Abstract Understanding morphological and physiological changes under different light conditions in native fruit species in juveniles stage is important, as it indicate the appropriate environment to achieve vigorous saplings. We aimed to verify growth and morphophysiological changes under shade gradient in feijoa (Acca sellowiana (O. Berg) Burret) to achieve good quality saplings adequate to improve cultivation in orchards. The saplings were grown for twenty-one-month under four shading treatments (0%, 30%, 50%, and 80%). Growth, photosynthetic pigments, gas exchanges, chlorophyll fluorescence, and leaf anatomy parameters were evaluated. Saplings under full sun and 30% shade had higher height and diameter growth and dry mass accumulation due to higher photosynthesis rate. As main acclimatization mechanisms in feijoa saplings under 80% shade were developed larger leaf area, reduced leaf blade thickness, and enhanced quantum yield of photosystem II. Even so, the net CO2 assimilation and the electron transport rate was lower and, consequently, there was a restriction on the growth and dry mass in saplings under deep shade. Therefore, to obtain higher quality feijoa saplings, we recommend that it be carried out in full sun or up to 30% shade, to maximize the sapling vigor in nurseries and, later, this light environment can also be used in orchards for favor growth and fruit production.


Resumo A verificação de mudanças morfológicas e fisiológicas sob diferentes condições luminosas em espécies frutíferas nativas em estágio juvenil é importante, uma vez que indicam o ambiente adequado para a formação de mudas com alto vigor. Objetivou-se verificar o crescimento e as alterações morfofisiológicas sob gradiente de sombreamento em mudas de feijoa (Acca sellowiana (O. Berg) Burret) para obter mudas de boa qualidade, adequadas para fomentar os plantios da espécie em pomares. As mudas foram cultivadas por vinte e um meses sob quatro tratamentos de sombreamento (0%, 30%, 50% e 80%). Foram avaliados parâmetros de crescimento, pigmentos fotossintéticos, trocas gasosas, fluorescência da clorofila e anatomia foliar. Mudas a pleno sol e 30% de sombra apresentaram maior crescimento em altura, diâmetro e acúmulo de massa seca, devido à maior taxa de fotossíntese. Como principais mecanismos de aclimatação sob 80% de sombra, as mudas desenvolveram maior área foliar, redução da espessura do limbo foliar e aumento do rendimento quântico do fotossistema II. Mesmo assim, a assimilação líquida de CO2 e a taxa de transporte de elétrons foram menores e, consequentemente, houve restrição ao crescimento e acúmulo de massa seca das mudas no maior nível de sombreamento. Portanto, para a obtenção de mudas de feijoa de maior qualidade, recomendamos que seja realizada a pleno sol ou até 30% de sombra, para maximizar o vigor das mudas em viveiros e, posteriormente, este ambiente de luz também pode ser utilizado em pomares para favorecer o crescimento e a produção de frutos.

Plants exposed to different light intensities generate physiological, morphological, and anatomical changes conducting to plasticity. Thus, this characteristic establishes the ability of plants to present phenotypic adjustments by the same genotype under different environmental conditions. The objective of this study was to verify the morphophysiological alterations in Campomanesia xanthocarpa (Mart.) O. Berg (guabiroba) seedlings cultivated in different shading levels. The seedlings were grown for 21 months under full sun or 30%, 50%, and 80% under shading. Growth, photosynthetic pigments, gas exchange rate, chlorophyll fluorescence, and leaf anatomy were evaluated. In all the treatments subjected to shading, plasticity mechanisms involved structural and physiological changes such as an increase in leaf area and chlorophyll content (total and Chl a), reduction in leaf thickness, and increased gas exchange and quantum yield of photosystem II. The guabiroba seedlings can be cultivated in full sun or different shading environments; even under high shading intensity (80%), the plants showed vigor similar to those produced in a sunny environment. These results confirmed our hypothesis about guabiroba acclimation capacity to shading, noteworthy information for nurseries, orchards, agroforestry systems, or forest restoration in a wide range of light environments.

