be preferred to SCO, and in the case of using SCO, SSR should be preferred to AH-Plus.

OBJECTIVES: Root canal preparation can lead to cracks on the roots by creating stresses on the root canal walls, which decreases the fracture resistance of the tooth. The present study compared the fracture resistance of the teeth prepared by the ProTaper Universal (PTU), ProTaper Next (PTN), and ProTaper Gold (PTG) rotary file systems. MATERIALS AND METHODS: Fifty-six single-canal premolar teeth were sectioned 14 mm from the root apex. The roots were standardized based on the buccolingual and mesiodistal diameter and randomly assigned to three experimental (n = 14) and one control group (n = 14). The teeth in three experimental groups were instrumented with PTU, PTN, and PTG rotary files. The roots in the control group were not instrumented. A vertical force was applied to each root in a universal testing machine until the root fractured. The data were statistically analyzed by one-way analysis of variance. RESULTS: There was no significant difference in the fracture resistance of the teeth between the control, PTU, PTN, and PTG groups (p = .115). CONCLUSIONS: Root canal preparation with ProTaper files manufactured with conventional NiTi (PTU) and heat-treated alloys (PTN and PTG) did not affect the fracture resistance of teeth.

Background. Calcium-enriched mixture (CEM) cement has been introduced and marketed as a biomaterial for use in furcal perforation repair and apexogenesis procedures, in which the compressive strength that indicates the material's resistance against crushing is of utmost importance. This study evaluated the effect of various liquid-to-powder ratios on CEM cement's compressive strength. Methods. One gram of the cement was mixed with 0.5, 0.34, and 0.25 mL of demineralized water and transferred to stainless steel molds (6 and 4 mm in height and diameter, respectively). Five cells in the mold were considered for each group. The compressive strength test was conducted using the universal testing machine after incubating for seven days under 95% humidity at 37°C. One-way ANOVA was applied for data analysis at P ≤ 0.05 significance level. Results. The mean compressive strength in the liquid-to-powder ratios of 0.5, 0.34, and 0.25 were 3.4456, 3.2960, and 3.3485, respectively, with no significant differences between them. Conclusion. Under this study's limitations, changing the liquid-to-powder ratio did not affect CEM cement's compressive strength.

Background . The aim of this in vitro study was to investigate the effect of zinc oxide (ZnO) and zirconium oxide (ZrO2) microparticles (MPs) and nanoparticles (NPs) in combination with white Portland cement (WPC) on odontogenic capacity of human dental pulp stem cells over a period of 21 days. Methods . Synthesized ZnO and ZrO2 particles were characterized using scanning electron microscopy and transmission electron microscopy. The viability of human dental pulp stem cells was measured by a 3-(4,5-dimethylthiazolyl-2-yl)-2,5- diphenyltetrazolium bromide assay at 7-, 14- and 21-day intervals after seeding on WPC disks enriched with ZnO and ZrO2 MPs and NPs. Odontogenic potential of ZnO and ZrO2 particles in combination with WPC was investigated by alkaline phosphatase (ALP) activity and ionized calcium level of supernatant culture media at different time intervals. Data were analyzed using one-way ANOVA and post hoc Tukey tests. Results . All the materials exhibited cell viability over a 21-day period, except for WPC with ZnO NPs on day 7, although it was not statistically significant (P>0.05). The ALP activity and ionized calcium level increased in all the groups compared to the control group (P<0.05). ZnO NPs had superior effect on odontogenic activity and calcium ion release compared to ZnO MPs (P=0.046). There was no significant difference between ZrO2 MPs and NPs in odontogenic activity (P>0.05). Conclusion . WPC enriched with ZnO and ZrO2 increased ALP activity and calcium ion release of human dental pulp stem cells over a period of 21 days in vitro.