×Butyagrus nabonnandii (Prosch.) Vorster is known as mule palm due to sterility, but recently, its pollen has been successfully used in backcrosses. It was first described as an artificial cross between Syagrus romanzoffiana (Cham.) Glassman and Butia odorata (Barb.Rodr.) Noblick in 1890. It has been considered rare in nature, and its morphology has been little studied. Thus, we aimed to verify if ×B. nabonnandii is sterile by studying its morphology and seed germination from different natural populations. The hybrid was sampled in four municipalities and is new to three of these. In one of the visited cities, 20 specimens were counted. The vegetative morphology showed less variation than the reproductive. However, part of the vegetative characters differed from previous descriptions relying solely on cultivated specimens. Contrary to previous reports, our data indicate that ×Butyagrus nabonnandii is neither rare nor infertile. Seed germination rates of ×B. nabonnandii are low due to seed predation by beetle larvae and seedless fruit production, which is also observed in the genera of the parental species. Furthermore, as in its parents, the morphology of the hybrid is complex, and future anatomical and molecular approaches are important for a better delimitation and understanding of the biology of ×B. nabonnandii.

With the new opportunities from DNA technology, multitier breeding schemes have the potential to become more effective and more integrated. Integrated breeding schemes can also be better adapted to account for potential genotype by environment interactions (G × E) between tiers. In this case, phenotypic and genotype information from lower tiers becomes more valuable as it involves measurement of traits that directly represent the breeding objective. The objective of this study was to compare scenarios that represented different selection strategies and their economic effectiveness in fine-wool commercial sheep operations that exploit multitier breeding structures. Genomic selection (GS) applied in the multiplier and the commercial tier presented the largest additional revenue among all scenarios, as it resulted in the largest amount of genetic progress. The largest benefits from GS were outweighed by the genotyping costs, which made DNA parentage the most feasible strategy for the multiplier tier, resulting in the highest cumulative net present value (CNPV). The benefits of phenotypes and genotype information from the commercial environment were larger in the presence of G × E between the nucleus and the commercial tier. The CNPV was larger with a 50% reduction in genotyping costs, which increased the returns of GS scenarios by 2.7-fold on average. Higher selection intensity when selecting multiplier rams also resulted in larger benefits. In this case, returns for the breeding scheme were 3.5-fold higher when 33% of multiplier males were selected based on commercial information, compared to scenarios selecting 50% of the available multiplier rams. The benefits of collecting commercial phenotypes and genotypes were long term, which means that return on investment often took more than 10 years to be achieved, and were largely dependent on two-stage selection to reduce cost while maintaining selection efficiency and on the cost of a genotype test.

RESUMO

The taxonomic limits of the Neotropical genus Dotocryptus Brèthes are reviewed. The genus is characterized mainly by the very long ovipositor, at least 2.2× as long as hind tibia; apex of ovipositor distinctly depressed, ventral valve expanded, and overlapping dorsal valve as a sheath; and mandible long with ventral tooth longer than dorsal one. A new species, Dotocryptus arcadius sp. nov., from Colombia is described and illustrated. The new species can be diagnosed mainly by the head and mesosoma with multiple whitish marks; epicnemial carina reaching 0.2-0.3 of distance to subalar ridge; legs marked with dark brown, orange, and whitish; and metasoma polished and glabrate. This is the northernmost record of the genus, previously known from Ecuador to southern Chile. An updated identification key to the species of the genus is provided, as well as diagnoses and new records for the previously known species.

This study comprises an update of the economic values for dairy traits for the Australian industry and the formulation of updated selection indices. An economic model, which calculates partial economic values for each trait individually, was developed to determine the economic implications of selective dairy breeding, based on the effect of trait changes on the profit of commercial dairy farms in Australia. Selection indices were developed from economic values, which were transformed into base economic weights by including the discounted genetic expressions coefficients. Economic weights (in Australian dollars) were 1.79, 6.92, -0.10, -5.44, 8.84, 7.68, 1.07, 4.86, 1.91, 3.51, 4.90, 0.31, 2.03, 2.00, and 0.59, for milk fat (kg), milk protein (kg), milk volume (L), body weight (kg), survival (%), residual survival (%), somatic cell count (cells/mL), fertility (%), mammary system [Australian Breeding Value (ABV) unit], temperament (ABV unit), milking speed (ABV unit), udder depth (%), overall type (%), fore udder attachment (%), and pin set (%), respectively. The updated economic weights presented in this study constituted the basis of the definition for 3 new indices. These indices were developed from combination of bioeconomic principles, patterns of farmer preferences for trait improvements, and desired gains approaches. The 3 indices, Balanced Performance Index, Health Weighted Index, and Type Weighted Index, have been released to the industry.

PURPOSE: Presentation of a new cranioplasty technique employing a combination of two technologies: rapid prototyping and surgical navigation. This technique allows the reconstruction of the skull cap after the resection of a bone tumor in a single surgical time. METHODS: The neurosurgeon plans the craniotomy previously on the EximiusMed software, compatible with the Eximius Surgical Navigator, both from the company Artis Tecnologia (Brazil). The navigator imports the planning and guides the surgeon during the craniotomy. The simulation of the bone fault allows the virtual reconstruction of the skull cap and the production of a personalized modelling mold using the Magics-Materialise (Belgium)-software. The mold and a replica of the bone fault are made by rapid prototyping by the company Artis Tecnologia (Brazil) and shipped under sterile conditions to the surgical center. The PMMA prosthesis is produced during the surgical act with the help of a hand press. RESULTS: The total time necessary for the planning and production of the modelling mold is four days. The precision of the mold is submillimetric and accurately reproduces the virtual reconstruction of the prosthesis. The production of the prosthesis during surgery takes until twenty minutes depending on the type of PMMA used. The modelling mold avoids contraction and dissipates the heat generated by the material's exothermic reaction in the polymerization phase. The craniectomy is performed with precision over the drawing made with the help of the Eximius Surgical Navigator, according to the planned measurements. The replica of the bone fault serves to evaluate the adaptation of the prosthesis as a support for the perforations and the placement of screws and fixation plates, as per the surgeon's discretion. CONCLUSIONS: This technique allows the adequate oncologic treatment associated with a satisfactory aesthetic result, with precision, in a single surgical time, reducing time and costs.

Breeding objectives and selection indexes underpin the direction, the extent and the economic implications of selection in livestock populations under specific production systems. The objective of this study was to describe the methodology to calculate correlations between national selection indexes and gain a deeper understanding of the factors influencing responses in economically important traits in both the New Zealand and Irish sheep industries. Moderate to strong correlations were calculated among indexes within and between countries, with the strongest correlation (0.86) between the New Zealand and Irish maternal indexes. In both countries, responses to selection in the maternal indexes are largely driven by growth traits; each index, however, has a different balance of traits. Ewe mature weight also accounts for an important proportion of overall response and has significant emphasis in both maternal indexes. The majority of emphasis in terminal indexes of both countries is on growth and meat traits. Results from this study indicate that differences between national breeding objectives are unlikely to be a barrier to exchange of gene stocks among countries. Future research should investigate the extent to which genotype-by-environment (G × E) interactions exist at the level of individual traits. The methodology presented in this study is robust and represents an opportunity to inform the potential merits of international exchange of germplasm.

Giving consideration to farmers' preferences for improvements in animal traits when designing genetic selection tools such as selection indexes might increase the uptake of these tools. The increase in use of genetic selection tools will, in turn, assist in the realization of genetic gain in breeding programs. However, the determination of farmers' preferences is not trivial because of its large heterogeneity. The aim of this study was to quantify Australian dairy farmers' preferences for cow trait improvements to inform and ultimately direct the choice of traits and selection indexes in the 2014 review of the National Breeding Objective. A specific aim was to analyze the heterogeneity of preferences for cow trait improvements by determining whether there are farmer types that can be identified with specific patterns of preferences. We analyzed whether farmer types differed in farming system, socioeconomic profile, and attitudes toward breeding and genetic evaluation tools. An online survey was developed to explore farmers' preferences for improvement in 13 cow traits. The pairwise comparisons method was used to derive a ranking of the traits for each respondent. A total of 551 farmers fully completed the survey. A principal component analysis followed by a Ward hierarchical cluster analysis was used to group farmers according to their preferences. Three types of farmers were determined: (1) production-focused farmers, who gave the highest preference of all for improvements in protein yield, lactation persistency, feed efficiency, cow live weight, and milking speed; (2) functionality-focused farmers with the highest preferences of all for improvements in mastitis, lameness, and calving difficulty; and (3) type-focused farmers with the highest preferences of all for mammary system and type. Farmer types differed in their age, their attitudes toward genetic selection, and in the selection criteria they use. Surprisingly, farmer types did not differ for herd size, calving, feeding system, or breed. These results support the idea that preferences for cow trait improvements are intrinsic to farmers and not to production systems or breeds. As a result of this study, and some bioeconomic modeling (not included in this study), the Australian dairy industry has implemented a main index and 2 alternative indexes targeting the different farmer types described here.