Introduction: This study aimed to compare the effects of different agitation techniques of sodium hypochlorite (NaOCL) as the final irrigation with XP-Finisher file, passive ultrasonic irrigation (PUI), Er:YAG laser and irrigation with conventional syringes, on penetration of Sure Seal Root bioceramic sealer into dentinal tubules. Methods and Materials: Forty freshly extracted single-rooted human teeth were included in the present in vitro study. All the teeth were prepared up to #40 (4%) with Bio Race rotary file system, using crown-down technique. After chemomechanical preparation, the teeth were randomly assigned to 4 groups based on the agitation protocol: conventional needle irrigation (CNI) with a 30-G needle in a syringe, PUI, Er:YAG laser and XP-Finisher file, Subsequent to the agitation procedure, the root canals were obturated with tapered bioceramic-covered gutta-percha point and, bioceramic sealer, using the single-cone technique. The maximum penetration of the sealer into the dentinal tubules was determined in the coronal, middle and apical thirds using the scanning electron microscope (SEM) technique. After confirmation of the normal distribution of data with Shapiro-Wilk test, Data were analyzed using the one-way ANOVA and Tukey tests. Results: The mean penetration depth of the bioceramic sealer into the dentinal tubules in the PUI group was greater than that of other groups; however, the difference between the two PUI and Er:YAG laser groups in the apical third was not significant (P=0.078). Er:YAG laser, PUI and XP-Finisher file agitation techniques resulted in significantly greater penetration of the bioceramic sealer into the dentinal tubules compared to the CNI (P<0.001). The XP-Finisher file technique in the apical (P=0.752) and middle thirds (P=0.339) and the Er:YAG laser technique in the apical thirds (P=0.086) were not significantly different from the conventional irrigation technique. Conclusion: The PUI technique resulted in significantly deeper penetration of the Bioceramic sealer into the dentinal tubules compared to the three other techniques.

Introduction: The aim of this study was to compare the flexural strength of mineral trioxide aggregate (MTA), calcium-enriched mixture (CEM), and BioAggregate (BA). Methods and Materials: In this study, the flexural strength of materials was measured using a 3-point bend test. After being prepared, MTA, CEM, and BA were inserted into the intra-putty molds using amalgam plugger. The specimens were covered with a sponge wetted with synthetic tissue fluid (STF) and incubated for 96 h. They were then subjected to a 3-point bend test using Universal Testing Machine. The Kruskal-Wallis and Mann-Whitney U tests were used to compare flexural strength in groups. In this study, P<0.05 was considered as the significant level. Results: There were significant differences between the three groups in terms of the flexural strength (P<0.001). The mean flexural strength in the BA, CEM, and MTA groups were 27.32±2, 9.09±1.16, and 10.25±1.6, respectively. Pairwise comparison showed significant differences between the three groups. Conclusion: This in vitro study showed that BA has the highest and CEM has the lowest flexural strength.

Background. of this study was to investigate the effect of different mixing methods (ultrasonic, amalgamator, and conventional) on the bacterial microleakage of white Portland cement (WPC) and white MTA (Tooth-colored Formula, Dentsply, Tulsa Dental, Tulsa, OK). Methods. A hundred human single-rooted permanent teeth were decoronated to obtain 14 mm of root length in all the samples. The root canals were cleaned, shaped and obturated. Three millimeters of each root apex were cut off and randomly divided into 6 groups of 15 each (3 groups for WMTA and 3 groups for WPC, each with 3 different mixing methods) and 2 positive and negative control groups (each containing 5 samples). Brain-heart infusion agar (BHI) suspension containing the bacterial species Enterococcus faecalis (ATCC 29212) was used for leakage assessment. Statistical analysis was carried out using descriptive statistics and Kaplan Mayer survival analysis with censored data and log rank test using SPSS 18. Statistical significance was set at P<0.05. Results. The survival means in PC for conventional method, amalgamator, and ultrasonic were 80.2±13.64, 78.5±13.46 and 84.667±11.42 days, with 49.13±12.96, 66±13.32 and 69.07±11.5 days for MTA, respectively. The log rank test showed no significant differences between the three methods in each material (P>0.05). Conclusion. Bacterial microleakage in the studied samples was not significantly different in terms of the type of the mixing method.