Verificaram-se a prevalência e as características anatomopatológicas de neoplasias espontâneas encontradas em camundongos, provenientes de biotério. Foram necropsiados 9.219 camundongos pertencentes a 13 linhagens, entre agosto de 2002 e janeiro de 2007, para monitoramento sanitário. Amostras de tecidos foram colhidas, fixadas em formol tamponado a 10 por cento e processadas pelas técnicas habituais para inclusão em parafina. Foram realizados 84 diagnósticos de neoplasmas, sendo acometidos 82 camundongos (0,9 por cento). As principais neoplasias diagnosticadas com as respectivas ocorrências foram: carcinoma mamário, 27,4 por cento; linfoma, 19,0 por cento; adenocarcinoma papilífero pulmonar primário, 17,9 por cento; carcinoma epidermoide, 8,3 por cento; osteossarcoma osteoblástico, 4,8 por cento; e outros com menor porcentagem de ocorrência. Houve predomínio de tumores malignos, sendo mais frequentes os carcinomas mamários. A linhagem BALB/c An foi a que apresentou a maior variedade de tipos de neoplasias, seguida das linhagens outbred Swiss Webster e NIH. Fibrossarcoma em camundongos C57BL/10 ScSn-Dmd mdx/J e carcinoma mamário na linhagem NOD foram relatados pela primeira vez.


The prevalence and the pathologic features of spontaneous neoplasms found in mice from a breeding colony were verified. Nine thousand two hundred and nineteen mice belonging to 13 strains were submitted to necropsy from August 2002 to January 2007 for health monitoring. Tissue samples were collected, fixed in 10 percent buffered formalin, and processed by the usual techniques for inclusion in paraffin. Eighty-four neoplasms were diagnosed and 82 mice had tumors (0.9 percent). The main neoplasms were diagnosed with the following occurrences: mammary carcinoma, 27.4 percent; lymphoma, 19.0 percent; primary papillary pulmonary adenocarcinoma, 17.9 percent; squamous carcinoma, 8.3 percent; osteoblastic osteosarcoma, 4.8 percent; and others with a lower percentage of occurrence. The malignant tumors predominated and the most common tumor was the mammary carcinoma. The wider variety of neoplasm types was found in the BALB/c An strain, followed by the outbred strains Swiss Webster and NIH. The fibrossarcoma in C57BL/10 ScSn-Dmd mdx/J mice and mammary carcinoma in NOD mice were reported for the first time.

This work aimed to evaluate the effects of simulated drift of glyphosate on the morphoanatomy of three eucalypt clones and to correlate the intoxication symptoms on a microscopic scale with those observed in this visual analysis. The effects of glyphosate drift were proportional to the five doses tested, with Eucalyptus urophylla being more tolerant to the herbicide than E. grandis and urograndis hybrid. The symptoms of intoxication which were similar for the different clones at 7 and 15 days after application were characterized by leaf wilting, chlorosis and curling and, at the highest rates, by necrosis, leaf senescence and death. Anatomically glyphosate doses higher than 86.4 g.ha-1 caused cellular plasmolysis, hypertrophy and hyperplasia, formation of the cicatrization tissue and dead cells on the adaxial epidermis. The spongy parenchyma had a decrease, and the palisade parenchyma and leaf blade thickness had an increase. The increased thickness in leaf blade and palisade parenchyma may be related to the plant response to glyphosate action, as a form of recovering the photosynthetically active area reduced by necroses and leaf senescence caused by the herbicide.