BACKGROUND: The aim of this study was to evaluate the discoloration induced by CEM cement, Portland cement (PC) and MTA mixed with propylene glycol (MTA-PG) in comparison to White MTA. MATERIAL AND METHODS: Ninety extracted premolar and canine teeth were resected 2 mm below the CEJ. The coronal part of crown was prepared with peeso reamer and Gates-Glidden drills, and the specimens were randomly divided into 4 experimental (n=20) and one control (n=10) groups. The tooth crowns in experimental groups 1 to 4 were filled with White MTA, PC, CEM cement and MTA-PG, respectively; and in group 5, the teeth were kept empty. After incubation, digital photographs of teeth were acquired at 4 time points (before, immediately after placing the materials, 3 and 6 months afterwards). Images were transferred to Adobe Photoshop CS4 and CIE L*a*b color space was used for tooth shade assessment. One-Way ANOVA and One-Sample t-test were used to compare discoloration of teeth between groups. RESULTS: Significant statistical discoloration was only observed in the cervical one third of all groups at each time points (except between 3 and 6 months). Tooth discoloration was greatest in PC and lower in MTA and MTA-PG at the end of 6 months. The tooth discoloration between immediately and 3 months after placing the materials had significant difference only between MTA and PC; and also the tooth discoloration between immediately and 6 months after placing the materials was observed only between PC and MTA, and PC and MTA-PG. CONCLUSIONS: All of the experimental biomaterials caused tooth discoloration after 6 months, of those, PC had the most and MTA and MTA-PG had the least discoloration effect. INTRODUCTION: The aim of this study was to evaluate the effect of adding propylene glycol (PG) to mineral trioxide aggregate (MTA) liquid with volume ratio of 20% on the compressive strength (CS) of MTA in two time periods (4 and 21 days) after mixing. METHODS AND MATERIALS: Four groups of steel cylinders (n=15) with an internal diameter of 3 and a height of 6 mm were prepared and MTA (groups 1 and 2) and MTA+PG (80% MTA liquid+20% PG) (groups 3 and 4) were placed in to the cylinders. In groups 1 and 3 the CS was evaluated after 4 days and in groups 2 and 4 after 21 days. Data were calculated using the two-ways ANOVA. The level of significance was set at 0.05. RESULTS: The highest (52.22±18.92 MPa) and lowest (4.5±0.67 MPa) of CS was obtained in 21-day MTA samples and 4-day MTA+PG specimen, respectively. The effect of time and PG were significant on the CS (P<0.05). Mixing MTA with PG significantly reduced the CS; but passing the time from 4 to 21 days significantly increased the CS. CONCLUSION: Considering the limitations of this study, PG had a negative effect on CS of MTA.

BACKGROUND: Calcium-Enriched Mixture Cement is used in the field of endodontics. It is similar to Mineral Trioxide Aggregate in its main ingredients.The present study investigated the effect of different mixing methods on the bacterial microleakage of CEM cement. METHODS: A total of 55 human single-rooted human permanent teeth were decoronated so that 14- mm-long samples were obtained and obturated with AH26 sealer and gutta-percha using lateral condensation technique. Three millimeters of the root end were cut off and randomly divided into 3 groups of 15 each (3 mixing methods of amalgamator, ultrasonic and conventional) and 2 negative and positive control groups (each containing 5 samples). BHI (Brain-Heart Infusion Agar) suspension containing Enterococcus faecalis was used for bacterial leakage assessment. Statistical analysis was carried out using descriptive statistics, Kaplan‒Meier survival analysis with censored data and log rank test. Statistical significance was set at P<0.05. RESULTS: The survival means for conventional, amalgamator and ultrasonic methods were 62.13±12.44, 68.87±12.79 and 77.53±12.52 days, respectively. The log rank test showed no significant differences between the groups. CONCLUSIONS: Based on the results of the present study it can be concluded that different mixing methods had no significant effect on the bacterial microleakage of CEM cement.

INTRODUCTION: The aim of this in vitro study was to evaluate the effect of calcium hydroxide (CH) on push-out bond strength of white mineral trioxide aggregate (WMTA) and calcium-enriched mixture (CEM) cement in simulated furcation perforations. METHODS AND MATERIALS: Furcation perforations, measuring 1.3 mm in diameter and 2 mm in height, were created in 80 human mandibular first molars. The teeth were then divided into 4 groups (n=20). In groups 1 and 3 CH was placed in the perforation for one week, before placement of WMTA and CEM. In groups 2 and 4 perforations were repaired without placement of CH. In groups 1 and 2 the perforation sites were repaired with WMTA and CEM cement was used in groups 3 and 4. After 7 days, the push-out test was carried out using a universal testing machine. Data were analyzed with two-way ANOVA. The level of significance was set at 0.05. RESULTS: The maximum and minimum bond strength values were recorded in the WMTA/CH (13.08±1.8 MPa) and CEM cement groups (8.03±0.98 MPa), respectively. There were significant differences in resistance to dislodgement between the WMTA/CH and other groups (P<0.05). CONCLUSION: Placement of CH before placement of WMTA in furcation perforation improves the push-out bond strength of this material.

Background. This study evaluated the antimicrobial activity of Tetraacetylethylenediamine-sodium perborate (TAED-SP) in comparison to 2.5% and 5% sodium hypochlorite (NaOCl) against Enterococcus faecalis. Methods. A standard suspension of E. faecalis was inoculated on 60 plates containing Mueller-Hinton agar culture medium. Four sterile disks of Beckman filtration paper were placed on each plate. TAED-SP, 5% and 2.5% NaOCl were placed on three disks. Sterile physiologic saline was placed on the fourth disk as negative control. After 24-hour incubation, the diameter of the inhibition zone around the disks was measured using a transparent ruler. One-way Analysis of Variance (ANOVA) was used to compare the mean zone of microbial growth in the groups. P-values less than 0.05 were considered statistically significant. Results. There was a significant difference in the diameter of the inhibition zones between groups (P < 0.05). The Tukey post hoc test showed a higher diameter of the inhibitory zone with TAED-SP than that of 2.5% NaOCl. However, there were no significant differences between the inhibitory zones of TAED-SP and 5% NaOCl. Conclusion. TAED-SP and 5% NaOCl have similar antibacterial activity against E. faecalis; however, TAED-SP has a greater antibacterial effect compared to 2.5% NaOCl.

BACKGROUND: According to the guideline of the United States center for disease control (CDC), the extracted teeth should be sterilized by autoclaving or storage in 10% formalin before using for educational or research purposes. The objective of this study was to evaluate the effect of this protocol on microhardness of dentin and enamel. MATERIAL AND METHODS: Thirty extracted single-root teeth were used in this study. The crowns were resected, and the roots were longitudinally sectioned into two halves. The Vickers microhardness (VHN) of specimens was measured on polished canal dentin and buccal enamel surfaces. The crowns were randomly divided into three groups (n=10). Group 1 and 2 were sterilized using autoclave and formalin, respectively while group 3 (control) was stored in synthetic tissue fluid. The root halves were also randomly divided into 3 groups (n=20) which were treated as mentioned above for crown samples. Following sterilization, VHN of samples was measured again. ANOVA and paired samples t-tests were used to analyze the data. RESULTS: Autoclaving caused a significant reduction in microhardness of dentin (P <0.001, 12.04% decreases in VHN). However, there were no significant differences for before and after sterilization within other groups. CONCLUSIONS: Based on the results of this study, the CDC protocol is recommended in studies related to enamel microhardness. However, Autoclaving is not an appropriate sterilization method in studies related to dentin microhardness. In these studies, two-week immersion in 10% formalin is recommended.