This work aimed to evaluate the effects of simulated drift of glyphosate on the morphoanatomy of three eucalypt clones and to correlate the intoxication symptoms on a microscopic scale with those observed in this visual analysis. The effects of glyphosate drift were proportional to the five doses tested, with Eucalyptus urophylla being more tolerant to the herbicide than E. grandis and urograndis hybrid. The symptoms of intoxication which were similar for the different clones at 7 and 15 days after application were characterized by leaf wilting, chlorosis and curling and, at the highest rates, by necrosis, leaf senescence and death. Anatomically glyphosate doses higher than 86.4 g.ha-1 caused cellular plasmolysis, hypertrophy and hyperplasia, formation of the cicatrization tissue and dead cells on the adaxial epidermis. The spongy parenchyma had a decrease, and the palisade parenchyma and leaf blade thickness had an increase. The increased thickness in leaf blade and palisade parenchyma may be related to the plant response to glyphosate action, as a form of recovering the photosynthetically active area reduced by necroses and leaf senescence caused by the herbicide.


Este trabalho teve como objetivo avaliar os efeitos da deriva simulada de glyphosate na morfoanatomia de três clones de eucalipto e correlacionar os sintomas de intoxicação em escala microscópica com aqueles observados à vista desarmada. Os efeitos da deriva do glyphosate foram proporcionais às doses testadas, sendo Eucalyptus urophylla mais tolerante ao herbicida que E. grandis e o híbrido urograndis. Os sintomas de intoxicação foram semelhantes para os diferentes clones testados, tanto aos 7 quanto aos 15 dias após a aplicação, sendo caracterizados, morfologicamente, por murcha, clorose e enrolamento foliar e, no caso das maiores doses, por necrose, senescência foliar e morte das plantas de eucalipto. Anatomicamente, doses de glyphosate superiores a 86,4 g.ha-1 provocaram plasmólise, hipertrofia e hiperplasia celular, formação de tecido de cicatrização e morte das células da face adaxial da epiderme. Observou-se diminuição na espessura do parênquima lacunoso e aumento na espessura do parênquima paliçádico e da lâmina foliar. O aumento na espessura da folha e do parênquima paliçádico podem estar relacionados à resposta das plantas ao glyphosate, como forma de compensar a área fotossinteticamente reduzida pelas necroses e senescência causadas pelo herbicida.

The aim of this study was to evaluate the viability in the effect of open pulled straw (OPS) vitrification procedure of sheep embryos after direct transference. Embryos were produced in vivo and cryopreserved in slow freezing or OPS vitrification. The survival rates of cryopreserved embryos were compared to non-frozen standard pattern. In a first set of experiments, embryos at morula and blastocyst stages were dived in ethylene glycol (1.5 M) and frozen in an automatic freezer. After being thawed, they were directly or indirectly transferred to ewes recipient. A second group of embryos were drawn into OPS and plunged into liquid nitrogen after being exposed at room temperature for 1 min and 45 s in 10% EG plus 10% dimethyl sulphoxide (DMSO), then again for 30 s in 20% EG + 20% DMSO + 0.5 M sucrose. After being warmed, embryos were also directly transferred using a French mini straw as the catheter for the transplantation process or after in vitro dilution of cryoprotectants (two-step-process). No significant difference was observed among fresh, frozen or vitrified embryos on pregnancy rate (50.0%, 38.6% and 55.8%). However, when we evaluated only the direct transference, the pregnancy rate of OPS vitrified embryos was higher than that of frozen embryos (57.1% vs 34.8%) (p = 0.07). In addition, vitrified morulae had a higher pregnancy rate than the one with frozen embryos (64.0% vs 38.9%) (p = 0.07). Finally, our results indicate that OPS vitrification technique in association with direct transference improves the viability of sheep embryos with potential applications to field conditions.

BACKGROUND: Hyperkalemia after transplantation is a common event, occurring in up to 70% of patients. It is usually asymptomatic but sometimes manifests as muscle weakness or cardiac arrhythmias. METHODS: Case report. RESULTS: At 102 days after a second cadaveric kidney transplantation, a 15-year-old boy, was admitted to the emergency room with severe muscle weakness. His examinations showed a serum potassium of 9.8 mEq/L; blood pH 7.1; serum bicarbonate 7.6 mmol/L; and creatinine 2.5 mg/dL. He was initially treated with sodium bicarbonate, calcium gluconate, and furosemide. Subsequent investigation showed hyperchloremic metabolic acidosis, urinary pH <5.5, positive urinary anion gap, reduced transtubular potassium gradient (TTKG, 1.5) and low levels of aldosterone (0.7 ng/mL), suggesting the presence of type 4 renal tubular acidosis (RTA). Other causes of hyperkalemia were excluded in the present case. Serum levels of potassium returned to normal when fludrocortisone was added to the bicarbonate supplementation. This case of severe hyperkalemic secondary to type 4 RTA after kidney transplantation only responded to the combination of alkali and mineralocorticoid therapies.

Both preclinical and clinical data link glutamate to the migraine pathophisiology. Altered plasma, platelets and cerebrospinal (CSF) glutamate levels have been reported in migraine patients. Chronic migraine is comorbid with several conditions. It has been recently shown chronic migraine comorbidity with fibromyalgia. The objective of this study was to study cerebrospinal fluid glutamate levels in chronic migraine patients with and without fibromyalgia. We studied 20 chronic migraine patients, with and without fibromyalgia, compared to age-sex matched controls. CSF glutamate levels were measured by HPLC. CSF glutamate demonstrated significantly higher levels in patients with fibromyalgia compared to those without fibromyalgia. Patients overall had higher CSF glutamate levels than controls. Mean pain score correlated with glutamate levels in chronic migraine patients. Tender points, the hallmark of fibromyalgia, can be considered as pressure allodynia, and is probably mediated by central sensitization, with increase in CSF glutamate levels. We postulate chronic migraine patients with fibromyalgia, in addition to have more disabling headaches, suffer from a more severe central sensitization process. This subtype of patients may respond to medications modulating glutamate receptors. Headache intensity correlate with glutamate levels in chronic migraine patients.

The serine peptidases, thrombocytin and PA-BJ, isolated from the venom of Bothrops atrox and Bothrops jararaca, respectively, induce platelet aggregation and granule secretion without clotting fibrinogen. The specific platelet aggregation activity of each enzyme was about 15 times lower than that of thrombin. This activity was blocked by monoclonal antibodies recognizing protease activated receptor 1 (PAR1) and by heparin, but not by hirudin nor thrombomodulin. Both enzymes induced calcium mobilization in platelets and desensitized platelets to the action of thrombin and the SFLLRN peptide. We compared the effect of thrombin, PA-BJ, and thrombocytin on the degradation of the soluble N-terminal domain of the PAR1 receptor. The major cleavage site by thrombin and both viper enzymes was Arg41-Ser42. In addition, a rapid cleavage of the peptide bond at Arg46-Asn47 by the viper enzymes was observed, resulting in the inactivation of the tethered ligand. PA-BJ and thrombocytin both cleaved at 41-42 and 46-47 peptide bonds, and fragment 42-103 disappeared rapidly. Both viper enzymes caused calcium mobilization in fibroblasts transfected with PAR4 and desensitized these cells to the thrombin action. In conclusion, both PAR1 and PAR4 mediate the effect of viper venom serine peptidases on platelets.

RESUMO

A insuficiência cardíaca congestiva foi definida por muito tempo como uma síndrome envolvendo profundas alteraçöes hemodinâmicas e neuro-hormonais. Nos últimos anos, uma reação inflamatória exuberante tem sido observada na presença de disfunção ventricular sistólica severa, tanto em modelos experimentais como em ensaios clínicos. Essa resposta inflamatória envolve a ativaçäo e a liberaçäo de uma série de substâncias peptídicas denominadas citocinas. Essas substâncias parecem ter papel relevante tanto na gênese como na manutencçäo do processo de insuficiência ventricular, e podem ser utilizadas inclusive como marcadores prognósticos da doença. O presente artigo apresenta, de forma resumida, os principais estudos clínicos nessa área, destacando a correlaçäo entre determinadas citocinas e variáveis clínico-laboratoriais já descritas em portadores de insuficiência cardíaca congestiva avançada.
